Growth of Tomato and Pepper Grafted Plug Seedlings under Different Shading Condition During Acclimatization after Graft-taking (접목활착 후 순화시 차광조건에 따른 토마토와 고추 접목묘의 생육)

  • Acclimatization after grafting and graft-take that in order to produce plug seedlings of high-quality are important plug seedling stage which not reduce the plug seedlings quality before shipment. Appropriate acclimatization environment can not only increase seedling quality before secondary growth period, but also effective in promoting the growth of plug seedlings. This study was conducted to determine the optimal the shading period and shading level for acclimatization of tomato and pepper grafted plug seedlings after graft-take. The tomato and pepper seedlings were used in this experiment, and after graft-take, a tunnel was installed on a bed in glasshouse of venlo type to started shading treatment. The shading levels were 35, 55, 75 and 95%, and the shading periods were 1 and 2 weeks, and non-treatment was set as the control. In the case of tomato grafted plug seedling, plant height, stem diameter, dry weight of root, leaf area were significantly higher at the shading period of 1 week and the shading level was 55%. In the case of pepper grafted plug seedling, plant height, stem diameter, and leaf area were the highest in the shading period of 2 weeks and the shading level was 35%. However, dry weight of root, compactness, and T/R ratio, which seedling quality indicators, were lower than in the shading period of 1 week and the shading level 55%. Therefore, considering the quality of seedlings, it is suggested that shading period of 1 week with shading level of 55% treatment can be suitable to produce high quality grafted plug seedlings of tomato and pepper.

Isolation of a Quinone-rich Fraction from Ardisia crispa Roots and its Attenuating Effects on Murine Skin Tumorigenesis

  • Ardisia crispa (Family: Myrsinaceae) is an evergreen, fruiting shrub that has been traditionally used as folklore medicine. Despite a scarcity of research publications, we have succeeded in showing suppressive effects on murine skin papillomagenesis. In extension, the present research was aimed at determining the effect of a quinone-rich fraction (QRF) isolated from the same root hexane extract on both initiation and promotion stages of carcinogenesis, at the selected dose of 30 mg/kg. Mice (groups I-IV) were initiated with a single dose of 7,12-dimethylbenz(${\alpha}$)anthracene (DMBA, $100{\mu}g/100{\mu}l$) followed by repeated promotion of croton oil (1%) twice weekly for 20 weeks. In addition, group I (anti-initiation) received QRF 7 days before and after DMBA; group II (anti-promotion) received QRF 30 minutes before each croton oil application; group III (anti-initiation/promotion) was treated with QRF as a combination of group I and II. A further two groups served as vehicle control (group V) and treated control (group VI). As carcinogen control, group IV showed the highest tumor volume ($8.79{\pm}5.44$) and tumor burden ($3.60{\pm}1.17$). Comparatively, group III revealed only 20% of tumor incidence, tumor burden ($3.00{\pm}1.00$) and tumor volume ($2.40{\pm}1.12$), which were significantly different from group IV. Group II also showed significant reduction of tumor volume (3.11), tumor burden (3.00) and tumor incidence (11.11%), along with prominent increase of latency period of tumor formation (week 12). Group I, nonetheless, demonstrated marked increment of tumor incidence by 40% with prompted latency period of tumor formation (week 7). No tumor formation was observed in groups V and VI. This study provided clear evidence of inhibitory effects of QRF during promotion period which was in agreement with our previous findings. The mechanism(s) underlying such effects have yet to be elucidated.

Influence of Ionophore Supplementation on Growth Performance, Dietary Energetics and Carcass Characteristics in Finishing Cattle during Period of Heat Stress

  • Forty-eight crossbred heifers ($378.1{\pm}18$ kg) were used in a 56-d feeding trial (four pens per treatment in a randomised complete block design) to evaluate the influence of ionophore supplementation on growth performance, dietary energetics and carcass characteristics in finishing cattle during a period of heat stress. Heifers were fed a diet based on steam-flaked corn (2.22 Mcal $NE_m/kg$) with and without an ionophore. Treatments were: i) control, no ionophore; ii) 30 mg/kg monensin sodium (RUM30); iii) 20 mg/kg lasalocid sodium (BOV20), and iv) 30 mg/kg lasalocid sodium (BOV30). Both dry matter intake (DMI) and climatic variables were measured daily and the temperature humidity index (THI) was estimated. The maximum THI during the study averaged 93, while the minimum was 70 (THI average = $79.2{\pm}2.3$). Compared to controls, monensin supplementation did not influence average daily gain, the estimated NE value of the diet, or observed-to-expected DMI, but tended (p = 0.07) to increase (4.8%) gain to feed. Compared to controls, the group fed BOV30 increased ($p{\leq}0.03$) daily gain (11.8%), gain to feed (8.3%), net energy of the diet (5%), and observed-to-expected DMI (5.2%). Daily weight gain was greater (7.6%, p = 0.05) for heifers fed BOV30 than for heifers fed MON30. Otherwise, differences between the two treatments in DMI, gain to feed, and dietary NE were not statistically significant (p>0.11). Plotting weekly intakes versus THI, observed intake of controls was greater (p<0.05) at THI values ${\leq}77$ than ionophore groups. When THI values were greater than 79, DMI of control and MON30 were not different (p = 0.42), although less than that of groups fed lasalocid (p = 0.04). Variation in energy intake was lower (p>0.05) in the ionophores group (CV = 1.7%) than in the control group (CV = 4.5%). Inclusion of ionophores in the diet resulted in relatively minor changes in carcass characteristics. It is concluded that ionophore supplementation did not exacerbate the decline of DM intake in heat-stressed cattle fed a high-energy finishing diet; on the contrary, it stabilised feed intake and favoured feed efficiency. Ionophore supplementation reduced estimated maintenance coefficients around 10% in finishing cattle during a period of heat stress. This effect was greatest for heifers supplemented with 30 mg lasalocid/kg of diet.

  • A long-term (160 days) feeding experiment was conducted to evaluate effects of dietary soybean meal (SBM) on growth of rainbow trout (Oncorhynchus mykiss). Fish fed diets containing up to $46\%$ of SBM showed the same growth rates with control during the first 60 days (p>0.05). However, growth showed a gradual decrease with the culture period. At the end of 160-days experiment, fish fed diets with above $34\%$ of SBM showed a decrease in growth rate compared to the control group. Fatness of experimental fish was reduced with an increase in dietary SBM levels and in feeding period. Hepatosomatic index was significantly lower from fish fed the diets containing above $46\%$ of SBM than that from fish fed the control diet throughout the 160 days of experimental period, and fish fed diet containing above $46\%$ of SBM appeared to make restricted utilization of carbohydrate sources. These results may suggest that soybean meal could be used up to $22\%$ in trout diet as a substitute for fish meal without unfavourable side effects.

Effects of Dietary $\omega$-Fatty Acids on Fatty Acids Composition and Storage Characteristics of Meats from Spent Hens (오메가 계열 지방산 급여가 산란노폐계육의 지방산 조성 및 저장성에 미치는 영향)

  • This study was conducted to investigate the influence of dietary $\omega$-fatty acids on fatty acids composition and storage characteristics in meat samples of spent hens. Spent hens were randomly assigned to one of the four dietary treatments : 1) Control (commercial feed), 2) T1 (commercial feed supplemented with 10% olive oil, 3) T2 (commercial feed with 10% canola oil), and 4) T3 (commercial feed with 10% sardine oil). They were fed one of the experimental diets for three weeks and slaughtered. The meat samples were stored at 4 ${\pm}$ 1 ˚C The storage characteristics and fatty acid contents were analyzed for meat samples stored over a period of 1, 3, 5, 7 and 9 days. The pH of all treatments significantly increased during the storage periods(P<0.05). The per-oxide values (POV) of all treatments were significantly increased up to 5 days ; after that the POV decreased significantly (P<0.05). The POV of treated groups were significantly higher than those of the Control in the thigh meat(P<0.05). The TBARS showed the highest POV among all treatments. Although the breast meat tended to he lower in POV than the thigh meat, no significant difference was detected between the two meats. The TBARS(thiobarbituric acid reactive substances) of all treatments were significantly increased as the storage period extended(P<0.05). After 3 days, the TB ARS of oil-treated groups were aignificantly higher than that of the Control (P<0.05). The T3 showed the highest TBARS among all treatments (P<0.05). The TBARS of the breast meat was lower than that of the thigh, but no statistical difference was found be- tween them. The unsaturated fatty acid content of the breast and thigh meats in all treatments were slightly decreased as the storage period extended. The oleic acid was higher in Ti, and the linoleic acid and linolenic acid were higher in T2 than the other treatments. The eicosapentaenoic acid and docosahexaenoic acid were higher in T3 than the other treatments. The unsaturated fatty acid contents of the breast meat were slightly lower than those of the thigh meat. The n-6 fatty acid contents of the breast and thigh meats were slightly increased as the storage periods ex-tended. The n-3 fatty acid content of T3 was the highest among all treatments. The n-6 fatty acid content of breast meat was lower than that of thigh meat. The n-6 fatty acid content of the breast meat was slightly lower than that of the thigh meat.

Production of Mozzarella Cheese Analogue by Ultrafiltration (한외여과를 이용한 Mozzarella Cheese Analogue 제조에 관한 연구)

  • The purpose of this study was to improve the production of Mozzarella cheese analogues manufactured using mixtures of soy milk and concentrated raw milk by performing ultrafiltration (UF) and to assess the quality of these cheeses during a 30-day storage period at $4^{\circ}C$, relative to that of Mozzarella cheese manufactured with the traditional method. The solid consistency of Mozzarella cheese analogue prepared from milk mixtures was lower than that of cheese manufactured from raw milk or soy milk and increased during storage, which is considered to be the result of decreasing water levels, as well as with increasing soy milk concentrations. In the Mozzarella cheese analogue generated using the milk mixtures, the fat content decreased with increase in the soy milk concentration, while it decreased during the storage period. Lactose levels were lowest in cheese composed of soy milk or raw milk and processed by UF, and decreased during storage in cheese produced using milk mixtures. In milk mixtures containing soy milk, the protein concentration increased with increasing amounts of raw milk and did not change during the storage period. The water-soluble nitrogen compound level was similar between cheeses and increased only slightly during storage. The amount of non-protein nitrogen compounds was higher in the cheese analogue than in the control cheese and tended to increase during storage. Analysis of the physicochemical traits of the Mozzarella cheese analogue yielded the following results: During storage, titratable acidity levels increased while pH tended to decrease. After analysis using electropherograms, it was classified as ${\alpha}$-, ${\beta}$-, or ${\kappa}$-casein. The results of rheometry tests showed that in the Mozzarella cheese analogue prepared from milk mixtures, with raw milk concentrated by UF, increases in concentration rate lead to lowered hardness, elasticity, cohesiveness, and brittleness. When cheese was produced from milk mixtures and concentrated by UF, meltability increased as the concentration rate increased, although to an extent that was less than that observed for the control cheese, and tended to increase during storage. Sensory evaluation showed that the analogue cheese was much better than the control cheese in terms of formation, appearance, and flavor.

Antimicrobial and clinical effects of mouthrinses of acid water prepared by an electrolysis apparatus on chronic periodontitis (산화전위수 양치용액 사용이 만성 치주질환에 미치는 효과에 대한 연구)

  • The purpose of this study was to assess the antimicrobial and clinical effects of acid water mouthrinse prepared by an electrolysis apparatus on chronic periodontitis and to evaluate the lasting period of these effects. The change' in the pattern of colonization of bacteria within the subgingival pockets was monitored by phase contrast microscopy, in 40 patients, over a period of 8 weeks. In addition, changes in the clinical parameters of the diseased sites were also monitored. Site of pocket ${\geq}$ 5mm was selected in each patient randomly divided into two groups. As a test group, acid water mouthrinse was used twice a day in 20 patients. As control, no mouthrinse was used in 20 patients The results were as follows : 1. The suppression of motile bacteria was maintained for up to 3-4 weeks at test group. 2. Two groups did not differ significantly in proportion of bacteria in subgingival plaque over a period of 8 weeks. 3. Loss of attachment showed a significant difference in test group and in test group compared with control group (P<0.05), but there was no significant difference in control group. 4. No statistical difference was shown in two groups concerning the gingival index, plaque index, bleeding index. The results suggest that acid water mouthrinse is effective for reducing subgingival bacteria. It can be concluded that acid water may be useful as an mouthrinsing agent.

The Effect of Soybean Galactooligosaccharides on Nutrient and Energy Digestibility and Digesta Transit Time in Weanling Piglets

  • Eight $12.4{\pm}0.6kg$ initial body weight crossbred barrows were used to determine the effect of soybean galactooligosaccharides on nutrient and energy digestibility, and digesta transit time. Four dietary treatments were utilized in this trial. Treatment one was a corn-soybean meal based diet (SBM) containing raffinose and stachyose at the levels of 0.16% and 0.75%, respectively. Treatment two (control) was a corn-HP300 (soybean concentrate protein) diet. In treatments three and four, 1.1% and 2.2% commercial stachyose was added to the control diet to provide total dietary stachyose at the levels of 1% and 2%, respectively. The soybean galactooligosaccharides (raffinose + stachyose) level in treatment one was slightly lower compared to that in treatment three. Three collection periods were run with two pigs for each treatment/period. There was a 4 d adjustment period followed by a 3 d collection period. The results showed that the nitrogen retention (86.79%) of pigs fed treatment two diet was higher than that of pigs fed treatment one by 5.2% (p<0.05). The nitrogen retention of treatment three was intermediate 83.09%. The apparent fecal digestibility of all amino acids in treatment two was numerically highest, followed by treatments three and four. However, there were no significant difference among groups (p>0.05). The dry matter (DM), organic matter (OM), crude protein (CP), and crude fiber (CF) digestibility numerically decreased as the soybean galactooligosaccharides level increased, but were not significantly different (p>0.05). Chromium content in feces (from the inclusion of 0.3% chromic oxide in the diets) differed among treatments (p<0.05) at 15 h, 18 h, and 21 h after eating. This showed that the digesta transit time was differed significantly among treatments. Treatment four was the shortest, followed by treatment three, SBM and control. The results demonstrated that in the absence of antinutritional factors and soybean antigen protein, inclusion of 1% and 2% stachyose in corn-HP300 diet has no significant effect on the digestibility of DM, OM, CP, CF and amino acids. When the soybean galactooligosaccharide level in diet one and diet three were adjusted to be almost the same, antinutritional factors such as trypsin inhibitor and soybean antigen protein could decrease the nutrient digestibility and nitrogen retention rate of diet. High levels of soybean galactooligosaccharides shortened the digesta transit time in the intestinal tract. This trial suggested that the total level of soybean galactooligosaccharides (stachyose+raffinose) in the weanling piglet diet is better not to exceed 1% when common soybean meal is used as main protein source.

Investigation of Biological Characteristics of Rice Stem Borer (Chilo suppressalis Walker) and Variation in its Occurrence Patterns Among Crops (작물별 이화명나방의 발생양상 변동과 생물적 특성 연구)

  • We analyzed the variation in occurrence time and intensity of attacks by the rice stem borer (Chilo suppressalis Walker) on several crops. This study aimed to understand how C. suppressalis damages crops and to utilize this basic data for the establishment of environment-friendly control methods like pheromone traps. This study surveyed the changes in occurrence patterns of C. suppressalis, biological characteristics of overwintering larvae, and the efficacy of different types of pheromone traps the years. We found similar occurrence patterns of C. suppressalis in different crops. In addition, occurrence time of the first generation was advanced. Overwintering larvae showed no difference in pupal period and weight compared to the non-overwintering ones. However, the larval period was reduced to 19.1 days in Miscanthus field, in contrast to that in the paddy fields. It was confirmed that larvae of C. suppressalis generally prefer the lower part of the stems of Miscanthus. Efficiency of the emergence trap was confirmed to be greater than that of the delta trap for capturing C. suppressalis adults. However, it is necessary to adjust the control period because of the advancement in occurring time of C. suppressalis in recent years. The larvae of C. suppressalis experience favorable environmental conditions for overwintering in Miscanthus fields. The major Miscanthus fields are generally located in the areas protected for sources of drinking water, owing to which spraying of chemical pesticides is very limited. The results of this study provide important inputs for the development of environment-friendly control methods.

Changes in Glucosinolate Component Content in Urine After Ingestion of Fresh and Cooked Broccoli (신선한 브로콜리와 조리된 브로콜리 섭취 후 소변으로 배설되는 Glucosinolates 대사물질의 함량 변화)

  • Sulforphane (SF) is a family of biologically active compound that is distributed widely in broccoli. Although studies in rodents have shown that these compounds are effective and versatile inhibitors of tumorigenesis, the role of dietary SF in protection against human cancers remains to be established. The objective of this study was to explore the quantitative relationship between the dietary intake of cruciferous vegetables and urinary excretion of SF. The effects of dietary broccoli on the body's ability to detoxify were studied in six male subjects between the ages of 22~30 years. Study included administering a glucosinolate-free diet for 8 days (control period). The broccoli diet was further subdivided into two periods; 250 g broccoli was fed per day during the first three days and 500 g broccoli was fed per day during the latter three days. After an 8-day washout period, a second experiment was conducted. The same protocol was used with the exception that uncooked broccoli was consumed. Urinary SF mercapturate was measured to determine the bioavailability of broccoli. The linear trend for mercapturate excretion was dose-dependent, resulting in 3.8- and 1.9-fold increase by the third and six days, respectively, compared to the control. Lower amount of SF-NAC conjugate was detected in cooked broccoli compared to fresh broccoli suggesting cooking may have caused a significant loss in glucosinolates in cruciferous vegetables. Therefore, SF can be used as a biomarker for intake of cruciferous vegetables.
