식이 Vitamin E가 급성 카드뮴중독 흰쥐 간조직의 Cytochrome $P_{450}$ 함량과 Xanthine Oxidase 활성에 미치는 영향 (Effects of Vitamin E on Liver Cytochrome $P_{450}$ Content and Xanthine Oxidase Activity in Acute Cadmium-Poisoned Rats)

  • The purpose of this study was to investigate the effects of vitamin E on liver microsomal cytochrome P450 contents and xanthine oxidase activity in acute cadmium poisoned rats. Sprague Dawley male rats weighing 100$\pm$10g were randomly assigned to one normal and three cadmium injected groups. Cadmium injected groups were fed vitamin E free diet(0E Cd group), 40mg vitamin E per kg diet(40E Cd group) or 400mg vitamin E per kg diet(400E Cd group). Vitamin E level of normal group was 40mg per kg diet. Animals were injected intraperitoneally with 2.0mg Cd2+/kg bw for 4 days after the rats were fed diets with three different levels of vitamin E for 2 and 4 weeks. Body weight, food intake and feed efficiency ratio of cadmium injected animals, were decreased compared with those of normal group. The weights of liver and kidney in cadmium injected groups were not different from those of normal group. Cadmium contents of liver in cadmium groups were 160 fold higher those that of normal group. Accumulation of cadmium poisoned rat liver was reduced by vitamin E supplementation. Contents of blood hemoglobin and hematocrit in 0E Cd groups were decreased to 2~ 14% of those of the other groups. Contents of serum triglyceride in all experimental groups were not significantly different each other. Levels of serum total cholesterol, LDL cholesterol and antherogenic index in 0E Cd and 40E Cd groups were higher than those of normal group, while the contents of HDL cholesterol in 0E Cd and 40E Cd groups were lower than those of normal group. Xanthine oxidase(XOD) activity and cytochrome P450 contents in the liver were significantly increased in cadmium injected groups but these were reduced by vitamin E supplementations. The present results indicate that acute cadmium poisoning in rats causes increasing free radical generation systems in the liver and that leads to liver tissue damage. But these abnormalities can be reduced by dietary vitamin E supplementations.

Butylated Hydroxytoluene첨가 식이 및 2-Acetylaminofluorene 투여가 식이지방을 달리한 쥐간의 Microsomal Mixed Function Oxidase계에 미치는 영향 (Effect of Butylated Hydroxytoluene and 2-Acetylaminofluorene Administration and Microsomal Mixed Function Oxidase System in Young Rats fed different Fats)

  • Sprague-dawley 숫쥐를 식이지방을 달리하여(I:Soybean oil p/s 4.0, II:beef tallow p/s 0.08) 이유후 8주동안 사육하였다. 이때 I,II군은 각각 기초식이군, 기초식이군에 0.3% butylated hydroxytoluene(BHT)를 첨가신킨 군, 생후 5~7주 사이에 4번의 2-Acetylaminofluorene(2-AAF)를 투여한 군 2-AAF를 투여하고 BHT도 먹인 군으로 나누었다. BHT는 이유 후부터 식이에 섞어 먹였으며 2-AAF를 투여하지 않은 군읜 2-AAF 주사에 의해 얻는 stress와 같은 효과를 주기위해 placebo로 polyethylene glycol 300을 투여하였다. Mixed function oxidase(MFO)계의 효소인 cytochrome p-450, cytochrome b$_5$및 cytochrome p-450 reductase와 과산화지질 등을 측정하였다. 성장기에 2-AAF의 투여는 성장지연을 초래하였으며 지질과산화물은 지방의종류, 2-AAF,BHT 등에 의해 큰 차는 없었다. Cytochrome p-450은 2-AAF에 의해 I-BHT-AAF와 II-AAF군에서 증가되었고 BHT에 의해서는 차이가 나지 않았다. 불포화지방을 먹인경우 cytochrome p-450과 cytochrome p-450 reductase가 2-AAF에 의해 증가되기보다는 오히려 감소하거나 I,II군에 비해 별 차이가 없었는데 2-AAF의 농도와 식이의 불포화도가 높아 세포막이 손상되었기 때문이라 사료된다. Cytochrome $b_5$는 각군 사이에 별 차가 없었다. Cytochrome p-450과 과산화지질(r=0.2475, p<0.05), cytochrome p-450 reductase와 cytochrome $b_5$ (r=0.2475, P<0.05)가 각각 양의 상관관계를 나타내었다. 따라서 2-AAF를 대사시키는 MFO계는 식이지방의 종류 및 BHT의 존재에 따라 영향을 받고, 특히 불포화지방식이인 경우 2-AAF를 대사시킬 수 있는 cytochrome p-450 유도 및 합성능력이 매우 저조함을 알 수 있으며 2-AAF는 어린 쥐의 성장을 지연시켰다.

Phylogenetic Analysis of Reticulitermes speratus using the Mitochondrial Cytochrome C Oxidase Subunit I Gene

  • Reticulitermes speratus is commonly found in Asia, including Korea and Japan. We recently analyzed the 5' region of mitochondrial cytochrome c oxidase subunit I to perform a phylogenetic analysis of R. speratus KMT1, isolated in Seoul, Korea. Our results, using COXI, suggest that the taxonomy of R. speratus should be reconsidered with regard to the subgenus group. A similar phylogenetic analysis by COXI and COXII demonstrated the reliability of COXI genetic information in a molecular phylogenetic analysis of termites.

Stock Characterization of the Fleshy Prawn (Penaeus chinensis) in the Yellow Sea by Intraspecific Sequence Variation of the Cytochrome c Oxidase Subunit I Gene

  • To determine the amount of genetic variation among populations of Penaeus chinensis (Osbeck) in the Yellow Sea, 342 bp region of the mitochondrial cytochrome c oxidase subunit I gene was amplified and sequenced. Six haplotypes, which differ by from one to four nucleotide sustitutions, were detected from 34 individuals of 4 populations examined. Mean sequence divergence between pairs of haplotypes was $0.68\%$. Most individuals from 4 populations were shared by the most common genotype. This genotype was distributed evenly in the Korean and Chinese populations. This result is in accordance with findings observed using RFLPs analysis of mtDNA (Hwang et al., 1997). Therefore, it is suggested that P. chinensis should be treated as one unit stock in the Yellow Sea.

Long-Term Feeding of Dietary Fat and Butylated Hydroxytoluene on The Hepatic Microsomal Mixed-Function Oxidase System in 2-Acetylaminofiuorene Treated Rats

  • This paper examines the effects of dietary polyunsaturated fatty acid/saturated fatty acid (p/s) ratios and butylated hydroxytoluene (BHT) on the hepatic microsomaI mixed-function oxidase sy. stem in 2~acetylaminofiuorene (2-AAF) treated rats. Sprague-Dawley male rats were fed the diet of beef tallow (p/s 0.08), beef tallow plus soybean oil (p/s 1.0), and soybean oil (p/s 4.0) at the level of 15%fat and with or without 0.3% BHT. After 2-AAF was injected twice at the ages of 23 and 27 weeks, cholesterol/phospholipid molar ratio, thiobarbituric acid reactive substances (TBARS) level, cytochrome P450, cytochrome $b_5$, NADPH-cytochrome $b_5$, and NADPH-cytochrome c reductase activity were measured from isolated hepatic microsomal fractions. In the beef tallow (p/s 0.08) and beef tallow plus soybean oil (p/s 1.0) groups, cholesterol/phospholipid molar ratio showed decreasing tendency by 2-AAF and BHT. Cytochrome P-450 content was decreased in the group of soybean oil (p/s 4.0) and NADPH-cytochrome c reductase activity was increased by 2-AAF and BHT in all the dietary groups. While TBARS levels were increased by 2-AAF in all the dietary groups, they were reduced by BHT in the soybean oil (p/s 4.0) group. These results suggest that long term intake of soybean oil (p/s 4.0) diet induced changes in the nature of microsomal membrane and induced less cytochrome P-450, low level feeding of BHT increased cytochrome c reductase activity and lowered microsomal lipid peroxidation levels, which were increased by 2-AAF treatment.

태충 침자가 간 미토콘드리아 내 Complex IV에 미치는 영향 (The Effects of Acupuncture at LR3 Acupoint on Mitochondrial Complex IV Oxidase activity in Liver)

  • Objectives : The liver is rich in mitochondria and it plays a key role in whole-body energy homeostasis. Mitochondria is double membrane-bound organelle that supplies energy for intracellular metabolism including Krebs cycle and beta-oxidation. Acupuncture is known to stimulate and regulate the flow of energy. To explore the effect of acupuncture on the mitochondrial respiratory chain activity in the rats' livers, the activity of mitochondrial respiratory chain complexes I to IV was observed. Methods : The rats were divided into 4 groups; Normal 1 (no acupuncture treatment and anesthesia for 5 min), Normal 2 (no acupuncture treatment and anesthesia for 10 min), MA1 (acupuncture treatment at bilateral LR3 under anesthesia for 5 min), and MA2 (acupuncture treatment at bilateral LR3 under anesthesia for 10 min). All rats were sacrificed and the livers were examined for respiratory chain change. Results : There was no difference in ubiquinon oxidoreductase, succinate dehydrogenase, and ubiquinol cytochrome C oxidoreductase after acupuncture at LR3. Acupuncture at LR3 for 10 min increased the activity of cytochrome C oxidase compared with no acupuncture groups. Conclusions : Acupuncture at LR3 mediated mitochondrial respiratory chain activity via the cytochrome C oxidase signaling pathway in the livers of rats.

Comparative Assessment of Diagnostic Performance of Cytochrome Oxidase Multiplex PCR and 18S rRNA Nested PCR

  • Malaria elimination and control require prompt and accurate diagnosis for treatment plan. Since microscopy and rapid diagnostic test (RDT) are not sensitive particularly for diagnosing low parasitemia, highly sensitive diagnostic tools are required for accurate treatment. Molecular diagnosis of malaria is commonly carried out by nested polymerase chain reaction (PCR) targeting 18S rRNA gene, while this technique involves long turnaround time and multiple steps leading to false positive results. To overcome these drawbacks, we compared highly sensitive cytochrome oxidase gene-based single-step multiplex reaction with 18S rRNA nested PCR. Cytochrome oxidase (cox) genes of P. falciparum (cox-III) and P. vivax (cox-I) were compared with 18S rRNA gene nested PCR and microscopy. Cox gene multiplex PCR was found to be highly specific and sensitive, enhancing the detection limit of mixed infections. Cox gene multiplex PCR showed a sensitivity of 100% and a specificity of 97%. This approach can be used as an alternative diagnostic method as it offers higher diagnostic performance and is amenable to high throughput scaling up for a larger sample size at low cost.

국소마취제가 Mitochondria에서의 전자이동 및 Superoxide Radicals의 생성에 미치는 영향 (Effects of Local Anesthetics on Electron Transport and Generation of Superoxide Radicals in Mitochondria)

  • 국소마취제가 mitochondria에서의 전자이동 및 superoxide라디칼의 생성 그리고 지질의 과산화에 따른 malondialdehyde생성에 미치는 영향을 관찰하였다. 국소마취제는 전자이동계 의 효소활성도에 영향을 나타내었다. NADH dehydrogenase, NADH oxidase와 NADH-ubiquinone oxidoreductase의 활성도는 lidocaine, procaine과 dibucaine에 의하여 효과적으로 억제되었고 cocaine에 의하여 약간 억제되었다. Succinate dehydrogenase, succinate cytochrome c oxidoreductase와 succinate-ubiquinone oxidoreductase 활성도는 lidocaine 과 dibucaine에 의하여 억제되었으나 succinate oxidase는 국소마취제에 의하여 활성화되었다. 국소마취제는 dihydroubiquinone-cytochrome c oxidoreducatse와 cytochrome c oxidase의 활성도를 억제하였다. 이와 같은 반응에서 국소마취제에 대한 complex I segment의 반응이 다른 complex segment보다 크게 나타났다. 국소마취제는 succinate 또는 NADH에 의한 superoxide 생성과 이에 대한 antimycin의 자극효과를 억제하였다. 또한 국소마취제는 산소라디칼에 의한 지질의 과산화를 억제하였다. 이상의 결과로부터 국소마취제는 mitochondria의 전자전달 과정 중 Complex I segment때 또는 인접한 부위에 작용하여 전자이동을 억제함으로써 superoxide 생성과 지질의 과산화를 억제할 것으로 시사되었다.

흰쥐에서 Treadmill 훈련에 따른 심근의 적응과 cytochrome oxidase 활성에 관한 세포화학적 연구 (Cytochemical Study on the Cytochrome Oxidase Activity and Myocardial Adaptation to Treadmill Exercise in Rat)

  • 흰쥐를 실험 대상으로 하여 Treadmill 운동을 실시하여 운동에 의한 심근의 적응과 cytochrome oxidase 활성에 관한 세포화학적 연구 알아보고자 실험을 실시하였다. 실험동물은 SD계 웅성 흰쥐를 이용하여 출생 3개월 정도의 체중 $150{\pm}10g$정도로 실험을 시작하였다. 실험군은 일상 대조군과 운동군 2개군으로 나누고, 각각 1일, 3일, 1주일, 4주일, 8주일 및 12주일 동안 각각 4마리씩 운동시켜 실험에 이용하였다. 형태 계측학적 방법으로 심장의 무게변화, 심근섬유와 모세혈관의 비, 그리고 CO 활성정도를 측정하였다. 광학및 전자현미경을 이용하여 관찰한 결과 다음과 같은 결론을 얻었다. 심장무게 변화는 모든 실험군에서 실험기간에 비례하여 증가하였다. 일상 대조군에 비해 운동군에서 유의한 증가를 보였다. 모세혈관과 근섬유 비율은 일상 대조군에서는 유의한 변화가 없었고, 운동군에서는 운동 4주일 이후부터 유의한 증가를 보였다. Cytochrome oxidase C 활성율은 일상 대조군에서는 유의한 변화가 없었고, 운동군에서는 운동 1주일 이후부터 유의한 증가를 보였다. 사립체에 CO활성 형태는 일상 대조군에서는 I형과 III형이, 운동군에서는 실험기간에 비례해서는 II형과 III형이 많이 관찰되었다. 이상과 같은 실험결과들을 미루어볼 때 Treadmill 운동결과 CO의 활성과 심근섬유와 모세혈관의 비율이 운동에 의해 변화를 보여, 흰쥐의 심근에서는 treadmill 운동은 약 4주 정도에서 심장의 적응성이 어느 정도 이루어지는 것으로 사료된다.

광합성 세균인 Rhodopseudomonas gelatinosa ATCC 17013에서 Cytochrome c-551의 정체 (Purification of Cytochrome c-551 from Photosynthetic Bacterium Rhodopseudomonas Gelatinosa ATCC 17013)

  • The soluble cytochrome c-551 of photosynthetic bacterium, Rhodopseudomonas gelatinosa ATCC 17013 was purified through a sequene of four step chromatography including CM-cellulose ion-exchange chromatography, DEAE-Sephacel chromatography, Sephacryl s-200 gel permeation chromatography, and HPLC (SP-5PW). The molecular weight of the purified cytochrome c-551 was 14, 600 Da, and this protein shows the absorption peak at 551 nm, 522 nm, and 417 nm as the reduced form, and at 412 nm as the oxidized form. The cytochrome c-551 seems to be a substrate for the terminal oxidase in the electron transport chain.
