오미자 유산균 발효물의 주름개선 효과 (Anti-Wrinkle Effect of Schizandra chinensis Baillon Fermented with Lactobacillus plantarum)

  • 새로운 주름개선제 성분을 찾기 위해서 본 연구에서는 사람 피부 섬유아세포의 세포독성, 콜라겐 생합성, matrix metalloproteinase-I (MMP-1) 및 elastase 저해활성에 대한 Lactobacillus plantarum으로 발효된 오미자 발효액의 주름개선 효과를 평가하였다. 먼저, 오미자 추출물은 L. rhamnosus으로 $37^{\circ}C$에서 1일 동안 발효하였다. 발효물의 세포독성은 cytopathic effect reduction 방법에 의해 평가하였다. 콜라겐 생합성에 대한 발효물의 영향은 procollagen type-IC peptide EIA kit에 의해 평가하였으며, matrix metalloproteinase-I(MMP-1)에 대한 발효물의 영향은 Matrix Metalloproteinase-1 Biotrack activity Assay Kit에 의해 평가하였다. Elastase inhibition assay는 기질로써 N-Suc-$(Ala)_3$-nitroanilide을 사용하여 기질 반응에 의해 평가하였다. 결과로써 오미자 발효물은 사람 피부 섬유아 세포에 대해 $100{\mu}g/mL$의 농도에서 세포독성을 나타내지 않았다. 또한 오미자 발효물은 콜라겐 생합성을 촉진시켰으며, MMP-1의 저해 효과를 나타내었다(p < 0.05). Elastase inhibition assay에서 오미자 발효물의 $IC_{50}$$36.4{\mu}g/mL$이었다. 그러므로 본 연구에서 오미자 발효물은 주름개선 효과를 보유하고 있으며, 이것은 피부의 주름개선을 위해 사용가능하리라 사료된다.

  • Mariculture of rainbow trout Onchorhynchus mykiss has been initiated in or around olive flounder Paralichthys olivaceus farms, where viral hemorrhagic septicemia virus (VHSV) is often detected in some fish. In the present study, we investigated VHSV infection in seawater-reared rainbow trout because VHSV has never been detected in salmonids in Korea. A total of 104 adult fish were tested for the presence of VHSV by reverse transcription-polymerase chain reaction, followed by virus isolation with the fathead minnow caudal trunk cell line. Cytopathic effects were observed in two samples but the virus was identified as infectious hematopoietic necrosis virus. Thus, VHSV was not isolated from seawater-reared rainbow trout.

  • Fourteen melandrin derivatives(I-XIV) were investigated on analgesic, anti-inflammatory and antiviral activities . Compound I [N-(p-hydroxybenzoyl)-5-hydroxyanthranilic acid methvl ester], Xll [N-(2-phenoxypropionyl)-5-hydroxy anthranilic acid propyl ester and XIV [N-(2-phenoxypropionyl)-5-hydroxyanthranilic acid exhibited analgesic activity in tail pressure and Randall-Selitto method. But no anti-inflammatory activity was shown. Compound I exhibited weak antiviral activity on Herpes simplex virus type I F strain by virus-induced cytopathic effect(CPE) assay and it's selectivity index(Sl) was 8.17.

  • An electron microscopic study was carried out on the morphogenesis of herpes simplex type 2 virus in BHK-21 cells BHK-21 cells was found susceptible to infection and replication of herpes simplex type 2 virus cytopathic effects of the herpes type appeared at approximately 1 day postinoculation. Foci consisting of rounded refractile cells and syncytia were observed. Projection of the nuclear membrane in the infected cells was also seen, Several infected cells showed a track-shaped structure which apparently consisted of multiple layered membranes of the nucleus.

  • Dehydroaltenusin (1) was isolated from the chloroform extract of the culture filtrate of a Streptomyces sp. and its structure was determined from spectral data as well as by comparison with published values. In an XTT-based in vitro anti-HIV assay, dehydroaltenusin effectively inhibited the cytopathic effects of HIV infection at a concentration of $1-5\;{\mu}g/mL$.

국내에서 분리한 닭 레오바이러스의 성상에 관한 연구 (Studies on Properties of Avian Reovirus Isolated in Korea)

  • Avian reoviruses have been implicated in respiratory disease enteric conditions including Cloacal pasting in young thicks, pericarditis, hydropericardium, anaemia with swollen spleen and liver and petechiation of skeletal muscle and viral arthritis. This study was conducted to examine properties of reovirus field 3 strains isolated from affected broiler from several farms. An infectious agent was isolated from leg tendons and intestine of broiler with clinical tenosynovitis. The agent grew well on the chicken embryo kideny cells(CEK). One of them produced cytopathic effects(CPE) of round type and formed intranuclear inclusions, and the other was characterized by CPE of syncytical type and cytoplasmic inclusion. The properties and serological classification of field strains were examined by hemagglutin test, virus neutralization test, agar gel precipitin, electropherotype. They showed no hemagglutination reactions and not well neutralization and to possess common antigens detectable by AGP test. RNA electropherotype presented 10 segment band as the previous report. These data suggest that the field strains and standard strains (1133, 1733) may be the same group.

감송향물추출물의 HO-1 발현 촉진을 통한 세포보호 작용 및 항염작용 (Cytoprotective and Anti-inflammatory Effects of Nardostachys jatamansi Water Extract Via Expression of HO-1)

  • Nardostachys jatamansi water extract (NJ) has long been used for the treatment of inflammation-and immune-mediated disorders in the oriental countries. However, its site of action and pharmacological mechanism are not fully investigated. In this study, the authors tried to explore the cytoprotective and anti-inflammatory actions of NJ. First of all, NJ has no harmful effects on viability of neuronal cell line HT22 cells in the dose range of 300 mg/ml. On the contrary, it shows cytoprotective effects on the cells treated with reactive oxygen species H2O2. Probably the cytoprotective effects of NJ might be caused by its ability to induce well known cytoprotective gene hem oxygenase-1 (HO-1). Furthermore, NJ shows inhibitory effects on the expression of inducible nitric oxide synthase (iNOS) and NO production which are known to destroy the integrity of both cells and tissues. It also inhibits potent proinflammatory cytokine tumor necrosis factor-alpha (TNF-a) production. The blocking effects of NJ on cytopathic and proinflammatory actions of LPS might be caused by the induction of cytoprotective and anti-inflammatory genes HO-1 in macrophages cell line RAW 264.7 cells. The results in this study suggest NJ could be used for the amelioration of inflammation which is underlying mechanism responsible for most chronic diseases.

Streptococcus thermophilus로 발효한 한약재 발효분말의 항염증 및 미백 효과 (Anti-Inflammatory and Whitening Effect of the Lyophilized Powder of Oriental Plant Extracts Fermented with Streptococcus thermophilus)

  • 새로운 항염증 및 미백 소재를 찾기 위해서, 본 연구에서는 6가지 한약재 추출물(유자, 상백피, 오미자, 율무, 당귀, 고삼)을 Streptococcus thermophilus에 의해 발효한 발효분말의 미백 및 항염증 효과를 피부섬유아세포에 대한 피부독성, 산화질소 생성, tyrosinase 활성, 멜라닌 형성의 저해 효과를 측정함으로써 평가하였다. 발효물은 $37^{\circ}C$에서 2일 동안 Streptococcus thermophilus에 의해 발효한 후, 동결건조에 의해 발효분말을 제조하였다. 발효분말은 피부섬유아세포에 대해 $500{\mu}g/mL$의 농도에서 cytopathic effect reduction 방법을 사용하면서 측정하였을 때 세포독성을 나타내지 않았다. 또한 발효분말은 Griess reagent system을 사용하면서 산화질소 생성에 대한 저해 효과를 나타내었다. 더욱이 발효분말은 tyrosinase 활성에 대한 저해 효과가 농도 의존적으로 나타났다. 발효분말은 배양액으로부터 멜라닌 생성에 대해 유의적인 저해를 나타내었다(p < 0.05). 그러므로 이러한 자료로부터 발효분말은 항염증 및 미백 효과를 갖는 것으로 나타났으며, 화장품을 위한 효과적인 성분으로써 사용가능할 것으로 사료된다.

  • The first outbreak of Aujeszky's disease(AD) was identified from piggery located at the southern part of Korea in July, 1987. This piggery suffered from a significant economic loss caused by unexpected piglet mortality and reproductive failure. Etiologic viral agents were isolated from tonsil and spleen of the infected piglets, and the isolates produced a typical cytopathic effects of herpesvirus with giant cell formation when inoculated in many different cells. Subsequently the field isolates were characterized as suid herpesvirus I by cross-neutralization test and indirect fluorescence assay utilizing specific monoclonal antibody, and were proved to be a pathogenic strain of AD virus(ADV).

  • An equine herpesvirus-4 (EHV-4) was isolated in nasal swabs collected in a horse showing respiratory clinical signs. Equine dermis cells inoculated with the sample were observed with characteristic viral cytopathic effects after 3 days of postinoculation and the infected cells exhibited bright intracelluar fluorescence by indirect immunofluorescence assay. At the nucleotide level, the partial glycoprotein B gene of the Korean EHV-4 isolate (K001) had 99.9% identity to 1942 strain (GenBank No. M26171). To author's knowledge, the report describes the first isolation and partial characterization of EHV-4 in Korea. The virus can be used for further study of EHV-4.
