Background: Inactivation in p53 tumor suppressor gene through a point mutation and deletion is one of the most frequent genetic changes found in human cancer, with 50% of an incidence. This high rate of mutation mostly suggests that the gene plays a central role in the development of cancer and the mutations detected so far were found in exons 5 to 8. Mutation of p53 locus produced accumulation of abnormal p53 protein, and negative regulation of cell proliferation and transcriptional activation as a suppressor of transformation were lost. In addition, inhibition of its normal cellular function of wild-type by mutant is an important step in tumorigenesis. Method: 4 colon cancer cell lines (SNU C1, C2A, C4, C5) were examined for mutation in exons 5 to 8 of the p53 tumor suppressor gene by PCR-SSCP analysis and expression pattern by western blotting and immunoprecipitation. p53-mediated transactivation ability were examined by CAT assay and base substitution of p53 in SNU C2A cell were detected by DNA sequencing. Results: 1) SNU C2A cell and SNU C5 cell were detected mobility shifts each in exon 5 and exon 7 of p53 gene by the PCR-SSCP method, implicating being of p53 mutation. 2) 3 colon cancer cell lines (SNU C1, SNU C2A, SNU C5) expressed wild type and mutant type p53 protein. 3) In northern blot experiment, SNU C2A and SNU C5 cell expressed high level of p53 mRNA. 4) Results of p53-mediated transactivation in colon cancer cell lines by CAT assay represented only SNU C2A cell has transcriptional activity. 5) DNA sequencing in SNU C2A cell showed missense mutation in codon 179 of one allele, histidine to arginine and wild type p53 in the other allele. Conclusion: Colon cancer cell lines showed correlation with mutation in p53 gene and accumulation of abnormal p53 protein. Colon cancer cell SNU C2A retained p53-mediated transactivation as heterozygous p53 with one mutant allele in 179 codon and the other wild-type allele.

Cellular response to ionizing radiation is affected by cell types, radiation doses, and post-irradiation time. Based on the trypan blue dye exclusion assay in normal oral mucosal cells (OM cells), a 48 h post-irradiation was sufffcient and an adequate time point for the evaluation of radiation sensitivity Its $LD_{50}$ was approximately 1.83 Gy To investigate possible biomarkers useful for the biological radiodosimetry of normal epithelial cells (p53, c-fos, cyclin D1, cdc-2, pRb) EGF receptor phosphorylation and Erk activation were evaluated at different radiation doses and different post-irradiation times. From 0.5 Gy, p53 was accumulated in the nucleus of basal cells of the OM raft culture at 4 h post-irradiation and sustained up to 24 h post-irradiation, which suggests that radiation-induced apoptosis or damage repair was not yet completed. The number of p53 positive cells and biosynthesis of p53 were correlated with radiation doses. Both cyclin D1 and c-fos were only transiently induced within 1 h post-irradiation. Cyclin D1 was induced at all radiation doses. However, cfos induction was highest at 0.1 Gy, approximately 7.3 fold more induction than the control, whose induction was reduced in a reverse correlation with radiation dose. The phosphorylation pattern of cdc-2 and pRb were unaffected by radiation. In contrast to A431 tails overexpressing the EGF receptor approximately 8.5 fold higher than normal epithelial, the OM cells reduced the basal level of the EGF receptor phosphorylation in a radiation dose dependent fashion. In conclusion, among radiation-induced biomolecules, the p53 nuclear accumulation may be considered for the future development of a useful marker far biological radiodosimetry in normal epithelial tissue since it was sustained for a longer period and showed a dose response relationship. Specific c-fos induction at a low dose may also be an important finding in this study It needs to be studied further for the elucidation of its possible connection with the low dose radio-adaptive response.

Quantitative precipitation estimation (QPE) is one of the most important elements in meteorological and hydrological applications. In this study, we adjusted the QPE from an S-band weather radar based on co-kriging method using the geostatistical structure function of error distribution of radar rainrate. In order to estimate the accurate quantitative precipitation, the error of radar rainrate which is a primary variable of co-kriging was determined by the difference of rain rates from rain gauge and radar. Also, the gauge rainfield, a secondary variable of co-kriging is derived from the ordinary kriging based on raingauge network. The error distribution of radar rain rate was produced by co-kriging with the derived theoretical variogram determined by experimental variogram. The error of radar rain rate was then applied to the radar estimated precipitation field. Locally heavy rainfall case during 6-7 July 2009 is chosen to verify this study. Correlation between adjusted one-hour radar rainfall accumulation and rain gauge rainfall accumulation improved from 0.55 to 0.84 when compared to prior adjustment of radar error with the adjustment of root mean square error from 7.45 to 3.93 mm.

Purpose: Charcot-Marie-Tooth disease (CMT) is a peripheral neuropathy mainly divided into CMT type 1 (CMT1) and CMT2 according to the phenotype and genotype. Although molecular pathologies for each genetic causative have not been revealed in CMT2, the correlation between cell death and accumulation of misfolded proteins in the endoplasmic reticulum (ER) of Schwann cells is well documented in CMT1. Establishment of in vitro models of ER stress-mediated Schwann cell death might be useful in developing drug-screening systems for the treatment of CMT1. Materials and Methods: To develop high-throughput screening (HTS) systems for CMT1, we generated cell models using transient expression of mutant proteins and chemical induction. Results: Overexpression of wild type and mutant peripheral myelin protein 22 (PMP22) induced ER stress. Similar results were obtained from mutant myelin protein zero (MPZ) proteins. Protein localization revealed that expressed mutant PMP22 and MPZ proteins accumulated in the ER of Schwann cells. Overexpression of wild type and L16P mutant PMP22 also reduced cell viability, implying protein accumulation-mediated ER stress causes cell death. To develop more stable screening systems, we mimicked the ER stress-mediated cell death in Schwann cells using ER stress inducing chemicals. Thapsigargin treatment caused cell death via ER stress in a dose dependent manner, which was measured by expression of ER stress markers. Conclusion: We have developed genetically and chemically induced ER stress models using Schwann cells. Application of these models to HTS systems might facilitate the elucidation of molecular pathology and development of therapeutic options for CMT1.

The surface characteristics of restoration such as surface roughness and droplet contact angle are important part for the process of bacterial adhesion. The purpose of this study is to compare plaque adhesion by measuring roughness, droplet contact angle, and amount of accumulated plaque on the surfaces of composite resins. Four kinds of composite resins, Z-100(Z1), Durafil(DF), Filtek supreme(FS), Clearfil AP X(CA) were used. Ten samples were divided into unpolished and polished group. Surface roughnesses and droplet contact angles were measured by profilometer and goniometer. Plaque weight gains are measured. The results were as follows: 1. The experimental group were rougher than the control group. Surface roughnesses were decreased in the following order; (Z1, DF, CA)>FS in the control group, and CA>Z1>(FS, DF) in the experimental group(P<0.05). 2 The control group showed larger contact angle than the experimental group. Contact angles were decreased in the following order; CA>(FS, DF, Z1) in the control group, and (CA, DF)>(FS, Z1) in the experimental group(P<0.05). 3. The experimental group showed more much plaque than the control group. The amounts of plaque accumulation in vitro were decreased in the following order; Z1>(DF, FS)>CA in the control group, and Z1>FS>(CA, DF) in the experimental group. The latter showed more much plaque than the former(P<0.05). 4. There were stronger correlation between plaque deposition and contact angle (P<0.05) than that of plaque deposition and surface roughness.

The aim of this study was to compare the efficiency of sonic and manual toothbrush in decreasing plaque accumulation in young orthodontic patients. Twenty one healthy orthodontic patients attending department of pediatric dentistry at Samsung Medical Center were chosen for the study. The subjects were randomly assigned to either sonic or manual toothbrush and instructed to brush for 2 minutes. Plaque score was assessed with the modified Visible Plaque Index (VPI) before and after brushing. Each brush was repeated twice. A mean plaque reduction of sonic toothbrush was 61.79%(${\pm}7.95$) compared to 69.19%(${\pm}10.08$) of manual toothbrush(p<0.05). With the sonic toothbrush, male presented a mean plaque reduction of 63.07%(${\pm}8.64$) while female presented 60.39%(${\pm}7.30$). For manual toothbrush, male presented 69.33%(${\pm}10.14$) and female presented 69.03%(${\pm}10.55$) reduction of plaque accumulation(p>0.05). There was no statistically significant correlation between age and plaque removal efficacy in this study(p>0.05). Manual toothbrush was significantly more efficient in removing plaque than the sonic toothbrush in young fixed orthodontic patients.
A pot experiment on paddy rice was carried out to investigate the effect of temperature after heading with application of phosphate, potash and silica on the yield and nutrient uptake. The results obtained are as follows: 1. The Grain yield was greatly affected by temperature during ripening. The yield at $20^{\circ}C$ and at $15^{\circ}C$ decreased to 62.3% and to 37.2% of that at $25^{\circ}C$ respectively. However, the application of phosphate, potash and silica at transplanting increased the grain yield even at low temperature of $15^{\circ}C$. 2. Percentage of ripened grain and weight of 1,000 grains decreased with decreasing temperature but they were increased by the application of phosphate, potash and silica at the low temperature of $15^{\circ}C$. 3. The grain filling rate was fast in the early stage of ripening at high temperature while it was fast in the later stage at low temperature. Low temperature damage was severe in the early stage of ripening. 4. Accumulation of carbohydrate in the grain was small at low temperature of $15^{\circ}C$ and the application of phosphate, potash and silica was effective to increase carbohydrate content in the grain at low temperature. 5. The content of T-N and $P_2O_5$ in straw grown at low temperature were high whereaas those of $K_2O$ and $SiO_2$ were low which were increased by the application of potash and silica. 6. There was a positive correlation between the grain yield and rate of ripening, 1,000 grain weight, contents of $K_2O$ and $SiO_2$, $K_2O/N$ and $SiO_2/N$ ratio, but a negative correlation between grain yield and contents of T-N and $P_2O_5$ in the straw at harvesting stage. 7. Ripening rate and weight of 1,000 grains showed a significant positive correlation with $K_2O$, $SiO_2$, $K_2O/N$ and $SiO_2/N$ ratio in the straw. Further, there was a negative correlation between ripening rate and the contents of T-N and $P_2O_5$ but a positive correlation between weight of 1,000 grains and these elements.
Twenty seafood samples, which are common edible species and commercially important items in Korea, were purchased at the local fisheries markets and were analyzed for polycyclic aromatic hydrocarbons (PAHs) using gas chromatography coupled to mass spectrometer detector (GC/MSD). The levels of sixteen PAHs in seafood from Korean coasts were 161 to 2,243 pg/g wet weight. The highest concentration was found at saury (Coloabis saira) and the lowest level was found at jacopever (Sebastes schlegeli). The concentrations of potentially carcinogenic PAHs of six species were in the range of 9 to 123 pg/g wet weight. The residues of PAHs in fishes from Korean coasts were slightly low or relatively moderate to other countries. There was no correlation between PAH residues and lipid contents in seafood samples. The predominant contributors in fish samples were lower-molecular-weight two and three ring aromatic PAHs such as naphthalene, acenaphthene, fluorene and phenanthrene. Filter-feeding organisms like shrimp, crab and topshell were dominated by three- and four-ring aromatic PAHs. The PAH profiles in marine sediments, bivalves, fishes, shrimp, crab and topshell according to exposure pathway were compared through factor analysis. The PAH profiles were clearly classified by the difference of species or environmental matrices. This result suggests that most of PAHs within the same samples behave identically in marine environment.
Electrochemical characteristics of nitrite ion were investigated at a poly(methylene blue)-modified glassy carbon electrode by cyclic voltammetry and differential pulse voltammetry. The poly(methylene blue)-modified glassy carbon electrode exhibited enhanced anodic signals for nitrite. The effects of key parameters on the detection of nitrite were evaluated at the modified electrode, such as pH, accumulation time, and scan rate. Under optimum condition, the chemically modified electrode can detect nitrite in the concentration range $2.0{\times}10^{-6}$ to $5.0{\times}10^{-4}$ M with the detection limit of $2.0{\times}10^{-6}$ M and a correlation coefficient of 0.999. The detection of nitrite using the chemically modified electrode was not affected by common ions such as $Na^+$, $K^+$, $Ca^{2+}$, $Cl^-$, $HPO_4^{2-}$ and $H_2PO_4^- $. The modified electrode showed good stability and reproducibility. The practical application of the present method was successfully applied to the determination of nitrite ion in cabbage samples.

Summer cabbages in an alpine area are very sensitive to the fluctuations in supply and demand. Yield variability due to weather conditions dictates the market fluctuations of cabbage price. This study reports an empirical relationship based on weather conditions to estimate the growth and harvestable biomass of cabbages, factors that are critical for supply of summer cabbages. Based on experimental results testing sowing date effects over the two years from 1997 to 1998, a logistic equation was parameterized to predict leaf area expansion of summer cabbages. This logistic model for leaf area expansion was then combined with an empirical allometric relationship to predict total biomass. The final equation for estimating fresh weight accumulation of Chinese cabbage is given by: $$Fresh\;weight=3500/(1+{\exp}(5.175-1.153{\times}(6/(1+{\exp}(6.367-0.0064{\times}PHU)))))$$ Where PHU is potential heat units ($^{\circ}C$). The model performance was tested using weather data from 2003 to 2006 to predict fresh harvestable biomass. Overall the model performance was satisfactory with the correlation efficient ranging between 0.89 and 0.94 for each year.
