The major target for drug discovery, G-protein coupled receptor (GPCR) is involved in many physiological activities and related to various diseases and disorders. Among experimental techniques relating to the GPCR drug discovery process, various cell-based screening methods are influenced by cell conditions used in the overall process. Recently, the utilization of frozen cells is suggested in terms of reducing data variation and cost-effectiveness. The aim of this study is to evaluate various conditions in cell freezing such as temperature conditions and storage terms. The stable cell lines for calcium sensing receptor and urotensin receptor were established followed by storing cultured cells at $-80^{\circ}C$ up to 4 weeks. To compare with cell stored at liquid nitrogen, agonist and antagonist responses were recorded based on the luminescence detection by the calcium induced photoprotein activation. Cell signals were reduced as the storage period was increased without the changes in $EC_{50}$ and $IC_{50}$ values $EC_{50}:3.46{\pm}1.36mM$, $IC_{50}:0.49{\pm}0.15{\mu}M$). In case of cells stored in liquid nitrogen, cell responses were decreased comparing to those in live cells, however changes by storage periods and significant variations of $EC_{50}/IC_{50}$ values were not detected. The decrease of cell signals in various frozen cells may be due to the increase of cell damages. From these results, the best way for a long-term cryopreservation is the use of liquid nitrogen condition, and for the purpose of short-term storage within a month, $-80^{\circ}C$ storage condition can be possible to adopt. As a conclusion, the active implementation of frozen cells may contribute to decrease variations of experimental data during the initial cell-based screening process.
Metabolic responses to the protein-free, high-carbohydrate diet and subsequent food restriction on the same diet at the level of 50% and 75% has been studied on the adult albino rats. The energy source was either corn starch or sugar. In experiment I, adult male rats weighing $509{\pm}8g$ were divided into two groups 10 rats each. Rats fed on the stock diet served as a control. Rats of restriction group received a protein free diet until they reduced their weight down to 400g and continue on a protein-calorie restriction diet until they reduced their weight down to 300g. In experiment II, 28 adult male rats and the same numbers of female rats weighing $329{\pm}5g$ and $223{\pm}4g$ respectively were divided into four groups, 7 males and females in each. Rats fed on a stock diet were sacrificed at the point when others started a protein free diet. These were served as the control. The protein free group received a protein free diet ad libitum for 4 weeks. The 50% restriction group and 75% restriction group were fed on a protein free diet coupled with food restriction at levels of 50% and 75% respectively for 3 weeks. In the result of this study: 1. The rate of body weight changes was similar between the males and the females. Feeding protein free diet ad lib. initiated a rapid weight lost of approximately 25% and protein free diet coupled with food restriction showed 37-43% reduction of their initial weight. 2. There was no significant differences in the value of the N concentration in liver, spleen, brain and muscle between controls and experimental groups. 3. Rats fed on protein free diet showed 1/10 value of the control in the nitrogen excretion in urine. However female showed less N excretion than male. 4. Observing blood picture, the effects of protein depletion and calorie restriction were not appeared any remarkable changes. 5. There was no sign of fatty liver which might result from protein depletion and calorie restriction. 6. Following semi-starvation, FAO and HMP-DH total enzyme activity was reduced, but activity per unit weight was relatively stable.

The large extracellular domain of glycoprotein hormone receptors is a unique feature within the G protein-coupled receptors (GPCRs) family. After interaction with the hormone, the receptor becomes coupled to Gs, which, in turn stimulates adenylyl cyclase and the production of cAMP. Potential phosphorylation sites exist in the C-terminal region of GPCRs. The experiments described herein represent attempts to determine the functions of the eel follicle-stimulating hormone receptor (eelFSHR). We constructed a mutant of eelFSHR, in which the C-terminal cytoplasmic tail was truncated at residue 614 (eelFSHR-t614). The eelFSHR-t614 lacked all potential phosphorylation sites present in the C-terminal region of eelFSHR. In order to obtain the eelFSHR ligand, we produced recombinant follicle-stimulating hormone ($rec-eelFSH{\beta}/{\alpha}$) in the CHO-suspension cells. The expression level was 2-3 times higher than that of the transient expression of eelFSH in attached CHO-K1 cells. The molecular weight of the $rec-eelFSH{\beta}/{\alpha}$ protein was identified to be approximately 34 kDa. The cells expressing eelFSHR-t614 showed an increase in agonist-induced cAMP responsiveness. The maximal cAMP responses of cells expressing eelFSHR-t614 were lower than those of cells expressing eelFSHR-wild type (eelFSHR-WT). The $EC_{50}$ following C-terminal deletion in CHO-K1 cells was approximately 60.4% of that of eelFSHR-WT. The maximal response in eelFSHR-t614 cells was also drastically lower than that of eelFSHR-WT. We also found similar results in PathHunter Parental cells expressing ${\beta}$-arrestin. Thus, these data provide evidence that the truncation of the C-terminal cytoplasmic tail phosphorylation sites in the eelFSHR greatly decreased cAMP responsiveness and maximal response in both CHO-K1 cells and Path-Hunter Parental cells expressing ${\beta}$-arrestin.
Even though the increase of greenhouse gases such as $CO_2$ is thought to be the main cause for global warming, its impact on global climate has not been revealed clearly in rather quantitative manners. However, researches using Genral Circulation Model(GCM) has shown that the accumulation of greenhouse gases increases the global mean temperature, which in turn impacts on the global water circulation pattern. A climate predictive capability is limited by lack of understanding of the different process governing the climate and hydrologic systems. The prediction of the complex responses of the fully coupled climate and hydrologic systems can be achieved only through development of models that adequately describe the relevant process at a wide range of spatial and temporal scales. These models must ultimately couple the atmospheres, oceans, and lad and will involve many submodels that properly represent the individual processes at work within the coupled components of systems. So far, there are no climate and related hydrologic models except local rainfall-runoff models in Korea. The purpose of this research is to predict the change of temperature in Korean Peninsula using regional scale model(IRSHAM96 model) and GCM data obtained from the increasing scenarios of $CO_2$ Korean Peninsula increased by $2.5^{\circ}C$ and the duration of Winter in $lxCO_2$ condition would be shorter the $2xCo_2$ condition due to global warming.

This study was performed to investigate the response of the lacewing Chrysopa cognata to both Aphis gossypii-induced plant volatiles and lacewing-derived volatiles. The results of a Y-tube olfactometer bioassay showed that more C. cognata males were attracted to green pepper plants infected with A. gossypii than to uninfected green pepper plants alone or clean air and C. cognata males were attractive to C. cognata females. Gas chromatography-electroantennographic detection (GC-EAD) analysis showed that the antennae of C. cognata females elicited EAD-active responses to the volatiles entrained from A. gossypii-infected green pepper plants. 4-Ethylacetophenone, 3-ethylbenzaldehyde, 3-ethylacetophenone, and 4-ethylbenzaldehyde from A. gossypii-induced green pepper volatiles, and (Z,Z)-4,7-tridecadiene, (Z)-4-tridecene, and (Z)-4-undecene from C. cognata female entrainment were elucidated by further analysis using GC coupled nuclear magnetic resonance spectroscopy. Of the A. gossypii-induced plant volatiles identified in this study, 4-ethylacetophenone and 3-ethylbenzaldehyde significantly increased the attraction of C. cognata males to nepetalactol, but (Z)-4-tridecene and (Z)-4-undecene did not. (Z,Z)-4,7-Tridecadiene significantly reduced the attractiveness of nepetalactol to C. cognata.

There has been an increasing number of studies on photovoltaic energy generation system in an offshore site with the largest energy generation efficiency, as increasing the researches and developments of renewable energies for use of offshore space and resources to replace existing fossil fuels and resolve environmental challenges. For installation and operation of floating photovoltaic systems in an offshore site with harsher environmental conditions, a stiffness of structural members comprising the total system must be reinforced to inland water spaces as dams, reservoirs etc., which have relatively weak condition. However, there are various limitations for the reinforcement of structural stiffness of the system, including producible size, total mass of the system, economic efficiency, etc. Thus, in this study, a floating breakwater is considered for reducing wave loads on the system and minimizing the reinforcement of the structural members. Wave reduction performances of floating breakwaters are evaluated, considering size and distance to the system. The wave loads on the system are evaluated using the higher-order boundary element method (HOBEM), considering the multi-body effect of buoys. Stresses on structural members are assessed by coupled analyses using the finite element method (FEM), considering the wave loads and hydrodynamic characteristics. As the maximum stresses on each of the cases are reviewed and compared, the effect of floating breakwater for floating photovoltaic system is checked, and it is confirmed that the size of breakwater has a significant effect on structural responses of the system.
The maintenance of poultry gut health is complex depending on the intricate balance among diet, the commensal microbiota, and the mucosa, including the gut epithelium and the superimposing mucus layer. Changes in microflora composition and abundance can confer beneficial or detrimental effects on fowl. Antibiotics have devastating impacts on altering the landscape of gut microbiota, which further leads to antibiotic resistance or spread the pathogenic populations. By eliciting the landscape of gut microbiota, strategies should be made to break down the regulatory signals of pathogenic bacteria. The optional strategy of conferring dietary fibers (DFs) can be used to counterbalance the gut microbiota. DFs are the non-starch carbohydrates indigestible by host endogenous enzymes but can be fermented by symbiotic microbiota to produce short-chain fatty acids (SCFAs). This is one of the primary modes through which the gut microbiota interacts and communicate with the host. The majority of SCFAs are produced in the large intestine (particularly in the caecum), where they are taken up by the enterocytes or transported through portal vein circulation into the bloodstream. Recent shreds of evidence have elucidated that SCFAs affect the gut and modulate the tissues and organs either by activating G-protein-coupled receptors or affecting epigenetic modifications in the genome through inducing histone acetylase activities and inhibiting histone deacetylases. Thus, in this way, SCFAs vastly influence poultry health by promoting energy regulation, mucosal integrity, immune homeostasis, and immune maturation. In this review article, we will focus on DFs, which directly interact with gut microbes and lead to the production of SCFAs. Further, we will discuss the current molecular mechanisms of how SCFAs are generated, transported, and modulated the pro-and anti-inflammatory immune responses against pathogens and host physiology and gut health.

Structural health monitoring for ships and offshore structures is important in various aspects. Ships and offshore structures are continuously exposed to various environmental conditions, such as waves, wind, and currents. In the event of an accident, immense economic losses, environmental pollution, and safety problems can occur, so it is necessary to detect structural damage or defects early. In this study, structural response data of multi-linked floating offshore structures under various wave load conditions was calculated by performing fluid-structure coupled analysis. Furthermore, the order reduction method with distortion base mode was applied to the structures for predicting the structural response by using the results of numerical analysis. The distortion base mode order reduction method can predict the structural response of a desired area with high accuracy, but prediction performance is affected by sensor arrangement. Optimization based on a genetic algorithm was performed to search for optimal sensor arrangement and improve the prediction performance of the distortion base mode-based reduced-order model. Consequently, a sensor arrangement that predicted the structural response with an error of about 84.0% less than the initial sensor arrangement was derived based on the root mean squared error, which is a prediction performance evaluation index. The computational cost was reduced by about 8 times compared to evaluating the prediction performance of reduced-order models for a total of 43,758 sensor arrangement combinations. and the expected performance was overturned to approximately 84.0% based on sensor placement, including the largest square root error.

The flexibility of base material gives considerable influences on seismic responses of a structure. The effects of relative stiffness between super-structure and base material on dynamic soil-structure interaction are evaluated by parametric studies. Two 5-story buildings are used for the study; one is shearwall structure with relatively higher fundamental frequency and the other is frame structure with relatively lower fundamental frequency. The structures are modeled as beam-sticks coupled with springs and dashpots representing the base material. Dynamic equilibrium equations of the soil-structure interaction system are sloved by mode superposition method using Rosset modal damping values. Soil-structure interaction effect is found to be major concern in seismic analysis of shearwall structure in most cases while it seldom becomes engineering problem in frame-type structure. It is also found that seismic responses at lower elevation of the super-structure are amplified though they decrease at higher elevation as soil-structure interaction effects of the system increase.

In this study, we proposed a pier of pontoon-type, "Very Large Floating Structure" (VLFS), with the length of 500m, breadth of 200 m and height of 2 m in Yeosu domestic port. Since this structure ought to endure wave loads for long periods at sea, it is essential to analyze the wave response characteristics. Direct-method is used to analyze the fluid-structure problem and the coupled motion of equation is used to obtain response results. The structural part is calculated by using finite element method (FEM) and the fluid part is analyzed by using boundary element method (BEM). Dynamic responses caused by the elastic deformation and rigid motion of structure are analyzed by numerical calculation. To investigate response characteristics of the pier in regular waves, several factors such as the wavelength, water depth, wave direction and flexural rigidity of structure are considered. As a result, wave response of pier changed at the point of $L/{\lambda}$ 1.5 and represented the torsional phenomenon according to the various incident waves. And the responses showed increasing tendency as the water depths increase at the incident point in case of $L/{\lambda}=8.0$ and peak point of vertical displacement amplitude moved from side to side as the flexural rigidity of structure changes.
