Antigenic protein fractions of Metagonimus yokogawci reacting with patient sera (요꼬가와흡충증 항체진단의 검토 및 특이 항원 단백질 분석)

  • Antibody test is sometimes necessary for the diagnosis of acute human metagonimiasis because eggs may not be detected in stool. The antibody test (ELISA) was evaluated for its significance by reacting human sera from clinically diagnosed metagonimiasis, fascioliasis, clonorchiasis and paragonimlasis with 4 crude extracts of Metosonimn vokognwai (metacercariael , adults of Fosciola hepatica Cronorchis sinenis and Paragonimus westermoni. By ELISA, 10 of 11 metagonimlasis sera showed the highest absorbance (abs.) to the homologous antigen. Cross reactions to M. yokogawai antigen occurred most frequently in clonorchiasis sera. The antigenic protein fractions in M. vokogawai metacercarial extract were observed by SDS-PAGElimmunoblot using patients and control sera together with experimental cat sera. Out of 14 protein bands In the extract, 11 bands were reacting. Cross reacting bands to other trematodiasis sera were frequently observed. Of the reacting bands, 66 and 22 kDa proteuls were recognized as specific for metagonimiasis.

Spectroscopic Analysis on the Michael Addition Reaction between Secondary Amino Group Containing Silica Nanoparticles with (Meth)acrylate Monomers (2차 아미노기를 갖는 실리카 나노입자와 (메타)아크릴레이트 단량체와의 마이클 부가반응에 대한 분광학적 분석)

  • In this study, we modified silica nanoparticles with bis[3-(trimethoxysilyl)propyl]amine (BTMA) silane coupling agent to introduce secondary amino groups on the silica surface. After modification of silica, we investigated effects of different types of (meth)acrylate group containing monomers on the Michael addition reaction to introduce reactive (meth)acrylate groups on the BTMA modified silica surface. We used two kinds of (meth)acrylate monomers, trimethylolpropane ethoxylate triacrylate (TMPET) which has three identical acrylate groups, and 3-(acryloyloxy)-2-hydroxypropyl methacrylate (AHM) which has one acrylate and one methacrylate group. We used fourier transform infrared spectroscopy (FTIR), elemental analysis (EA) and solid state cross-polarization magic angle spinning (CP/MAS) nuclear magnetic resonance spectroscopy (NMR) to understand reactions between NH groups on the silica surface with (meth)acrylate groups of TMPET and AHM monomers. We found almost complete Michael addition reaction between all three acrylate groups of TMPET with NH groups on the BTMA modified silica. But, for the AHM treatment of BTMA modified silica, we found Michael addition reaction occurred only between acrylate groups of AHM and NH groups of silica surface, not between methacrylate groups of AHM and NH groups of BTMA modified silica surface.

A Clonorchis sinensis-specific antigen that detects active human clonorchiasis (간흡충 현증감염 특이항원)

  • A Czonorchis sinensis-specific antigen in excretory-secretory product of C. sinensis (CsE) was assessed in human clonorchiasis by immunoblot. Thirty and 7 kDa antigens of CsE2, one of four different batches of CsEs reacted strongly with infection sera from clonorchiasis patients; however, the antigens reacted weakly with 6-month post- treatment sera from praziquantel-cured cases, but were still highly detected by the sera from praziquantel∼failed patients, indicating that the 30 and 7 kDa antigens can detect antibodies during an active infection. The 30 kDa antigen showed some cross reactions with sera from patients with Pcragonimus westemani and Metcfonimw vokogcujci, while the 7 kDa antigen did not, suggesting that the 7 kDa antigen has high specificity. The 30 kDa antigen reacted with some past clonorchiasis sera, whereas the 7 kDa antigen did not, supporting that antibodies to the 7 kDa antigen are not present in sera from past clonorchiasis patients. In an endemic area, 92% (23/25) of active clonorchiasis patients and 91% (10/11) of mixed infection patients with C. sinensis and M. Wokosawai had IgG antibodies to the 7 kDa antigen, while 40% (6/15) of past clonorchiasis individuals and 43% (3/7) of metagonimiasis patients cross-reacted to the antigen. These data suggest that the 7 kDa antigen in an excretory-secretory antigen may serve as a marker of an active clonorchiasis with reliable specificities in past clonorchiasis, paragonimiasis and metagonimiasis.

A Study on Exothermic Properties of TiO2/Epoxy Nanocomposites (TiO2/Epoxy 나노복합재의 발열 특성에 관한 연구)

  • Recently, various nanoparticles have been used for filler in polymer matrices. The particles of nano size are whether high or not cross-link density in polymer affects the thermal and mechanical properties of one. The properties change as a result of chemical reactions between the nanoparticles and the surface of the polymer. There are two models for nanocomposites: "repulsive interaction" and "attractive interaction" between the nanoparticles and matrix. In this study, the variation in the curing mechanism was examined when nano-size $TiO_2$ was dispersed into an epoxy (Bisphenol A, YD-128) with different curing agents. The results of this study showed that the exothermic temperature and Tg in the case of the nanoparticles used (Jeffamine) (D-180) at room temperature were reduced by an increase in the $TiO_2$ contents because of the "repulsive interaction" between the nanoparticles and the matrix. The tensile strengths were increased by increasing amounts of $TiO_2$ until 3 wt% because of a dispersion strengthening effect caused by the nanoparticles, because of the repulsive interaction. However, such tensile properties decreased at 5 wt% of $TiO_2$, because the $TiO_2$ was agglomerated in the epoxy. In contrast, in the case of the nanoparticles that used NMA and BDMA, the exothermic temperature and Tg tended to rise with increasing amounts of $TiO_2$ as a result of the "attractive interaction." This was because the same amounts of $TiO_2$ were well dispersed in the epoxy. The tensile strength decreased with an increase in the $TiO_2$ contents. In the general attractive interaction model, however, the cross-link density was higher, and tensile strength tended to increase. Therefore, for the nanoparticles that used NMA, it was difficult to conclude that the result was caused by the "attractive model."

A Viscoelastic Study of Glass Transition and Degradation Processes of Phenolic Resin/Carbon Fiber Composites (페놀수지/탄소섬유 열경화성 복합재료의 유리전이와 고온 분해과정에서 관찰되는 점탄성 특성 연구)

  • Viscoelastic characteristics of cured phenolic resin/carbon fiber composite materials were investigated through glass transition and degradation reaction processes in the high temperature region up to $400^{\circ}C$. A typical glass transition of the cross-linked thermoset polymer was followed by irreversible degradation reactions, which were exhibited by the increasing storage modulus and loss modulus peak. A degradation master curve was constructed by using the vertical and horizontal shift factors, both of which complied well with the Arrhenius equation in light of the kinetic expression of degradation rate constants. Using an analogy to the Havriliak-Negami equation in dielectric relaxation phenomena, a viscoelastic modeling methodology was developed to characterize the frequency- and temperature-dependent complex moduli of the degrading thermoset polymer composite systems. The temperature-dependent relaxation time of the degrading composites was determined in a continuous fashion and showed a minimum relaxation time between the glass transition and degradation reaction regions. The capability of the developed modeling methodology was demonstrated by describing the complex behavior of the viscoelastic complex moduli of reacting phenolic resin composite systems.

Outcome of inflammatory response after normothermia during cardiopulmonary bypass surgery in infants with isolated ventricular septal defect

  • Purpose: A recent study analyzing several cytokines reported that long cardiopulmonary bypass (CPB) time and long aortic cross clamp (ACC) time were accompanied by enhanced postoperative inflammation, which contrasted with the modest influence of the degree of hypothermia. In this present study, we aimed to examine the effect of CPB temperature on the clinical outcome in infants undergoing repair of isolated ventricular septal defect (VSD). Methods: Of the 212 infants with isolated VSD who underwent open heart surgery (OHS) between January 2001 and December 2010, 43 infants were enrolled. They were classified into 2 groups: group 1, infants undergoing hypothermic CPB ($26^{\circ}C-28^{\circ}C$; n=19) and group 2, infants undergoing near-normothermic CPB ($34^{\circ}C-36^{\circ}C$; n=24). Results: The age at the time of the OHS, and number of infants aged<3 months showed no significant differences between the groups. The CPB time and ACC time in group 1 were longer than those in group 2 (88 minutes vs. 59 minutes, P =0.002, and 54 minutes vs. 37 minutes, P =0.006 respectively). The duration of postoperative mechanical ventilation was 1.6 days in group 1 and 1.8 days in group 2. None of the infants showed postoperative neurological and developmental abnormalities. Moreover, no postoperative differences in the white blood cell count and C-reactive protein levels were noted between two groups. Conclusion: This study revealed that hypothermic and near-normothermic CPB were associated with similar clinical outcomes and inflammatory reactions in neonates and infants treated for simple congenital heart disease.

Development of Rapid Immune-gold Strip Kit for On-Site Diagnosis of Tomato spotted wilt virus

  • A rapid, user-friendly and simple immune-chromatographic dipstick kit named 'rapid immune-gold strip' (RIGS) kit was developed in a novel single strip format to detect on-site detection of Tomato spotted wilt virus (TSWV). Immunoglobulin G (IgG) from polyclonal antisera raised in rabbits against TSWV was purified through protein-A affinity chromatography and then the purified TSWV-IgG was conjugated to colloidal gold nano-particles which served as a test line on nitrocellulose membrane. Protein A that non-specifically binds to TSWV antibody was used as a control line on the same strip. The diagnosis process with the TSWV-RIGS involves simply grinding the suspect plant sample in a bag that contains the extraction buffer and inserting the strip the bag. Results can be seen in 2-5 minutes. The flow of the complexes of gold particles coated with TSWV-IgG and a crude sap from TSWV-infected pepper, tobacco and tomato plants resulted in intensive color formed on the test lines proportional to the concentrations of TSWV. The RIGS-TSWV kit did not show any cross-reactions against other tomato-infecting viruses unrelated to TSWV. These results indicate that the TSWV-RIGS kit is highly sensitive and is not required for laboratory training and experience prior to testing. The TSWV-RIGS kit is suitable for on-site detection of suspect TSWV-infected plants as well as for laboratory diagnosis.

Effect of Solvent on the Grafting Polymerization of Polyethylene Wax with Maleic anhydride (폴리에틸렌 왁스와 무수 말레인산의 그라프팅 중합 반응에서 용매가 미치는 영향)

  • In this study, we have investigated the influence of the nature of solvent on the grafting reaction of maleic anhydride onto polyethylene wax obtained as a by-product in a high density polyethylene plant. The results show that the grafting ratio in xylene as a solvent was higher than toluene. This is because xylene has excellent monomer solubility, swelling property and miscibility. It has been also observed that grafting degree shows an initial jump in percentage of grafting with increasing amount of solvent, from 0% v/w to 200% v/w giving maximum grafting in 200% v/w and then slightly decreases on further increase in the amount of solvent and becomes almost constant. It can be also seen that gel content was not formed under the use of solvent. It means that solvent prevented cross-linking reaction due to chain transfer reactions to solvent molecules. Studies of melt viscosity at $140^{\circ}C$ showed that viscosity increased after grafting of maleic anhydride onto polyethylene wax.

Kinetics and Mechanism of the Pyridinolysis of Aryl Ethyl Chlorothiophosphates in Acetonitrile

  • The nucleophilic substitution reactions of Y-aryl ethyl chlorothiophosphates with X-pyridines are studied kinetically in acetonitrile at $55.0^{\circ}C$. The Hammett and Bronsted plots for substituent X variations in the nucleophiles exhibit biphasic concave upwards with a break point at X = 3-Me. The substituents of X = 4-CN and 4-Ac show great positive deviations from both the Hammett and Bronsted plots. The Hammett plots for substituent Y variations in the substrates exhibit biphasic concave upwards with a minimum point at Y = H. The obtained values of the cross-interaction constants (${\rho}_{XY}$) are all in spite of the biphasic free energy correlations for both substituent X and Y variations, since the ${\rho}_X$values with both the strongly and weakly basic pyridines are almost constant. A stepwise mechanism with a rate-limiting leaving group departure from the intermediate is proposed where the distance between X and Y does not vary from the intermediate to the second transition state. A frontside attack is proposed with the strongly basic pyridines based on the considerably great magnitudes of ${\rho}_X$ and ${\beta}_X$ values and a backside attack is proposed with the weakly basic pyridines based on the relatively small magnitudes of ${\rho}_X$ and ${\beta}_X$. The positive deviations of the two strong ${\pi}$-acceptor parasubstituents, X = 4-Ac and 4-CN, from both the Hammett and Bronsted plots are rationalized by the great extents of bond formation and breaking.

Kinetics and Mechanism of the Benzylaminolysis of O,O-Diphenyl S-Aryl Phosphorothioates in Dimethyl Sulfoxide

  • Kinetic studies of the reactions of O,O-diphenyl Z-S-aryl phosphorothioates with X-benzylamines have been carried out in dimethyl sulfoxide at 55.0 $^{\circ}C$. The Hammett (log $k_2$ vs ${\sigma}_X$) and Bronsted [log $k_2$ vs $pK_a(X)$] plots for substituent X variations in the nucleophiles are biphasic concave downwards with a maximum point at X = H, and the unusual positive ${\rho}_X$ and negative ${\beta}_X$ values are obtained for the strongly basic benzylamines. The sign of the cross-interaction constant (${\rho}_{XZ}$) is negative for both the strongly and weakly basic nucleophiles. Greater magnitude of ${\rho}_{XZ}$ value is observed with the weakly basic nucleophiles (${\rho}_{XZ}$ = -2.35) compared to with the strongly basic nucleophiles (${\rho}_{XZ}$ = -0.03). The deuterium kinetic isotope effects ($k_H/k_D$) involving deuterated benzylamines [$XC_6H_4CH_2ND_2$] are primary normal ($k_H/k_D$ > 1). The proposed mechanism is a concerted $S_N2$ involving a frontside nucleophilic attack with a hydrogen bonded, four-center-type transition state for both the strongly and weakly basic nucleophiles. The unusual positive ${\rho}_X$ and negative ${\beta}_X$ values with the strongly basic benzylamines are rationalized by through-space interaction between the ${\pi}$-clouds of the electron-rich phenyl ring of benzylamine and the phenyl ring of the leaving group thiophenoxide.
