Effect of growth condition on mycelial growth and fruiting body cultivation of Cordyceps militaris wild strain

  • Cordyceps militaris is widely used in China, Korea, and other Asian countries as both a traditional medicinal ingredient and an edible fungus. This study aimed to optimize the growth conditions and fruiting body production of C. militaris by investigating various culture media and physical parameters such as pH, aeration, illumination, temperature, spawn materials, and oat-sawdust-based substrate formulations. After a 7-day incubation period, oats with a pH of 6.0, under sealed and illuminated conditions at 32℃, demonstrated the most effective mycelial growth. Substrates consisting of 70% oat and 30% sawdust had the shortest incubation time of 30.5 days for fruiting body formation. The basidiospores showed a typical germination pattern where the sporidium produced a single germ tube that elongated, and branched to form monokaryotic primary mycelia. In conclusion, using oats as a substrate in the cultivation of C. militaris could reduce production costs and help protect the environment.

  • The water in the crop cultivation shows difference according to the variety of crop, cultivations period and climatic condition. The growth and development, quantity and fruit enlargements are affected by soil water conditions. In previous study, leaf area and photosynthesis are decreased by lower soil moisture. Other research reported that excess moisture condition at vegetative and reproductive growth period in cultivation of soybean caused highest reduction in crop growth rate (CGR) and dry weights of plant parts. In particular, the damage was bigger during vegetative growth stage than reproductive growth period. Soybean (Glycine max (L.) Merill) is useful and popular crop throughout the world. It is very popular crop in Korea, China, Japan and other Asian countries. Soybeans used in various way including soybean sprouts, paste, soymilk, oil and tofu. Two soybean cultivars grown in four different irrigation conditions were determined for physiological responses. In this study, we examined leaf area (LA), leaf dry weight (LDW), specific leaf area (SLA), root dry weight (RDW) and shoot height (SH) in different water conditions. 50mL/9day irrigation periods showed the lowest contents in LA, LDW, RDW, SH. Water deficit caused increase of leaf Water saturation deficits (WSD), Cheongjakong 3 and Taekwangkong showed increase of leaf water saturation deficits (WSD) in drought conditions and leaf water potential and stomatal conductance were decreased. Photochemical efficiency was decreased in 50mL/1day irrigation condition while, there was decrease of growth and development in 50mL/9day with drought.

Proteomic Changes in Odae Polished White Rice Grown at Different Cultivation Conditions (재배환경에 따라 변화하는 오대벼 백미의 단백질체 분석)

  • It has been known that the proteome profiles in the period of growth and development of rice are changed by the growth conditions including temperature, soil, and fertilization. In this study, the proteome profiles of Odae polished white rice grown in Chulwon and Chilgog were compared on 2-dimensional(D) gels. The differentially expressed proteins were selected from the 112 identified total proteins and classified into functional groups. The most significantly differentially expressed proteins were stress responsive proteins; Ent-kaur-16-ene synthase, which is responsible for synthesizing a plant hormone gibberellin, was expressed in Chulwon rice and heat shock proteins were in Chilgog rice, respectively. Xylanase inhibitor protein, which inhibits the enzyme xylanase produced by pathogenic fungi and Bacilli, was expressed significantly high in Chilgog rice grown at high temperature. Differential expressions of transporter proteins were observed both in Chulwon and Chilgog rice. Regarding the facts that Chilgog rice contained relatively higher amount of proteins than Chulwon rice and Chulwon rice showed large number of proteins were differentially expressed, it can be concluded that different cultivation conditions could change the protein expression profiles in rice in various ways, including elevation of protein amount or differential expressions of specific proteins, etc. The results suggest that the characteristics of the profiles of the proteome in the polished white rice are definitely changed by the environmental factors including high temperature. The results can be utilized for the development of the proper cultivation conditions for the production of high quality rice with good palatability.

  • In this paper, we propose a system that can provide a environment for plant cultivation in connection with LED plant factories and enable users to participate in plant cultivation remotely to engage in personal hobbies. The proposed system can monitor the growth conditions of plants through various sensors and remotely adjust the cultivation environment required for plant growth through the Arduino system, so that users can feel the satisfaction of plant cultivation and harvesting as a hobby. On the other hand, we suggest a mutual benefit structure for plant factory and users by securing a certain amount of income source to factory, by paying the idle space to the individual online. This paper demonstrates the feasibility of the proposed system by making the prototype of the remote plant cultivation consignment system using the Arduino and Android application(App.), and contributes to popularize the LED plant factories and expand the business area in future.

Assessment of Safety Cultivation Zones for Sweet Persimmon by Warmth Index Change in South Korea (남한 온량지수의 변화와 단감의 안전재배에 관한 연구)

  • The monthly mean air temperature datasets of 61 stations in South Korea from 1973 to 2012 were collected to calculate trends in the warmth index (WI) and to analyze the potential enlargement of safety cultivation limit for sweet persimmon. The WI averaged over the last 40 years was 104.1 (℃·Month) at 61 stations, with the highest at Seogwipo station (WI=137.9) and the lowest at Daegwallyeong station (WI=60.9). It has increased by 1.8 (℃·Month) per 10 years over the last 40 years, with the highest in the year 1994 (WI=112.0) and the lowest in the year 1976 (WI=94.7). When the possible stations for sweet persimmon cultivation were classified by the basis on WI≥100, 38 out of the 61 weather stations were included in the safety cultivation zone for sweet persimmon for the last 40 years. On the other hand, the number of weather stations within the safety cultivation zones for sweet persimmon for the last 10 years (from 2003 to 2012) were 47 by adding additional 9 stations (Socho, Wonju, Chungju, Seosan, Uljin, Yangpyeong, Icheon, Cheonan, and Geochang stations). A further study of the climate conditions and soil characteristics is required for a better assessment of the safety cultivation zones for sweet persimmon.

Cultural Characteristics and Log-Mediated Cultivation of the Medicinal Mushroom, Phellinus linteus

  • The optimal conditions for mycelial growth of Phellinus linteus ATCC 26710 were determined to be a log length of 20 cm, temperature of $30^{\circ}C$ and pH of 6.0. Mycelial growth was excellent on the mushroom complete medium, and was optimal when sucrose, man nose and glucose were supplied as carbon sources. Potassium nitrate and sodium nitrate as nitrogen sources supported good mycelial growth. To evaluate P. linteus mycelial colonization on logs, sterilized short log inoculation, drilling inoculation and log-end sandwich inoculation techniques were used. Only sterilized short log inoculation produced good mycelial colonization. Initial mycelial growth and full mycelial colonization were best on 20 cm logs having 42% moisture content. The initial mycelial growth of P. linteus was accelerated over 12hr of sterilization. Basidiocarp formation was optimal using a burying method of logs after $5{\sim}6$ months, and fruiting body formation was superior in cultivation house conditions of $31{\sim}35^{\circ}C$ and in excess of 96% relative humidity.

  • The water requirement of transplanting culture(TC) consisting of 16 plots and direct seeding culture in a well-drained paddy field(DC) also having 15 plots were measured in order to clarify the influence factors on water requirement. Each plot was maintained under different cultivation conditions. Those varied cultivation conditions were classified into 7 items. Subsequently, the relationship between water requirement and 7 items were analyzed by evaluating the average water requirement in each condition to clarify the items influencing water requirement. From the analysis of the results, the water requirement of DC was determined to be 3.5 times of the TC. Furthermore, water requirement in DC and TC were influenced mainly by ponding depth and locational condition respectively.

  • The optimal cultural conditions for production of the bluish purple prgment by the cultivation of Streptomyces californicus KS-89 were determined with various substrates. The carbon and nitrogen sources on the production of pigment indicated that soluble strarch and glycerol as carbon sources and sodium glutamate sodium nitrate as nitrogen sources given a mzimum yield of the pigment at 3$0^{\circ}C$ for 7 days. The addition of ferrous sulfate was essential. The highest production of pigment was observed with cultivation in a medium containing 2.0% soluble starch 1% glycerol 0.5% sodium glutamate 0.05% sodim nitrate 0.001% L-proline 0.025% K2HPO4 0.005% MgSO4 .7H2O, 0.04% FeSO4.7H2O, 0.001% thiamine.HCl and pH7.0.

Development of Cultivation Facility Models to Reduce High Temperature Damage in Oak Mushroom (Lentinula edodes) Cultivation Using Bed-logs (고온피해 경감을 위한 표고 원목재배사 모델개발)

  • Five empirical farmhouses were selected to reduce the high temperature damage in oak mushroom cultivation using bed-logs. The cultivation facilities were categorized as follows: those having two blackout curtains or one blackout curtain and outdoor oak mushroom cultivation. The inequality of the indoor condition, oak mushroom hyphae rampant ratio, and fruit body production in each test condition was evaluated. $3^{\circ}C$ was lower in indoor temperature of cultivation facility having two blackout curtains than one blackout curtain. Specifically, the indoor air humidity average of cultivation facilities having one or two blackout curtains was 10% lower than that of outdoor oak mushroom cultivation. This condition is not ideal for oak mushroom cultivation as continuous indoor humidity control is essential for producing good fruit bodies. The Inoculated bed-log surface and oak mushroom hyphae rampant ratio of bed-logs cultivated with two blackout curtains was superior to other tested conditions. The mushroom production ratio observed in facilities with two blackout curtains was 117-204% higher than those cultivated in facilities with only one blackout curtain. Furthermore, the mushroom production ratio increased in based on these findings, we recommended five cultivation facility models to reduce high temperature damage in oak mushroom cultivation using bed-logs.

  • The medium used for bottle cultivation of mushrooms is made according to a specific workflow; this includes mixing the raw materials, adjusting the moisture content, placing the mixture into bottles, and sterilizing and cooling the medium. Medium composition should be kept constant throughout the year. placed in the bottle, choice of materials, moisture content, weight of the medium, bulk density, nitrogen content, three-phase medium, such as suitable conditions. To achieve constant medium composition a comprehensive model has been established in accordance with the technology developed in the work process with respect to medium compositions piecemeal. By comprehensively applying the various methods to establish medium composition, our country can expect to grow at over four seasons and overcome the many difficulties in mushroom mycelial growth periods, cultivation periods, quantity, and quality.
