Agrobacterium-mediated transformation of Bacillus thuringiensis cry1Ac gene in chrysanthemum (Dendranthema grandiflorum Kitamura) 'Linneker Salmon' (국화(Dendranthema grandiflorum Kitamura) 'Linneker Salmon'에 Agrobacterium을 이용한 Bacillus thuringiensis cry1Ac 유전자의 형질전환)

  • Cry1Ac gene was introduced into chrysanthemum (Dendranthema grandiflorum Kitamura) 'Linneker Salmon' through Agrobacterium-mediated gene transformation to develop new lines showing resistance to tobacco cutworm (Spodoptera litura). Cry1Ac gene was transferred into chrysanthemum by Agrobacterium C58C1 containing pCAMBIA2301. After infection of Agrobacterium C58C1 with leaf segments, the segments were cultured on regeneration medium (MS + 1.0 mg/L BA + 0.5 mg/L IAA) containing 10 mg/L kanamycin for the first selection, on the same medium containing 20 mg/L kanamycin for the second selection, and on rooting medium (MS basal medium) containing 20 mg/L kanamycin for the third selection. Until the third selection, sixty nine plantlets (1.6%) were survived and rooted. Thirty six ones (0.8%) among them were confirmed as putative transformants with nptll gene by nptll primer PCR, and 35 (0.8%) of 36 ones as transformants with nptll gene and cry1Ac gene by Southern analysis. The gene transformation efficiency of cry1Ac gene was favorable with 0.8%. The resistance of tobacco cutworm (Spodoptera litura) in chrysan-themum transformant introduced cry1Ac gene was tested in green house. Three transformants were confirmed to have resistance to tobacco cutworm.

  • We report the draft genome sequence of Bacillus thuringiensis serovar aizawai AS23, an insecticidal strain targeting lepidopteran pests, which was isolated from the rhizosphere of Korean melon (Cucumis melo L.). The genome of strain AS23 comprising 6,846,584 bp with a G + C content of 34.83% was assembled to 11 contigs obtained using hybrid assembly. Additionally, we mined the genome for pesticidal genes, identifying several insecticidal genes, including Cry1Aa3, Cry1Ca9, Cry1Da2, Cry1Ia44, Cry2Ab41, Cry9Ea9, Spp1Aa1, and Vip3Aa86.

  • 언어 발달의 조기 단계를 이해하기 위한 일환으로 crying은 언어전 발달의 기초 단계로서 여러 학문적 분야에서 많은 연구가 있어왔다. 그러나 구순구개열(CLP))환아의 경우는cry-producing/control mechnism에 variation이 많은 이유로 이 분야의 연구는 거의 없는 실정이다. 이에 본 연구에서는 다음과 같은 의문점을 가지고 CLP환아의 cry feature에 대한분석을 하였다. 첫째, 정상아와 CLP환아의 cry에 전형적인 차이가 있는가? 둘째, CLP환아의 술전, 술후 cry feature에 변화가 있는가? 셋째, cry분석이 CLP환아의 이후 speech disorder에 대한 언어전 평가로서의 가치가 있는가? 넷째, 특정 parameter가 언어전 평가에 적절한 도구로 작용할 수 있는가? 생후 15개월 이내의 CLP 환아 3명과 유사한 나이대의 정상아 8명의 cry에 대한 공기역학 및 음향음성학적 분석을 통해 CLP 환아와 정상아, CLP환아의 술전, 술후 cry특성을 비교 분석하였다. 결과는 다음과 같다. 1 공기역학적 분석 1) airflow는 CLP 환아의 경우 정상아보다 약간 높았고 술 후 약간 증가하였다. 2)폐활량을 나타내는volume에서는 정상아보다 술전 CLP환자의 경우 보상적으로 더 큰 수치를 보였고 술후 약간 증가하였다. 3)강도를 나타내는 parameter(SPL)에서는 정상아 보다 술전 CLP환자의 계측치가 약간 작았으나 술 후 증가하는 양상을 보였다. 2. 음향음성학적 분석 1)기저 주파수 분석시 정상아에 비해 술 전 CLP환자의 경우 계측치가 약간 낮았으나 술 후 증가하여 정상군의 계측치에 근접하였다. 2)강도를 나타내는energy 측정시 정상아에 비해 술 전 CLP계측치가 보상성으로 약간 큰수치를 나타내었고 술 후 약간 더 증가하였다. 3) Shimmer에서는CUI환자의 술후계측치가술전에 비해 현저히 감소하여 정상군의 수치에 근접하였다.

Expression of a Fusion Protein with Cry1Ac Protein and a Scorpion Insect Toxin in Acrystalliferous Bacillus thuringiensis Strain

  • Expression of a fusion protein between B. thuringiensis crystal protein, Cry1Ac1 and a scorpion insect toxin (AaIT, Androctonus australis Hector insect toxin) in acrystalliferous B. thuringiensis strain (Cry-B strain) was examined. The cry 1Ac1 gene was cloned in B. thuringiensis-E coli shuttle vector, pHT3101, under the control of the native cry 1Ac1 gene promoter (pProAc) and a gene encoding AaIT was inserted in XhoI site in the middle of the cry 1Ac1 gene (pProAc-ScoR). B. thuringiensis Cry-B strain carrying pProAc-ScoR (PyoAc-ScoR/CB) produced an inclusion body of irregular shape and the expressed fusion protein is approximately 65 kDa in size. Sporulated cells and spore-crystal mixtures of ProAc-ScoR/CB had insecticidal activity against Plutella xylostella larvae, showing $LT_50$ of ProAc-ScoR/CB (22.59 hrs) lower than that of ProAc/CB (30.06 hrs) at $1{\times}{10^7} {CEU/cm^2}$. These results suggest that the fusion protein including a B. thuringiensis crystal protein and an AaIT may be functionally expressed in B. thupingiensis. Moreover, we verified the additive toxicity of AaIT, which is a new feasible candidate for insect control.

  • The characteristics of parasporal inclusion body from Bacillus thuringiensis subsp. kurstaki KB099 isolate which is high bioactive to the tobacco cutworm, Spodoptera litura, were examined. Parasporal inclusion of B. thuringiensis subsp. kurstaki KB099 isolate showed only 1 band at 130 kDa compared with B. thuringiensis subsp. kurstaki HD-l isolate producing 2 protein bands at 130 kDa and 60 kDa from by SDS-PAGE analysis without any enzyme treatment. Also, we confirmed that gut extract of sensitive S. litura KB099 isolate had digested only 60 kDa ${\delta}$-endotoxin protein. When the digestive enzyme of sensitive insect responsible for parasporal inclusion from KB099 and HD-l isolate was treated to each of them, protein band 60 KDa of KB099 was maintained up to 12 hours but all bands of HD-l were disappeared within 6 hours. In KB099 isolate, 6 genes (Cry1Aa, Cry1Ab, Cry1Ac, Cry1C, Cry1D and Cry1I) were identified by PCR analysis. Also, $Cry^-$ mutant of KB099 isolate was investigated by phase- contrast microscope, SDS-PAGE and PCR.

  • The cryIB, truncated cryIB$[cryIB({\alpha})]$, cryIA(b), and cytA genes, encoding 135-, 89-, 131-, and 27-kDa proteins, respectively, from Bacillus thuringiensis were cloned into a shuttle vector pBES and expressed in E. coli and Bacillus species. The morphology and solubility in alkaline buffer of the insecticidal crystal proteins were investigated. Transformation of intact cells of E. coli and Bacillus species was achieved by electroporation. High field strength of 11.0 kV/cm and resistance of 129 ohms were required for efficient transformation of E. coli strains and 4.5 kV/cm and 48 ohms for Bacillus species. Strains of recombinant E. coli and Bacillus species produced the insecticidal crystal proteins and accumulated as the same bipyramidal and irregular structures as those of CryIB and IA(b) and CytA of B. thuringiensls, respectively. The insecticidal crystal proteins accumulated in recombinant E. coli wire smaller in size than those in recombinant Bacillus species. The solubility in alkaline buffer of the insecticidal crystal proteins of recombinant E. coli increased gradually as the pH increased, whereas in the case of Bacillus species the solubility increased gradually as the pH increased up to 9 and then the solubility increased greatly up to two times higher than that of E. coli proteins.

Detection of cryIB Genes in Bacillus thuringiensis subsp. entomocidus and subsp. subtoxicus

  • To find new crystal protein genes, we screened 42 Bacillus thuringiensis strains of serovar standards by Southern hybridization with a cryI-specific probe which was amplified from B. thuringiensis subsp. kurstaki HDl by polymerase chain reaction (PCR). Two strains, B. thuringiensis subsp. entomocidus HD9 and subsp. subtoxicus HD109, generated weak signals under the low-stringency hybridization conditions. Further analysis with Southern hybridization revealed that the two strains contained cryIB genes which are slightly different from those of B. thuringiensis subsp. thuringiensis HD2. These results were confirmed by PCR with cryIB-specific primers followed by the restriction analysis of PCR products.

Determination of Plasmids Encoding Crystal Toxic Protein Gene in Bacillus thuringiensis var kurstaki HD-1 (Bacillus thuringiensis var kurstaki HD-1의 내독소 단백질 유전에 관여하는 plasmid의 결정)

  • The objective of this study is to identify plasmids of Bacillus thuringiensis var. kurstaki HD-1(B. t k HD-1) toxic to lepidopteran larvae. The results from agarose gel electrophoresis indicated that the bacterium contained 9 plasmids with approximate sizes of 1.4, 4.9, 5.4, 9.3, 10, 29, 44, 52, and 150 megadaltons(Md). By treating the wild type of B. t k HD-1 with either SDS or EtBr as curing agent, 26 cured mutants of the bacterium were obtained, 9 of them were crystallifereous(cry+) and the others acrystallifereous(cry-). Plasmids from B. t k HD-1 were transferred to B. cereus 569 strR cry- recipients(Bc569 M1). Among 13 isolates of Bc569 M1 transcipient, 11 of them were capable of producing the crystal toxic proteins. The plasmid patterns of Bc569 M1 transcipients and partially curved mutants of B. t k HD-1 on agarose gel electrophoresis suggested that the 29 and 44Md plasmids should be involved in the production of crystalline toxic proteins.

A Highly Pathogenic Strain of Bacillus thuringiensis serovar kurstaki in Lepidopteran Pests

  • In order to detect and identify the most toxic Bacillus thuringiensis strains against pests, we isolated a B. thuringiensis strain (Bn1) from Balaninus nucum (Coleoptera: Curculionidae), the most damaging hazelnut pest. Bn1 was characterized via morphological, biochemical, and molecular techniques. The isolate was serotyped, and the results showed that Bn1 was the B. thuringiensis serovar, kurstaki (H3abc). The scanning electron microscopy indicated that Bn1 has crystals with cubic and bipyramidal shapes. The Polymerase Chain Reactions (PCRs) revealed the presence of the cry1 and cry2 genes. The presence of Cry1 and Cry2 proteins in the Bn1 isolate was confirmed via SDS-PAGE, at approximately 130 kDa and 65 kDa, respectively. The bioassays conducted to determine the insecticidal activity of the Bn1 isolate were conducted with four distinct insects, using spore-crystal mixtures. We noted that Bn1 has higher toxicity as compared with the standard B. thuringiensis subsp. kurstaki (HD-1). The highest observed mortality was 90% against Malacosoma neustria and Lymantria dispar larvae. Our results show that the B. thuringiensis isolate (Bn1) may prove valuable as a significant microbial control agent against lepidopteran pests.
