RNA accumulating strain of Torulopsis versatilis MT-1 was cultured in molasses medium for higher contents of RNA in cell. Yeast cells were harvested at logarithmic phase on synchronous culture. Yield of cells on dry base to input sugar was 59.5%. Crude protein content was 55.1% in cell. RNA content was 13.9%. Some problems found in the process for the preparation of yeast extracts were improved by the addition of culture broth of Streptomyces faecalis MSF which secrete RNase (5' nuclease and 5' adenylic acid deaminase). When the culture broth of S. faecalis MSF was added in autolysis process 46% of RNA in cell was converted to I and G(5' inosinic acid and 5' guanylic acid) in extract. By addition of 3-7% culture broth of S.faecalis MSF in autolysis or enzymolysis process at the start or early stage, RNA in extract was converted easily to I and G and protein in cells was easily extracted and hydrolyzed to amino acid. Taste of those yeast extracts was delicious. The yeasty smell in yeast extracts was removed. And cell debris was easily removed from extract.
The display industry is transitioning from traditional rigid products such as flat panel displays to flexible or wearable ones designed to be folded or rolled. Accordingly, colorless and optically transparent polyimide (CPI) films are one of the prime candidates to substitute traditional cover glass as a passivation layer to accommodate product flexibility. However, CPI films subjected to repetitive pure bending loads inevitably entail an accumulation of residual strain that can eventually cause wrinkles or delamination in the underlying component after a certain number of static and cyclic loading. The purpose of this study is to establish an experimental method to systematically evaluate the bending residual strain of CPI films. Films were monotonically and cyclically wrapped on mandrels of various diameters to ensure a constant strain in each. After unwrapping the wound CPI film, the residual radius of curvature remaining on the film was measured and converted into residual strain. The critical radius of curvature at which residual strain does not remain was about 5 mm, and the residual strain decreased in proportion to the log time. It is expected that flexible displays can be reliably designed using the data between the applied bending strain and the residual strain.
Estimation of correct ice load under various operating conditions is important during the design and the operation stages of an icebreaker. Normal operating conditions are expected from the official field ice trials and also from general ice transit action. In this paper ice load for the Korean icebreaking research vessel, ARAON, under normal operating condition, is discussed. Published ice load data from full-scale sea trials of six icebreakers were analysed to derive an empirical ice load prediction formula. The IBRV ARAON had sea ice trials during 2010 and 2012 summer season. Strain gauge signal were recorded during her icebreaking voyage and the measured strain data were converted to the equivalent hull stress values. The effect of ARAON's speed in ice and the hull stresses are investigated. By comparing the empirical formula and ice load calculation based von measured data, it is recommended to use the empirical ice load estimation formula for the initial design stage.
We discuss the experimental procedure for extracting reliable phase information from a defocus series of transmission electron microscopy (TEM) dark-field images using the transport of intensity equation (TIE). Taking InGaN/GaN multi-quantum well light-emitting diode as a model system, various factors affecting the final result of reconstructed phase such as TEM sample preparation, TEM imaging condition, image alignment, the correction of defocus values and the use of high frequency pass filter are evaluated. The obtained phase of wave function was converted to the geometric phase of the corresponding lattice planes, which was then used for the two-dimensional mapping of lattice strain following the dark-field inline holography (DIH) routine. The strain map obtained by DIH after optimized image processing is compared with that obtained by the geometric phase analysis of high resolution TEM (HRTEM) image, manifesting that DIH yields more accurate and reliable strain information than HRTEM-based GPA.

This study conducted four field measurements of local ice pressure during the icebreaking voyage of the icebreaking research vessel "ARAON" in the Chukchi and Beaufort seas from July to August of 2010. For measurements, 14 strain gauges, including 8 strain gauge rosettes, were set on the bow of the port side. Influence coefficients were determined using a finite element model of the instrumented area and they were used to convert the measured strains on the hull structure to local ice pressures. The converted maximum pressure was calculated as 2.12 MPa on an area of $0.28m^2$. Pressure-area curves were developed from the surveyed pressure data and the results were compared with previously measured data. The study results are expected to provide an understanding of local ice pressures and thus be useful in the structural design of ice class ships.

A mid-size RC test apparatus (MRCA) equipped with a program is developed that can test samples up to D=10 cm diameter and H=20 cm height which are larger than usual samples used in practice. Using the developed RC test apparatus, two types of crushed trackbed foundation materials were tested in order to get the shear modulus reduction curves of the materials with changing of shear strain levels. For comparison purpose, large repetitive triaxial compression tests (LRT) with samples of height H=60cm and diameter D=30 cm were performed also. Resilient modulus obtained from the LRT was converted to shear modulus by considering elastic theory and strain level conversion and were compared to shear modulus values from the MRCA. It is found from this study that the MRCA can be used to test the trackbed foundation materials properly. It is found also that strain levels of $E_{v2}$ mostly used in the field should be verified considering the shear modulus reduction curves and proper values of $E_{v2}$ of trackbed foundation must be used considering the strain level verified.
The primary function of the trackbed in a conventional railway track system is to decrease the stresses in the subgrade to be in an acceptable level. A properly designed trackbed layer performs this task adequately. Many design procedures have used assumed and/or are based on critical stiffness values of the layers obtained mostly in the field to calculate an appropriate thickness of the sublayers of the trackbed foundation. However, those stiffness values do not consider strain levels clearly and precisely in the layers. This study proposes a method of computation of stiffness that can handle with strain level in the layers of the trackbed foundation in order to provide properly selected design values of the stiffness of the layers. The shear modulus values are dependent on shear strain level so that the strain levels generated in the subgrade in the trackbed under wheel loading and below plate of Repeated Plate Bearing Test (RPBT) are investigated by finite element analysis program ABAQUS and PLAXIS programs. The strain levels generated in the subgrade from RPBT are compared to those values from RC (Resonant Column) test after some consideration of strain levels and stress consideration. For comparison of shear modulus G obtained from RC test and stiffness moduli $E_{v2}$ obtained from RPBT in the field, many numbers of mid-size RC tests in laboratory and RPBT in field were performed extensively. It was found in this study that there is a big difference in stiffness modulus when the converted $E_{v2}$ values were compared to those values of RC test. It is verified in this study that it is necessary to use precise and increased loading steps to construct nonlinear curves from RPBT in order to get correct $E_{v2}$ values in proper strain levels.
A new aniline-utilizing microorganism, strain HY1 obtained from an orchard soil, was characterized by using the BIOLOG system, an analysis of the total cellular fatty acids, and a 16S rDNA sequence. Strain HY1 was identified as a Burkholderia species, and was designated Burkholderia sp. HY1. GC and HPLC analyses revealed that Burkholderia sp. HY1 was able to degrade aniline to produce catechol, which was subsequently converted to cis,cis-muconic acid through an ortho-ring fission pathway under aerobic conditions. Strain HY1 exhibited a drastic reduction in the rate of aniline degradation when glucose was added to the aniline media. However, the addition of peptone or nitrate to the aniline media dramatically accelerated the rate of aniline degradation. A fatty acid analysis showed that strain HY1 was able to produce lipids 16:0 2OH, and 11 methyl 18:1 ${\omega}7c$ approximately 3.7-, 2.2-, and 6-fold more, respectively, when grown on aniline media than when grown on TSA. An analysison the alignment of a 1,435 bp fragment. A phylogenetic analysis of the 16S rDNA sequence based on a 1,420 bp multi-alignment sowed of the 16s rDNA sequence revealed that strain HY1 was very closely related to Burkholderia graminis with 95% similarity based that strain HY1 was placed among three major clonal types of $\beta$-Proteobacteria, including Burkholderia graminis, Burkholderia phenazinium, and Burkholderia glathei. The sequence GAT(C or G)${\b{G}}$, which is highly conserved in several locations in the 16S rDNA gene among the major clonal type strains of $\beta$-Proteobacteria, was frequently replaced with GAT(C or G)${\b{A}}$ in the 16S rDNA sequence from strain HY1.
A new strain of Pseudomonas aeruginosa growing in a rice field contaminated with herbicide and effluents of a factory manufacturing explosives was isolated. This isolate showed excellent growth in unusually high concentration of acrylamide (60 mM). It utilized acrylamide as the sole source of carbon and nitrogen for growth. Other amides such as acetamide, butyramide, isobutyramide, and methacrylamide were also utilized for the growth by this isolate. Acrylamide was degraded into acrylic acid and ammonia by the enzyme amidase. More than $65\%$ of added acrylamide (40 mM) was converted into acrylic acid after 40 h of growth of the culture. Amidase activity was inducible, the highest activity being observed with isobutyramide ($12.5{\mu}M$ ammonia/mg protein/min). These results demonstrate that this bacterium can degrade a variety of amides.
The buckling of two elastic layers bonded to a semi-infinite substrate under a transverse compressive plane strain is investigated. Incremental deformation theory is employed to describe the buckling behavior of both two isotropic layers and the semi-infinite substrate. The problem is converted to an eigenvalue-eigenvector case, from which the critical buckling strain and the wavelength of the buckled shape are obtained. The results are presented on the effects of the layer geometries and material properties on the buckling behavior.
