Two-Stage Biological Hydrogen Production by Rhodopseudomonas palustris P4 (Rhodopseudomonas palustris P4에 의한 이 단계(Two-stage) 생물학적 수소생산)

  • The integrated or the two-stage (dark anaerobic and photosynthetic) fermentation processes were compared for the hydrogen production using purple non-sulfur photosynthetic bacteria, Rhodopseudomonas palustris P4. Cell growth, pH changes and organic acids and bacteriochlorophyll contents were monitored during the processes. Culture broth of Rps. palustris P4 exhibited dark-red during the photosynthetic culture condition, while yellow under the anaerobic condition without light. Rps. palustris P4 grown at the photosynthetic condition evolved 0.38 and 1.33 ml $H_2$/mg-dcw during the dark and the light fermentation, respectively, which were totally 1.71 ml $H_2$/mg-dcw at the two-stage fermentation. The rate of hydrogen production using Rps. palustris P4 grown under the dark anaerobic condition was 2.76 ml $H_2$/mg-dcw which consisted of 0.46 and 2.30 ml $H_2$/mg-dcw from the dark and the photosynthetic fermentation processes, respectively. Rps. palustris P4 grown under dark anaerobic conditions produced $H_2$ 1.6 times higher than that of grown under the photosynthetic condition. However, total fermentation period of the former was 1.5 times slower than that of the latter, because the induced time of hydrogen production during the photosynthetic fermentation was 96 and 24 hours when the seed culture was the dark anaerobic and photosynthetic, respectively. The integrated fermentation process by Rps. palustris P4 produced 0.52 ml $H_2$/mg-dcw(1.01 mol $H_2$/mol glucose), which was 20% of the two-stage fermentation.

The Simple and Rough Screening Method of Phosphorus Deficient Tolerance Rice

  • Even though phosphorus (P) is essential element for plant growth and development, it is not enough for crop production in soil. To breed more P deficient tolerance rice, screening and selection in rice population is needed. We tried to develop more simple and rough screening method for breeding of P deficient tolerance rice. In P deficient condition, tiller number was dramatically decreased among yield components in rice. Though this result, we confirmed tiller number could be the best marker in screening of P deficient tolerance rice. 480 rice genetic resources were cultivated in rice bed tray filled with P deficient soil for four weeks and each dry weight was measured. Among them, the 55 kinds of genetic resource were selected then cultivated in paddy field with 3 fertilizer conditions. Plant dry weight and tiller number in ripening stage were shown significant difference according to P condition. Plant dry weight and tiller number in ripening stage was highly correlated especially in P deficient condition. Furthermore, the tiller number in ripening stage and plant dry weight in rough screening were shown high degree correlation. Though these results, we might expect measuring of plant dry weight after cultivation in rice bed tray filled with P deficient soil could be a simple and effective screening method in selection of P deficient tolerance rice.

  • Numerical analysis using 3D finite element program (PLAXIS 3D) evaluated the interaction of soil - pile structure under dynamic surface loading. The dynamic p-y curve of the 1-g shaking table experiment by numerical analysis was calculated, and the parametric studies were presented by considering the pile-soil condition, the pile tip condition, and the loading condition. The frequency of 1.4 Hz is almost equal to the natural frequency of the pile - soil system. The p and y values of resonance phenomenon are significantly different from the results of other frequencies. The results can be summarized by a third order polynomial function representing the trend line in the p-y curve. In the case of a single pile, the shape of the dominant curve was found to be an ellipse by mathematical proof. The elliptic equation can be used for the dynamic design or analysis of soil-pile system.

Characterization of Optimum Conditions Affecting on Multi-Color Dyeing of Cut Rose (Rosa hybrida 'Akito') (절화장미(Rosa hybrida 'Akito') 복색염색에 미치는 몇 가지 요인의 최적조건 구명)

  • This experiment was conducted to examine the ideal condition for multiple colors dyeing of cut rose, 'Akito'. Four different colors of dye were applied to make multiple flower colors in cut rose by absorption method. Acidic condition around pH 3.5 showed the fastest absorption speed in all four colors. There is no difference at each pH levels, at part of leaves dyeing, it is more affected colors rather than pH levels in purple and blue colors. From vary concentration of each dyes, $10g{\cdot}L^{-1}$ concentration of dyeing solution gave the best color definition. But it's not significant and result was different by each color in a same concentration. 10 minutes of dyeing performed pastel color and 30 minutes performed the best color definition, but the 30 minutes treatment accompanied with significant dyeing on the leaves also. As a result of this experiment, optimum condition for dyeing cut rose, 'Akito' requires pH 3.5, $5-10g{\cdot}L^{-1}$ of dye concentration and 10-30 minutes treatment.

  • In this paper, a new asymptotic stability condition of continuous fuzzy system is proposed. The new stability condition considers the nonquadratic stability by using the P-matrix measure. Later the relationship of the suggested stability condition and the well-known stability condition is discussed and it is shown in a rigorous manner that the proposed criterion includes the conventional conditions.

  • Enzyme activity and expression of cytochrome P450 gene involved in the pathway of capsidiol biosynthesis were compared in five different solanaceae plants such as red pepper, green pepper, tobacco, potato and egg plant. Base on genomic DNA and/or RT-PCR results, four solanaceae plants such as red pepper, green pepper, tobacco and egg plant possess P450 gene in the genome and specifically expressed by elicitor treatment. However, potato was appeared to have neither P450 nor cyclase gene in the genome. P450 genes did not show any expression in the plants under normal condition, but showed highly specific expression under elicitation condition in various organs and tissue such as leaf, root, stem and culture cells.

  • The problem of vertex labeling with a condition at distance two in a graph, is a variation of Hale's channel assignment problem, which was first explored by Griggs and Yeh. For positive integer $p{\geq}q$, the ${\lambda}_{p,q}$-number of graph G, denoted ${\lambda}(G;p,q)$, is the smallest span among all integer labellings of V(G) such that vertices at distance two receive labels which differ by at least q and adjacent vertices receive labels which differ by at least p. Van den Heuvel and McGuinness have proved that ${\lambda}(G;p,q){\leq}(4q-2){\Delta}+10p+38q-24$ for any planar graph G with maximum degree ${\Delta}$. In this paper, we studied the upper bound of ${\lambda}_{p,q}$-number of some planar graphs. It is proved that ${\lambda}(G;p,q){\leq}(2q-1){\Delta}+2(2p-1)$ if G is an outerplanar graph and ${\lambda}(G;p,q){\leq}(2q-1){\Delta}+6p-4q-1$ if G is a Halin graph.

Effect of Lime Amount and Application Time on Soil pH Change, Yield, and Quality of Leaf Tobacco (석회시용량과 시용시기가 경작지 토양산도 변화와 잎담배 수량 및 품질에 미치는 영향)

  • This study was carried out to improve the tobacco field condition and to determine the effect of lime amount and application time on soil pH, yield, and quality of leaf tobacco. Lime was applied to the tobacco field by determined amounts according to different pH level. The changes of soil pH, growth of tobacco, yield, and quality of KF109 and Br21 tobacco were surveyed by time lapse. The target pH value in tobacco field soil can be reached at 6 weeks after lime application, and then the soil pH was lowered slightly after that time. The lime amount needed to reach target pH was decreased 40 % in the same tobacco field after 1 year. Though the initial growth rate of flue-cured tobacco in the field of pH 7.0 was lower than that of conventional tobacco field, the field of pH 7.0 showed the highest yield after the maximum growth stage. The quality of cured leaf tobacco in the field of pH 7.0 applied lime at spring season was slightly lowered compared with that in conventional. This results indicated that the best pH condition in tobacco field for the best tobacco growth was 6.5 and the proper time of lime application was fall season of previous year by application of the whole quantity.

  • This study was carried out to establish an effective and practical system for commercialization of embryo production techniques by analyzing several factors influencing in vivo embryo production on condition of donors treated FSH-P and SUPER-OV in Korean native cattle. In vivo embryos were flushed 226 times from 128 donors. The results obtained for the factors influencing in vivo embryo production by conditionof donors treated FSH-P and SUPER-OV were as follows :1. Age and parity of donors did not affect the transferable and freezable embryos among the treatments in FSH-P but the transferable and freezable embryos were decreased after 6 years old and 4th parity in SUPER-OV(P<0.01). 2. The production of embryos on the weight of donors was higher in over 400kg of body weight in FSH-P(P<0.01) and was higher in over 450kg than 400~450kg of body weight in SUPER-OV(P<0.05). For FSH-P embryo production was better responded in 350~450kg of body weight with 30~32mg doses, and showed a better result in over 450kg body weight with 32~34mg doses.(Key Words : in vivo embryo, donors, FSH-P SUPER-OV)
