Sclerotinia Twig Blight on Trees and Cottony Rot on Fruits of Satsuma Mandarin Caused by Sclerotinia sclerotiorum

  • Sclerotinia twig blight on trees and cottony rot on fruits of Satsuma mandarin were observed in Cheju, Korea in 1996 and 1997. The causal fungus causing citrus twig blight and cottony rot was identified as Sclerotinia sclerotiorum which had cup-shaped apothecia bearing hyaline and clavate asci and periphyses on sclerotium. Symptoms were produced on twigs, developing young leaves, fully developed new leaves, and fruits 2-9 days after wound inoculation, but only on twigs with young lateral buds and developing young leaves by unwound inoculation. The fungus grew well between 10 and $27^{\circ}$, but optimum temperature was $24^{\circ}$ on potato dextrose agar. Most varieties were highly susceptible to S. sclerotiorum, whereas some varieties such as Ichinan were relatively resistant among twelve Satsuma mandarin varieties in the field. This is the first record of Scledrotinia twig blight on trees and cottony rot on stored fruits of Satsuma mandarin in Korea.

First Report of Sclerotinia sclerotiorum Causing Sclerotinia Rot on Ixeridium dentatum in Korea

  • Sclerotinia rot was observed on Ixeridium dentatum cultivated as a succeeding crop in a garlic field in Seosan-si, Korea during the growing season in 2016 and 2017. Symptoms progressed from the initial irregular, water-soaked spots on main stems to wilting and eventually to plant death. White, cottony mycelia and black, irregular sclerotia formed on the basal stem and on soil surfaces. The optimal temperature of hyphal growth and sclerotia germination were $20^{\circ}C$ and $25^{\circ}C$, respectively. Sequence analysis of the internal transcribed spacer (ITS) regions revealed that the three strains isolated from Ixeridium dentatum are grouped with Sclerotinia sclerotiorum. Three strains were identified as Sclerotinia sclerotiorum based on morphological features, ITS sequence, and pathogenicity test. To the best of our knowledge, this work is the first report of Sclerotinia sclerotiorum causing sclerotinia rot on Ixeridium dentatum in Korea.

Rhizopus Soft Rot on Pear (Pyrus serotina) Caused by Rhizopus stolonifer in Korea

  • Rhizopus soft rot caused by Rhizopus stolonifer occurred on pears (Pyrus serotina) in the Jinju City Agricultural Products Wholesale Market in Korea from 2004 to 2005. The infection usually started from wounds due to cracking at harvest time. The lesions started as water-soaked, rapidly softened, then gradually expanded. The mycelia grew vigorously on the surface of the fruits and formed stolons. Colonies on potato dextrose agar at $25^{\circ}C$ were white cottony to brownish black. Sporangia were globose, black and $90{\sim}120\;{\mu}m$ in size. Sporangiophores were light brown and $480{\sim}2600{\times}12{\sim}18\;{mu}m$ in size. Sporangiospores were globose to oval, brownish, streaked, and $8{\sim}14{\times}6{\sim}10\;{\mu}m$ in size. Columella were light brownish gray, hemispherical and $70{\sim}80\;{\mu}m$ in size. On the basis of these symptoms, mycological characteristics and pathogenicity tests on host plants, the fungus was identified as Rhizopus stolonifer (Ehrenb.) Vuill. This is the first report of rhizopus soft rot on pear (P. serotina) caused by R. stolonifer in Korea.

Occurrence of Rhizopus Soft Rot on Peach (Prunus persica var. vulgaris) Caused by Rhizopus nigricans in Korea

  • A soft rot of fruits caused by Rhizopus nigricans occurred on peach (Prunus persica var. vulgaris) in The Chinju City Agricultural Products Wholesale Marke during in summer season of 2000. The disease infection usually started from wounding after harvest fruits, and then moved to outside. At first, the lesions started with water soaked and rapidly softened and diseased area gradually expanded. In severely infected film house, the rate of infected fruits reached 65.2%. Numerous sporangiospores were produced on the diseased fruits. Most of the sporangiospores were appeared to be readily dispersed in the air. The mycelia grew surface of fruits and produced stolons. Colonies on potato dextrose agar at $25{\sim}30^{\circ}C$ white cottony at first becoming heavily speckled by the presence of sporangia and the browinish black at maturity, spreading rapidly by means of stolons fired at various points to the substrate by rhizoids. Sporangia were $85.3{\sim}243.5{\times}53.4{\sim}219.2\;{\mu}m$ in size and were globose or sub-globose with. somewhat flattened base. The color of sporangia was white at first and then turned black with many spores, and never over-hanging. Sporangiophores were $8.9{\sim}36.6\;{\mu}m$ in width, smooth-walled, non-septate, light brown, simple, long, arising in groups of $3{\sim}5$ from stolons opposite rhizoids. Sporangiospores was $9.7{\sim}24.8{\times}5.9{\sim}15.8\;{\mu}m$, irregular, round, oval, elongate, angular, and browinish-black streaked. Columella was $70.2{\times}149.7{\mu}m$. brownish gray, and umberella-shaped when dehisced. The causal organism was identified as. Rhizopus nigricans Lind on the basis of the morphiogical characteristics of the fungus. Rhizopus soft rot on peach (Prunus persica) caused by the fungi has not been reported in Korea. This is the first report of rhizopus soft rot on peach caused by Rhizopus nigricans in Korea.

Rhizopus stolonifer에 의한 여주 무름병 (Rhizopus Soft Rot on Momordica charantia Caused by. Rhizopus stolenifer in Korea)

  • 2003년부터 2005년까지 경남 창원시 대산면 북부마을 여주재배 농가에서 Rhizopus stolonifer에 의한 여주 무름병이 발생하였다. 병징은 처음 과실껍질 부분이 수침상으로 물러지면서 빠르게 부패되었다. 균사생육 적온은 $25^{\circ}C$ 내외로 처음 흰색에서 연한 갈색으로 되며 검은색의 포자낭을 많이 형성한다. 포자낭경은 처음 흰색에서 회색을 나타내며 포자낭을 균사 끝에 형성하고 폭은 $15\~30{\mu}m$ 이었다. 포자낭은 처음에 흰색에서 나중에 검은색으로 되었고 모양은 반구형으로 크기는 $90\~180{\mu}m$이었다. 주축의 모양은 반구형으로 크기는 $80\~150{\mu}m$이었다. 포자낭포자는 담갈색으로 단포이며 구형 또는 타원형으로 불규칙한 것이 많으며 크기는 $7\~18{\times}6\~12{\mu}m$이었다. 이상과 같은 병원균의 균학적 및 병원학적 특성조사 결과로 본 병해를 Rhizopus stolonifer (Ehrenberg ex. Fr.) Lind에 의한 여주 무름병으로 명명할 것을 제안한다.

Sporangiophores were $2.6{\sim}5.8{\times}12.3{\sim}24.2{\mu}m$ in width, smooth-walled, non-septate, tight brown, simple, long, arising in groups of $3{\sim}5$ from stolons opposite rhizoids. Sporangiospores were $8.2{\sim}18.8{\mu}m$ long, irregular, round, oval, elongate, angular, and browinish-black streaked. Columella was $64.1{\times}136.3{\mu}m$. brownish gray, and umberella-shaped when dehisced. The causal organism was identified to be R. stolonifer. This is the first report of Rhizopus soft rot on cherry tomato caused by R. stolonifer in Korea.

Relatedness Among Indiginous Members of Sclerotinia sclerotiorum by Mycelial Compatibility and RAPD Analysis in the Jordan Valley

  • Sclerotinia sclerotiorum attacks most of the vegetable crops in the Jordan valley. Twenty-five samples/isolates were obtained in a complete coverage of that region. They were characterized for their mycelium incompatibility, and specific gene amplified using the primer SSREV/SSFWD. All isolates gave similar single band around 278 bp. Thirteen isolates were completely incompatible with the other 12 ones. The latter ones fell into four subgroups of mycelium incompatibility. RAPD analysis using three primers (OPA-2, OPA-10, and OPA-18) clustered the 25 isolates into subgroups in agreement with their morphological separation, indicating close correlation between amplified gene(s) and the gene(s) of incompatibility. All highly virulent isolates were among the group of 13, indicating a well established genomic type pathogen in this region.

  • 우리나라에서 Phytophthora capsici균에 의한 가지의 병해는 아직 그 발생이 보고되어 있지 않다. 근래 수원을 포함한 중부지방에 P. capsici균에 의한 피해과가 채집되어 기록병해인 P. parasitica균에 의한 솜털역병과 그 병징, 병원성 및 병원균의 형태등을 비교 검토하였다. 본 병은 9월 강우가 있은 후 발생이 심하였으며 처음 과면에 원형갈색의 병반이 나타나 점차 확대되면서 그 위에 짧은 균계와 분생자경이 밀생하고 유주자양이 풍부히 형성되어 솜털모양으로 공중균계가 길게 자라고 유주자양이 거의 형성되지 않는 솜털병역과는 구별되었다. 그리고 병원균의 형태 및 병원성에 있어서도 뚜렷한 차이를 볼 수 있었다. 따라서 본 병원균은 P. capsica임을 확인할 수 있었으며 미기록인 본병해에 대하여 가지갈색썩음병이라는 이름을 붙이기로 하였다.

Sporangia were 125.3${\times}$294.2 ${\mu}$m. globose or sub-globose with somewhat flattened base. White at first the black, many spored, and are never overhanging. Sporangiophores were 2.7-6.8${\times}$12.9-33.9 ${\mu}$m, smooth-walled, non-septate, light brown, simple, long, arising in groups of 3-5 from stolons opposite rhizoids. Sporangiophores were 8.6-21.1${\times}$6.41-1.7 ${\mu}$m, irregular, round, oval, elongate, angular and brownish-black streaked. Columella were 63.8${\times}$140.4 ${\mu}$m. brownish gray, umberella-shaped when dehisced. The causal organism was identified as Rhizopus stolonifer Lind on the basis of the morphological characteristics of the fungus. Rhizopus soft rot on squash (Cucurbita moschata) caused by the fungi has not been previously reported in Korea.

Biological Control of Sclerotinia sclerotiorum Using Indigenous Chitinolytic Actinomycetes in Jordan

  • The white cottony stem rot pathogen Sclerotinia scierotiorum was subjected to 70 different isolates of actinomycetes indigenous to Jordan as biological control agents. Forty of them demonstrated chitinase activity on crab shell chitin agay (CCA) media and they were segregated into three groups: 14 highly active, 12 moderately active, and 14 with low activity, with average clearing zones of (4.7-8.3), (3.7-4.3), and (2.3-3.3) mm surrounding colonies on CCA, respectively. Further, these isolates were able to inhibit radial mycelium growth of the pathogen and were categorized into three antagonistic groups: 13 strong, 13 moderate, and 14 weak antagonists, with antibiosis inhibition Bones of (32.0-45.7), (22.7-31.3), and (3.7-22.3) mm, respectively. High levels of chitinase activity of the isolates Ma3 (8.3 mm), Jul (7.7 mm), and Sa8 (7.7 mm) with their antagonistic activity against mycelium growth of 45.7, 44.3, and 40.7 mm were observed, respectively. These isolates exhibited fungicidal activity against sclevotia of S. sclerotiorum. On the other hand, isolates Na5, Aj3, and Aj2 that produced no chitinase showed fungistatic effect only.
