오미자의 부위별 일반성분과 무기질함량에 관한 연구 (A Study on the General Components and Minerals in Parts of Omija (Schizandra Chinensis Baillon))

  • This study was carried out to comparison of the contents of general and mineral components between the original sample and water extract in each part of omija. The content of Carbohydrate was highest in endocarps, and that the crude protein and crude lipid in seeds. The contents of K and Mn in the fruits, endocarps, and seeds were all higher than those of the other cations. The content of K and Zn in endocarps were three to four times as much as those of seeds. and the content of Na, Ca, and Cu in endocarps were 1.5 to 1.75 times of seeds. Mineral contents by water extract in each part were ordered as K, Mg, Ca, and Fe. As the percentage of each ion in water extract on the basis of original sample, Fe was the highest ratio of behavior, and Mn was lowest.

Acacia melanoxylon 잎 메탄올 추출액의 생물활성 (The Bioefficacy of Methanol Crude Leaf Extract of Acacia melanoxylon)

  • The methanolic crude leaf extract of Acacia melanoxylon shows inhibitory effects on seed germination, shoot growth, root growth and biomass of black gram (Phaseolus mungo L. Co 1) and weeds (Cassia occidentalis L. and Cyperus rotundus L.). The post emergence treatment was given on the 8th day after sowing and 4-6 days after seedling emergence. After postemergence treatment with 20% extract concentration, wilting symptoms appeared in P. mungo and C. occidentalis while necrosis occurred in C. rotundus. The seedling growth was inhibitory at all concentrations (5%, 10%, 20%) as compared to control plants. This inhibitory effect may be due to the presence of allelochemicals like phenolics in the A. melanoxylon leaves. Different types of phenolic acids were identified.

  • We previously reported the antimutagenicity, anti-lipid peroxidation and antiinflammtory properties of the three Chinese herb medicine, Kalopanax pictus, Pueraria thunbergiana and Rhus verniciflua, which have been used as therapeutics for diabetes mellitus and inflammation in Korea. Since these drugs have been used for the prescriptions consisted of several crude drugs, we investigated whether the extract of the three drugs could increase the individual drugs or not. The three extracts were independently prepared from K. pictus, P. thunbergiana, and R. verniciflua, which extracts were named the K-1, P-1 and R-1 in this order and the extract (KPR-1) was also prepared from the combined plant materials with each same weight of the three drugs. Enhancement of Polyphenol Content and Antioxidant Activity of Brown Alga Eisenia bicyclis Extract by Microbial Fermentation

  • The objective of this study was to select an effective microbial strain to improve the functional qualities of Eisenia bicyclis water extract by fermentation. For this purpose, several microorganisms isolated from traditional Korean fermented foods were inoculated and cultivated in E. bicyclis water extract. Ultimately, yeast strain YM-1 was selected for further study based on its total phenolic compound (TP) content and antioxidant activity, which were enhanced by microbial fermentation. The extract fermented by YM-1 exhibited a superior TP content and 2,2'-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity compared to extracts fermented by other microbes. The highest TP content and DPPH radical scavenging activity were observed after one day of YM-1 fermentation. Yeast strain YM-1 was identified as Candida utilis based on an analysis of its physiological characteristics. During fermentation of the extract by C. utilis YM-1, no significant difference was observed in the proximate composition, including moisture, crude lipid, crude protein, and crude ash. Fermentation by C. utilis YM-1 resulted in enhanced biological activity, including increases in the TP content and antioxidant activity. Thus, fermentation by C. utilis YM-1 is an attractive strategy for developing value-added food ingredients.

In vitro system에서 청국장 메탄올 추출물의 신경세포 보호효과 (Neuronal Cell Protective Effects of Methanol Extract from Cheonggukjang Using in vitro System)

  • 청국장 메탄올 추출물의 $H_2O_2$로 유도된 신경세포 독성에 대한 보호 효과를 조사하였다. 청국장 분말의 일반성분을 분석한 결과 조단백질 40.95%, 조지방 22.49%, 가용성 무질소물 15.99%, 수분 7.91%, 조회분 6.74% 및 조섬유 5.92% 순으로 나타났으며, 총 페놀성 화합물 함량은 28.43 mg/g이었다. $H_2O_2$로 유발된 산화적 손상에 의한 ROS 축적량을 조사한 결과 $H_2O_2$ 단독 처리구보다 청국장 메탄올 추출물 처리구에서 낮은 ROS 축적량을 보였다. $H_2O_2$로 유도된 PC12 신경세포에 대한 보호효과를 MTT 방법을 이용하여 측정한 결과 모든 시료에서 40%정도의 신경세포 보호효과를 보였고, LDH release 실험 결과 5 mg/mL 농도에서 15%정도의 LDH 방출량 저해 효과를 나타내었다. 청국장 메탄올추출물의 아세틸콜린에스터레이스 저해활성은 농도 의존적인 경향이었다. 본 연구 결과를 종합해 볼 때, 청국장 메탄올 추출물은 산화적 스트레스로부터 신경세포 보호효과 및 아세틸콜린에 스터레이스 저해활성을 나타내어 알츠하이머성 신경질환의 예방을 위한 식품으로서의 활용 가능성이 높다고 판단된다.

수확시기에 따른 구기자 품종의 성분변화 (Changes in Composition of Gugija (Lycii Fructus) Species According to Harvest Time)

  • 시험장에서 육성한 품종인 명안, 불로, 청대, 장명, 청운과 육성중인 청양6호, 청양7호 및 청양재래의 가용성 무질소물, 조단백질, 조지질, 회분, 총 폴리페놀, 엑스, 베타인의 함량을 8, 9, 10 및 11월의 수확시기별로 조사한 결과, 가용성 무질소물 및 엑스는 육성중인 청양6호가 가장 높은 함량을 보였고, 조단백질, 총폴리페놀 및 베타인의 함량에서는 육성중인 청양 7호가 가장 높은 함량을 보였으며, 조지질은 청대에서, 조회분은 청운에서 가장 높았다. 건과비율은 육성중인 청양6호에서 가장 높았고, 건과비율이 높을수록 가용성 무질소물의 함량은 증가 (r = 0.70)하는 반면 조단백질 (r = -0.83) 및 조회분 (r = -0.71)등은 감소하였다. 일반성분 중에는 서로 일정한 상관을 보였는데, 가용성 무질소물의 함량이 증가하면 엑스의 함량 (r = 0.81)은 높아지는 반면, 단백질 (r = -0.88), 지질 (r = -0.82) 및 회분 함량 (r = -0.81)은 감소하고, 총폴리페놀 함량이 증가하면 베타인 함량이 증가 (r = 0.90)하는 경향이었다. 품종특성과 성분을 분석한 결과 육성중인 청양 6호는 가용성 무질소물 및 엑스분이 높게 관찰되었을 뿐만 아니라, 건과비율이 높아 타 품종에 비해 쉽게 건조될 수 있어 건조비용을 줄일 수 있는 것으로 예상되었고, 청양 7호는 조단백질, 총폴리페놀 및 베타인 함량이 높아 기능성 품종으로서의 가치를 보여주었다.

Antioxidant and analgesic activity of Clerodendrum visconsum leaf

  • The crude ethanolic extract of the leaves of Clerodendrum viscosum (Family: Verbenaceae) was evaluated for its antioxidant and analgesic activities to investigate the scientific basis of the traditional uses. The antioxidant property of the extract was assessed by 1, 1-diphenyl -2- picryl hydrazyl (DPPH) free radical scavenging assay. The extract showed prominent antioxidant activity ($IC_{50}about{\sim}16{\mu}g/ml$) which was comparable to standard drug ascorbic acid ($IC_{50}about{\sim}15{\mu}g/ml$).The extract produced significant (P<0.001) writhing inhibition in acetic acid induced writhing in mice at the dose of 125 mg, 250 mg and 500 mg/kg body weight respectively, which were comparable to the standard drug diclofenac sodium. The results tend to suggest that the crude leaves extract at the above doses have antioxidant and analgesic activities and indicate that it might possess biologically active constituents having free radical scavenging and analgesic activities respectably.

Analgesic, cytotoxic and antioxidant activities of Trewia polycarpa bark

  • The crude ethanol extract of the stem bark of Trewia polycarpa (Family: Euphorbiaceae) was subjected to acetic acid induced writhing inhibition, Brine Shrimp lethality bioassay and 1, 1-diphenyl-2-picryl hydrazyl free radical scavenging assay for screening of analgesic, cytotoxic and antioxidant activity respectively. The extract produced significant (P < 0.001) writhing inhibition in acetic acid induced writhing in mice at the dose of 125, 250 and 500 mg/kg body weight respectively, which were comparable to the standard drug diclofenac sodium. The extract showed significant lethality to Brine Shrimp and the $LC_{50}$ value was $8\;{\mu}g/ml$. The extract showed prominent free radical scavenging activity ($IC_{50}$ about ${\sim}10\;{\mu}g/ml$) compare to standard drug ascorbic acid ($IC_{50}about\;{\sim}15\;{\mu}g/ml$). The results tend to suggest that the crude ethanol extract of the bark might possess analgesic, cytotoxic and antioxidant activities or active constituent(s) responsible for the activities.

Variation of Nutritional and Antioxidant Characteristics of Extract of Lycium barbarum produced by using Different Extraction Processes

  • Lycium barbarum extract has a high potential to be developed as a health functional food due to the various health-promoting effects of Lycium barbarum. This study analyzed changes in nutritional and functional components depending on the extraction solvent (purified water and a mixture of purified water and alcohol) and the condition of the sample. The nutritional components (carbohydrates, protein, fat, ash), organic acids, amino acids, total phenolic compounds, and total flavonoids of the extract produced during the extraction process were analyzed. The nutritional composition and functional substances of the extracts showed some differences depending on the type of solvent and the condition of the sample. The amounts of crude protein (7.61%), crude fat (1.63%), carbohydrate (90.22%), and ash (0.54%) of dried Lycium barbarum extract using purified water as a solvent were similar to those of the powder sample extract. The highest content of citric acid was 4.31 mg/mL, similar to the case of acetic acid, when the powder sample used a mixture of purified water and alcohol as a solvent. The highest amino acid content was 357.39 mg/mL when the powder sample was mixed with purified water and alcohol as a solvent. The total amount of phenolic compounds was 686.16 g/L when the powder sample was extracted with a mixture of purified water and alcohol as a solvent. The highest total flavonoid content was 111.32 g/L when the powder sample was extracted with a mixture of purified water and alcohol as a solvent.

황색포도상구균을 포함한 식중독 미생물들에 대한 홍삼의 항균효과 (Antimicrobial Effect of Red Ginseng Against Major Food Poisoning Microorganisms Including Staphylococcus aureus)

  • S. aureus, E. coli, C. albicans, A. niger 등 4종의 식중독균에 대해 agar diffusion법을 이용하여 홍삼(Panax ginseng C.A.Meyer)으로 부터 조제한 홍삼농축액, 조사포닌, 비수용성 분획에 대한 항균활성을 조사하였다. 그 결과, 홍삼농축액 및 비수용성분획은 E. coli, C. albicans, A. niger에 대해서는 항균활성을 나타내지 않았고 조사포닌도 고농도인 30%를 제외한 모든 농도에서 항균활성을 나타내지 않았다. S. aureus는 Gram positive 세균으로서 화농성 식중독의 원인균이면서 동시에 아토피성 피부염의 원인균으로 알려져 있는데, 홍삼농축액은 30% 농도에서 이 균에 대해 항균활성을 나타내었고 조사포닌도 7.5%에서 항균활성을 나타내었다. 홍삼으로부터 조제한 비수용성 분획도 10~200 mg/mL 농도로 실험한 결과 모든 분획에서 항균효과를 나타내었다. 조사포닌 및 홍삼농축액의 미생물 생육저해양상을 조사하기 위해 미리 S. aureus를 접종한 0.85% 생리식염수에 농축액 및 조사포닌을 농도별로 첨가하고 35℃, 12시간 배양한 후 생균수를 측정한 결과, 홍삼농축액은 10% 이상의 농도에서, 조사포닌은 2% 이상의 농도에서 각각 균의 생육을 억제하였다. 그러나 이러한 농도에서도 생균수는 완전히 사멸되지 않아서 홍삼농축액 및 조사포닌의 S. aureus에 대한 생육억제작용은 살균작용이 아닌 정균 작용으로 추정되었다. 사포닌의 항균활성 유무를 확인하기 위해 순수 분리된 ginsenoside 6종(PT saponin, PD saponin, ginsenoside-Rb2,-Rc,-Rd,-Rf,-Rg2)의 항균활성을 50~200 ㎍/mL의 농도에서 조사한 결과 모두 항균효과가 관찰되지 않아서 ginsenoside는 S. aureus에 대해서는 항균효과가 없는 것으로 사료된다. 한편 사포닌을 제외한 비사포닌 성분인 비수용성분획에 대해 상기의 4종 병원성미생물을 대상으로 항균활성을 조사한 결과 E. coli, C. albicans, A. niger에 대해서는 항균효과가 관찰되지 않았고 S. aureus에 대해서만 선택적인 항균활성을 나타내었다. 항균활성 발현 비수용성분획 중 15% methanol분획(MF-1)이 가장 높은 항균활성을 나타내어 이에 대한 최소생육저해농도를 조사한 결과 0.625 mg/mL 이었다. MF-1 분획을 질량분석기(HPLC-MS)로 조사한 결과 주요한 활성성분은 분자량 179.55 및 187.55를 가지는 물질로 추정되었다.
