Optimization of Extraction Conditions for Soluble Ginseng Components Using Microwave Extraction System under Pressure (가압형 마이크로웨이브 추출장치를 이용한 가용성 인삼성분의 추출조건 최적화)

  • Microwave extraction system equipped with closed vessels was applied to confirm its capability of extracting ginseng solubles including saponin, which was monitored to optimize extraction con ditions by response surface methodology. Total yield increased with the decrease in particles size and more extraction steps. Soluble components were completely extracted by operating the system within 6 min per one step, which should be repeated by 3 or 4 times. Optimized conditions for maxi mum extraction of response variables(total yield, crude saponin) were 40~50% of ethanol concentration, about 140oC of extraction temperature, and within 6 min of the extraction time. The extraction effi ciency of total yield was dependent on the decrease of ethanol concentration, while crude saponin content was favored on the higher degree of ethanol concentration. According to the temperature guideline, below 90oC of extraction temperature, the optimal ranges of extraction conditions were predicted as 30~50% of ethanol concentration and 2.5~6.5 min of extraction time. Estimated values of total yield and crude saponin were in good agreement with experimental values.

Quality Characteristics of 4 Year-old Ginseng by Enzymatic Hydrolysis Conditions (4년근 인삼의 효소적 가수분해 조건에 따른 품질특성)

  • This study investigated quality characteristics of 4 year-old ginseng by enzymatic hydrolysis conditions to increase utilization. Ginseng was ground after steaming and was each treated with hydrolase A, B, C and D. When quality characteristics by the enzymes were examined, no significant difference was observed with pH of 5.5~5.6 and the sugar content of 4.0~4.33. The crude saponin content was the highest in ginseng treated with D, followed by B, C and A. The crude saponin, the reducing sugar and the total sugar contents increased until 0.3% (w/w) concentration in enzyme D with no significant difference by its concentration. Although active ingredients increased with time passage of hydrolysis, no significant change was found after three hours and the crude saponin content was the highest when ginseng was treated at $60^{\circ}C$. From these results, optimum conditions for 4 year-old ginseng were $60^{\circ}C$ for 3 hours with 0.3% (w/w) enzyme D, and under these conditions the reducing sugar, the total sugar and the crude saponin contents recorded 18.11, 36.21 and 4.23 mg/g, respectively. Therefore, enzymatic hydrolysis was found to be effective in increasing active ingredients of 4 year-old ginseng with various usages expected.

Comparison of Phytochemical Constituents According to the Cultivation Method (Paddy Field Cultivation, Upland Field Cultivation) of Korean Fresh Ginseng (한국산 수삼의 재배환경(논, 밭)에 따른 식물화학성분 비교)

  • Objectives: This study aimed to examine the changes in ingredients according to its cultivation method by examining the content of saponin and non-saponin components of ginseng. Methods: Ginseng saponin component analysis was compared and reviewed using the high-performance liquid chromatography method, and acidic polysaccharide component was measured using the carbazole sulfuric acid method. Results: The comparative analysis of ginseng saponin content of 4 and 6 years old fresh ginseng showed the following results. According to the cultivation method, upland field cultivation fresh ginseng showed higher average content of crude saponin than paddy field cultivation fresh ginseng. Whereas, paddy field cultivation fresh ginseng showed higher average content of total saponin than upland field cultivation fresh ginseng. Ginsenoside Rb1 showed higher content of paddy field cultivation fresh ginseng than upland field cultivation fresh ginseng in 6 years old ginseng. However, it showed higher content of upland field cultivation fresh ginseng than paddy field cultivation fresh ginseng in 4 years old ginseng. Additionally, ginsenoside Rg1 showed higher content of paddy field cultivation fresh ginseng than upland field cultivation fresh ginseng in 6 years old ginseng, whereas upland field cultivation fresh ginseng showed higher content of paddy field cultivation fresh ginseng in 4 years old ginseng. The effect on the content of ginseng saponins and acidic polysaccharides according to paddy field and upland field cultivation methods is considered to be small. Conclusions: The paddy field cultivation method, which is more efficient in production cost due to depletion of farmland and long-distance cultivation, is considered to be an economical cultivation method.

Proximate Analysis and Antioxidant Activity of Cultivated Wild Panax ginseng (연근별 산양삼의 일반성분 및 항산화 효과)

  • Proximate analysis and antioxidant activity of cultivated wild Panax ginseng (CWPG) were investigated to provide fundamental information of CWPG with different ages and to increase its industrial application. Proximate analyses of CWPG with different ages were performed. Extraction of CWPG with different ages was carried out using heat-reflux extraction, and their extraction yield, crude saponin content, ginsenoside content, and antioxidant activity were analyzed. Moisture content decreased, but crude fat and crude protein were increased with aging. Extraction yield and crude saponin contents did not show a specific pattern while 5-year-old CWPG revealed the highest extraction yield and crude saponin content. All CWPGs showed typical ginsenoside profiles containing C-K and Rh2 ginsenosides, which are not found in ginseng. The 3-year-old CWPG showed the highest antioxidant activity including total phenolic content, total flavonoid content, and DPPH and ABTS radical scavenging activities. Moreover, 3-year-old CWPG also revealed the highest acidic polysaccharide content. Therefore, these results suggested that 3-year-old CWPG, which is the cheapest, can be used in industrial application due to its high antioxidant activity and acidic polysaccharide content with similar ginsenoside profile compared to 5- and 7-year-old CWPGs.

Monitoring of Chemical Changes in Explosively Puffed Ginsengvand the Optimization of Puffing Conditions

  • Response surface methodology was used to predict the optimum conditions of explosive puffing process for ginseng. A central composite design was used to monitor the effect of moisture content and puffing pressure on dependent variables such as functional compounds (extract yield, crude saponin, acidic polysaccharide, and total phenolic content) and sensory properties. Correlation coefficients $(R^2)$ of models for crude saponin, acidic polysaccharide, and total phenolic content were 0.9176 (p<0.05), 0.9494 (p<0.05), and 0.9878 (p<0.001), respectively. Functional compounds increased with decreasing moisture content and increasing puffing pressure. Overall palatability was high at 15-20% moisture content and 98-294 kPa of puffing pressure. On the basis of superimposed contour maps for functional compounds and overall palatability of puffed ginseng, the optimum ranges of puffing conditions were 10-17% moisture content and 294-392 kPa puffing pressure.

Extraction of Ginseng Saponin by the Treatment of Microbial Macerating Enzyme (미생물이 생성한 식물조직부양효소를 이용한 인삼 Saponin의 추출)

  • The purpose of this study was to extract saponin efficiently from ginseng leaves and peelings by macerating them with microbial enzyme. To begin with, we selected G-211 strain having the highest macerating activity among several rotting molds of fresh ginseng. Crude macerating enzyme was prepared from this G-2l1 strain by ammonium sulfate precipitation, and was applied to macerating leaves and peelings of ginseng. The optimal pH of the enzyme for maceration was 5.0 in both leaves and peelings of ginsen g. The optimal pH for the extraction of soluble matters and saponins was 4.5 and 5.5 in ginseng leaves and ginseng peelings, respectively. When this enzyme was treated together with crude cellulase from Trichoderma viride (To4), the extract content of saponin was 3.45% for ginseng leaves and 3.90% for ginseng peelings. Their yields were 39.8 % and 39.3 % of total saponin amounts in ginseng leaves and ginseng peelings, respectively. The ginsenoside patterns of saponins extracted with the treatment of enzymes were also studied by HPLC technics.

Antioxidant and Hepatoprotective Effects of Hydroponic-cultured Ginseng Folium by fermentation (발효에 의한 수경재배 인삼 잎의 항산화 및 간 보호 효과)

  • Objectives : Positive effects of Ginseng has great research attentions such as anticancer, anti-diabetic, antiaging, liver, immune function, CNS, etc. In this study, we investigated Hydroponic-cultured Ginseng Folium fermented byBacillus subtilisto establish fermentation conditions for enhancing functionality.Methods : Ginseng Folium were cultivated hydroponic-cultured and were extracted with methanol. We inoculateBacillus subtilisfor fermentation by adding to 0%, 3% and 5% sugar respectively and checked antioxidant activities, total phenolic content and total saponin content in 2 days intervals during 11 days. The antioxidant activities were studied by the 1,1-diphenyl-2-picryl hydrazyl(DPPH) radical, 2, 2'-Azino-bis(3-ethylbenzothiazoline-6 sulfonic acid) diammonium salt(ABTS) radical scavenging assay and Reducing power assay. We analyzed the Total phenol content, crude saponin content and ginsenoside content. Moreever, Hepatoprotective effects by Glutamic oxaloacetic transaminase(GOT) and Glutamic pyruvic transaminase(GPT) in Sprague-Dawley rat.Results : The results of DPPH and ABTS were 66.89% and 96.72%, respectively. The reducing power was resulted in optical density of 0.7312 with 3% sugar after 9 days of fermentation. and the concentration at 200 ㎍/㎖. Total phenol content was 36.92㎎/g with 3% sugar after 9 days of fermentation, in which crude saponin content wasn't changed, and ginsenoside content such as Rg3, Re and Rb was increased. Activities of GOT and GPT concentration were decreased in rat.Conclusions : This study suggests that hydroponic-cultured Ginseng Folium fermented byBacillus subtilisin 9 days showed significant efficacy of hepato-protection as well as antioxidant compared to the others. In addition, it shows not only improved value but also utilized hydroponic-cultured Ginseng Folium by fermentation.

Effect of Processing Methods on the Saponin Contents of Panax ginseng Leaf-Tea (고려인삼엽차의 제조방법에 따른 사포닌 성분의 함량 및 조성)

  • Panax ginseng leaf tea was developed for the functional benefit of health, preference and convenience. The leaves of 4-year-old ginseng were selected in July and August. The ginseng leaf was treated by three methods : heat processed tea(HPT), aged tea(AGT) and hot-air dried tea(DRT). The contents and compositions of their crude saponin of ginseng leaves were measured. 1. The content of crude saponin of HPT was the higher than other treatments. The content of HPT was 18.72∼18.82%, ACT 18.24∼18.29% and DRT 17.02∼17.17%. 2. The harvest time and treatment methods were not affect the composition of ginsenoside in ginseng leaf tea. The ginsenoside-Re was shown the highest value as 1.97∼2.15. And ginsenoside-Rd was 1.48∼1.79, -Rg$_1$ 1.33∼1.58 and -Rb, -Rb$_2$, -Rc in the order. 3. The content of protopanaxadiol(PD) and protopanaxatriol(PT) was shown that DRT was 1.11∼1.13, HPT 1.09~l.12 and AGT 0.92∼1.02. The content of PD and PT were shown similar result at any harvest time. 4. The contents of crude saponin extracted by hot-water at 5 min was the higher ratios in HPT and harvested in July than other treatments. The content of crude saponin of ginseng leaf harvested in July was 15.88% and HPT was 16.88%. The order of contents of ginsenoside were -Re, -Rd, -Rg$_1$, -Rb$_1$, -Rb$_2$, and - Rc. The extraction ratio of crude saponin extracted by the circulated extraction method in 8 hours and 5 min extraction were 81.74∼84.38%. And HPT of ginseng leaf harvested in July was the highest value 84.3% but the extraction ratio of ginsenoside was 78.00~88.13%. But the extraction ratio of ginsenoside was similar trend in all treatments.

  • The purpose of this study was to investigate the anti-obesity effects of crude saponin (CS) in Korean red ginseng (KRG) on rats fed a high fat (HF) diet. Methods: Male Sprague-Dawley (SD) rats were divided into a normal diet group and a high-fat (HF) diet group. The HF diet group became obese from being fed a HF diet over five weeks. The normal diet group were fed a normal diet. Each CS group of the normal diet group and HF diet group was administered CS (200 mg/kg, i.p.) for three weeks, the control group of both types was administered normal saline (1ml/kg, i.p.) instead of CS. Each group had 5 rats. Results: After administration of CS, the body weight, food consumption, adipose tissues, and expression of appetite peptides such as leptin and neuropeptide-Y (NPY) were investigated in the HF diet group as well as the normal diet group. Administration of CS reduced body weight, food intake, and fat content in the HF and normal diet groups. After CS administration, NPY expression and leptin were lower in the HF diet group. Conclusions: Our results suggest that CS may be useful in the treatment of obesity, especially of type qixu (氣虛).

New Methods for Separation of Crude Ginseng Saponins (인삼 조사포닌의 새로운 분리 방법)

  • In order to increase ginsenoside content, to reduce chemical change, to shorten extracting procedure, new methods of extraction and fractionation of crude ginseng saponin were established and compared for their chemical composition. Those are hot MeOH extraction/n-BuOH fractionation (BuOH method) and hot MeOH extraction/Diaion HP-20 adsorption/MeOH elution (HP-20 method), which are already known methods, and additional three new methods: hot MeOH extraction/cation AG 50W $adsorption/H_2O$ elution/n-BuOH extraction (AG 50W method), cool MeOH extraction/Diaion HP-20 adsorption/MeOH elution (cool extraction method) and direct extraction with EtOAc/n-BuOH (direct extraction method). AG 50W method provided a crude saponin showing the highest content of ginsenosides of 61.5% and the lowest contents of protein and free amino acids of 0.93% and 0.19%, respectively. The protein content was the highest as 14.18% in the crude saponin by HP-20 method, while free sugar content was the highest as 13.5% by BuOH method, indicating that these are factors that lower the rate of ginsenoside in crude saponins by those methods. On the other hand, it was revealed that AG 50W method produced large amount of prosapogenins during the pass through the cation exchange resin (AG 50W) column being strongly acidic. Crude saponin from direct extraction method showed relatively higher composition of ginsenoside $Rg_1$ and Re. The results suggest that contents and composition of ginsenosides and other chemical components in crude ginseng saponin greatly depend on the condition of the extraction and fractionation.
