  • This study investigates the damages of and analyzes the social and economic impacts of volcanic ash eruptions in the world in order to estimate the potential volcanic ash impacts in South Korea when Mt. Baekdusan volcano erupts in the future. First, we build a comparison chart called "the impact of volcanic ash" on each economic and social sector by using major volcanic eruptions and we compare the damage with respect to volcanic ash thickness/weights. Secondly, we analyze the social and economic impact from volcanic ash. The economic damage is not likely to occur in South Korea, unless Mt. Baekdusan erupts in winter. However, the potential damage should not be overlooked because the volcanic ash may have a global impact around the world. If Mt. Baekdusan volcano erupts when the wind blows from north or northeast, the volcanic ash may then significantly affect South Korea of which economy is highly dependent on exports. Particularly when the volcanic ash moves to the densely populated metropolitan areas or agricultural areas, the damage can be significant. In preparation for the potential volcanic disasters, the volcanic ash forecast table suitable for South Korea should be prepared. In addition, building a Korean volcanic ash hazard map in advance will have a strategic significance.

  • $CO_2$ leakage from a geological formation utilized for $CO_2$ storage could result in failure of the facility and threaten the environment, as well as human safety and health. A reactive transport model of a $CO_2-H_2O$-cement reaction was constructed to understand chemical changes in the case of $CO_2$ leakage through a cement crack in an injection well, which is the most probable leakage pathway during geological storage. The model results showed the dissolution of portlandite and CSH (calcium silicate hydrate) within the cement paste, and the precipitation of secondary CSH and calcite as the $CO_2$ plume migrated along the crack. Calcite occupied most of the crack after 3 year of reaction, which could be maintained until 30 years after crack development. The present results could be applied in the development of technology to prevent $CO_2$ leakage and to enhance the integrity of wells constructed for $CO_2$ geological storage.

  • The rapidly changing 21st-century knowledge and information society is emphasizing converged education that crosses various academic fields. In particular, the society expected the cultivation of the talent who balance scientific creativity and artistic sensitivity by adding arts to the existing converged education revolving around science and technology. However, at present, most STEAM education has been actively conducted with a focus on science and technology, whereas the subject of arts has been regarded or utilized as a supplementary means. Its problem is that the educational characteristics and values of art education have not been effectively utilized in educational terms and this could lead to superficial integrated education. In this respect, this study had the knowledge of various fields, such as science, technology, and mathematics, utilized usefully during the process of experiencing and creating arts. Accordingly, this study designed an education programs as with the case of Nam-Jun Baek who expanded the dominion of arts by creatively utilizing his own time's scientific technologies. In this educational process, the target program was developed in a manner that enables EPL to be utilized essentially as the study's knowledge-based tool and medium. The results of applying this educational program in 5th-grade elementary school students showed that the program has positive effects on the creative attributes of the students.

  • Lately, intensity and frequency of natural disasters such as flood are increasing because of abnormal climate. Casualties and property damages due to large-scale floods such as Typhoon Rusa in 2002 and Typhoon Maemi in 2003 rapidly increased, and these show the limits of the existing disaster prevention measures and flood forecasting systems regarding irregular climate changes. In order to efficiently respond to extraordinary flood, it is important to provide effective countermeasures through an inundation model that can accurately simulate flood inundation patterns. However, the existing flood inundation analysis model has problems such as excessive take of analysis time and accuracy of the analyzed results. Therefore, this study conducted a flood inundation analysis by using the Gerris flow solver that uses quadtree grid, targeting the Baeksan Levee in the Nakdong River Basin that collapsed because of a concentrated torrential rainfall in August, 2002. Through comparisons with the FLUMEN model that uses unstructured grid among the existing flood inundation models and the actual flooded areas, it determined the applicability and efficiency of the quadtree grid-based flood inundation model of the Gerris flow solver.

  • There has been an increase for the landslide areas and restoration expenses due, in large part, to the increased locally heavy rains caused by recent climate change as well as the reckless development. This study carried out a slope stability analysis by the application of distributed wetness index, using the GIS-based infinite slope stability model, which took the root cohesion effect into consideration, for part of Mt. Umyeon in Seoul, where landslide occurred in July 2011, in order to compensate the defects of existing analysis method, and subsequently compared its result with the case on the exploitation of lumped wetness index. In addition, this study estimated the distributed wetness index by methodology, applying three methods of specific catchment area calculation: single flow direction (SFD), multiple flow direction (MFD), and infinity flow direction (IFD), for catchment area, one of the variables of distributed wetness indices, and finally implemented a series of comparative analysis for slope stability by methodology. The simulation results showed that most unstable areas within the study site were dominantly located in cutting-area surroundings along with the residential area and the mountaintop and unstable areas of IFD and lumped wetness index method were similar while SFD and MFD provided smaller unstable areas than the two former methods.

  • Numerical modeling is commonly used to reproduce the physical phenomena of dam-break and to compile resulting flood hazard maps. The accuracy of a dam-break model depends on the physical structure that describes the volume of storage, breach formation and progress, input variables, and model parameters. Model input and parameters are subjective in that they are prescribed; hence, caution is needed when interpreting the results. This study focuses on three parameters (breach degree ${\theta}$, shape factor P, and collapse rate k) used when the dam-break model is coupled with FLO-2D (a two-dimensional flood simulation model) to estimate flood coverage and depth etc. The results show that the simulation is sensitive to the shape factor P and the collapse rate k but not to the breach degree ${\theta}$. This study will contribute to reducing flood damage from dam-break disasters in the future.

  • There are various types of real world signals. For example, an electrocardiogram(ECG) represents myocardium activities (contraction and relaxation) according to the beating of the heart. ECG can be expressed as the fluctuation of ampere ratings over time. A signal is a composite of various types of signals. An orchestra (which boasts a beautiful melody) consists of a variety of instruments with a unique frequency; subsequently, each sound is combined to form a perfect harmony. Various research on how to to decompose mixed stationary signals have been conducted. In the case of non-stationary signals, there is a limitation to use methodologies for stationary signals. Huang et al. (1998) proposed empirical mode decomposition(EMD) to deal with non-stationarity. EMD provides a data-driven approach to decompose a signal into intrinsic mode functions according to local oscillation through the identification of local extrema. However, due to the repeating process in the construction of envelopes, EMD algorithm is not efficient and not robust to a noise, and its computational complexity tends to increase as the size of a signal grows. In this research, we propose a new method to extract a local oscillation embedded in a signal by utilizing the second derivative.

  • Type I Interferons (IFNα) are known for their role as biological anticancer agents owing to their cell-apoptosis inducing properties. Development of an appropriate, cost-effective host expression system is crucial for meeting the increasing demand for proteins. Therefore, this study aims to develop codon-optimized IFNα-2b in L. lactis NZ3900. These cells express extracellular protein using the NICE system and Usp45 signal peptide. To validate the mature form of the expressed protein, the recombinant IFNα-2b was screened in a human colorectal cancer cell line using the cytotoxicity assay. The IFNα-2b was successfully cloned into the pNZ8148 vector, thereby generating recombinant L. lactis pNZ8148-SPUsp45-IFNα-2b. The computational analysis of codon-optimized IFNα-2b revealed no mutation and amino acid changes; additionally, the codon-optimized IFNα-2b showed 100% similarity with native human IFNα-2b, in the BLAST analysis. The partial size exclusion chromatography (SEC) of extracellular protein yielded a 19 kDa protein, which was further confirmed by its positive binding to anti-IFNα-2b in the western blot analysis. The crude protein and SEC-purified partial fraction showed IC50 values of 33.22 ㎍/ml and 127.2 ㎍/ml, respectively, which indicated better activity than the metabolites of L. lactis NZ3900 (231.8 ㎍/ml). These values were also comparable with those of the regular anticancer drug tamoxifen (105.5 ㎍/ml). These results demonstrated L. lactis as a promising host system that functions by utilizing the pNZ8148 NICE system. Meanwhile, codon-optimized usage of the inserted gene increased the optimal protein expression levels, which could be beneficial for its large-scale production. Taken together, the recombinant L. lactis IFNα-2b is a potential alternative treatment for colorectal cancer. Furthermore, its activity was analyzed in the WiDr cell line, to assess its colorectal anticancer activities in vivo.
