The importance of cornified envelope (CE)-bound lipids to epidermal barrier function is increasingly being recognized. In the present study, we intentionally damaged the cornified layer of hairless mice by ultraviolet irradiation and sodium dodecyl sulfate (SDS) treatment, and assessed the changes in epidermal barrier function by measuring Trans Epidermal Water Loss (TEWL). We also measured changes in the amount of CE-bound lipids using thin layer chromatography (TLC). The results showed that both treatments increased TEWL and decreased CE-bound lipids (omega-hydroxy cerami de and omega-hydroxy acid). In addition, investigation of the chronological changes in TEWL revealed an inverse relationship between TEWL and CE-bound lipids, and a correlation between CE-bound lipids and epidermal barrier function. We then measured the amount of CE-bound lipids in the cheek and the medial side of the upper arm in humans. The results showed that because the cheek receives external stimulation on a daily basis, the amount of CE-bound lipids was significantly lower, while the level of TEWL was higher. These observations, together with those from the animal study, indicate that CE-bound lipids are related to epidermal barrier function.

HaCaT cells are the immortalized human keratinocytes and have been extensively used to study the epidermal homeostasis and its pathophysiology. T helper cells play a role in various chronic dermatological conditions and they can affect skin barrier homeostasis. To evaluate whether HaCaT cells can be used as a model cell system to study abnormal skin barrier development in various dermatologic diseases, we analyzed the gene expression profile of epidermal differentiation markers of HaCaT cells in response to major T helper (Th) cell cytokines, such as $IFN{\gamma}$, IL-4, IL-17A and IL-22. The gene transcriptional profile of cornified envelope-associated proteins, such as filaggrin, loricrin, involucrin and keratin 10 (KRT10), in HaCaT cells was generally different from that in normal human keratinocytes (NHKs). This suggests that HaCaT cells have a limitation as a model system to study the pathophysiological mechanism associated with the Th cell cytokine-dependent changes in cornified envelope-associated proteins which are essential for normal skin barrier development. In contrast, the gene transcription profile change of human ${\beta}2$-defensin (HBD2) in response to $IFN{\gamma}$, IL-4 or IL-17A in HaCaT cells was consistent with the expression pattern of NHKs. $IFN{\gamma}$ also up-regulated transglutaminase 2 (TGM2) gene transcription in both HaCaT cells and NHKs. As an alternative cell culture system for NHKs, HaCaT cells can be used to study molecular mechanisms associated with abnormal HBD2 and TGM2 expression in response to $IFN{\gamma}$, IL-4 or IL-17A.
본 연구에서는 두충 추출물이 RBL-2H3 세포의 ${\beta}$-hexosaminidase의 분비 억제와 HaCaT keratinocytes 피부장벽의 회복과 관련한 filaggrin, transglutaminase-1 (TGase-1), cornified cell envelope(CE)의 발현에 미치는 영향에 관하여 연구하였다. ${\beta}$-hexosaminidase 방출 억제 능은 13% 효능을 확인하였고, 피부장벽기능의 회복과 관련된 인자들은 발현과 활성의 정도가 매우 우수한 것을 확인하였다. 각질형성세포의 분화를 판단할 수 있는 CE 측정에서는 두충추출물이 양성대조군보다 더 좋은 효능을 나타내기도 하였다. 따라서 두충 추출물은 ${\beta}$-hexosaminidase 분비 억제에 효과가 있으며, 손상된 피부장벽강화에 영향을 미치는 각질형성세포의 분화 촉진에 효과가 있음을 확인하였다.
본 연구는 울릉도 돌외(Gynostemma pentaphyllum) 잎으로부터 캘러스 유도 및 추출한 후, 피부장벽 기능과 관련된 항염증, 항알러지, 각질세포외막 형성, S. aureus에 의한 각질형성세포 손상 완화 효능을 확인하고자 수행되었다. 돌외 캘러스 추출물의 피부에 대한 항염증 효능을 확인하기 위해, PAR-2 agonist로 활성화된 primary epidermal keratinocyte (HEKa)에서 염증성 사이토카인들의 발현을 확인한 결과, 돌외 캘러스 추출물은 IL-8, IL-25, TSLP 발현을 저해하는 효능이 있는 것을 확인하였다. RBL-2H3 세포를 이용한 β-hexosaminidase assay 시험을 통해 항알러지 효능을 확인한 결과, β-hexosaminidase 방출을 억제하는 효과를 보였다. 또한, 돌외 캘러스 추출물은 HaCaT 세포에서 각질세포외막(cornified envelope) 형성 효과가 있음을 확인하였고, HaCaT 세포와 S. aureus 공배양 실험을 통해 S. aureus에 의한 각질형성세포 생존율 감소를 완화하는 효능이 있음을 확인하였다. 본 연구 결과를 종합해보면, 울릉도 돌외 캘러스 추출물은 항염, 항알러지, S. aureus에 의한 각질세포 손상 완화 효능에 유효한 소재의 가능성을 확인하였고, 피부장벽 개선을 위한 화장품 소재로서 활용 가치가 있을 것으로 판단된다.
본 연구에서는 다양한 식물추출물의 항산화성 및 미용관련 기능성을 알아보기 위하여 기존의 용매추출에 비해 안전성이 높은 것으로 알려진 초임계 추출방식을 적용하여 구절초(Chrysanthemum zawadskii), 수세미(Lufa cylindrica), 작약(Paeonia lactiflora), 치자나무(Gardenia jasminoides) 및 황금(Scutellaria baicalensis)의 천연식물 초임계 추출물을 확보하고, 이들의 항산화력 및 피부장벽 향상 효과를 조사하였다. 초임계 식물추출물의 피부장벽기능 향상 효과를 조사하기 위하여 항산화 활성 외 proliferator-activated receptor(PPAR)-${\alpha}$ 활성, cornified envelope(CE)에 관련된 단백질인 involucrin의 발현량을 측정하였다. 이들 추출물 중 황금추출물이 가장 높은 항산화 활성을 보였으며, 모든 추출물에서 대조군과 비교하여 유의적으로 높은 수준의 PPAR-${\alpha}$ 활성을 나타내었다. 또한, 피부장벽 기능 지표 단백질인 involucrin 발현량도 모든 추출물에서 높게 나타났으며, 황금추출물이 가장 높은 단백질 발현 양상을 보였다. 따라서 본 연구에서 조사한 5개 식물의 초임계 추출물은 항산화 및 피부장벽 기능개선과 같은 기능성 생물활성 소재로 활용 될 수 있을 것으로 판단된다.
Triterpenoid, saponin, 전분 등이 함유되어 있는 것으로 알려진 Adenophorae radix (A. radix)의 뿌리 추출물을 이용하여 각질형성세포의 분화와 피부장벽기능에 대한 연구를 수행하였다. A. radix의 뿌리 추출물은 CV-1 세포를 이용하여 $PPAR{\alpha}$ 발현을 살펴본 결과, Wy-14,643 $0.5-1.0{\mu}M$ 수준의 발현양을 나타내었다. 인체 각질형성 세포주(HaCaT)와 각질형성세포(nomal human keratinocyte)에 대한 각질형성능(cornified envelop formation, CE)은 대조군에 비해 통계적으로 유의한 증가를 보였다. HaCaT 세포에 A. radix의 뿌리 추출물 처리하였을 때, transglutaminase (TGase-1)의 유의적 증가를 보였다. A. radix의 뿌리 추출물을 함유한 간단한 화장품 제형을 약 2주간에 걸쳐 임상시험을 실시한 결과, TEWL의 유의적 감소와 수분량의 증가를 살펴볼 수 있었으며, 하박 내측에서 지질을 추출하여 세라마이드를 분석한 결과 통계적으로 유의한 증가를 관찰할 수 있었다. 이를 통하여 A. radix의 뿌리 추출물을 건조피부나 아토피 등의 피부질환과 관련된 질환의 예방 및 치료제로 사용될 수 있을 것이다.
In 2022, the domestic production performance of functional cosmetics in South Korea reached 4.6 trillion won, accounting for 33.85% of the total cosmetics production. The number of functional cosmetics reviewed increased by about 7.5% from the previous year, totaling 974 items. Especially with the increasing importance of the skin barrier function due to skin sensitivity caused by various environmental pollutants, domestic cosmetic companies are showing interest in the development of new ingredients and products related to this area. This study aims to analyze academic research trends related to in vitro experiments for the development of cosmetics improving the skin barrier, to provide practical information for the cosmetic industry. The findings are as follows: Academic research mainly focused on the efficacy of natural ingredients in improving the skin barrier, but there is a significant lack of quantitative accumulation of research. For the development of skin barrier-improving cosmetic ingredients, efficacy evaluation indicators were set, including hyaluronic acid production, expression of filaggrin gene, loricrin, formation of cornified envelope (CE), and expression of ceramide synthesis enzyme genes. Moreover, effective cosmetic ingredients for improving the skin barrier included lemongrass and perilla leaf extracts, flavonoids, Lactococcus lactis subsp. lactis, Exosomelike Nanovesicles derived from apple callus, Eleutherococcus sessiliflorus, Acanthopanax sessiliflorus, Eleutherococcus gracilistylus, Acer okamotoanum extracts, Aloe vera adventitious root extract, ethanol extract of Aruncus dioicus, and organic solvent fraction of Dracocephalum argunense.
The skin plays a key role in protecting the body from the environment and from water loss. Cornified envelope (CE) and natural moisturizing factor (NMF) are considered as the primary regulators of skin hydration and barrier function. The CE prevents loss of water from the body and is formed by cross-linking of several proteins. Among these proteins, filaggrin is an important protein because NMF is produced by the degradation of filaggrin. Proteases, including matriptase and prostasin, stimulate the generation of filaggrin from profilaggrin and caspase-14 plays a role in the degradation of filaggrin. This study elucidated the effects of an ethanol extract of Boesenbergia pandurata (Roxb.) Schltr., known as fingerroot, and its active compound panduratin A on CE formation and filaggrin processing in HaCaT, human epidermal keratinocytes. B. pandurata extract (BPE) and panduratin A significantly stimulated not only CE formation but also the expression of CE proteins, such as loricrin, involucrin, and transglutaminase, which were associated with $PPAR{\alpha}$ expression. The mRNA and protein levels of filaggrin and filaggrin-related enzymes, such as matriptase, prostasin, and caspase-14 were also up-regulated by BPE and panduratin A treatment. These results suggest that BPE and panduratin A are potential nutraceuticals which can enhance skin hydration and barrier function based on their CE formation and filaggrin processing.
피부장벽을 포함한 표피층은 인체의 조직 가운데에서도 가장 역동적인 기관이다. 다시 말해서 끊임없이 새로운 표피세포의 형성, 분화 및 탈각과정이 반복되면서 표피항상성(epidermal homeostasis)을 유지한다. 표피항상성은 피부기능 가운데 가장 주요한 기능인 permeability barrier homeostasis의 확립으로 연결된다. Permeability barrier homeostasis는 각질층에서 이루어지며 이를 형성하고 유지하기 위해 매우 정교하게 조절되어야 한다. 표피항상성을 조절하는 핵심 조절인자로서 nuclear hormone receptor(NHR)가 중심에 있음이 최근 다양한 연구를 통해 입증되었다. 이들은 각질세포 특이적인 단백질, 즉, involucrin, loricrin 및 trans-glutaminase 1(TG 1) 등의 발현을 유전자 수준에서 조절할 뿐 아니라 표피 지질성분의 생합성을 증가시키는 등 피부장벽을 구성하는 brick 및 mortar의 생성과 유지에 핵심적 역할을 하는 것으로 알려졌다. NHR 가운데 peroxisome proliferator activator receptor(PPAR)와 liver X receptor(LXR)의 activator/ligands가 리놀레인산 등 지방산, leukotriene, prostanoid 및 oxygenated sterol 등이 지질대사과정에서 형성된 지질 종류인 까닭에 liposensor로도 알려지고 있다. 따라서 liposensor들을 비롯한 PPAR과 LXR activator/ligands들은 피부장벽기능이 저해된 아토피성 피부를 포함하여 건조피부를 관리하는 epidermotherapy의 수단으로서 잠재적 가능성이 있다고 생각된다.
본 연구에서는 프로바이오틱스 유래 엑소좀 유사 나노베지클을 분리하고, 피부에 대한 여러 가지 생리활성을 평가했다. 프로바이오틱스의 한 종인 Lactococcus lactis subsp. lactis (LL)를 배양하고 고압균질기와 한외여과를 통해 70 ~ 200 nm 크기를 갖는 LL 유래 엑소좀 유사 나노베지클(LVs)을 분리했다. 나노입자추적분석 결과 1.81 × 1011 particles/mL로 나타났다. LVs를 섬유아세포와 피부각질세포에 처리하여 피부 주름과 장벽 개선과 관련된 효능을 확인했다. 우선 섬유아세포에서 fibrillin (FBN1) 유전자 발현량이 23%, 피부각질세포에서 fibronectin (FN1)과 filaggrin (FGN) 유전자 발현량이 각각 65%, 400% 증가했다. 그리고 각질형성능은 대조군 대비 30% 증가함을 확인할 수 있었다. 또한, UV 조사한 피부각질세포에 LVs를 처리했을 때 collagen type I alpha 1 (COL1A1)이 대조군 대비 약 83% 증가하는 결과를 보여주었다. 이로써 프로바이오틱스 유래 엑소좀 유사 나노베지클은 장벽 개선과 관련하여 화장품 및 의약품 소재로 이용할 수 있음을 확인했다.
