Characterization of Antidiabetic Compounds from Extract of Torreya nucifera (비자나무 추출물의 항당뇨 활성물질의 특성 연구)

  • Natural products have gained increasing attention due to their advantage of long-term safety and low toxicity for a very long time. Torreya nucifera is widespread in southern Korea and Jeju Island and its seeds are commonly used as edible food. Oriental ingredients have often been reported for their insecticidal, antioxidant and antibacterial properties, but there have not yet been any studies on their antidiabetic effect. In this study, we investigated several biological activities of T. nucifera pericarp (TNP) and seeds (TNS) extracts and proceeded to characterize the antidiabetic compounds of TNS. The initial results suggested that TNS extract at 15 and 10 ㎍/ml concentration has inhibitory effects on α-glucosidase and protein tyrosine phosphatase 1B, that is 14.5 and 4.35 times higher than TNP, respectively. Thus, the stronger antidiabetic TNS was selected for the subsequent experiments to characterize its active compounds. Ultrafiltration was used to determine the apparent molecular weight of the active compounds, showing 300 kDa or more. Finally the mixture was then partially purified using Diaion HP-20 column chromatography by eluting with 50~100% methanol. Therefore we concluded that the active compounds of TNS have potential as therapeutic agents in functional food or supplemental treatment to improve diabetic diseases.

A Study on Infiltration Process and Physicochemical Influence in the Unsaturated and the Saturated Zone of the Bottom Ashes from Thermal Power Plant (화력발전소 배출 바닥재의 불포화대와 포화대 침투과정과 물리화학적 영향에 대한 연구)

  • This study focused on the physicochemical effects of bottom ash dissolved precipitation on the soil and groundwater environment. The iced column and percolation experiments showed that most of the bottom ash particles were drained as the ash-dissolved solution, while the charcoal powder was filtered through the soil. Ion species of Al, As, Cu, Cd, Cr, Pb, Fe, Mn, Ca, K, Si, F, NO3, SO4 were analyzed from the eluates collected during the 24 h column test. In the charcoal powder eluates, a high concentration of K was detected at the beginning of the reaction, but it decreased with time. The concentrations of Al and Ca were observed to increase with time, although they existed in trace amount. In the bottom ash eluates, the concentrations of Ca and SO4 decreased by 30 mg·L-1 and 67 mg·L-1, respectively, over 24 h. It is regarded that the infiltration patterns of the bottom ash and biochar in the unsaturated zone were different owing to their particle sizes and solvent properties. It is expected that a significant amount of the bottom ash will mix with the precipitation and percolate below the water table, especially in the case of thin and highly permeable unsaturated zone. The biochar was filtered through the unsaturated zone. The biochar did not dissolve in the groundwater, although it reached the saturation zone. For these reasons, it is considered that the direct contamination by the bottom ash and biochar are unlikely to occur.

Effects of Traffic Volume and Air Quality on the Characteristic of Urban Park Soil (교통량과 대기질이 도시 공원 토양 특성에 미치는 영향)

  • This study aims to understand how mobile and stationary air pollution sources affect the air quality and soil properties in urban parks. We selected three sites of urban parks in Seoul as follows: Ha-neul Park in Mapo-gu (Site_M), Ill-won Eco-Park in Gangnam-gu (Site_G), and Yangjae Citizen's Forest in Seocho-gu (Site_Y), and compared the results of each site's traffic volume, air quality concentration, and soil analysis. Traffic volume was high in Site_M, followed by Site_G and Y; Site_M and G were closer to the resource recovery facility than Site_Y. Hence, we hypothesized that PM and NO2 concentrations in the atmosphere were higher in Site_M than Site_G and Y, causing different soil nitrogen content among sites due to different atmospheric deposition. Consistent with our hypothesis, the concentrations of PM2.5 and NO2 were higher in Site_M and G than Site_Y, while Site_Y had higher PM10 than other sites. The soil NO3- contents showed no significant difference among three sites, whereas the soil NH4+ content was extremely high in Site_Y. This high content of soil NH4+ is thought to be due to acidification from excessive fertilization. Lower soil pH of Site_Y further supported the evidence of heavy fertilization in this site. Overall nitrogen dynamics implies that soil nitrogen status is more influenced by park management such as fertilization rather than atmospheric deposition. Despite of lower soil NH4+ content of Site_M and G than Y, vegetation vitality looked similar among three sites. This indirectly indicates that excessive fertilizer input in urban park management needs to be reconsidered. This study showed that even if the air quality was different due to mobile and stationary sources, it did not directly affect the soil nitrogen nutrient status of the adjacent urban park.

Comparative study on efficacies and ingredient contents of different solvent extracts of Insampaedok-san (인삼패독산의 추출용매에 따른 성분함량 및 효능연구)

  • Objectives : This study evaluated activities and ingredient contents concerning extracts according to extraction solvents of Insampaedok-san (IS, Renshen bai du-san). Methods : The herbal constituents of IS were extracted with water and 70% ethanol at $100^{\circ}C$ for 2 hr. Using the HPLC system, the six ingredient contents of different solvent extracts of IS were analyzed. The nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$) production and proinflammatory cytokines were measured in RAW264.7 cells stimulated with lipopolysaccharide (LPS). The macrophage-derived chemokine (MDC/CCL22) and regulated on activation normal T-cell expression and secreted (RANTES/CCL5) production were measured in HaCaT and BEAS-2B cells stimulated tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and interferon-${\gamma}$ (IFN-${\gamma}$). The activities of glycerol-3-phosphate dehydrogenase (GPDH) and leptin level were measured in 3T3-L1 cells. Results : The calibration curves showed good linearity ($r^2$=1.0000) for different concentration ranges. The contents of liquiritin, naringin, hesperidin, neohesperirin and glycyrrizin in 70% ethanol extracts of IS were relatively higher than that of water extract, however the content of ferulic acid in 70% ethanol and water extract of IS were similar. The extraction solvents of water and 70% ethanol were evaluated inhibitory effect on the production of NO, $PGE_2$, TNF-${\alpha}$ and IL-6 in RAW 264.7 cells. Their extractions were inhibitory effect on production of MDC/CCL22 and RANTES/CCL5 in HaCaT cell and BEAS-2B cell, respectively. In addition, evaluated reduced on GPDH activity and leptin level in 3T3-L1 preadipocyte cell. Conclusions : Our results suggest that IS extracts were inhibitory effects of disease such as inflammation, allergies and obesity.

Determination of Antifungal Activity on Pepper Anthracnose and Plant Growth Promoting Activity of Pleurospermum camtschaticum Root Extract (누리대 뿌리 추출물의 고추 탄저병에 대한 항균 및 생장 촉진 활성 검정)

  • This study was conducted to confirm the utilization of Pleurospermum camtschaticum root extract as an organic agricultural material. Antioxidant activity of P. camtschaticum root extract, closely related to antibacterial activity, increased in a dose-dependent manner. In mycelial growth inhibitory activity, 100% P. camtschaticum root extract supressed over 70% for Colletotrichum coccodes and over 68% for Colletotrichum dematium. In the pepper fruit anthracnose development test, the size of the lesion decreased in a dose-dependent manner, which showed the same tendency as the previous results in inhibitory activity on mycelial growth. In the pepper seed germination and red pepper growth promotion test of P. camtschaticum root extract, oposite results was confirmed. The lower the concentration, the more the seed germination and growth promotion effects were shown. The phenol content of pepper leaves was also measured after pepper growth promotion test have been completed. The phenol content related to antibacterial activity increased in all treated groups compared to the untreated group. Therefore, the results of this study showed the possibility of development as an organic material.

Phytoplankton Variability in Response to Glacier Retreat in Marian Cove, King George Island, Antarctica in 2021-2022 Summer (하계 마리안 소만 빙하후퇴에 따른 식물플랑크톤 변동성 분석)

  • Rapid climate change has resulted in glacial retreat and increased meltwater inputs in the Antarctic Peninsula, including King George Island where Marian Cove is located. Consequently, these phenomena are expected to induce changes in the water column light properties, which in turn will affect phytoplankton communities. To comprehend the effects of glacial retreat on the marine ecosystem in Marian Cove, we investigated on phytoplankton biomass (chlorophyll-a, chl-a) and various environment parameters in this area in December 2021 and January 2022. The average temperature at the euphotic depth in January 2022 (1.41 ± 0.13 ℃) was higher than that in December 2021 (0.87 ± 0.17 ℃). Contrastingly, the average salinity was lower in January 2022 (33.9 ± 0.10 psu) than in December 2021 (34.1 ± 0.12 psu). Major nutrients, including dissolved inorganic nitrogen, phosphate, and silicate, were sufficiently high, and thus, did not act as limiting factors for phytoplankton biomass. In December 2021 and January 2022, the mean chl-a concentrations were 1.03 ± 0.64 and 0.66 ± 0.15㎍ L-1, respectively. The mean concentration of suspended particulate matter (SPM) was 24.9 ± 3.54 mgL-1 during the study period, with elevated values observed in the vicinity of the inner glacier. However, relative lower chl-a concentrations were observed near the inner glacier, possibly due to high SPM load from the glacier, resulting in reduced light attenuation by SPM shading. Furthermore, the proportion of nanophytoplankton exceeded 70% in the inner cove, contributing to elevated mean fractions of nanophytoplankton in the glacier retreat marine ecosystem. Overall, our study indicated that freshwater and SPM inputs from glacial meltwater may possibly act as main factors controlling the dynamics of phytoplankton communities in glacier retreat areas. The findings may also serve as fundamental data for better understanding the carbon cycle in Marian Cove.

Leaf Mineral Contents and Growth Characteristics of Strawberry Grown in Aquaponic System with Different Growing Media in a Plant Factory (식물공장형 아쿠아포닉스 시스템에서 배지 종류에 따른 딸기 잎의 무기이온 함량과 생육 특성)

  • This study was aimed to determine the effects of grow media on the mineral contents of the leaves and growth characteristics of strawberry grown under aquaponics system in a plant factory. For aquaculture, 12 fish (Cyprinus carpio) (total weight, 2.0 kg) were raised in an aquaponics tank (W 0.7 m × L 1.5 m × H 0.45 m, 472.5 L) filled with 367.5 L of water at a density of 5.44 kg·m-3 and total 34 of strawberry seedlings were transplanted in the pots filed with 200 g of orchid stone, hydroball or polyurethane sponge in the growing bed (W 0.7 m × L 1.5 m × H 0.22 m) laid out with holly acrylic sheet (140×60 mm, Ø80) on the top of the system. The pH and EC of the aquaponic solution was ranged from 7.6 to 4.9 and 0.24-0.91 dS·m-1, respectively. The concentration of NO3-N was about 28% lower than that of the hydroponic standard solution, and K, Fe and B were 10, 27 and 3.8 times lower, respectively; however, the mineral contents of strawberry leaves were in the appropriate ranges with lower contents in the leaves grown with sponge media. The organic content (OM), nitrogen (N), phosphorus (P), and potassium (K) of the sludge were 61.5, 5.72, 8.92, and 0.24%, respectively. The leaf area, leaf number, and dry and fresh weights of shoot at 81 DAT were significantly higher in the hydroball, and the average number of fruits per plant was significantly higher in both the orchid stone and hydroball. There was no significant difference in the fresh and dry weights of fruits. Integrated all the results suggest that the orchid stone and hydroball media are more effective to utilize nutrients in solid particles of aquaponic solution, compared to the polyurethane sponge.

Anti-tumor Effect of a Combination of Hongyoung Ethanol Extract and Cisplatinin YD-10B Oral Cancer Cells (YD-10B 구강암세포에서 홍영 에탄올 추출물과 시스플라틴 병용에 의한 항암 효과)

  • Solanum tuberosum Linnaeus cv Hongyoung, which represents red potato, was developed in Korea. Hongyoung is known to have anti-oxidant, anti-inflammatory, anti-viral, and anti-tumor properties, but no research has been conducted on the growth inhibition and apoptosis effects of hongyoung in YD-10B oral cancer cells. In this study, the combined treatment of hongyoung ethanol extract (HEE) and cisplatin were examined to determine its ability to inhibit cancer cell growth, induce apoptosis, and inhibit matrix metalloproteinases (MMP)-2 and MMP-9 cancer metastasis. The cell viability was investigated using a 2-(2-methoxy-4-nitrophenyl)-3-(4-nitrophenyl)-5-(2,4-disulfophenyl)-2H- tetrazolium monosodium salt (MTS) assay, and the ability to induce apoptosis was analyzed using an FACS analyzer. The mRNA expression and protein activity of MMP-2 and MMP-9 were measured via RT-PCR and zymography. The YD-10B oral cancer cells showed an increase in growth inhibition as the concentration of HEE increased. The combination of 200 µM cisplatin and 500 ㎍/ml HEE reduced the growth of the YD-10B oral cancer cells by more than 50% compared to cisplatin alone. When phorbol 12-myristate 13-acetate (PMA)-treated YD-10B oral cancer cells were co-treated with 200 µM cisplatin and 500 ㎍/ml HEE, both the mRNA expression and protein activity of the MMP-2 and MMP-9 decreased. In addition, the percentage of the sub-G1 phase, which indicates apoptosis ability, more than doubled when treated in combination with 200 µM cisplatin and 500 ㎍/ml HEE than when cisplatin alone was used. The results of this study therefore suggest the possibility of using a combination of HEE and cisplatin in the development of effective drugs to treat oral cancer.

Study on VOCs Emission Characteristic of Taxidermied Mounting Techniques (박제표본 제작방법에 따른 휘발성유기화합물 방출 특성 연구)

  • Biological materials, such as stuffed specimens, can release various acids or volatiles. There has been no research carried out on the emission characteristics of organic compounds generated from the preservatives used in taxidermy specimens or associated manufacturing materials and methods. Therefore, in order to identify the organic compounds generated from taxidermy specimens, a degradation experiment was conducted on specimens for each material and for storage specimens. To produce Ogye chicken specimens, naphthalene and borax were used as preservatives, and planer sawdust, newspaper, and polystyrene foam were used as the core body materials. The deterioration experiment was conducted for 2 weeks in a high-temperature environment(50℃) and a high-humidity environment (95%), with an Ogye chicken specimen (year 2015) kept in an animal storage facility. Results indicated that the concentration of organic compounds generated by the specimen in the high-temperature environment tended to be greater than that in the high-humidity environment. The preservatives benzene, toluene, xylene, and p-dichlorobenzene were detected in the specimens using naphthalene, confirming that naphthalene is a major organic compound release factor, and the specimens that used sawdust, newspaper, and polystyrene foam also exhibited organic compounds. This appears to have been due to degradation of the material. In addition, ammonia was detected in the specimens for each material due to decay. In particular, the specimens using borax at high temperature were subject to approximately 9 times higher rates of ammonia-related deterioration than the specimens using naphthalene. These results can be considered to result from the prevention of biological damage through insecticidal effects by accelerating the sublimation of naphthalene in a high-temperature environment. Naphthalene is a potentially carcinogenic substance, and when used as a preservative, proper use management is required. Taxidermy specimens can release various organic compounds depending on the manufacturing techniques used, so a systematic preservation management plan is required that depends on conditions such as the applicable manufacturing materials and preservatives.

Effect of Manganese Sulfate Concentration in Media on Production Speed of Insecticidal Crystal by Bacillus thuringiensis (배지 중 Manganese sulfate 농도가 Bacillus thuringiensis의 곤충독소 생성 시간에 미치는 영향)

  • In this study, the effect of MnSO4 on the insecticidal crystal (IC) produced by Bacillus thuringiensis for a rapid detection medium was analyzed. The strains used included one B. thuringiensis reference (KCTC 1511) and nine wild-type strains. The IC in B. thuringiensis was detected following the method published by the Ministry of Food and Drug Safety in Korea. In the nutrient agar to which 0.005% MnSO4 was added, IC was observed on two of the three plates after 48 hours of incubation and on all three plates after 120 hours. In AK agar, IC was observed on one and two of the three plates after 48 and 96 hours of incubation, respectively. These results indicated that 0.005% MnSO4 nutrient agar is more appropriate than AK agar for production of IC in B. thuringiensis. The effect of various MnSO4 concentrations on IC production was studied after 24 hours of incubation. IC was produced on 1 of the 10 plates with 0.000% MnSO4 nutrient agar, 2 of the 10 plates with 0.001% MnSO4 nutrient agar, and 3 of the 10 plates with 0.002% MnSO4 nutrient agar. IC was not observed for the other nutrient agars containing 0.003%-0.009% MnSO4. These results indicated that nutrient agar with 0.002% MnSO4 led to the most rapid production of IC by B. thuringiensis after 24 hours of incubation. However, the conditions for IC production by B. thuringiensis depended on the incubation conditions and strain activity. Therefore, further studies are needed to verify the effects of 0.002% MnSO4 on the production of IC by various Bacillus thuringiensis strains.
