Effect of Grape Intake on Cadmium Metabolism of Rats during Aging (포도의 부위별 섭취가 흰쥐의 노화 과정 중 Cadmium의 대사 및 중독에 미치는 영향)

  • This study was carried out to investigate the effect of whole grape, grape pomace and grape juice intake on cadmium (Cd) metabolism during aging in thirteen-month-old Sprague-Dawley male rats. One hundred and twenty rats weighing 548.8 $\pm$ 4.3 g were assigned to eight groups according to body weight and were raised for 3, 5 or 7 months on diets containing 2 % (w/w) dried powders of three different parts of the grape (Campbell Early) and 0.02 % (w/w) CdC12. Food intake tended to decrease with aging, and body weight and epididymal fat pad (EFP) weights of Cd-exposed groups were lower than those of Cd-free groups. Cadmium accumulated in the blood and tissues and Cd concentration was the lowest in the pomace group among Cd-exposed animals. Metallothionein (MT) concentration in the tissues increased through Cd administration. Grape diets, especially grape pomace diets, were effective in decreasing Cd absorption in the tissues by increasing Cd excretion through feces. The intake of grape pomace alleviated the decrease in bone density caused by Cd administration and prevented a decrease in glomerular filtration rates (GFR) with aging. Among the parts of grape, grape pomace, which had highest content of dietary fiber and flavonoids, was the most effective. The results of this study suggest the possibility of using grape pomace as a functional food material, a prospect that previously has been discarded.

  • Choline chloride-glycerol (1:2 mol), a natural deep eutectic solvent (NADES) is examined as a draw solution in forward osmosis (FO) for dewatering application. The NADES is easy to prepare, low in toxicity and environmentally benign. A polyamide thin film composite membrane was used. Characterization of the membrane confirmed porous membrane structure with good hydrophilicity and a low structural parameter (722 ㎛) suitable for FO application. A dilute solution of 20% (v/v) NADES was enough to generate moderate water flux (14.98 L m-2h-1) with relatively low reverse solute flux (0.125 g m-2h-1) with deionized water feed. Application in dewatering industrial wastewater feed showed reasonably good water flux (11.9 L m-2h-1) which could be maintained by controlling the external concentration polarization and fouling/scaling mitigation via simple periodic deionized water wash. In another application, clarified sugarcane juice could be successfully concentrated. Recovery of the draw solute was accomplished easily by chilling utilizing thermo responsive phase transition property of NADES. This study established that low concentration NADES can be a viable alternative as a draw solute for dewatering of wastewater and other heat sensitive applications along with a simple recovery process.

Fermentation Quality of Ensiled Water Hyacinth (Eichhornia crassipes) as Affected by Additives

  • A lab-scale ensiling study was carried out to investigate the fermentation quality of water hyacinth (WH) supplemented with molasses, rice bran, as an absorbent, and an inoculant in the form of fermented vegetable juice and their combinations. After wilting the water hyacinths for 7 h to a dry matter (DM) content of 240 to 250 g/kg, the following treatments were applied: i) Control (C), WH only; ii) WH with sugarcane molasses at 40 g/kg WH (CM); iii) WH inoculated with fermented vegetable juice at 10 ml/kg WH (CI); iv) CM and CI (CMI) combined; v) WH with 150 g rice bran/kg WH (CA); vi) CA and CI combined (CAI); vii) CA and CM combined (CAM); and viii) CA, CM and CI combined (CAMI). After application of additives, the differently treated forages were mixed and ensiled in triplicates in 1,500-ml polyethylene jars. After ensiling for 3 d, pH values in all treatments, except C and CI, had decreased to approximately 4.0 and remained low till 14 d. After 56 d, pH had increased between 0.4 to 0.9 pH-units compared to those at 14 d. The ammonia nitrogen ($NH_3$-N) concentration ranged from an acceptable level in treatment CM (8 g/kg N) to a high $NH_3$-N value in treatment CMI (16 g/kg N). Lactic acid formation was higher in CI than in all other treatments. Butyric acid contents, which indicate badly fermented silages, were low in all silages (<2 g/kg DM). There were two-way interactions (p-values from <0.001 to 0.045) for almost all fermentation end-products and pH, except for the molasses${\times}$inoculant interaction on $NH_3$-N (p = 0.26). Significant 3-way interactions were found on all observed variables except for weight losses of silages. It is concluded that conserving wilted WH as silage for ruminants may be improved by the addition of molasses or rice bran.

  • Potential mutagenicity of ten heated edible mountain herbs were examined with spore recassay, Ames test and DNA breaking test. Samples of edible mountain herbs were prepared with water extraction at $100^{\circ}C$ for 20 minutes. With the rec-assay, no significant mutagengic activity could be obtained from all of the samples, but among the eight of metal ions added to sample solution, $Pb^{2+}$ to R. crispus heated juice, $Zn^{2+}$ to L. fischeri and S. bracycarpa heated juice increased mutagenic activity of the samples. With the Ames test and DNA breaking test, all of the samples did not show mutagenicity. However, breaking action was activated on heated L. fischeri, P. japonicus. A. triphylla and A. tataricus juices in the presence of 25mM $Cu^{2+}$. But heated A. elata, H. aurantiaca, A. triphylla, S. bracycarpa and A. scaber juices were inactivated in the presence of 25mM $Fe^{2+}$. Desmutagenic activities against benzo$({\alpha})$pyrene significantly increased as increasing concentration of the heated edible mountain herb juices.

The Relationshin between ACE Inhibitory Activity and Degradations of Sulfur Containing Materials in Dolsan Leaf Mustard Juice

  • This Study was tarried out to investigate the relationship ACE inhibitory activity and degradations of sulfur containing materials in Dolsan leaf mustard juice (DLMJ). The changes of sulfur containing materials which were treated with autolysis, myrosinase, ascorbate and papain were studied, as well as the changes of ACE inhibitory activity in DLMJ. At $37^{\circ}C$, sulfur contain-ing materials by autolysis decreased most rapidly from $0.43\%$ to $0.13\%$ in the second day. Conversely. ACE inhibitory activity increased most from $66\%$ to $87\%$. in the second day at $37^{\circ}C$. As myrosinase concentrations increased more, sulfur containing materials in DLMJ decreased more. The ACE inhibitory activities at 0, 0.5, 1, 2, and 4 Units of myrosinase for 240 min later were 70, 74, 75, 82, and $85\%$, respectively. At 1 mM ascorbate. concentrations of Sulfur containing materials in DLMJ decreased more significantly on the second day than on the other days. At 1 mM ascorbate for 6 days, ACE Inhibitory activity reached a maximum of about $92\%$. And, an increase of papain concentration was noted in accordance with a decreased sulfur containing materials. The maximum rate of AEC inhibitory activity at control, 3, 6, and 12 Units of papains treatments was shown as 70, 70, 75, and $78\%$ at 60 min, respectively. These results suggested that the degradation of sulfur containing materials led to the increase of ACE inhibitory activity. Consequently, it was suggested that ACE inhibiting was significantly related to the degradatives of sulfur containing materials.

  • We investigated the optimal condition for production of bacterial cellulose with Gluconacetobacter persimmonus KJ145. For bacterial cellulose production, optimal medium composition and culture conditions were conducted to determine. Apple juice (10$^{\circ}$Brix) medium was suitable than Hestrin & Schramm medium which is generally used for the bacterial cellulose production. When 1% pyruvate as carbon source was added to apple juice, bacterial cellulose production rose to high level. The effect of various nitrogen sources was investigated: CSL was found to be essential to high cellulose yields and the optimal CSL concentration was 10%. Optimal temperature and culture time for the bacterial cellulose production was 35$^{\circ}C$ and 16 days, respectively At the optimal condition Gluconacetobacter persimmonus KJ145 produced 8.96g/L of bacterial cellulose (dry weight), which was much higher than reported values.

Isolation and Identification of an Antioxidant Substance from Heated Garlic (Allium sativum L.)

  • The objectives of this study were to identity antioxidant substance in heated garlic juice (HGJ). We evaluated the antioxidant activities of heated garlic juice exposed to 120, 130, and $140^{\circ}C$ for 2 hr. The HGJ was partitioned using the solvents of hexane, chloroform, ethyl acetate, butanol, and water. The ethyl acetate fraction of HGJ treated at $130^{\circ}C$ for 2 hr showed strong antioxidant activity; this extract was isolated and purified using silica gel column chromatography and semi-preparative high-performance liquid chromatography. The structure of the purified compound was determined using spectroscopic methods, i.e., ultraviolet, mass spectrometry, infrared, $^1H$ NMR, $^{13}C$ NMR, DEPT, HMBC, and HMQC. The isolated compound was identified as thiacremonone (2,4-dihydroxy-2,5-dimethyl-thiophene-3-one). Thiacremonone showed strong 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging activity, with a 50% inhibition concentration ($IC_{50}$) of $22.25{\pm}0.44\;{\mu}g/mL$, which is much higher than that of the antioxidants ascorbic acid ($30.06{\pm}0.42\;{\mu}g/mL$), ${\alpha}$-tocopherol ($71.30{\pm}0.97\;{\mu}g/mL$), and butylated hydroxyanisole ($50.54{\pm}0.94\;{\mu}g/mL$).

Selection of Starter Cultures and Optimum Conditions for Lactic Acid Fermentation of Onion

  • Lactic acid bacteria (LAB) isolated from various fruits and vegetables were screened in order to determine appropriate fermentation starters for manufacturing functional fermented onion juice. From the initial screening test comprising more than 700 isolated LAB, 16 isolates were selected based on their acid production rate. Among the selected isolates, the fermentation broth of KC-007 exhibited the highest electron donating and nitrite scavenging activities, with values at pH 1.2 of 95.6 and 68.7%, respectively. From the overall results obtained in this study, we finally selected the bacterium KC-007 as a fermentation starter. This bacterium was identified and named as Pediococcus pentosaceus based on its morphological and physiological characteristics, carbon-utilization pattern (as assessed using an API 50CHL kit), and molecular genetic characteristics (as assessed using the nucleotide sequence of the 16S rRNA gene). The optimal temperature, pH, and starter inoculation concentration (v/v) required for growth of the isolated strain were $40^{\circ}C$, pH 4.0-6.0, and 2%(v/v), respectively.

Effect of Adrenergic Nervous System on Pancreatic Exocrine Secretion in Rats (흰쥐에서 췌장의 외분비 기능에 미치는 adrenaline 동작성 신경계의 영향)

  • The present study was performed to investigate a possible influence of the adrenergic nervous system on pancreatic exocrine secretion stimulated by intraduodenal acid perfusion. Pancreatic secretion was collected in rats anesthetized with urethane after 24 hours fasting. The duodenal lumen was perfused (0.2 ml/min) with HCI solution in a concentration of 0.005, 0.01, 0.02, 0.05 or 0.1 N When the volume of panceratic juice secreted for IS min became constant phentolamine (1 mg/kg), $noradrenaline\;(10\;{\mu}g/kg),\;Propranolol\;(1\;mg/kg),\;and \;isoproterenol\;(1\;{\mu}g/kg)$ were administered through the jugular vein in bolus. The secretory volume and protein output were measured in the pancreatic juice collected for 15 min. 1) HCI, perfused intraduodenally in graded concentrations from 0.005 N to 0.1 N, increased the pancreatic secretory volume and protein output dose-dependently. 2) In the basal state as well as in the stimulated state by the duodenal acid perfusion, phentolamine increased the pancreatic secretory volume and protein output while propranolol inhibited the volume and protein output. 3) In the basal state, noradrenaline did not change the pancreatic secretory volume but increased the protein output while isoproterenol increased both of the secretory volume and the protein output. These results strongly suggest that ${\alpha}-adrenoceptors$ in the rat pancreas exert an inhibitory influence on the pancreatic exocrine secretion including volume and protein output in the basal state as well as in the stimulated state by the intraduodenal acid perfusion while ${\beta}-adrenoceptors$ play a stimulatory role in the pancreatic exocrine secretion. However, in the physiological situation, adrenergic excitation may stimulate the protein output through ${\beta}-adrenoceptors$ without change in the secretory volume in the rat pancreas.

A Clinical study - The Effect of Chongmyungtang(聰明湯) added Grape Juice on Power of Attention (포도즙가총명탕(葡萄汁加聰明湯)의 집중력(集中力)에 미치는 임상적(臨床的) 연구(硏究))

  • Clinical studies were carried out 25 cases through administration of Chongmyungtang added Grape Juice(CMT+GJ) from December 1st 2002 to December 31th 2002. The results were summarized as follows; 1. There were significant decreases in physical symptoms of fatigue from $4.64{\pm}2.71$ to $2.52{\pm}1.78$ by administration of CMT+GJ. 2. There were significant decreases in mental symptoms of fatigue from $5.44{\pm}2.38$ to $3.88{\pm}2.03$ by administration of CMT+GJ. 3. There were significant decreases in neuro-sensory symptoms of fatigue from $3.84{\pm}2.03$ to $2.88{\pm}2.07$ by administration of CMT+GJ. 4. There were significant decreases in total subjective symptoms of fatigue from $13.92{\pm}5.42$ to $9.28{\pm}4.21$ by administration of CMT+GJ. 5. There were not significant changes in VAS of fatigue and attention ratio by administration of CMT+GJ. According to the results, we could suggest that CMT+GJ is able to be applied to the development of concentration.
