Characterization and Isolation of Mutants Involved in Cell Cycle Progression and Regulation in Saccharomyces cerevisiae (Saccharomyces cerevisiae에서 세포주기의 진행과 조절에 관련된 변이주들의 분리 및 특성화)

  • These studies were carried out to understand the mechanisms of genes which are related in cell cycle progression at G1/S phase. Mutants involved in cell cycle progression and regulation in Saccharomyces cerevisiae were isolated and characterized. To isolate new mutants, we screened the sensitivity to ciclopirox olamine (CPO) which inhibits the cell cycle traverse at or very near the G1/S phase boundary in HeLa cell and budding yeast. As results, we isolated 30 mutants and named cos(ciclopirox olamine sensitivity: cos27∼cos57) mutants. To determine the phenotype of mutants, we examined the sensitivity to methyl-methane sulfonate (MMS) and hydroxyurea (HU). Several mutants were sensitive to MMS and HU. According to these Phenotypes, cos mutants were grouped into four. Group I mutants are cos27, cos28, cos32, cos33, cos36, cos37, cos40, cos42, cos46, cos50, cos52 and cos53 which show MMS, HU sensitivities and might act at a checkpoint pathway during S phase. Group II mutants are cos43 and cos48 which show MMS sensitivities and might act at a checkpoint pathway during Gl or G2 phase. Group III mutants are cos35, cos47, cos54, cos55 and cos56 which show HU sensitivities and might act at a progress pathway during S phase. Finally, Group IV mutants are cos29, cos30, cos31, cos34, cos38, cos39, cos41, cos44, cos45, cos49, cos51 and cos57 which show only CPO sensitivities. Moreover, we examined the terminal phenotype of mutants under fluorescent microscope and then found one of S phase checkpoint related mutant(cos37). Furthermore, we constructed the heterozygote strain between mutant and wild type haploid strains to study their genetic analysis of cos mutants.

Effect of Antimicrobial Activity by Chitosan Oligosaccharide N-Conjugated with Asparagine

  • Since the number of amino groups which are exposed by deacetylation of acetyl-D-glucosamine influences antimicrobial activity, a chitosan oligosaccharide (COS) derivative by N-conjugation of COS with asparagine, an amino acid with two amino groups, was synthesized and the antimicrobial effect on E. coli growth was compared with other COS derivatives which were N-conjugated with glycine, alanine, aspartic acid, cysteins, an methionine, and unmodified COS. The structure of asparagine N-conjugated COS (Asn-COS) derivative was identified by using a FT-IR, $^{13}C\;FT-NMR$, and an elemental analyzer. The antimicrobial activity of Asn-COS against E. coli growth was significantly improved as compared to the other COS derivatives as well as COS itself. This means that Asn-COS with two positive charges strongly interacts with the carboxyl negative charges on the bacteria cell wall. The results for Asn-COS were as follows: 100% bactericidal activity, 0.002% MIC, and no growth of E. coli during 3 days of culture time, suggesting that Asn-COS may be useful as a new antibiotic agent.

Comparison of Properties of Polyclonal Anti-N-Acetylchitooligosaccharides and Anti-Chitooligosaccharides Antibodies Produced for ELISA

  • To develop the enzyme-linked immunosorbent assay (ELISA) for the analysis of N-acetylehitooligosaccharides (NACOS) and chitooligosaccharides (COS), specific antibodies (Abs) were produced, and their properties were compared. N-acetylehitohexaose (NACOS6), chitohexaose (COS6), and COS mixture (COSM) conjugated to bovine serum albumin (BSA) were used to immunize rabbits. By the use of specific Abs and NACOS6-horseradish peroxidase (HRP), COS6-HRP, and COSM-HRP conjugates, competitive direct ELISA (cdELISA) was developed. The detection limits of NACOS6 by the anti-NACOS6 Ab and COS6 by the anti-COS6 and the anti-COSM Abs in the cdELISAs were about 0.2, 2, ana 2 ng/ml (ppb), respectively. In the cdELISA, the anti-NACOS6 Ab was found to recognize NACOS3-NACOS6, but not N-acetyl-D-glucosamine (GlcNAc), NACOS2, and COSs; the anti-COS6 Ab recognized COS2-COS6 and COSM, but not glucosamine (GlcN) and NACOSs. The recognition pattern of the anti-COSM Ab was almost the same as that of the anti- COS6 Ab, except that the former recognized COS2 and COS3 slightly better than the latter.

  • This study goes into the field of CosPlay that takes place in Korea, and directly observes the people who engage in CosPlays. Based on in-depth interviews, this research identified the reasons why the CosPlay aficionados participate in this practice and their cultural preferences. Ethnography methodology was used to understand the behavior of the cultural entities of CosPlays. Moreover, this research attempted to understand their daily formalities through their own perspective and language instead of superficial language. In general, CosPlay form that they manifest is classified into two main categories: those who enjoy posing by becoming their favorite characters and those like to wear the clothes of their favorite characters and go up on the stage to perform an act of animation or game where the applicable character appears. Some of these groups are: 1) 'Bishojo CosPlay' that CosPlays mostly Bishojo cartoon characters and wants to just stand out, 2) 'Aekyo' group that CosPlays merely for the sake of the enjoyment of wearing costumes instead of having the mania-like characteristics, 3) 'CosPlayer Group' who enjoy doing organized team CosPlay in order to present a performance, 4) 'J-Rock CosPlay' group that CosPlay Japanese visual rock groups and prepare to be among a professional CosPlay team, and 5) 'People related to Cospre.com' who try to make CosPlay rooted in as one of new cultures.

An Ethnographic Study on CosPlay Group in Korea II - Analysis on CosPlay Culture in Korea and Japan - (한국 코스프레 집단의 문화기술지적 연구 II - 한국과 일본의 코스프레 문화에 대한 비교 분석 -)

  • This study examines the social meaning of the CosPlay, the growth potential of CosPlay culture and its effect on the related industry through the perspectives and language of the youths who enjoy CosPlay, based on the ethnographic research. Also, this study presents a comparative description of Korea and Japan CosPlay culture by the ethnographic methodology whose purpose is to define relationship of cause and effect with phenomenon. For further step, this study plans to emphasize the need to link culture, clothes and related industry in order to create a cultural environment where diversity co-exists. CosPlay is the mania culture of Japan that emulated the Halloween party of the West and that developed the party into a unique form. In Korea, this practice was accepted for the first time among a handful of youths, starting from the 1990s, after which, it was introduced to the masses while holding CosPlay related events. While CosPlay is succeeded as an industry in Japan, CosPlay in Korea is considered childish play due to the Korean culture of considering cartoon as a childish and low class genre which is enjoyed by youths. CosPlay in Korea faces the following changes: aging of the members who comprised the CosPlay culture at the initial stage; population increase, centered on middle and high school students; interest of the government and the businesses that wish to produce economic wealth by organizing CosPlay events into events for youths; and changes in the environment that comprised the surrounding of the CosPlay culture. CosPlay is an honest play that demonstrates one's effort on the stage through performance. Moreover, most of the middle and high school students who comprise the CosPlay culture demonstrate similar characteristics as mania type of people when it came to the reason that they enjoy CosPlay. However, they did not consider CosPlay culture as an important aspect of their lives. Instead, most of them said that they participate to relieve stress. Thus, they have the potential to move onto another form of youth culture that may appear more attractive to them. To them, it is not the CosPlay culture that is important, but the fact that CosPlay provides a forum where they can freely engage in play.

Chitooligosaccharide Prolongs Vase Life of Cut Roses by Decreasing Reactive Oxygen Species

  • Chitooligosaccharide (COS), as antioxidant, extensively applied to food and juice preservation. In the present study, influences of COS on vase life and ornamental value of cut roses were investigated. Results showed that vase life of cut roses treated by COS was longer 6.4 days than one of control and ornamental character of cut roses was improved effectively by COS. The increase of vase life and ornamental value were chiefly governed by that COS improved water absorption capacity of cut roses. Besides that, COS decreased the contents of superoxide anion and hydrogen peroxide and lowered the levels of malondialdehyde in turn during the senescence process of cut roses. That was because that COS not only enhanced activities of antioxidant enzymes glutathione reductase, but also improved reduced glutathione contents in petals of cut rose. Therefore, COS could be used in commercial preservatives to improve the longevity of cut roses.

A Development of COS Holder to Reduce an Impact Ambient Noise (충격성 소음 감소용 COS 홀더 개발)

  • We are using COS to block the excess current and to protect the transformer. But the fuse of COS is melt due to the overload resulting from the excess current flows and it destroys an air severing relations to clear as strong arc happens. Such phenomenon induces an impact ambient noise and it gives the circumstance area resident or pedestrian the fear. Thus, it is necessary that we urgently need to control the ambient noise from the actual circumstances. In this study, we investigate the characteristic of an impact ambient noise associate with the melting state of the COS fuse and suggest the method to reduce the noise.

In Vitro Construction and Characterization of the Bacteriophage P4 Derivative, P4 sid71 cosP2, Containing the Bacteriophage P2 cos Region (박테리오파지 P2의 cos 지역을 함유하는 박테리오파지 P4 유도체인 P4 sid71 cosP2의 In vitro 조성과 정성 연구)

  • Bacteriophage P2 sir mutants are inefficient helpers for their satellite bacteriophage P4. The term, "P2 sir-associated helper inefficiency" has been used to define this phenomenon and it has been suggested that the DNA sequence difference between the cos region of P2 and that of P4 is responsible. To test this hypothesis, P4 derivative phage, P4 sid71 cosP2, containing the cos region of P2 and sid71 allele was constructed through several in vitro DNA manipulation steps. Its burst size was determined using a one-step growth experiment. The results showed that the substitution of the cos region of P2 for the cos region of P4 in P4 sid71 cosP2 overcame "P2 sir-associated helper inefficiency". P4 sid71 cosP2 stock phages prepared with P2 wild type helper and P2 sir helper were analyzed using a CsCl buoyant equilibrium density gradient experiment. The results revealed that the phage particles containing three copies of the P4 genome were the predominant particles in both cases.

Development of Current Limiting COS Fuse Link with Improved Overcurrent and Protection Coordination performance (과전류 차단과 보호협조 성능이 향상된 한류형 COS 퓨즈링크 개발)

  • A Cut Out Switch (COS) is used for line protection and pole transformer protection in power systems. The COS used to protect the pole transformer is installed on the power side of the pole transformer to protect the electric equipment from fault currents. The COS is composed largely of a body and a fuse holder, and when the fault current is energized, the element of the fuse link in the fuse holder is melted to block the fault current. The arc generated when the COS fuse link is blown causes fire and noise, causing discomfort to residents in the surrounding area, and the arc flame can cause secondary damage to the peripheral device. In this study, a current-limiting COS fuse with improved overcurrent blocking performance rather than explosion type was developed to solve the arc and noise problems during COS operation. The overcurrent breaking performance of the current-limiting COS improves the reliability by developing a striker and COS fuse bracket. In addition, this study aimed to verify the performance of the developed current-limiting COS fuse through a test at an authorized institution.

A Study on the Dextran Formation and Inner Structure of Jeung-Pyun during the Fermentation by the Added Chitosan-Oligosaccharide (키토산 올리고당 첨가 증편 발효 중 Dextran 형성과 증편의 내부구조에 관한 연구)

  • This study was carried out in order to investigate the dextran formation and the internal structure of the chitosan-oligisaccharide(COS) Jeung-Pyun during the fermentation. To develop a functional Jeung-Pyun, COS was added to the rice at the level of 0, 2, 4, and 6% to the 2-hour-soaked rice. The pH, dextran and reducing sugar contents of COS batter, specific volume and the internal structure of COS were analyzed as a function of fermentation time. The specific volume of COS peaked at the 3 hour fermentation. The pH values of COS batters reduced with the fermentation time. The dextran content of the control batters peaked at the 7 hour fermentation. In contrast, the dextran contents of COS batters reduced up to 7th hour of fermentation, and then increased slowly until 20th hour of the fermentation process. From the air pore size and distribution of COS observed by SEM, the COS fermented fur 3 hour was judged as the best. It was concluded that COS prevented the fermation of dextran at the early stage of fermentation process.
