Assay development and HTS on microfluidic Lab-on-a-chip

  • Microfluidic lab-on-a-chip (LOC) systems have enabled a new generation ofassay technologies in chemical and biomedical sciences. Caliper's microfluidic LOC systems contain a network of microscopic channels through which fluids and chemical are moved in order to perform experiments. The main advantages of these continuous-flow devices are integration and automation of multiple steps in complex analytical procedures to improve the reproducibility of the results, and eliminated the manual labor, time and pipetting errors involved in analyses. The present talk is devoted to give a brief introduction of microfluidic basics and to present in applying continuous-flow microchips to drug screening with model enzyme assays.

The Consequence Analysis for Unconfined Vapor Cloud Explosion Accident by the Continuous Release of Gas-Liquid Flow (기-액흐름 연속누출에 의한 개방공간 증기운 폭발사고의 영향평가)

  • For the unconfined vapor cloud explosion accident by the continuous release of gas-liquid flow of various saturated liquids in a vessel at ground level, overpressures were estimated and analyzed with various release conditions and materials by TNT equivalency model with vapor dispersion. We found that at same release conditions, overpressure showed n-heptane > xylene > n-hexane > toluene > n-heptane > benzene, respectively and that overpressure was increased with increasing the hole diameter and the storage pressure, but it was increased with decreasing the wind speed, the interested distance, and the vessel thickness.

Effects of Flow in Line Dance Class on Happiness and Continuous Participation Intention in Elementary Student. (초등학생의 라인댄스 수업몰입에 따른 행복감 및 지속적 참여의사에 미치는 영향)

  • The purpose of this study was to know the effects of flow in line dance class on happiness and continuous participation intention in elementary student. By using purposive sampling 271 questionnaires had been used for practical analysis. Collected data were put to frequency analysis, exploratory factor analysis reliability analysis, correlation analysis, and multiple regression analysis using the SPSS Win Ver. 22.0 program. The results of these analyses were as follows. First, the autotelic experience, the concentration, the sense of control and the sense of achievement which are the subordinate variables of the flow, were all found to have significant effects on the confidence, the autotelic experience, the sense of control and the sense of achievement which are the subordinate variables of the flow, were found to have significant effects on the self-realization, the autotelic experience, the concentration, and the sense of achievement which are the subordinate variables of the flow, were found to have significant effects on the enjoyment. Second, the autotelic experience, the concentration, and the sense of achievement which are the subordinate variables of the flow, were found to have significant effects on the continuous participation intention. Third, the confidence and the enjoyment, which are the subordinate variables of the happiness, were found to have significant effects on the continuous participation intention.

  • Starting from the review of signal flow graphs and flow graphs, this paper gives an example of sampled-data systems for Sedlar & Bekey's formulation. In this purpose it discussed the difference between Mason's signal-flow graphs and Coates flow graphs for drawing th flow graph of a linear system, and then a new flow-graph symbol introduced in order to distinguish between continuous and discrete systems. Thus, the paper is analysed and compared with a sampled-data systems between conventional methods and new method of signal flow graphs.

  • This study was performed to evaluate the effects of air flow rate and aeration mode on the treatment of bioventing for diesel-contaminated soil. Initial concentrations of diesel-contaminated soils were about 2,500 and 9,000mg/kg. Air flow rates were 30, 60, and 100mL/min, and air was injected in the continuous and the intermittent modes. TPH removal efficiency of intermittent aeration mode was higher than that of continuous aeration mode. Greater air flow rate than the value of guidance book was needed for bioventing.

  • A side-extrusion model, meant for deeper understanding of the material flow in the CONFORM (continuous extrusion forming) of trub shaped aluminum profiles is presented. In order to get the desirded straight shape of the extrudate,every part of its cross-section must exit the die with the same velocity. Problem is assumed by plane strain UBET-model to analyze it in a simplified way. This has been done by studying the side-extrusion through a two -hole die face. The flow is balanced by determining the optimum lengths of the bearing lands, i.e., those lengths which result in equal exit velocities of the extrudates. Furthermore, the material flow, as influenced by the punch velocity, has been investigated.

  • Compression test has been used to measure material flow stress due to limited capability of tensile test at the fast strain rate. Since the frictional stress unavoidable during compression test should be properly eliminated from the measured stress, calibration of the measured stress by using friction factor has been made for the flow stress measurement. Also, calibrated stresses by interrupted and continuous compression tests have been compared with the true stress measured by tensile test at $0.2\%$ carbon steel.

  • The combustion characteristics using forced pulsating flow were experimentally investigated with confined premixed flames stabilized by a reward-facing step. The intermittent combustion has many merits, for instance, such as high load combustion, high heat transfer, low emission gas, compared with those of continuous combustion. For these purposes, data processing of binary image was conducted to reveal the differences between intermittent and continuous combustion. As the results, it was possible to calculate the reaction zone using OH-emission band and, therefore, showed that forced pulsating flow was useful in combustion technology.

  • In this paper, flow characteristics of plasma flow in a micro-tube were investigated experimentally using micro particle image velocimetry(micro-PIV). For comparison, the experiments were repeated for deionized(DI) wale. instead of plasma. Both velocity profiles of plasma and do-ionized water are well agreed with the theoretical velocity distribution of newtonian fluid. We also carried out generating plasma-in-oil droplet formation at a Y-junction microchannel. In order to clarify the hydrodynamic aspects involved in plasma droplet formation, Rhodamine-B were mixed with plasma only for visualization of plasma droplet. With oil as the continuous phase and plasma as the dispersed phase, plasma droplet can be generated in a continuous phase flow at a Y-junction. For given experimental parameters, regular-sized droplets are reproducibly formed at a uniform flow conditions.

  • A numerical analysis has been performed solidification problem using the fixed grid-enthalpy method with enthalpy-porosity relation. A modified standard $k-\varepsilon$ model was applied to describe the influence of turbulent flow. Computational procedures are based on the finite volume method and the non-staggered grid system. Comparisons with the different three experimental results show that applying a modified standard $k-\varepsilon$model in mushyzone is better than the previous computation results. This paper includes another EMBR's influences such as change of velocity field, Increasement of temperature and dispersion of flow out of nozzle into the flow field. These EMBR's influences are compared to case without EMBR.
