World-wide crop loss caused by insect pest and nematode reaches critical level. In Korea, similar crop loss has been gradually augmented in the field and greenhouse due to continuous crop rotation. The current methods on controlling herbivorous insects and plant parasitic nematodes are mostly depended on agro-chemicals that have resulted additional side-effect including occurrence of resistant insects and nematodes, environmental contamination, and accumulation in human body. To overcome the pitfalls, microbe-based control method have been introduced and applied for several decades. Here, we revised biological control using by the bacteria, fungi, and virus in order to kill insect and nematode and to attenuate its virulence mechanism. The introduced microbes mainly secreted out the hydrolysing enzymes and toxic compounds to target host membrane or cell wall directly. Indirectly, the microbe-triggered plant innate immunity against insects and nematodes was also reported. In conclusion, we provide a new frontier of microbe-based environmentally friendly procedure and effective methods to manage insects and nematodes.
This study was conducted to establish a practical masspropagation system of Phalaenopsis clones from basal shoot segments. The frequency of PLB (protocorm like body) induction was compared with various explants. Basal shoot segments showed the most successful result of 45%, while root tips, stalk node segments, stalk leaves and mature leaves represented low frequency (below 5%). The PLB induction ratio in the culture of basal shoot segments was examined with 11 different Phalaenopsis varieties, and the majority of varieties, including pink flower lines, showed an about 30% rate of PLB formation. Especially, when whole basal shoot parts without cutting were inoculated onto PLB induction medium, giant PLB was induced from explant. This giant PLB was green color and big in size compared with normal PLB. When dissected giant PLB segments inoculated onto PLB multiplication medium, only normal size of PLBs were induced from them. PLBs induced by basal shoot culture were transferred onto proliferation medium and then shooting medium, from which normal plants were formed. Therefore, this culture method is considered as effective and practical protocol for Phalaenopsis mericlone production. In addition, it is suggested that clones of an infinite number can be produced consecutively by this culture system through repeated cycles of PLB induction and proliferation using the basal shoot segment of flask plant.
This study developed a real-time dam's hydrologic variables prediction model (DHVPM) and evaluated its performance for simulating historical dam inflow and outflow in the Chungju dam basin. The DHVPM consists of the Sejong University River Forecast (SURF) model for hydrologic modeling and an autoreservoir operation method (Auto ROM) for dam operation. SURF model is continuous rainfall-runoff model with data assimilation using an ensemble Kalman filter technique. The four extreme events including the maximum inflow of each year for 2006~2009 were selected to examine the performance of DHVPM. The statistical criteria, the relative error in peak flow, root mean square error, and model efficiency, demonstrated that DHVPM with data assimilation can simulate more close to observed inflow than those with no data assimilation at both 1-hour lead time, except the relative error in peak flow in 2007. Especially, DHVPM with data assimilation until 10-hour lead time reduced the biases of inflow forecast attributed to observed precipitation error. In conclusion, DHVPM with data assimilation can be useful to improve the accuracy of inflow forecast in the basin where real-time observed inflow are available.
Hectorite was synthesized by a two-step hydrothermal process from $Mg(OH)_{2}$, water glass (${\sim}30\;wt%\;SiO_{2}$) and Li-compound at $90{\pm}5^{\circ}C$. The product shows excellent dispersion and swelling properties. The mixture of the starting materials was heated in a glass vessel for the first reaction with continuous stirring and the pH of the solution was adjusted to $6{\sim}8$, resulting in the formation of a precursor of hectorite. The excess salt components were washed out from the resulting slurry and then was matured in the glass vessel for the 2nd reaction. Li compound was added during the reaction. After a 10 h retention, the gel of hectorite was formed. The XRD pattern of the synthesized one was coincided with that of natural hectorite and SEM study revealed uniform grains 50 m in diameter. The d001 basal spacing of the product moved from 12 to $17.4\;{\AA}$ after glycolation treatment. The measured value of CEC and the swelling capacity was 90 cmol/kg and $60{\sim}70\;mL/2\;g$, respectively.
This study was undertaken to investigate the differentiation, from spore germination to hyphae growth and phialide formation, of Aspergillus niger through the method of synchronous and submerged culture. Through continuous experiments by shake culture with potassium acetate medium, we observed the formation of spores at appropriate concentration and pH. Potassium acetate medium was set pH 5.5, 6.0, 6.5 and 7.0 on each scale, and control, 20 mM, and 40 mM, 80 mM and 160 mM concentrations on the other scale. Aspergillus niger was cultured in the defined media at $28^{\circ}C$, and mycelial dry weight, changes of pH and the onset of sporulation were checked. The mycelial dry weight, increased in potassium acetate medium, and pH increased during mycelial growth and gradually decreased after the spore formation. When pH increased excessively in Potassium acetate medium with pH 7.0, the mycelia could not adapt and mycelial dry weight decreased gradually. At pH 5.5, the onset of sporulation was done within one day at 20 mM it took, at 80 mM three days and at 160 mM concentration. in two days, at 40 mM one to four days were taken, 80 mM concentration respectively. At pH 6.5, the onset of sporulation was done in three days and four days at 80 mM concentrations respectively. Spore formation was not shown at pH 7.0. In controlled medium with all levels of pH, spore formation was not shown.

Purpose: In-situ monitoring of water quality is fundamental to most environmental applications. The high cost and long delays of conventional laboratory methods used to determine water quality, including on-site sampling and chemical analysis, have limited their use in efficiently managing water sources while preventing environmental pollution. The objective of this study was to develop an on-site water monitoring system consisting mainly of an Arduino board and a sensor array of multiple ion selective electrodes (ISEs) to measure the concentration of $NO_3$ ions. Methods: The developed system includes a combination of three ISEs, double-junction reference electrode, solution container, sampling system consisting of three pumps and solenoid valves, signal processing circuit, and an Arduino board for data acquisition and system control. Prior to each sample measurement, a two-point normalization method was applied for a sensitivity adjustment followed by an offset adjustment to minimize the potential drift that could occur during continuous measurement and standardize the response of multiple electrodes. To investigate its utility in on-site nitrate monitoring, the prototype was tested in a facility where drinking water was collected from a water supply source. Results: Differences in the electric potentials of the $NO_3$ ISEs between 10 and $100mg{\cdot}L^{-1}$$NO_3$ concentration levels were nearly constant with negative sensitivities of 58 to 62 mV during the period of sample measurement, which is representative of a stable electrode response. The $NO_3$ concentrations determined by the ISEs were almost comparable to those obtained with standard instruments within 15% relative errors. Conclusions: The use of the developed on-site nitrate monitoring system based on automatic sampling and two-point normalization was feasible for detecting abrupt changes in nitrate concentration at various water supply sites, showing a maximum difference of $4.2mg{\cdot}L^{-1}$ from an actual concentration of $14mg{\cdot}L^{-1}$.

Korea introduced the cooperation charge on conservation of ecosystem for minimizing damage of ecosystem due to development projects and their effects and for preparing resources for natural environment conservation projects. Advanced countries have made efforts by expanding investment in natural environment conservation and restoring projects to promote prevention of global warming and improvement of biological diversity, are establishing nationwide strategies and plans. To examine the reality of projects by returns of the cooperation charge on conservation of ecosystem, microsite projects in schools and public facilities take the largest share while their project budgets are only about 100~300 KRW, relatively small, which might be attributable to budget restrictions in accordance with the calculating method of levying cooperation charge on conservation of ecosystem and problems of project proceeding in the system of returning fund for projects in general. The conclusion which this study suggests on invigoration of cooperation charge on conservation of ecosystem and its operation are as followings. First, although the cooperation charge on conservation of ecosystem has been introduced in 2001, the amount of imposition per unit area remains unchanged. It is desirable to increase the amount into $1,400KRW/m^2$ as of August, 2011 as the price index has been continuously rising for the past 10 years and the upward adjustment of imposition per unit area should be notified by the decree of the Ministry of Environment every January. Second, the ceiling amount of the cooperation charge on conservation of ecosystem should be abolished. Now the ceiling amount is defined as 1 billion KRW but it was found that there was not any ceiling amount specified according to the comparative analysis of similar systems with the Korean environmental improvement fund. The ceiling should be abolished so that medium level businesses are carried out and ecosystem recovering projects in the true sense of the word can be made smoothly. Third, weight should be introduced in calculating amounts in accordance with ecologic and economic values. Harmony between development and environment can be achieved by applying differentiated weights of constant regional coefficient by use zone and ecologic and economic values. Continuous efforts of improving cooperation charge on conservation of ecosystem should be made more than anything else so that projects by returns of cooperation charge on conservation of ecosystem get effectiveness.
To improve the lubricity of ultra low sulfur diesel, vegetable oil-based alkanol amide derivatives were prepared and their lubricity properties were studied. To synthesize the alkanol amides, we conducted the amidation reaction of diethaolamine High Frequency Reciprocating Rig (HFRR) and the fatty acid methyl esters, obtained by the continuous transesterification of methanol and several vegetable oil, such as soybean oil, palm oil and coconut oil. The synthesized amides were soluble in ultra low sulfur diesel in the concentration range of ca. 1 wt%; the lubricating properties of ultra low sulfur diesel containing 120 ppm of amides were measured using an HFRR method. It was found that the wear scar diameter in the pure ultra low sulfur diesel decreased significantly from 581 ${\mu}m$ to 305~323 ${\mu}m$ upon the addition of the amides, indicating that lubricating properties of the diesel were improved. On the other hand, the types of vegetable oils did not affect the wear scar diameters, implying that lubricating properties of the diesel did not depend strongly on the structures of alkyl groups of alkanol amide derivatives. When we measured the lubricating properties of the one type of diesels containing various amounts of alkanol amide, we observed that the wear scar diameter decreased drastically with increasing the amide concentration, meaning that the lubricity improved with the amide concentration.
Background: Supraglottis and glottis have a different embryologic origin. Supraglottic cancer is characterized by high incidence of cervical lymph node metastasis at initial diagnosis, and favored surgical management of the early supraglottic cancer was partial supraglottic laryngectomy, however the procedure resulted in frequent incidences of postsurgical aspiration and voice disabilities. Objectives: We retrospectively analyzed the problems and the advantages of the endoscopic laser assisted supraglottic partial laryngectomy as a part of surgical management for early supraglottic cancer. Materials and Methods: During the past nine years 25 cases of supraglottic cancer(Tl 10 cases, T2 15 cases) were treated by tracheotomy and laser assisted supraglottic partial laryngectomy(KTP532, 15 Watt, continuous type) and in 10 cases with cervical lymph node metastasis, they were additionally managed by neck dissection one week later, and all cases received postoperative irradiation therapy. Results: At present, 19 cases are alive with no evidence of disease. During the follow up period total of six cases(primary failure: three cases, nodal failure: three cases) were recurred. In relation to tumor staging, One of the 10 Tl cases and two of the 15 T2 cases recurred showing 88% locoregional recurrence rate for early supraglottic cancer. Postoperative com-plication included bleeding in three cases who were controlled by electrocautery under general anesthsia, one case of longstanding aspiration and two cases of laryngeal stenosis as a delayed complication. Conclusion: High control rate suggests that the endoscopic laser assisted supraglottic partial laryngectomy may be a good initial management method for early supraglottic cancer, however it is difficult to determine the resection margin, therefore, accurate tumor staging must be done prior to surgery. In order to prepare for postoperative bleeding, edema and aspiration, the tracheotomy must be performed prior to surgery.

To monitor the wind-induced responses of buildings, conventional real-time kinematic (RTK) methods based on two global positioning system (GPS) receivers (e.g., a reference and a rover) are widely applied. However, these methods can encounter problems such as difficulty in securing and maintaining a space for a reference station. With the recently developed virtual reference station (VRS)-RTK approach, the position of a structure can be measured using only a rover receiver. In this study, to evaluate the applicability of VRS-RTK methods in monitoring the lateral structural responses of frame structures, we performed free vibration tests on a one-story frame model (the first natural frequency of 1 Hz) and a three-story frame model (the first natural frequency of 0.85 Hz). To assess the reliability of the displacement and acceleration responses measured by the GPS, we performed a concurrent measurement using laser displacement sensors and an accelerometer. The accelerometer results were consistent with the GPS measurements in terms of the time history and frequency content. Furthermore, to derive an appropriate sampling rate for the continuous monitoring of buildings, the errors in the displacement responses were evaluated at different GPS sampling rates (5, 10, 20 Hz). The results indicate that as the sampling rate increased, the errors in the displacement responses decreased. In addition, in the three-story model, all modal components (first, second, and third modes) could be recorded at a sampling rate of 20 Hz.
