잉어(Cyprinus carpio)와 붕어(Carassius auratus)간 잡종의 세포유전학적 분석 (Cytogenetic Analysis of Induced Hybrid between Common Carp (Cyprinus carpio) and Crucian Carp (Carassius auratus))

  • 잉어(Cyprinus carpio) 암컷과 붕어(Carassius auratus) 수컷간 유도된 잡종에 대한 세포 유전학적 연구를 수행하였다. 유도된 잡종의 세포 및 핵의 크기는 양친의 중간값을 나타내었고, 염색체의 경우 잉어, 붕어 및 유도된 잡종 모두 2n=100 으로 나타났다. Flow cytometry를 이용, 잡종의 genome size를 분석한 결과, 유도된 잡종은 3.7pg/cell를 갖음으로서 잉어(3.6pg/cell)와 붕어(3.8pg/cell)의 중간치를 나타내었다.

  • 한국의 예산과 당진에서 각각 채취된 붕어 (Carassius auratus)와 떡붕어 (Carassius cuvieri)로부터 genomic DNA를 분리 추출하여 반복해서 PCR로 증폭시켰다. 선택된 7개의 RAPD primer를 이용하여 primer 당 total loci, shared loci by each species, polymorphic 및 specific loci를 얻어냈다. 2종의 붕어로부터 primer와 2지역간에 banding patterns의 복잡성이 두드러지게 나타났다. DNA fragment의 분자적 크기는 150bp에서부터 1,600bp까지 커다란 차이를 나타내었다. 본 연구에서 CCY 붕어 종에서는 458개의 loci가 나타났고, CCD 떡붕어 종에서는 358개의 loci가 확인되었다. 또한 CCY 붕어 종에서는 84개의 polymorphic loci (18.3%)가 확인되었고, CCD떡붕어 종에서는 48개의 polymorphic loci (13.4%)가 확인되었다. CCY 붕어 종에서는 154개의 shared loci가 나타났으며, 이는 primer당 평균적으로 22개의 loci로 확인되었다. 또한 CCD떡붕어 종에서는 187개의 shared loci가 확인되었고, 평균해서 primer 당 26.7개의 loci가 나타났다. CCY붕어 종과 CCD 떡붕어 종의 polymorphic loci는 각각 84개와 48개로 확인되었다. 모든 붕어와 떡붕어 시료의 평균적인 BS value를 기초로 해서 CCY 붕어 종의 similarity matrix를 조사해 본 결과 0.434로부터 0.868까지 나타났고, CCD 떡붕어 종의 값은 0.449로부터 0.924까지 확인되었다. CCY 붕어 종내의 평균적인 BS value는 0.641±0.013이고, CCD 떡붕어 종내의 BS value의 평균값은 0.684±0.013을 나타내었다. 결과적으로 CCD 떡붕어 종내의 개체의 BS value 평균값이 CCY 붕어 종내의 평균값보다 높게 나타났다. 2 붕어와 떡붕어간의 평균적인 BS value은 0.484±0.007 (0.307~0.682)를 나타내었다. 7개의 primer를 사용하여 얻어진 dendrogram은 cluster 1 (AURATUS no. 01~AURATUS no. 11), cluster 2 (CUVIERI no. 12~CUVIERI no. 21) 및cluster 3 (CUVIERI no. 22)와 같이 3개의 유전적 클러스터로 나뉘어졌다. CCY 붕어 종내의 8번째 개체 (AURATUS no. 08)와 9번째 개체 (AURATUS no. 09) 사이가 가장 가까운 유전적 관계 (0.064)를 나타내었다. 또한 CCY붕어 종의 11번째(AURATUS no. 11)와 CCD떡붕어 종의 17번째 (CUVIERI no. 17) 사이가 가장 먼 유전적 거리 (0.477)를 나타내었다. 결과적으로 볼 때 한국 및 대서양산 lobster (0.612), 갈치 (0.708), 동자개(0.714)에 비해서 상대적으로 낮은 유전적 거리를 나타내었다.

중국에서 수입된 잉어 및 붕어의 잉어부종바이러스병 및 허피스바이러스성 조혈기괴사증 모니터링 (Monitoring of carp edemavirus disease and herpesviral hematopoietic necrosis in Cyprinus carpio and Carassius auratus imported from China)

  • In this study, carp edemavirus disease and herpesviral hematopoietic necrosis were examined in carp (Cyprinus carpio) and crucian carp (Carassius auratus) imported from China. From August 2016 to May 2017, 100 carp and 300 crucian carp were examined. There were no clinical symptoms in all the tested fish, and no internal or external parasitic infections were identified except for the detection of Argulus sp. in one leather carp. Carp edema virus disease was examined by PCR in C. carpio, but all were negative. In C. auratus, herpesviral hematopoietic necrosis was tested by PCR and 4 out of 300 were positive. The results of this study are expected to be useful as basic data for conducting risk analysis on the relevant organisms and counterpart countries or for designating new pathogens for quarantine.

포르말린 침지에 의한 붕어(Carassius auratus)의 혈액학 및 혈액화학치에 대한 영향 (Effects of formalin on haematology and blood chemistry in crucian carp (Carassius auratus))

  • To determine the effects of exposure to formalin on the secondary stress indices, changes in haematology and blood chemistry were monitored in healthy crucian carps (Carassius auratus). Fishes were separately exposed in a concentration range of 125 to 500 ppm formalin for 60 min. After exposure, red blood cell (RBC) count and packed cell volume (PCV) were elevated in the 500 ppm formalin exposed group. However, mean corpuscular volume (MCV) and mean corpuscular haemoglobin (MCH) were decreased significantly in the 500 ppm formalin exposed group. Total protein, albumin, aspartate aminotransferase (AST), alanine aminotransferase (ALT), creatinine, total bilirubin, inorganic phosphorus (IP) and magnesium were significantly increased at a concentration of 500 ppm. Alkaline phosphatase (ALP) and glucose were increased at a concentration of 500 ppm, but this was not significant. Lactate dehydrogenase (LDH) and calcium were significantly decreased at concentrations of 250 and 500 ppm. AST, ALT, glucose and magnesium were significantly increased in the 250 ppm formalin exposed group. These results suggests that formalin exposure might cause some damage in the liver and kidney of crucian carp.

  • Histopathological changes and cadmium accumulations in the gills were investigated in crucian carp (Carassius auratus) exposed to 0, 0.01, 0.1, 0.5 mg/1 cadmium (Cd) concentrations for 25 days. After 25 days of exposure. cadmium accumulations increased in each of the different exposure concentrations. Histopathological changes in the gills of curcian carp exposed to cadmium included the acidification of mucous cell, the terminal clubbing of lamellae, the hyperplasia of epitherial cell and the curved of secondary lamellae. The similar histopathological changes were observed once the accumulations of cadmium reached the similar levels found in the present study with the different exposure schemes. Also the acidification of mucous cell in gills appeared when cadmium accumulations reached 5 $\mu\textrm{g}$/g and more. These results suggested that histopathological changes in the gills, an appearance of mucous cell acidification. be used to estimate the relation between toxic effect and cadmium accumulations in the gills.

  • To determine the effects of anesthesia to clove oil on the secondary stress indices, changes in haematological and blood chemistry were monitored in healthy crucian carps (Carassius auratus). 24 fishes were divided into three groups, and blood was collected before anaesthesia (control group), immediately after anaesthesia at a concentration of 30 mg/l clove oil (anesthetized group), and 24 h after anaesthesia (recovered group). The anesthesia to clove oil significantly increased glucose in anaesthetized group, and constantly decreased lactate dehydrogenase in anaesthetized group and recovered group. However, clove oil had not effect on other biochemical indices. These results suggests that the anesthetic use of clove oil at a concentration of 30 mg/l does not cause irreversible damage in carp.

  • 살충제 imidacloprid(1-(6-chloro-3-pyridylmethyl)-N-nitroimidazolidin-2-ylideneamine)의 붕어(Carassius auratus L.) 체내 행적과 장기에 미치는 영향을 구명하기 위하여 imidacloprid를 환경추정농도인 0.064 mg $kg^{-1}$로 처리한 시험수에 붕어를 4일간 경시적으로 노출시켰다. 수중의 imidacloprid는 붕어체내에 흡수되어 노출 2일차에 최대 흡수량을 나타내었으며, 다른 장기에 비해 쓸개에서 농도가 가장 높았다. 붕어 사육수에서 미확인 주대사산물과 imidacloprid urea가 대사산물로 검출되었다. 또한 붕어체내에 흡수된 imidacloprid의 주요 배설경로는 담도이었으며, 장기에 대한 조직학적 이상증상은 발견되지 않았다.

  • This study investigated cytogenetic and hematological and histological characteristics between diploid and spontaneous triploid on Cyprinidae (Crucian carp, Carassius auratus; crucian carp, C. cuvieri and common carp, Cyprinus carpio) in four major rivers of Korea. In our results, DNA contents of triploid Cyprinidae were 50% more than those of diploid Cyprinidae. Also, erythrocyte size of triploid Cyprinidae was 50% larger than those of diploid Cyprinidae. In all sampling rivers, sex ratios of C. auratus were biased to female, and especially, triploid groups of C. auratus were all female groups (p<0.05). In principle, sex ratios of C. cuvieri and common carp were equivalent between male and female.

붕어 수정란을 이용한 다이옥신의 초기발생단계 독성평가 (Early Life Stage Toxicity for 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) in Crucian Carp (Carassius auratus))

  • Crucian carp (Carassius auratus) has been used as the sentinel species for POPs (Persistent Organic Pollutants) monitoring in aquatic environment. However, there is little information for dioxin toxicity and especially, early life stage toxicity in crucian carp have been never carried out. In this study, we investigated several toxic effects for 2,3,7,8-tetrachlorodibenzo-p-dioxin (2,3,7,8-TCDD) in fertilized egg obtained by natural fertilization from crucian carp. The embryos at 3 h post-fertilization (hpf) were treated with 0.039, 0.156, 0.625, and 2.5 (${\mu}g/L$) TCDD by waterborne exposure for 60 minutes and changed with fresh water 2 times per day. Fertilized eggs started hatching at 51 hpf and TCDD exposed embryo showed decrease of hatching rate in a dose-dependent manner at 75 hpf. Pericardial edema was continuously observed in larvae exposed to TCDD from hatching start time (51 hpf), followed by the onset of mortality. In addition, AhR-related gene, CYP1A was clearly increased by TCDD in a dose dependent manner. These results indicated that fertilized eggs obtained from crucian carp have the TCDD related gene regulation and a distinct TCDD developmental toxicity syndrome by TCDD exposure. Therefore, we suggested that early life stage test in crucian carp could be used as test methods on dioxins toxicity.
