무자갈버섯 (Hebeloma crustuliniforme) 배양 균사체로부터 분리한 단백다당체 분호기 HCA의 항암 및 백혈구감소증 억제효과 (Antitumor and Antileukopenic Activity of HCA, the Protein-polysaccharide Fraction of Cultured Mycelia of Hebeloma crustuliniforme)

  • A protein-polysaccharide fraction was isolated from the mycelial culture of a Korean basidiomycetous fungus, Hebeloma crustuliniforme, and was named HCA (Hebeloma crustulineforme fraction A). When intraperitoneally injected at 200 mg/kg/day once daily for 10 days, HCA inhibited the growth of sarcoma 180 solid tumor in ICR mice by 65.1% (p<0.05). HCA also exerted protective effect against leukopenia induced by cyclophosphamide with the protection ratio of 39.7% (p<0.01).

  • To find antitumor components from the higher fungi of Korea, the mycelia of Favolus alveolarius (Fr.) Quelet were cultured in a liquid medium. The cultured mycelia were extracted with hot water twice, and a high molecular weight fraction was obtained by adding two volumes of ethanol to the extract. Two grams of Fraction A were obtained by dialyzing it. It was further separated into four fractions by gel filtration with Sepharose CL-4B, and they were designated Fractions B, C, D, and E. The results of the antitumor test showed that Fractions A, B, C, D and E had tumor inhibition ratios of 92.3, 78.5, 59.6, 77.4 and 62.2%, respectively. Anthrone test was carried out to determine the contents of total polysaccharide of the five fractions, and they had 46.3, 27.3, 65.3, 64.6, and 46.1%, respectively. The contents of the total protein of the five fractions were 29.4, 13.9, 14.3, 14.3, and 29.1%, respectively. Monosaccharide subunits of each fraction were analyzed with gas chromatography, and glucose, xylose, mannose, galactose and fucose were identified. Fraction A was examined for immunological effects. It increased the count of hemolytic plaque forming cells 12.8 times to that of the control group, and the population of macrophage in peritoneal cavity 3.2 times to that of the control group.

  • 한국산 담자균류의 항종양성 성분을 탐색하기 위하여 턱수염버섯의 균사를 분리하여 인공배지에서 진탕배 양후, 그 배양균사체로부터 수용셩 단백성 다당류를 추출하였다. 이 성분이 ICR마우스에 이식한 육종 180에 대하여 54.3%의 종양억제율을 나타내었다. 이 성분은 다당류 42%와 단백질 28%로 구성되어 있었다. 그 중 다당류는 glucose (57.8%), mannose(19.3%), galactose(10.8%), xylose (6.8%), 및 fucose (5.7%)를 함유하고 있었다.

The Fruiting Body Formation of Armillaria mellea on Oak Sawdust Medium Covered with Ground Raw Carrots

  • To produce an artificial fruiting body of Armillaria mellea on the oak sawdust medium, seven strains of A. mellea were used. The top surface of oak sawdust medium covered with ground raw carrot was inoculated with each of 7 strains and cultured for 30 days at $25^{\circ}C$ in the dark condition until the mycelia of A. mellea completely colonized the medium from top to bottom. Then, the mycelia which were fully covered on the top surface of the medium were scratched slightly with a spatula and filled with tap water for 3 hours. To induce the primordial formation, the 7 strains of A. mellea were transferred to the growth chamber under the illumination (350 lux) of 12 hours and relative humidity of $85{\pm}5%$ in a day and then cultured at $16{\pm}1^{\circ}C$. Only A. mellea IUM 949 could form primordia on the sawdust medium, but the other strains did not make primordia at the same condition. The primordia of A. mellea IUM 949 were formed 10 days after complete colonization of the medium and the fruiting bodies were produced 7 days after a primordial formation. The experimental results suggested that IUM 949 strain might be a good candidate for mass production of fruiting bodies of A. mellea.

Purification and Characterization of an Acid Deoxyribonuclease from the Cultured Mycelia of Cordyceps sinensis

  • A new acid deoxyribonuclease (DNase) was purified from the cultured mycelia of Cordyceps sinensis, and designated CSDNase. CSDNase was purified by $(NH_4)_2SO_4$ precipitation, Sephacryl S-100 HR gel filtration, weak anion-exchange HPLC, and gel filtration HPLC. The protein was single-chained, with an apparent molecular mass of ca. 34 kDa, as revealed by SDS-PAGE, and an isoelectric point of 7.05, as estimated by isoelectric focusing. CSDNase acted on both double-stranded (ds) and single- stranded (ss) DNA, but preferentially on dsDNA. The optimum pH of CSDNase was pH 5.5 and its optimum temperature 55. The activity of CSDNase was not dependent on divalent cations, but its enzymic activity was inhibited by high concentration of the cation: $MgCl_2$ above 150 mM, $MnCl_2$ above 200 mM, $ZnCl_2$ above 150 mM, $CaCl_2$ above 200 mM, NaCl above 300 mM, and KCl above 300 mM. CSDNase was found to hydrolyze DNA, and to generate 3-phosphate and 5-OH termini. These results indicate that the nucleolytic properties of CSDNase are essentially the same as those of other well-characterized acid DNases, and that CSDNase is a member of the acid DNase family. To our knowledge, this is the first report of an acid DNase in a fungus.

가시오가피 버섯균사체 발효물의 기능성 식품 소재 개발 (Development of Functional Food Materials from Acanthopanax senticosus-Fermented Mushroom Mycelia)

  • 가시오가피(Acanthopanax senticosus)의 잎과 줄기를 천연물배지로 발효미생물인 영지버섯, 노루궁뎅이버섯 및 상황버섯균사체를 각각 최적조건으로 발효하여 3종의 가시오가피-버섯발효물인 가시오가피 영지버섯균사체 발효물, 가시오가피 노루궁뎅이버섯균사체 발효물, 가시오가피 상황버섯균사체 발효물을 얻었으며 이를 건강기능식품 소재로서 이용하고자 하였다. 우선적으로 배양용 시험관과 실험실용 배양병을 사용하여 버섯균사체별 가시오가피의 천연배지로서 최적혼합비율을 검토한 결과 영지버섯균사체, 노루궁뎅이버섯균사체 및 상황버섯균사체의 가시오가피 줄기와 잎의 혼합비율을 각각 80:20, 50:50, 50:50으로 확립하였으며, 이 조건을 가지고 산업용 배양포트(2,000 mL)에서의 최적배양시간을 검토한 결과 영지버섯균사체 14일, 노루궁뎅이버섯균사체 40일, 상황버섯균사체 30일로 확립하였다. 상기와 같이 최적화된 조건으로 배양된 3종의 가시오가피 버섯균사체 발효물들을 물과 70% 에탄올로 추출, 여과, 농축, 동결건조를 수행하여 가시오가피 버섯균사체 발효물 추출분말인 열수추출물(FM-5111, FM-5121, FM-5131)과 70% 에탄올추출물(FM-5112, FM-5122, FM-5132)을 얻었다. 이 가시오가피 버섯균사체 발효물 추출분말의 eleutheroside B와 eleutheroside E를 분석한 결과, eleutheroside B는 FM-5111 $0.050{\pm}0.003mg/g$, FM-5121 $0.067{\pm}0.001mg/g$, FM-5131 $0.026{\pm}0.001mg/g$, FM-5112 $0.038{\pm}0.002mg/g$, FM-5122 $0.053{\pm}0.005mg/g$, FM-5132 $0.025{\pm}0.001mg/g$의 함량을 나타내었고, eleutheroside E는 FM-5111 $0.052{\pm}0.014mg/g$, FM-5121 $0.047{\pm}0.003mg/g$, FM-5131 $0.078{\pm}0.003mg/g$, FM-5112 $0.045{\pm}0.002mg/g$, FM-5122 $0.040{\pm}0.002mg/g$, FM-5132 $0.080{\pm}0.011mg/g$의 함량을 보였으며, ${\beta}$-glucan의 경우 FM-5111 $4.47{\pm}0.14%$, FM-5121 $3.41{\pm}0.15%$, FM-5131 $4.15{\pm}0.06%$, FM-5112 $5.18{\pm}0.36%$, FM-5122 $3.74{\pm}0.40%$, FM-5132 $4.37{\pm}0.32%$의 함량이 측정되었다.

액체배양에서의 홍국균의 생장, 색소 및 monacolin K 생산 특성 (Characteristics of Growth, Pigment and Monacolin K Production by Monascus strains in Liquid Culture)

  • The hyphal growth, production of color pigments and monacolin K by Monascus strains were investigated in liquid medium. Thirty five different strains were collected and cultured in potato dextrose yeast extract broth (PDYB), potato dextrose broth (PDB) and malt extract broth (MEB) medea at $25^{\circ}C$ for 7 days. The growth rates of most of strains were highest in PDYB medium. Growth rate as well as pigment production were influenced by suspension conditions of mycelia during liquid cultivation. Most of strains producing monacolin K corresponded to strains producing red pigment highly and showing more pH changes of liquid media. Monacolin K produced from strains was detected in culture broth as well as mycelia. Any citrinin was not detected in monacolin K producing strains. These results imply that the selection of the strains producing red pigment highly and showing more pH changes in liquid cultivation could be applied for primary screening of Monascus strains for preparation of red mold rice.

  • 1. 분화과정에 따른 균체로부터 추출한 총 RNA량은 포자형성 직전에 증가함을 알았다. 2. 추출한 RNA를 가수분해하여 P.E.I. Cellulose TLC를 전개한 결과 AMP유도체와 IMP가 존재함을 확인하였다. 3. 포자형성을 전후하여 IMP와 AMP의 RNA에 대한 함량비율은 일정함을 알았다. 4. 포자형성 전기의 균체에 아데노신, 구아노신, 이노신, 크산토신을 처리한바 포자형성이 10배 이상 촉진되었다. 5. 포자형성 전기의 균체에 A. bisporus, F. velutipes, L. edodes의 열수추출액을 처리한바 포자형성이 $1.5{\sim}2$배 촉진되었다.

버섯류의 원형질체 나출을 위한 고효율 효소 선발 (Selection of High Efficient Enzyme for Protoplasts Isolation from Mushrooms)

  • 현재 시판되고 있는 세포벽 분해 효소 중 cellulase onozuka R-10(Yakult Honsha, Japan), yatalase(Takara), $Glucanex^R$ 200G(Novo Industry, Denmark)를 조합하여 Flammulina velutipes를 대상으로 가장 효율적인 방법을 선발하고, 선발된 방법을 Pleurotus ostreatus, P. eryngii, Hypsizygus marmoreus에 적용하였다. F. velutipes의 포자, 셀로판지에 배양한 균사, homogenizer로 마쇄한 균사체를 대상으로 공시한 세포벽 분해효소를 단독 혹은 조합하여 처리하였다. 그 결과, 포자현탁액이나 셀로판지에 배양한 균사를 사용하는 것에 비해 homogenizer로 마쇄한 균사체를 사용하는 것이 높은 수율로 원형질체를 분리할 수 있었다. 또한 시험에 사용한 효소 중선발된 $Glucanex^R$ 200G와 cellulase onozuka R-10 효소의 혼합 처리에서 고효율의 원형질체가 분리되었다. 분리된 원형질체를 대상으로 재생률을 조사한 결과, 0.39~0.51% 범위의 재생률을 나타내었다
