Enhancement of BDNF Production by Co-cultivation of Human Neuroblastoma and Fibroblast Cells

  • It has been proved that co-cultivation of human neroblastoma cells and human fibroblast cells can enhance nerve cell growth and the production of BDNF in perfusion cultivation. In batch co-cultivation, maximum cell density was increased up to 1.76${\times}$106 viable cells/mL from 9${\times}$105 viable cells/mL of only neuroblastoma cell culture. The growth of neuroblastoma cells was greatly improved by culturing both nerve and fibroblast cells in a perfusion process, maintaining 1.5${\times}$106 viable cells/mL, which was much higher than that form fed-batch cultivation. The nerve cell growth was greatly enhance in both fed-batch and perfusion cultivations while the growth of fibroblast cells was not. It strongly implies that the factors secreted from human fibrobast cells and/or the environments of co-culture system can enhance both cell growth and BDNF secretion. Specific BDNF production rate was not enhanced in co-cultures; however, the production period was increased as the cell growth was lengthened in the co-culture case. Competitive growth between nerve cells and fibroblast cells was not observed in all cases, showing no changes of fibroblast cell growth and only enhancement of the neuroblastoma cell growth and overall BDNF production. It was also found that the perfusion cultivation was the most appropriate process for cultivating two cell lines simultaneously in a bioreactor.

Solid media based on Monochamus alternatus to growth and physiologically active substance of Paecilomyces tenuipes fruiting bodies

  • Paecilomyces tenuipes (P. tenuipes) is a fungus cultivated artificially by South Korean researchers, utilizing rice bran as its substrate. The increased demand for this fungus has not been met with successful cultivation methods for fruiting body production in natural environments. Therefore, we tested the effect on the growth of P. tenuipes using a Solid media based on pests. In this results, the Solid media based on M.alternatus was effective in increasing the growth of P. tenuipes and the content of cordycepin. Moreover, we confirmed the conditions for manufacturing a Solid media based on M.alternatus for P. tenuipes growth. We suggested that the growth-promoting compounds offers valuable insights for optimizing fungal cultivation conditions, thereby enhancing productivity and contributing to a broader understanding of fungal physiology in varying nutritional environments.

The Role of Visitor's Positive Emotions on Satisfaction and Loyalty with the Perception of Perceived Restorative Environment of Healing Garden

  • Background and objective: The purpose of this study is to investigate the effects of visitors' positive emotions on satisfaction and loyalty with the perception of restorative environment of a healing garden created in an urban agriculture expo. Methods: The psychological indicators to the images of the healing garden were analyzed by the visitors' demographic variables and the three factors of plant cultivation activity level: plant cultivation experience, plant preference, and plant-related event. Results: Between age groups and occupational groups, significant differences were found statistically. The Perceived Restorativeness Scale(PRS) showed significantly differences between age groups in repose, fascination and legibility. The Positive Affect & Negative Affect Schedule(PANAS) showed statistically significant differences between age groups in positive emotions. In addition, we investigated the correlation between the PANAS and the three factors of plant cultivation experience level, the four factors of the PRS, satisfaction and loyalty. The three factors of plant cultivation experience level, the four factors of the PRS, satisfaction and loyalty showed a positive correlation with positive emotions and were inversely correlated with negative emotions significantly. Multiple regression analysis with dummy variables was conducted to examine the effects of plant cultivation activity level, attention restoration, and the PANAS on healing garden visitors' satisfaction and loyalty. As a result, among the four factors of the PRS, fascination and positive affectivity were significant variables that affect healing garden visitors' satisfaction and loyalty. Conclusion: The results indicated that the higher the attention restoration of visitors due to the fascination of the healing garden and the higher their positive affectivity and the more they have plant-related memories, the higher their impact on healing garden visitors' satisfaction and loyalty. Therefore, fascinating natural environments or greenery landscapes like healing gardens where people can contact plants would reduce negative emotions such as anger and sadness but to increase positive emotions such as pleasure, joy and satisfaction.

Impact of Different Environmental Conditions and Management Techniques on Productivity of Grassland in Central and Southern Area of Korea (중부 및 남부지역에서 초지환경과 관리기술의 차이가 초지의 생산성에 미치는 영향)

  • This experiment was carried out to study the effects of different environmental conditions and cultivation techniques on productivity of grasslands in central and southern area of Korea on 2017 and 2018. Average dry matter yield of grasslands at 10 actual production sites was 7,496 kg/ha. that was ranged from 4,652 to 13,292 kg/ha with least significant difference(LSD) of 1,577kg/ha between grasslands (p<0.05) on 2017. Average dry matter yield of grasslands at 10 actual production sites was 7,914 kg/ha. that was ranged from 3,927 to 12,372 kg/ha with LSD of 1,577kg/ha between grasslands(p<0.05) on 2018. Dry matter(DM) yield of grasslands have positive correlation with soil fertility (p<0.01) but not correlated with rainfall and air temperature among cultivation environments. And also DM yield of grasslands have positive correlation with grassland management techniques(p<0.01). These results suggest that practices of grassland management techniques and improvement of soil fertility are more important than cultivation environments by climate change for increasing the DM yield of grassland in central and southern area of Korea.

  • The Quality of human living depends on the environmental quality of the region sustaining the life. The environmental deterioration of the modern society is due to mechanical environmentalism. For the better quality of the life, The changes of recognition and attitude on the environments are required. These changes of mind are also important in environmental education for teenagers. The 7th national curriculum, officially anounced December 1998, focuses on the change of attitude to environments and practical behavior in real life for “Environments”, the environmental education curriculum in middle school. Basic elements of the curriculum are cultivation of the pro-environmental thinking, multi-levelling of teaching materials and methods, and encouraging of student participating activity. Actually, the curriculum construction is composed of stepped-levelling of teaching and learning, reasonable contents volume, encouraging of student practice, and suggesting of evaluation standards of textbook writing. Three main subjects of environmental education for middle school consist of (1) man and environment, (2) recognition of environmental problem, and (3) protection activity for environment. Methodology of environmental education can include multi-disciplinary approaches, variable teaching methods, and continuing evaluation of student practice and participation attitude. Environmental education for teenagers relating to the 7th national curriculum focuses on recognition of the environmental problems and practice activity in daily life. The recognition includes considering relationship of human life to environment, solving environmental problems in regional context, and development of comprehensive understanding concept of the environments. For the practice education, variable teaching methods, such as field survey and application of multi-media, are needed.

  • Processing techniques of remote sensed image data using computer have been recognized very necessary techniques to all social fields, such as, environmental observation, land cultivation, resource investigation, military trend grasp and agricultural product estimation, etc. Especially, accurate classification and analysis to remote sensed image da are important elements that can determine reliability of remote sensed image data processing systems, and many researches have been processed to improve these accuracy of classification and analysis. Traditionally, remote sensed image data processing systems have been processed 2 or 3 selected bands in multiple bands, in this time, their selection criterions are statistical separability or wavelength properties. But, it have be bring up the necessity of bands selection method by data distribution characteristics than traditional bands selection by wavelength properties or statistical separability. Because data sensing environments change from multispectral environments to hyperspectral environments. In this paper for efficient data classification in multispectral bands environment, a band feature extraction method using the Rough sets theory is proposed. First, we make a look up table from training data, and analyze the properties of experimental multispectral image data, then select the efficient band using indiscernibility relation of Rough set theory from analysis results. Proposed method is applied to LANDSAT TM data on 2 June 1992. From this, we show clustering trends that similar to traditional band selection results by wavelength properties, from this, we verify that can use the proposed method that centered on data properties to select the efficient bands, though data sensing environment change to hyperspectral band environments.

  • The management of non point sources was marked by the need for clean water environments. It was proposed the fundamentals to promote the reasonable land management in this study. We monitored rainfall events at two non point sources with different crop cultivations such as a sweet potato and a cherry tree for three years. Because the most important factor was rainfall, the rainfall runoff and pollutant loads were generated 100% in the case of rainfall ranges with 50 < rainfall (mm). However the frequency of rainfall runoff was interacted with the crop cultivation and soil characteristics in the case of rainfall ranges such as 30 < rainfall (mm) ${\leq}50^a$ and 10 < rainfall (mm) ${\leq}30^b$. The frequency of rainfall runoff was a : 60% and b : 5% in the cherry tree cultivation with growing significantly and pollutant loads were lower than that of the sweet potato cultivation. Meanwhile the frequency of rainfall runoff was a : 60% and b : 5% in the sweet potato cultivation.

  • In cope with insufficient agricultural labor and requirement of high quality product Hydroponics is a really good method. It makes the high density agriculture possible and all the growing environments controllable. So its research is so much progressing to maximize the quantity and quality of farm products. Furthermore, the big progress, in the research of a future agriculture, is systematically conducted for the automatic controlled system. In this paper, a new approach to the automation of the cultivation in a green house is suggested and a practical automatic control cultivation system is implemented. To automatically control and optimize the very nonlinear and time-varying growth of farm products, a hybrid strategy(FECA; Fuzzy Expertized Control Algorithm) is proposed which serially combines a fuzzy expert system with the fuzzy logic control. The fuzzy expert system(FMES; Fuzzy Model-based Expert System) is intended to overcome the non-linearity of the growth of farm products. The part of fuzzy controller is incorporated to solve the time-variance of the growth of farm products. Finally, the efficiency and the effectiveness of the implemented agricultural automation system is presented through the cultiviation results.

  • The above results indicate that vegetable crop production using vinyl houses continously increases. However, technical support and facility improvement are still insufficent to establish a full production system. In order to improve the cultural environment in facilities for better vegetable culture, several facters should be considered. 1. Facility design and arrangement should be made to improve micro-environments for crop growth and development because direction of facility and covering materials can affect the amount of light trasmitted into vinyl houses. 2. Cultivation of several leading varieties in each crop may not provide stable production and profit especially under undesirable environmental conditions. Thus, it is required to grow not only leading varieties but also other cultivars to decrease the economic losses. 3. Crops should be carefully managed when planted in early season not to experience low temperatures which induce growth retards. Early season cultivation requires proper selection of cultivars which are resistant to low temperture. 4. Active control of micro-environment in facilities should be made via improving the ventilation and irrigation systems.

Heading Ecology of Rice Varieties Adaptable to the Temperature and Day-Length Conditions in North Korean Regions (북한 지역 기온과 일장 환경 적응 벼 품종의 출수생태 특성 분석)

  • We investigated the basic vegetative phase (BVP), photoperiod-sensitivity (PS), and thermo-sensitivity (TS) of 40 rice varieties to characterize their heading ecology that is adaptable to North Korean rice production areas. The ranges were 12 - 43 days for BVP, 0 - 74 days for PS, and 15 - 33 days for TS, depending on the variety. The number of days from transplanting to the heading stage (DTH) was significantly correlated with PS in the 13 major rice production regions where all 40 varieties (including early-, middle-, and mid-late-maturing varieties) were tested. DTH was significantly correlated with BVP and TS but not with PS in the 10 low-temperature regions where only 28 early-maturing varieties were tested. The heading ecology of the adaptable varieties for each region could be characterized by an exponential equation between the BVP and PS of varieties that headed at the border of the safe marginal heading date (SMHD) for each of the seven regional environments (Kaesong, Haeju, Yongyon, Singye, Sariwon, Nampo, and Pyongyang). A PS of 25 - 30 days or less was an additional adaptive trait in the Sariwon and Pyongyang environments. The varieties that reached the heading stage not later than the SMHD in six regional environments (Anju, Kusong, Sinuiju, Changjon, Wonsan, and Supung) and those that reached the heading stage not later than the late marginal heading date (LMHD) in four regional environments (Hamhung, Pyonggang, Huichon, and Kanggye) had both a PS of 26 days or less and a BVP of 25 - 28 days or less. In the Yangdok, Sinpo, and Chunggang environments, varieties that reached the heading stage not later than the LMHD for each region had a BVP of 15 - 20 days or less. The results suggested that a shortened BVP trait should be introduced to existing early-maturing rice varieties to reduce the duration of growth period to reach the heading stage.
