Antimicrobial activities of various extracts of Coriolus versicolor against oral bacteria (구강세균에 대한 구름버섯 추출물의 항균효과)

  • Objectives: Coriolus versicolor is an edible mushroom with physiological activities that has been used in traditional medicine. The aim of this study was to investigate the antimicrobial activity of extracts obtained from Coriolus versicolor against oral pathogens. Methods: The antimicrobial activities of various extracts of Coriolus versicolor were examined by disc diffusion assay, and minimum inhibitory concentration (MIC) of these extracts were also was determined by broth dilution method. The growth inhibition effect of extracts was measured at 600 nm for 12 hrs against Streptococcus ratti, Streptococcus criceti, Aggregati--bacter actinomycetemcomitans, Actinomyces viscosus, and Actinomyces israelii. Results: Coriolus versicolor extracts showed antimicrobial activities against all nine oral pathogens through disc diffusion assay. The ethanol extract and ethyl acetate extract differed significantly compared with acetone extract against Streptococcus ratti, Streptococcus criceti, Actinomyces viscosus, Actinomyces israelii and Aggregatibacter actinomycetemcomitans (p<0.05). These extracts exhibited MIC ranges of 2.63 to >10.50 mg/ml against the tested bacteria. The ethanol extract from Coriolus versicolor showed lower MIC values of 2.63 to 5.25 mg/ml. According to the obtained growth curve, the extracts of Coriolus versicolor were more effective against Actinomyces viscosus. Conclusions: The acetone, ethanol, and ethyl acetate extracts from Coriolus versicolor showed antimicrobial activities against Streptococcus mutans, Staphylococcus aureus, Streptococcus sanguinis, Streptococcus sobrinus, Streptococcus ratti, Streptococcus criceti, Aggregatibacter actinomycetemcomitans, Actinomyces viscosus, and Actinomyces israeli.i Therefore, they could be considered as natural oral antimicrobial agents against oral pathogens.

Optimization of Media Composition and Culture Conditions for the Mycelial Growth of Coriolus versicolor and Lentinus edodes (Coriolus versicolor와 Lentinus erodes의 영양배지 조성 및 배양조건의 최적화)

  • The media compositions and culture conditions were optimized for mycelial growth of Coriolus versicolor and Lentinus edodes. Media composition for optimal growth of Coriolus versicolor was 2.0% glucose 0.4% peptone and 0.6% yeast extract. Media composition for optimal growth of Lenttnus edodes was 2.0% glucose 2.0% starch 0.4% bacto-soytone and 0.6% yeast extract. The media supplemented with KH2PO4, 0.046% KH2PO4 0.1% and MgSO4, .7H2O 0.05% supported better mycelial growth than the media without mineral salts. The optimum temperature for mycelial growth ranged from $25^{\circ}C$-28$^{\circ}C$. The optimum pH range for mycelial growth of Coriolus versicolor was 5.2~5.6 while that of Lentinus edodes appeared to be 5.75.

  • Protoplast fusion between isoleucine-, argihine- and thymidine-requiring auxotroph $(Ile^{-}, Arg^{-}, Thy^{-})$ of Lentinula edodes and arginine-requiring auxotroph $(Arg^-)$ of Coriolus versicolor has been achieved using 30% polyethylene glycol (M.W.4000) in 10 mM $CaCl_2$-glycine solution (pH 8.0). Fusion hybrids were selected in the 0.6 M sucrose supplemented minimal media on the basis of nutritional complementation with fusion frequency of $7.4{\times}10{-6}$ The hybrids included both parental and non-parental types in colony morphology, growth rate and isozyme patterns. We succeeded inter-order protoplast fusion between the auxotrophs of Lentinula edodes and Coriolus versicolor overcoming the natural barriers of incompatibility. We examined the characteristics of the hybrids and clarified the fusion rocess using electron microscopy.

  • Present expriments were conducted to determine the possibility of cultivation of Coriolus versicolor selected among the higher fungi growing in Korea. Dried apple sawdust used in this study consisted of 31.5% C, 0.54% N, 2.41 % CaO, 0.05% $P_2O_5$, 0.48% MgO and 0.81% $K_2O$(pH 5.6). Coriolus versicolor mushrooms were cultivated on the sawdust media; apple sawdust; rice bran=80:20 in 850cc polypropylene bottles. The isolate of Coriolus versicolor used was YCV collected from an Apple farm in Youngchun district. It took 34 days to make fruitbody from spawning. The fruit bodies produced the total fresh weight 28 g in a bottles, and converted to 64% of fresh weight.

Generation of Nucler Hybrids Overcoming the Natural Barrier of Incompatibility: Transfer of Nuclei from Lentinula edodes into Protoplasts of Coriolus versicolor

  • Heterokaryotic nuclear hybrids overcoming the natural barriers of incompatibility have been studied in basidiomycetes. To produce these nuclear hybrids between incompatible mushrooms, which have several potent pharmacological effects, nuclear transfer was performed between Lentinula edodes and Coriolus versicolor. Nuclei from serine auxotrophs of Lentinula edodes, $LE207(Ser^{-})$ were transferred into the protoplasts of arginine auxotrophs of Coriolus versicolor, $CV17(Ser^{-})$using 30% polyethylene glycol 4000 in 10 mM $Cacl_{2}$-glycine solution (pH 8.0). Nulcear transfer progenies were selected by nutritional complementation on minimal media supplemented with 0.6 M sucrose. The progenies were classified based on colony morphology to L. edodes-like, C, versicolor-like and non-parental type. Most of the progenies grew slower than either parent. The number of nuclei per cell was similar but the DNA content varied between progenies. The isozyme patterns of nuclear hybrids resembled either of the parent porfiles or showed a mixed profile.

  • Coriolus versicolor, is one of the most popular medicinal mushrooms due its various biologically active components. This study was conducted to obtain basic information regarding the mycelial culture conditions of C. versicolor. Based on the culture, and MCM media were suitable for the mycelial growth of the mushroom. The optimum carbon and nitrogen sources were dextrin and yeast extract, respectively, and the optimum C/N ratio was 10 to 2 when 2% glucose was used. Other minor components required for optimal growth included thiamine-HCl and biotin as vitamins, succinic acid, lactic acid and citric acid as organic acids, as well as $MgSO_4{\cdot}7H_2O$ as mineral salts.

  • The purpose of the study was to verify if or not as funtional food by estimating antitumor activity and antimicrobial activity of Coriolus versicolor(Fr.)Quel and Ganoderma Lucidum (Fr.)Karst. 1. The contents of moisture. crude fiber and crude protein of GL were higher $(18.28\%,\;10.3\%,\;78.4\%)$ than that of CV, but the content of crude lipid GL was higher than that of CV. 2. Inhibitory effects of on the growth of AGS human gastric adenocarcinoma cells and HT-29 human colon adenocarcinoma cell were increased by increasing of concentration in added methanol extracts of CV and GL. 3. Antimicrobial activity on Escherichia col1; Staphylococcus aureus, Salmonella typhimurium existed methanol extracts of CV. Therefore. it is suggested that CV is worth as functional food.

  • The carpophores of three Korean mushrooms, Coriolus versicolor, Pleurotus ostreatus and Lentinus edodes were respectively extracted with hot water and the extracts were dialyzed through Visking tube. They were found to exert an antitumor activity against sarcoma-180 implanted in mice. Especially, the inhibition ratio of the extract of Coriolus versicolor (100 mg/ Kg.i.p) was almost 100% . But all the extracts did not affect the growth of leukemia L517Y cells in vitro. Therefore these facts indicate that the extracts appear to stimulate cell-mediated immunity.
