Effects of Bopheyangyoungjeon(BYJ) on airway smooth muscle, airway inflammation, IgE and Interleukin-4 in mouse model of allergic bronchial asthma (보폐양영전(保肺養營煎)이 흰쥐의 기관지평활근(氣管支平滑筋) 장력(張力)과 면역에 미치는 영향(影響))

  • It has been reported that Bopheyangyoungjeon(BYJ) has an effect on deficiency asthma(喘虛) clinically. The aim of this study was to determine an appropriate dosage of BYJ to treat asthma. In order to study the effects of orally administered BYJ on allergic asthma, mice were pretreated with three oral doses of the herbal solution of BYJ before antigen sensitization. 2 days later Mice were actively sensitized with a subcutaneous injection of ovalbumin and 13 day later ovalbumin aerosols were used to provoke asthmatic reaction. Serum level of IgE, IL-4, WBC, RBC, HGB, cell numbers in bronchoalveolar lavage fluid(BALF), and in vitro isometric contractile responses of the isolated tracheal smooth muscle(TSM) to acetylcholine(ACh, 0.1-1000uM), KCl were measured. The results were as follows ; 1. Contractile responses of TSM to ACh significantly increased in C group at Ach 0.3, 1, 3, 10, 30, 100, 300, 1000uM(P<0.05, P<0.01) and increased in D at 0.1, 0.3, 3, 30, 30, 100, 300, 1000uM. 2. The sensitivity of TSM to Ach increased more in A, B group, but it was not significant. 3. The maximal contractile response of TSM to ACh decreased more significantly in C group(P<0.01) and D group(P<0.05) the control group. 4. The maximal contractile response of TSM to KCI decreased more significantly in B group and C group(P<0.001) than in the control group. 5. The counts of lymphocytes in BALF decreased more significantly in B group and D group(P<0.05) than in the control group. 6. The counts of macrophages in BALF decreased more significantly in B group, C(P<0.05) than in the control group. 8. Serum IgE level increased more significantly in B group and C group(P<0.05) than the control group. 9. The counts of WBC, RBC, HGB in blood increased more significantly in A group than the control group. The above results support a role for BYJ orally administered in treatment of deficiency allergic Asthma.

  • Objectives : It has been reported that Cordyceps militaris Mycelia(CMM) has an effect on deficiency allergic asthma(虛喘) clinically. The aim of this study was to determine an appropriate oriental treatment and the doses of CMM to treat asthma. Methods : In order to study the effect of herbal acupuncture solution of the CMM on allergic asthma, the mouse were pretreated by CMM herbal acupuncture at BL13, LU4 before antigen sensitization. 2 days later Mice were actively sensitized with a subcutaneous injection of ovalbumin(OA) and 13 days later they were provoked with OA aerosols. IL-4, lymphocyte, macrophage in bronchoalveolar lavage fluid(BALF), IgE in serum, WBC, RBC, HGB in blood, and in vitro isometric contractile responses of the isolated tracheal smooth muscle(TSM) to acetylcholine$(ACh,\;0.1-1000\;{\mu}M)$, KCl were measured. Results : Contractile responses of TSM to ACh were significantly increased in CMM herbal acupuncture 1 group $(Ach\;1000\;{\mu}M)$, CMM herbal acupuncture 2 group $(ACh\;1,\;10\;{\mu}M)$, CMM herbal acupuncture 3 group $(Ach\;0.3,\;1,\;30,\;300\;{\mu}M)$. The sensitivity of TSM to ACh was significantly decreased in CMM herbal acupuncture 3 group. The maximal contractile response of TSM to ACh was significantly decreased in CMM herbal acupuncture 1, 3 group. The maximal contractile response of TSM to KCl was significantly decreased in CMM herbal acupuncture 1, 2, 3 group. The counts of lymphocytes in BALF was significantly increased in CMM herbal acupuncture 3 group. The counts of macrophages in BALF was significantly decreased in CMM herbal acupuncture 3 group. Interleukin-4 level in BALF was significantly increased in CMM herbal acupuncture 1,3 group. and it was increased in CMM herbal acupuncture 2 group, but there was no significance. Serum IgE level was significantly decreased in CMM herbal acupuncture 1, 2, 3 group. The counts of WBC in blood was significantly increased in CMM herbal acupuncture 1, 3 group Conclusion : Based on the above results it is assumed that CMM herbal acupuncture at BL13, LU4 can help the treatment of deficiency allergic Asthm.

Studies on Digitalis Receptor Desensitization in Rat Ventricle

  • $^3$H〕Ouabain binding parameters(K$\_$D/ and B$\_$max/,) in homogenates prepared fpom control rat ventricular strip and Langendorff preparations which were not previously exposed to ouabain were compared to those in homogenates from ventricular strip and Langendorff preparations that had been first exposed to a complete ouabain dose-response curve(10$\^$-7/M to 10$\^$-4/ M). In rat ventricular strips and Langendorff perfused rat heart preparations, cumulative dose-response cruves of ouabain revealed biphasic positive inotropic effects, a "low-dose" and a "high-dose" effect with ED$\_$50/ values of 0.5${\mu}$M and 35${\mu}$M ouabain, respectively- The "low-dose" effect in rat ventricular strips disappeared or was diminished significantly when the ouabain dose-response curve wag repeated after the washout of the effects of the first curve, whereas the maximal "high-dose" effect was identical in both exposures to oubain. However, there was no change in the "low-dose" effects in both sets of the Langendorff perfused hearts. The contractile activity of the pre-exposed strips did not indicate the presence of residual ouabain since their basal contractile force was decreased 10% compared to initial control. 〔$^3$H〕Ouabain binding parameters, K$\_$D/ and B$\_$max/, were not changed comparing homogenate of control ventricular strips with that of strips pre-exposed to ouabain. These results suggest that digitalis receptor desensitization in the rat ventricular strip may due to the change of post-receptor events induced by ouabain binding to a high affinity site(${\alpha}$$_2$ isoform).

  • Superprecipitation of actomyosin has been considered to be an in vitro model of the muscle contraction. The superprecipitation and ATPase activity (which supplies the energy for contraction) are influenced by several factors which are the large amount of changes in ionic strength, Mg and ATP concentrations. But those behaviors are found to be promptly influenced by the change in a small range of calcium concentration which can be controlled by the cellular function of muscle physiologically only in the presence of the modullatory proteins, tropomyosin and troponin. In order to elucidate the precise roles of calcium in the muscle contraction and relaxation, the effects of calcium on the actin- myosin interaction was observed in the presence of tropomyosin and troponin using the superprecipitation system. The results are summarized as follows: 1. EGTA (glycol ether diaminetetraacetic acid)prolonged the initiation of the superprecipitation of natural actomyosin. 2. Superprecipitation curve was declined by adding EGTA at the time when tile curve reached the half- maximum. The degree of declining was proportional to the amount of EGTA added. Especially, upon adding 0.25 mM EGTA the curve was lowered to the level before the protein superprecipitated. But addition of EGTA did not affect the curve after attaining the maximum. 3. Superprecipitation of Perry myosin B was not affected by EGTA added both before and during the course of the reaction. 4. Tropomyosin did not change the response of Perry myosin B to EGTA added at any time of the reaction. 5. Troponin also did not change the response of Perry myosin B to EGTA. 6. Both tropomyosin and troponin together rendered the Perry myosin B to obtain the same response as natural actomyosin to EGTA. 7. It was concluded that actin-myosin interaction was influenced by the minute change of calcium concentration only in the presence of both tropomyosin and troponin. We could reproduce the contraction and relaxation of the muscle in vitro under the presence of ATP by changing the calcium concentration.

  • The purpose oi this study is to identify clearly the physiologic significance of autonomic nervous system. This study is to find the loose of endogenous neurotransmitter while using the neural response of the neural excitatory action which is distributed to the perivascular smooth muscle through the electrical stimulation of the smooth muscle of coronary artery of pig. The effects of perivascular nerve stimulation were investigated on isolated coronary artery of pig.1 . The magnitude of contractile response to perivascular nerve stimulation increased with increasing frequency (2-80 Hz) of stimulation. 2. The contractions to perivascular nerve stimulation(40V, 40Hz. 0.5msec, 1 min) were increased greatly by pretreatment of the cholinestrase inhibitor physostigmine. 3. The contraction to perivascular nerve stimulation(40V,40Hz, 0.5msec, 1min) was antagonised markedly by the muscarinic antagonist atropine. 4. The contraction to perivascular nerve stimulation(40V, 40Hz, 0.5msec, 1 min) was blocked by the neural blocker tetrodotoxin. 5. The contractions to perivascular nerve stimulation(40V. 40Hz, 0.5msec, 1 min) were not affected significantly by the -adrenergic antagonist phentolamine or - adrenergic antagonist propranolol. 6. The contractile response by the acetylcholine was increased by the pretreatment of cholinestrase inhibitor physostigmine. The finding suggest that it is powerful excitatory action linked to muscarinic receptor by cholinergic nerve in coronary artery of pig.

  • The relationship between structure and contractile activity was studied on the three neuropeptide $\gamma$ (mammalian-, bowfin-, shark-NP$\gamma$) that were synthesized by the solid-phase method. Circular dichroism spectra showed that mammalian-, bowfin- and shark-neuropeptide $\gamma$ took an unordered structure in buffer solution and artificial liposome. The intestinal motility response was investigated with guinea-pig ileum, rat duodenum and carp intestine. In case of carp intestine, contractile activity was as follows; bowfin-NP$\gamma$> shark-NP$\gamma$>mammalian-NP$\gamma$, On the other hand, the contractile activity of mammalian-neuropeptide $\gamma$ was more potent than those of bowfin-, shark-neuropeptide $\gamma$ in the guinea-pig ileum and rat duodenum. These results suggest that NP$\gamma$ show the species-specific activity.

  • To elucidate the action of $P_2$-purinoceptor, effects of adenosine triphosphate(ATP) and perivascular nerve stimulation were investigated from polygraph in the isolated renal artery of rabbit 1. ATP caused the relaxation on the precontraction with noradrenaline$(10{\mu}M)$ on the presene and absence of endothelium in the isolated renal artery of rabbit, the relaxative response was increased between 0.1 and $30{\mu}M$ on dose-dependent manner. 2. The relaxative response induced by ATP$(10{\mu}M)$ on precontraction with noradrenaline$(10{\mu}M)$ was blocked by the pretreatment with reactive blue 2$(10{\mu}M)$. 3. ATP inhibited the contractile response by perivascular nerve stimulation(0.3ms, 80V, 50Hz, 1 sec), the inhibitory action was blocked by the pretreatment with 8-phenyltheophylline$(10{\mu}M)$.

Influence of ${\beta}-Eudesmol$ on Blood Pressure

  • The present study was undertaken to investigate the effects of ${\beta}-eudesmol$, one of various ingredients isolated and identified from the bark of Magnolia obovata Thunberg, on arterial blood pressure and vascular contractile responses in the normotensve rats and to establish its mechanism of action. ${\beta}-Eudesmol\;(30{\sim}300\;{\mu}g/kg)$ given into a femoral vein of the normotensive rat produced a dose-dependent depressor response. These ${\beta}-eudesmol-induced$ hypotensive responses were markedly inhibited in the presence of chlorisondamine (1.0 mg/kg, i.v.) or phentolamine (2.0 mg/kg, i.v.). Interestingly, the infusion of ${\beta}-eudesmol$ (1.0 mg/kg/30min) into a femoral vein made a significant reduction in pressor responses induced by intravenous norepinephrine. Furthermore, the phenylephrine $(10^{-5}\;M)-induced$ contractile responses were depressed in the presence of high concentrations of ${\beta}-eudesmol\;(10{\sim}40\;{\mu}g/ml)$, but not affected in low concentration of ${\beta}-eudesmol\;(2.5{\sim}5\;{\mu}g/ml)$. Also, high potassium $(5.6{\times}10^{-2}\;M)-induced$ contractile responses were greatly inhibited in the presence of ${\beta}-eudesmol\;(10{\sim}40\;{\mu}g/ml)$ in a dose-dependent fashion. Taken together, these results obtained from the present study demonstrate that intravenous ${\beta}-eudesmol$ causes a dose-dependent depressor action in the anesthetized rat at least partly through the blockade of vascular adrenergic ${\alpha}_1-receptors$, in addition to the some unknown mechanism of direct vasorelaxation.

Neurotensin Enhances Gastric Motility in Antral Circular Muscle Strip of Guinea-pig

  • Many reports suggest that neurotensin (NT) in the gastrointestinal tract may play a possible role as a neurotransmitter, a circulating hormone, or a modulator of motor activity. NT exerts various actions in the intestine; it produces contractile and relaxant responses in intestinal smooth muscle. This study was designed to investigate the effect of NT on motility of antral circular muscle strips in guinea-pig stomach. To assess the role of $Ca^{2+}$ influx in underlying mechanism, slow waves were simultaneously recorded with spontaneous contractions using conventional intracellular microelectrode technique. At the concentration of $10^{-7}$ M, where NT showed maximum response, NT enhanced the magnitude $(863{\pm}198%,\;mean\;SEM,\;n=13)$ and the frequency $(154{\pm}10.3%,\;n=11)$ of spontaneous contractions. NT evoked a slight hyperpolarization of membrane potential, tall and steep slow waves with abortive spikes $(278{\pm}50%,\;n=4).$ These effects were not affected by atropine $(2\;{\mu}M),$ guanethidine $(2\;{\mu}M)$ and tetrodotoxin (0.2μM). NT-induced contractile responses were abolished in $Ca^{2+}-free$ solution and reduced greatly to near abolition by $10\;{\mu}M$ of verapamil or 0.2 mM of $CdCl_2.$ Verapamil attenuated the effects of NT on frequency and amplitude of the slow waves. Taken together, these results indicate that NT enhances contractility in guinea-pig gastric antral circular muscle and $Ca^{2+}$ influx through the voltage-operated $Ca^{2+}$ channel appears to play an important role in the NT-induced contractile mechanism.

Ethylacetate Fraction of Rubus coreanum Causes Vascular Relaxation and Hypotensive Action

  • The present study was designed to investigate whether ethylacetate (EtOAc) fraction extracted from Rubus coreanum affect the contractility of the isolated thoracic aortic strips and blood pressure of normotensive rats. The EtOAc fraction ($400{\mu}g/mL$) significantly depressed both phenylephrine (PE, $10{\mu}M$)- and high $K^+$ (56 mM)-induced contractile responses of the isolated thoracic aortic strips in a concentration-dependent fashion. In the simultaneous presence of L-NAME (an inhibitor of NO synthase, $300{\mu}M$) and EtOAc ($400{\mu}g/mL$), both PE- and high $K^+$-induced contractile responses were recovered to the corresponding control level in comparison with inhibition of EtOAc-treatment alone. Moreover, in the simultaneous presence of EtOAc after pretreatment with 0.4% CHAPS, both PE- and high $K^+$-induced contractile responses were recovered to the corresponding control level compared to the inhibitory response of EtOAc-treatment alone. Also, in anesthetized rats, EtOAc fraction (0.3~3.0 mg/kg) injected into a femoral vein dose-dependently produced depressor responses. This hypotensive action of EtOAc fraction was greatly inhibited after treatment with phentolamine (1 mg/kg), chlorisondamine (1 mg/kg), L-NAME (3 mg/kg/30 min) or sodium nitroprusside ($30{\mu}g/kg/30 min$). Intravenous infusion of EtOAc fraction (1.0~10.0 mg/kg/30 min) markedly inhibited norepinehrine-induced pressor responses. Taken together, these results demostrate that EtOAc causes vascular relaxation in the isolated rat thoracic aortic strips as well as hypotensive action in anesthetized rats. These vasorelaxation and hypotension of EtOAc seem to be mediated at least by the increased NO production through the activation of NO synthase of vascular endothelium, and the inhibitory adrenergic modulation.
