Effects of magnesium sulfate ($MgSO_4$) and magnesium hydroxide $[Mg(OH)_2]$, which have different chemical characteristics, on soil pH, electrical conductivity (EC), and exchangeable cation distributions were investigated. Using plastic columns packed with a loam soil, the two Mg-fertilizers were treated at the rate of $300kg\;MgO\;ha^{-1}$ and water was applied on the soil surface four times at every 7 days. Soil samples were taken at 5, 10, 15, and 20 cm depth after 7 days of each water application. Magnesium hydroxide could increase soil pH, but due to the low solubility of $Mg(OH)_2$, the effect on pH was limited on the surface soil. Soil pH was lowered in the $MgSO_4$ treatment and the effect was found through the 20 cm depth. Since the pH decrease in $MgSO_4$ treated soil was due to the salts from $MgSO_4$, after leaching of most salts from the investigated soil depth pH was not significantly different from that of non-treated soil. Soil EC was increased in $MgSO_4$ treatment through the soil depth, but in $Mg(OH)_2$ treatment EC was slightly increased only at the surface layer. Exchangeable Mg was increased in both of the treatments at surface layer after the first water application. In $Mg(OH)_2$ treatment, the increase of exchangeable Mg was found only at 5 cm depth through the experiment, but leaching down of Mg in the $MgSO_4$ treatment was very apparent. High concentration of Mg in the $MgSO_4$ treated soil could effectively replace exchangeable Ca through the investigated soil depth, but the effect of $Mg(OH)_2$ on exchangeable Ca was not significant.
This study was conducted to improve the useful components and biological activity of Lentinula edodes fermented by lactic acid bacteria (LAB). Three LAB strains (Lactobacillus brevis KCCM 11904, L. plantarum KCCM 354469, and L. fermentum KCCM 12116) were inoculated and used for L. edodes hot water extract (10%, 20%, 30%) fermentation. LAB fermentation of L. edodes hot water extracts decreased pH and thus were more acidic than non-fermented L. edodes hot water extract. β-glucan and ergothioneine contents were increased by L. edodes in a concentration-dependent manner. The ergothioneine and β-glucan contents were highest in fermented with 30% L. edodes hot water extract fermented by L. plantarum and L. brevis (40.48 mg/100 g and 13.94%, respectively). The hepato-protective effect of fermented L. edodes hot water extracts by the three LAB were tested using Sprague-Dawley rat primary hepatocytes. In primary hepatocytes obtained following liver injury induced by acetaminophen, fermented L. edodes hot water extracts by the three LAB showed protective effects, as evident by reduction of the aspartate aminotransferase, alanine aminotransferase, and lactate dehydrogenase liver markers. The collective results indicate that the fermented L. edodes hot water extracts obtained using LAB are potentially valuable in preventing or treating liver disease.
Litsea populifolia, a plant species of the Lauraceae family, is widely distributed in the tropical and subtropical areas of Asia. The phylogenetic relationships and botanical characteristics of L. populifolia have been reported; however, its anti-oxidative and anti-cancer activities remain unclear. In this study, we evaluated the anti-oxidative and anti-cancer effects of ethanol extracts of L. populifolia (EELP) together with the molecular mechanism of its anti-cancer activity in human lung adenocarcinoma A549 cells. EELP showed significant anti-oxidative effects with a 50% inhibitory concentration at $11.71{\mu}g/ml$, which was measured by the 2,2-diphenyl-1-picrylhydrazyl radical scavenging assay. EELP exhibited cytotoxic activity and induced cell cycle arrest at the G1 phase in A549 cells in a dose-dependent manner, whereas EELP did not have the cytotoxic effect on the normal human lung cell line IMR90. Treatment with EELP also resulted in a decreased expression of G1/S transition-related molecules-including cyclin-dependent kinase (CDK) 2, CDK6, cyclin D1, and cyclin E-both for the transcription and translation levels. EELP-induced G1 arrest was associated with the phosphorylation of checkpoint kinase 2 (CHK2), p53, cell division cycle 25 homolog A (CDC25A), and the reduction of CDC25A expression in A549 cells. Collectively, these results suggest that EELP may exert an anti-cancer effect by cell cycle arrest at the G1 phase through both p53-dependent and p53-independent (ATM/CHK2/CDC25A/CDK2) pathways in A549 cells.
Effects of environmental factors on phytoplankton succession and community structure were studied in Lake Chuncheon located in Bukhan River, South Korea. The data were sampled at three sites such as CC1 (lower side), CC2 (middle side), and CC3 (upper side of Lake Chuncheon) from 2014 to 2017. The annual average precipitation in Lake Chuncheon was 992 mm during the study period (2014~2017), and the annual precipitation was lower than 800 mm in 2014 and 2015. The annual average water temperature, total phosphorus (TP), and total nitrogen (TN) ranged from 17.0 to $21.1^{\circ}C$, 0.012 to $0.019mg\;L^{-1}$, and 1.272 to $1.922mg\;L^{-1}$, respectively. The TN concentration was relatively high in 2015 compared with the other study years, as a drought continued from 2014 to 2015. When comparing the correlation between precipitation and environmental factors, water temperature (p<0.01) and TP(p<0.05) showed positive correlations with rainfall. The average numbers of phytoplankton cells by branch were 2,094, 2,182, and $3,108cells\;mL^{-1}$ in CC1, CC2, and CC3, respectively. CC3 is considered advantageous for phytoplankton growth, even in small pollution sources due to low water depth. As a result of analyzing the relationship between precipitation and phytoplankton, the correlation between the two was shown to be high for 2016 (p<0.01) and 2017 (p<0.05), which is when precipitation was high. However, the correlation was not clear to 2014 and 2015. The relationship between water temperature and phytoplankton indicated a negative correlation with diatoms (p<0.01), yet positive correlations with green algae (p<0.01) and cyanobacteria (p<0.01). Diatoms increased in spring and autumn, which are characterized by low water temperature, and green algae and cyanobacteria increased in summer, when the water temperature is high. Our findings provide a scientific basis for characteristics of phytoplankton and water quality and management at the Lake Chuncheon.

This study was undertaken to establish optimum medium compositions for cost-effective mannitol production by Leuconostoc mesenteroides SRCM201425 isolated from kimchi. L. mesenteroides SRCM21425 from kimchi was selected for efficient mannitol production based on fructose analysis and identified by its 16S rRNA gene sequence, as well as by carbohydrate fermentation pattern analysis. To enhance mannitol production by L. mesenteroides SRCM201425, the effects of carbon, nitrogen, and mineral sources on mannitol production were first determined using Plackett-Burman design (PBD). The effects of 11 variables on mannitol production were investigated of which three variables, fructose, sucrose, and peptone, were selected. In the second step, each concentration of fructose, sucrose, and peptone was optimized using a central composite design (CCD) and response surface analysis. The predicted concentrations of fructose, sucrose, and peptone were 38.68 g/l, 30 g/l, and 39.67 g/l, respectively. The mathematical response model was reliable, with a coefficient of determination of $R^2=0.9185$. Mannitol production increased 20-fold as compared with the MRS medium, corresponding to a mannitol yield 97.46% when compared to MRS supplemented with 100 g/l of fructose in flask system. Furthermore, the production in the optimized medium was cost-effective. The findings of this study can be expected to be useful in biological production for catalytic hydrogenation causing byproduct and additional production costs.

This study examined the effects of 2-methoxy-1,4-naphthoquinone (MQ) on the monocyte chemoattractant protein-1 (MCP-1)-induced migration of monocytes, which is an important phenomenon for the body defense and immune response. MQ is a major component extracted from Impatiens balsamina leaves, which have been used for many years in Asian medicine for the treatment of a range of diseases and pain. The cytotoxicity of MQ began to appear at a concentration of $10{\mu}M$, and approximately 50% cytotoxicity was confirmed at $100{\mu}M$. The MCP-1 induced migration of the THP-1 monocyte cell line increased after MQ treatment in a dose dependent manner and the largest increase was observed at $0.1{\mu}M$. The level of cAMP expression decreased after a treatment with $0.1{\mu}M$ MQ. The phosphorylation of extracellular signal-regulated kinases 1/2 (Erk1/2), a key signaling protein involved in the signaling pathway of C-C motif chemokine receptor 2 (CCR2), a receptor for MCP-1, was increased by the simultaneous treatment of $0.1{\mu}M$ MQ. These results show that MQ increases the MCP-1-induced migration of THP-1, decreases the level of cAMP expression, and increases the level of Erk1/2 phosphorylation.
The geological CO2 sequestration in underground geological formation such as deep saline aquifers and depleted hydrocarbon reservoirs is one of the most promising options for reducing the atmospheric CO2 emissions. The process in geological CO2 sequestration involves injection of supercritical CO2 (scCO2) into porous media saturated with pore water and initiates CO2 flooding with immiscible displacement. The CO2 migration and distribution, and, consequently, the displacement efficiency is governed by the interaction of fluids. Especially, the viscous force and capillary force are controlled by geological formation conditions and injection conditions. This study aimed to estimate the effects of surfactant on interfacial tension between the immiscible fluids, scCO2 and porewater, under high pressure and high temperature conditions by using a pair of proxy fluids under standard conditions through pendant drop method. It also aimed to observe migration and distribution patterns of the immiscible fluids and estimate the effects of surfactant concentrations on the displacement efficiency of scCO2. Micromodel experiments were conducted by applying n-hexane and deionized water as proxy fluids for scCO2 and porewater. In order to quantitatively analyze the immiscible displacement phenomena by n-hexane injection in pore network, the images of migration and distribution pattern of the two fluids are acquired through a imaging system. The experimental results revealed that the addition of surfactants sharply reduces the interfacial tension between hexane and deionized water at low concentrations and approaches a constant value as the concentration increases. Also it was found that, by directly affecting the flow path of the flooding fluid at the pore scale in the porous medium, the surfactant showed the identical effect on the displacement efficiency of n-hexane at equilibrium state. The experimental observation results could provide important fundamental information on immiscible displacement of fluids in porous media and suggest the potential to improve the displacement efficiency of scCO2 by using surfactants.

Euonymus porphyreus, a species of plant in the Celastraceae family, is widely distributed in East Asia, especially in Southern China. The botanical characteristics of E. porphyreus have been reported, but its antioxidative and anticancer activities remain unclear. In this study, we evaluated the antioxidative and anticancer effects of ethanol extracts of E. porphyreus (EEEP) and the molecular mechanism of its anticancer activity in human lung adenocarcinoma A549 cells. The total polyphenol and flavonoid compound contents from EEEP were 115.42 mg/g and 23.07 mg/g, respectively. EEEP showed significant antioxidative effects with a concentration at 50% of the inhibition (IC50) value of 11.09 ㎍/ml, as measured by 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay. EEEP showed cytotoxic activity by increasing the SubG1 cell population of A549 cells in a dose-dependent manner. Apoptosis in A549 cells treated with EEEP was evident due to increased apoptotic cells and apoptotic bodies, as detected by Annexin V and 4,6-diamidino-2-phenylindole (DAPI) staining, respectively. EEEP-induced apoptosis resulted in increased expression of the First apoptosis signal (Fas), p53, and Bax, with decreased expression of Bcl-2 and subsequent activation of caspase-8, -9, and caspase-3, leading to cleavage of poly (ADP-ribose) polymerase (PARP). Collectively, these results suggest that EEEP may exert an anticancer effect by inducing apoptosis in A549 cells through both intrinsic and extrinsic pathways.

This study examined the effects of a hydrothermal pretreatment (HP) on the nutritional values and in vitro fermentation characteristics of tall fescue and corn silage. This study was conducted through a factorial design of 2 (control or HP) × 2 (hay; tall fescue or silage; corn). For the HP, forage was placed into a glass bottle with 20% w/v of water, and the glass bottle was sealed and heated to reach a temperature of 121℃ (0.12 MPa). The solid residue and liquid were collected and oven-dried at 65℃ for three days. The dried materials were tested for in vitro fermentation at 39℃ for 24 and 48 h. The content of ADF increased significantly regardless of the forage type. After in vitro incubation for 24 h, the total VFA content was significantly lower after HP, regardless of the forage type (p ≤ 0.05), and the propionate concentration was increased in corn silage with HP (p ≤ 0.05). After 48 hours of in vitro incubation, the propionate content increased significantly (p ≤ 0.03) in corn silage with HP (p ≤ 0.05), but the butyrate content decreased significantly (p ≤ 0.05). There was no change in the in vitro dry matter and neutral detergent fiber digestibility by HP regardless of the forage type. Therefore, the use of hydrothermally pretreated corn silage could be advantageous for the supply of energy for ruminants.

Gingival inflammation is one of the main causes that can be related to various periodontal diseases. Human gingival fibroblast (HGF) is the major constituent in periodontal connective tissue and secretes various inflammatory mediators, such as nitric oxide (NO) and prostaglandin E2 (PGE2), upon lipopolysaccharide stimulation. This study is aimed at investigating the anti-inflammatory and antioxidative activities of Lotus Root extract (LRE) in Porphyromonas gingivalis derived lipopolysaccharide (LPS-PG)-stimulated HGF-1 cells. The concentration of NO and PGE2, as well as their responsible enzymes, inducible NO synthase (iNOS), and cyclooxygenase-2 (COX-2), was analyzed by Griess reaction, ELISA, and western blot analysis. LPS-PG sharply elevated the production and protein expression of inflammatory mediators, which were significantly attenuated by LRE treatment in a dose-dependent manner. LRE treatment also suppressed activation of Toll-like receptor 4 (TLR4)/myeloid differentiation primary response gene 88 (MyD88) and nuclear factor-κB (NF-κB) in LPS-PG-stimulated HGF-1 cells. In addition, one of phase II enzyme, NAD(P)H quinone dehydrogenase (NQO)-1, and its transcription factor, Nuclear factor erythroid 2-related factor 2 (Nrf2), were significantly induced by LRE treatment. Consequently, these results suggest that LRE ameliorates LPS-PG-induced inflammatory responses by attenuating TLR4/MyD88-mediated NF-κB, and activating NQO-1/Nrf2 antioxidant response element signaling pathways in HGF-1 cells.
