High Production of L-Threonine using Controlled Feeding of L-Methionine and Phosphate by Escherichia coli Mutant (L-Methionine과 Phosphate의 제한 공급에 의한 Escherichia coli MT201로부터의 고농도 L-Threonine 생산)

  • L-Threonine fermentation process was constructed on batch and fed-batch culture by using Escherichia coli MT201. The production type of L-threonine was observed as growth-associated production in batch culture. In fed-batch culture studying optimal concentration of yeast extract in feeding media, when 600 g/l of glucose and 60 g/l of yeast extract were added in feeding media, 87 g/$\ell$ of L-threonine was produced. To improve cell growth and L-threonine production, the culture of high cell density was performed in fed-batch culture with oxygen enriched air and feeding media containing L-methionine and phosphate. Under the conditions, we could achieve the highest L-threonine production of98 g/$\ell$ at 60 h. The highest productivity of L-threonine was about 3.85 g/$\ell$/h.

  • The present study examined the import routes of distilled rice liquor soju and how soju developed among the royal family and the upper classes using celadon inscribed with poetry related to alcohol beverages in the 12th century, Maebyeong style vases inscribed with government office name in charge of alcohol beverages of the royal family in the 14th century during the Koryo Dynasty. Distilled rice liquor was imported from the southwestern region to Koryo by Arabian merchants through direct and indirect routes in the Yuen Dynasty during the age of King Chungsuk and King Chunghye in around the 14th century. As soju was added to existing takju and cheongju, the three major types of alcohol beverages were completed during the late Koryo Dynasty. Celadon pitcher inscribed with poetry shows the delicate sentimentalism, aristocratic prosperity, and poetic sentiment. In particular, it is valuable in that it reflects Koryo people's mind, view of nature, and attitude toward alcohol beverages, and their inner world was also described with celadon patterns. Maebyeong style vases Yangonseo, Saonseo, Deokcheongo, Euiseonggo and Saseonseo, which are real celadon antiques inscribed with government office name, were used for rice liquor preservation. In particular, Maebyeong style vase has the exact year of creation, so it is a historically important celadon in research not only on alcohol food culture but also on art history. This shows that alcohol beverages were important foods that there were controlled and stored in celadon by the government offices for the royal family's related alcohol ceremonies. Through celadon inscribed with poetry and government office name displaying Koryo people's unique imagination and cultural consciousness, we can read their mind and lifestyle based on historical and social alcohol food culture in the Koryo Dynasty.

Inulae Flos Induces Cell Cycle Arrest and Autophagy of HT-1080 Human Fibrosarcoma Cells in 2D and 3D Cell Culture Model (선복화의 2D 및 3D 세포배양모델에서 HT-1080 인간 섬유육종 세포의 세포주기정지 및 자가포식 유도 효과)

  • Objective : Inulae Flos(IF) has been used to treat arthritis, sever furuncle, fear and palpitation, vomiting, stroke, asthma and cough in Korean Medicine. Although the anticancer activity of IF has been reported, the molecular mechanism is still not well understood. In this study, we investigated the growth inhibitory activity of an ethanol extract of IF in HT-1080 human fibrosarcoma cells and its underlying mechanisms using two-dimensional (2D) and three-dimensional (3D) cell culture system. Methods : HT-1080 cells were cultured with IF for 9 days in 3D cell culture. To check an inhibition of cell prolifelation by IF, MTT assay was performed. DNA contents were measured using flow cytometry. Western blotting was used to evaluate the regulation of cell cycle- and autophagy-related proteins. Acridine orange staining was performed to confirm autophagy, and DCF-DA staining was performed to confirm the occurrence of ROS. Results : IF controlled a spheroid formation and decreased a cell viability in 3D cell culture. IF-induced cell proliferation inhibition was associated with a distinct increase of S and G2/M phase cell distribution in 2D cell cultre. In addition, IF significantly induced autophagy and generated reactive oxygen species(ROS). Interestingly, IF-induced cell cycle arrest and autophagy were recovered after pre-treatment of N-acetyl-L-cysteine, ROS scavenger. Conclusion : Our results indicate that IF induced ROS-mediated cell cycle arrest and autophagy and it may potentially useful for human fibrosarcoma treatment.

Cultural Backgrounds of Reviewers, Negative Emotions, and Review Helpulness: Based on Impression Management Theory (리뷰어의 문화적 배경, 부정적 감정표현, 그리고 리뷰유용성: 인상관리 이론을 중심으로)

  • Online consumer reviews provide a variety of information from the customer perspective in terms of satisfaction and dissatisfaction. Negative emotional expression is a potential antecedent of review usefulness, and can also influence potential consumers' attitudes and decisions. In addition, because national culture provides a perspective from which individuals view the world and act, it is highly likely that differences in negative emotional expression will occur depending on culture. This study explores the relationship between national culture and negative emotional expression based on impression management theory and ultimately analyzes the impact on review usefulness. For empirical analysis, 16,076 reviews of 140 hotels located in Seoul were collected and analyzed using the PLS-SEM method. As a result of the analysis, it was found that power distance and masculinity culture dimensions had a positive effect on reviewers' negative emotional expressions, while uncertainty avoidance and long-term orientation had a negative effect. In addition, negative emotional expression was analyzed to have a positive effect on review usefulness even when review ratings were controlled.

  • The experiment was carried out to investigate the productivity of freshwater rotifer, Brachionus calyciflorus and marine rotifer, B. rotundiformis at various temperatures, initial inoculation and pH in a 5 L semi-continuous high density culture. Rotifers were fed by commercial condensed freshwater Chlorella. When pH was not controlled, average daily productions of freshwater and marine rotifers increased with temperature. The highest production, $44\times10^6$ rotifers, was achieved of B. calyciflorus at $32^{\circ}C$ and the possible production lasting period of B. calyciflorus was shorter than that of B. rotundiformis. Under the adjustment of pH at 7, the possible production lasting periods of B. calyciflorus and B. rotundiformis inoculated with 5,000 inds./mL were longer than those of rotifers inoculated with 10,000 inds./mL, and the daily production rate of the former was higher than that of the latter. The results from this study indicated that optimum density of the initial inoculation for the cultivation of B. calyciflorus and B. rotundiformis was 5,000 inds,/mL under the controlled conditions of pH 7 and at $32^{\circ}C$ in a semi-continuous high density culture, in terms of production rate and food cost aspects.

Isolation and Characterization of Hydrogen Producing Bacterium (수소생산균 Enterobacter cloacae YJ -1의 분리 및 특성)

  • The hydrogen-producing bacterium was isolated from fresh water and identified as Enterobacter cloacae. The isolated was named Enterobacter cloacae YJ-1. In batch culture, The optimum cultivation temperature and pH of strain YJ-1 was 35℃ and 7.5, respectively. All of the added glucose was consumed completely during fermentation even though pH was not controlled. Amount of hydrogen produced on each condition of 2% glucose, 4% sucrose and 5% fructose was 950, 1000 and 948 mL/L, respectively and resulted in increasing hydrogen production approximately 2.5-times more than controlled condition. The maximum hydrogen production was obtained when 50 mM phosphate was added. In repeated-batch culture, hydrogen gas of 1920 mL/L was totally produced for 48. The maximum hydrogen was produced on the condition of 0.5% yeast extract, but the production amount was not changed on the condition of over 0.5%. Most of the organic acids produced during the fermentation were formic and acetic acid, and propionic acid was moiety also generated.

Analyses of Transpiration and Growth of Paprika (Capsicum annuum L.) as Affected by Moisture Content of Growing Medium in Rockwool Culture

  • Since the moisture content (MC) of growing medium closely related with the crop transpiration, the MC should be included to the environmental factors to be considered for irrigation control in soilless culture. The objective of this study was to analyze the transpiration of paprika plants using daily mean solar radiation (RAD) and vapor pressure deficit (VPD) as well as the growth of the plants at different MCs of rockwool growing media. The starting points of irrigation were controlled by a moisture sensor with minimum set points of 40%, 50%, and 60% of MCs. The canopy transpirations were measured for 80 to 120 days after transplanting and analyzed. The transpirations were well regressed with a combination of both RAD and VPD rather than daily mean RAD only under the controlled MCs. The transpiration at 60% MC was higher than those at 50% and 40% MCs. Leaf area, leaf fresh and dry weights at 60% MC were higher than those at 50% and 40% MCs while the number of leaves had no significant difference among the MCs. There were no significant differences in number of fruits and fruit size among all the MCs, while fruit weight was significantly lower at 40% MC than other treatments. Fresh and dry fruit yields were the highest at 60% MC. Therefore it was concluded that the transpiration was affected by the MC of rockwool growing medium and the minimum set point of 50-60% MC of rockwool growing medium gave better effects on the growth of the paprika plants.

The Analysis of Research Trends on Forest Therapy in the Korean Journal (산림치유 연구의 국내동향 분석)

  • Objectives The purpose of this study is to understand the research trend of reports on forest therapy so far and analyze the Korean medicine therapy being applied in forest therapy programs. Methods We ran a keyword search on domestic databases with the following keyword 'forest therapy, forest healing, forest treatment, recreational forest, forest bath, forest experience'. The search took place in December 2014 and there was no limit to search time. A total of 334 forest therapy articles have been selected. Results The number of research on forest therapy continued to rise from 1985, with 334 articles being published from 84 journals. When those 188 articles were sorted by their contents and methods, except 146 articles of survey on simple satisfaction, recognition and visting, 94 were clinical studies, 79 were literature studies, 15 were experimental studies. Of the 94 clinical researches, there were 52 CCTs (Controled Clinical Trials), 39 ODs (efficacy studies with either a controlled or an Other than controlled Design) and 3 RCTs (Randomized Clinical Trials). Among the clinical researches, there were a total of 21 studies that used Korean Medicine programs, and meditation was the most popular, being used in 18 studies. Herbal food and tea therapy and Qigong were used in 3 studies each, and Korean medicine music programs were used in 2 studies. Conclusions A systematic and standardized Korean medicine forest therapy program must be developed, and based on the program, more research treating diseases should be conducted.

Study on the effect of self-improvement programs in military (군에서의 자기계발 프로그램의 효과성에 관한 연구)

  • Recently the military is improving the policies of military culture for soldiers to lead a better military life. In this effect, 'division A' introduced 'goal-directed self development program'. This program is guiding the soldiers to set up personal mission in life not only in the military but also linked to the future, after being discharged from military service. Therefore the purpose of this research is to test the effect of this program on soldier's objective management, self esteem and satisfaction and to propose political strategies for creating healthier military culture. The research method was to compare and analyze the difference between two groups, 'division A'(experimental group), which operated the program, and 'division B'(controlled group) which did not operated the program. research targets were 488 soldiers in total, 257 soldiers in experimental group and 231 in controlled group. T-test(verification) and analysis of covariance was done with SPSS WIN 11.5 program with collected information. The result showed that the program has meaningful effect in every aspect mentioned above to the soldiers.

Dual Mode Feedback-Controlled Cycling System for Upper Limb Rehabilitation of Children with Cerebral Palsy

  • Background/Objectives: This paper proposes a dual mode feedback-controlled cycling system for children with spastic cerebral palsy to rehabilitate upper extremities. Repetitive upper limb exercise in this therapy aims to both reduce and analyze the abnormal torque patterns of arm movements in three- dimensional space. Methods/Statistical analysis: We designed an exercycle robot which consists of a BLDC motor, a torque sensor, a bevel gear and bearings. Mechanical structures are customized for children of age between 7~13 years old and induces reaching and pulling task in a symmetric circulation. The shafts and external frames were designed and printed using 3D printer. While the child performs active/passive exercise, angular position, angular velocity, and relative torque of the pedal shaft are measured and displayed in real time. Findings: Experiment was designed to observe the features of a cerebral palsy child's exercise. Two children with bilateral spastic cerebral palsy participated in the experiment and conducted an active exercise at normal speed for 3 sets, 15 seconds for each. As the pedal reached 90 degrees and 270 degrees, the subject showed minimum torque, in which the child showed difficulty in the pulling task of the cycle. The passive exercise assisted the child to maintain a relatively constant torque while visually observing the movement patterns. Using two types of exercise enabled the child to overcome the abnormal torque measured in the active data by performing the passive exercise. Thus, this system has advantage not only in allowing the child to perform the difficult task, which may contribute in improving the muscle strength and endurance and reducing the spasticity but also provide customizable system according to the child's motion characteristic. Improvements/Applications: Further study is needed to observe how passive exercise influences the movement characteristics of an active motion and how customized experiment settings can optimize the effect of pediatric rehabilitation for spastic cerebral palsy.
