미세결정화키틴과 팜유의 수준을 달리한 식이가 흰쥐의 지방대상에 미치는 영향 (The Effects of Micro Crystalline Chitin and Palm Oil on the Lipid Metabolism in the Rat)

  • The effects of MCC and palm oil at different levels on lipid metabolism were assessed in adolescent rat. Dietary fat levels were 20% and 40% (kcal/kcal) and MCC level were 0%, 2%, and 4% (wt/wt). The experimental period took 8 weeks. During the experiment, weight gain and food efficiency ration were not affected by dietary factors. The weight of thymus however, was lower in high fat groups than in middle fat groups. The contents of total lipid and total cholesterol in plasma and HDL-cholesterol in plasma and HDL-cholesterol were significantly affected by dietary factors. The concentrations of cholesterol in LDL and VLDL , and the triglyceride content of VLDL was higher in high fat groups than in middle fat groups. Adding MCC lowered the LDL triglyceride in the high fat groups. The analysis of plasma fatty acids generally reflected the composition of the dietary palm oil. MCC had significant effects on total lipid and triglyceride of feces, but not on total cholesterol . In conclusion, the level of MCC had little hypolipodemic effects on lipid metabolism in rats fed a high fat diet. the middle fat group that contained 4% MCC showed increased contents of fecal triglyceride than the others, indicating that MCC had an effect on lipid absorption. Therefore, the other physiological functions of MCC need to be tested for their useful applications.

Effect of Chitin Sources on Production of Chitinase and Chitosanase by Streptomyces griseus HUT 6037

  • The advantage of using Streptomyces griseus HUT 6037 in the production of chitinase or chitosanase is that the organism is capable of hydrolyzing amorphous or crystalline chitin and chitosan according to the type of the substrate used. We investigated the effects of the enzyme induction time and chitin sources, CM-chitosan and deacetylated chitosan (degree of deacetylation 75-99%), on production of chitosanase. We found that this strain accumulated chitosanase when cells were grown in the culture medium containing chitosanaceous substrates instead of chitinaceous substrates. The highest chitosanase activity was obtained at 4 dyas of cultivation with 99% deacetylated chitosan. The specific activities of chitinase and chitosanase were 0.91 and 1.33 U/mg protein at 3 and 5 days, respectively. From the study of the enzymatic digestibility of various degrees of deacetylated chitosan, it was found that (GlcN)$_3$, (GlcN)$_4$and (GlcN)(sub)5 were produced during the enzymatic hydrolysis reaction. The results of this study suggested that the sugar composition of (GlcN)$_3$was homogeneous and those of (GlcN)$_4$and (GlcN)(sub)5 were heterogeneous.

Effective Production of Chitinase and Chitosanase by Streptomyces griseus HUT 6037 Using Colloidal Chitin and Various Degrees of Deacetylation of Chitosan

  • The advantages of the organism Streptomycs griseus HUT 6037 is that the chitinase and chitosanase using chitinaceouse substrate are capable of hydrolyzing both amorphous and crystalline chitin and chitosan. We attempted to investigate the optimization of induction protocol for high-level production and secretion of chitosanase and the influence of chitin and partially deacetylated chitosan sources (75∼99% deacetylation). The maximum specific activity or chitinase has been found at 5 days cultivation with the 48 hours induction time using colloidal chitin as a carbon source. To investigate characteristic of chitosan activity according to substrate, we used chitosan with various degree of deacetylation as a carbon source and found that this strain accumulates chitosanase in the culture medium using chitosanaceous substrates rather than chitinaceous substrates. The highest chitosanase activity was also presented on 4 days with 99% deacetylated chitosan. The partially 53% deacetylated chitosan can secrete both chitinase and chitosanase which was defined as a soluble chitosan. The specific activities of chitinase and chitosanase were 0.89 at 3 days and 1.33 U/mg protein at 5 days, respectively. It indicate that chitosanase obtained from S. griseus HUT 6037 can hydrolyze GlcNAc-GlcN and GlcN-GlcN linkages by exo-splitting manner. This activity increased with increasing degree of deacetylation of chitosan. It is the first attempted to investigate the effects of chitosanase on various degrees of deacetylations of chitosan by S. griseus HUT 6037. The highest specific activity of chitosanase was obtained with 99% deacetylated chitosan.

Intein을 이용한 대장균에서의 Trichoderma reesei 유래의 Cellobiohydrolase I 섬유소 결합 도메인의 발현 (Intein-mediated expression of Trichoderma reesei Cellobiohydrolase I Cellulose Binding Domain in E. coli)

  • Cellulose binding domains (CBDs) of cellulases are thought to assist in the hydrolysis of insoluble crystalline cellulose. To gain sufficient amount of CBDs, the self-cleavable intein tag was used for expression and purification of Trichoderma reesei cellobiohydrolase I CBD in E. coli. Synthetic CBD genes, CBD or linker-CBD were cloned into expression vector pTYB11. Recombinant CBDs were successfully purified by intein mediated purification with an affinity chitin-binding domain. The final yields of recombinant CBD and linker-CBD were 3.2 mg/L and 1.4 mg/L, respectively. The functional bindings of recombinant CBDs were confirmed by Avicel binding experiments. The simple and easy purification method using self-cleavable intein tag can be further used in pretreatment of crystalline cellulose or characterization of engineered CBDs.

  • 셀룰로오스 아밀로오스 키토산, 키틴, 알긴산, 풀루란 또는 아밀로펙틴을 6- [4- (4'- (니트로페닐아조)펜옥시)]펜타노일 클로라이드(NA6C)와 반응시켜 전치환 또는 거의 전치환 6- [4- (4'- (니트로페닐아조)펜옥시카보닐)] 펜타노화 다당류 유도체들을 합성함과 동시에 이들의 열방성 액정의 거동들을 검토하였다. NA6C의 경우와 같이, 모든 다당류 유도체들은 단방성 네마틱 상들을 형성하였다. 이러한 사실은 다당류 유도체들의 액정 구조는 다당류 골격에 의해 결정되는 것이 아니고 mesogenic 곁사슬 그룹들에 의해 결정됨을 시사한다. 이것이 셀룰로오스 유도체들을 제외한 다당류 유도체들이 열방성 네마틱 상을 형성한다고 하는 최초의 보고이다. 다당류 유도체들에 있어서 관찰되는 네마틱 상들의 열적 안정성과 질서도는 아조벤젠 그룹들을 유연한 스페이서를 통하여 유연한 혹은 강직한 골격들에 도입시켜 얻은 고분자들에 대해 보고된 결과들과 현저히 다르다. 이들의 결과를 주사슬과 곁사슬의 배열 그리고 주사슬의 유연성의 차이와 관련하에서 검토하였다.

(8-콜레스테릴옥시카보닐)헵타노화 다당류들의 열방성 액정 특성 (Thermotropic Liquid Crystalline Properties of (8-Cholesteryloxycarbonyl)heptanoated Polysaccharides)

  • 셀룰로오스, 아밀로오스, 키토산, 키틴, 알긴산, 풀루란 또는 아밀로펙틴을 (8-콜레스테릴옥시카보닐)헵타 노일 클로라이드 (CH8C)와 반응시켜 전치환 또는 거의 전치환 (8-콜레스테릴옥시카보닐) 헵타노화 다당류 유도체들을 합성함과 동시에 이들의 열방성 액정의 거동들을 검토하였다. CH8C 의 경우와 같이, 아밀로펙틴 유도체를 제외한 모든 다당류 유도체들은 좌측방향의 나선구조를 지니며 온도상승에 의해 광학피치들 $({\lambda_m}'s)$이 감소하는 단방성 콜레스테릴 상들을 형성하였다. 아밀로펙틴 유도체도 좌측방향의 나선구조를 지닌 단방성 콜레스테릴 상을 형성하나 다른 다당류 유도체들과 달리 콜레스테릴 상의 전 범위에서 반사색깔들을 나타내지 않았다. 이러한 사실은 콜레스테릴 그룹에 의한 나선의 비틀림력은 아밀로펙틴 중의 분기구조에 민감하게 의존함을 시사한다. 다당류 유도체들에서 관찰되는 액정 상의 열적 안정성과 질서도, 동일한 온도에서의 ${\lambda}_m$ 의 크기 그리고 ${\lambda}_m$의 온도 의존성은 콜레스테릴 그룹들을 유연한 스페이서들을 통하여 유연한 흑은 반강직한 골격들에 도입시켜 얻은 고분자들에 대해 보고된 결과와 전혀 다르다. 이들의 결과를 주사슬과 곁사슬의 화학구조 그리고 주사슬의 유연성의 차이와 관련하에서 검토하였다.
