cis-Dichlorodiammineplatinum (II) (cis-Platin), a metallic compound, has widely been used as an effective anticancer chemotherapeutic agent. The precise mechanism of action of this agent is still unknown, but it is postulated that cis-Platin may act on the cancer cell like bifunctional alkylating agents. Although this agent is very beneficial to the patients with cervical cancer, germinoma of testis, neuroblastoma and others, it may also damage to the normal cell so that many side effects; severe hemorrhagic enterocolitis, bone marrow depression, renal damage and liver damage will develope. This experiment has been undertaken to pursue the cytotoxic effects of the cis-Platin on the ultrastructures of the interalveolar septum in the mouse lung. A total of 55 healthy male mice of ICR strain were used as experimental animals and divided into 5 mice of normal control group and 50 mice of cis-Platin treated group. The mice of cis-Platin treated group were sacrificed by carotid exsanguination at 6, 12, 24 hours, 3 days and 7 days after intraperitoneal injection of 6.0 mg of cis-Platin ($Abiplatin^R$ Abic Co. Ltd.) per kg of mouse body weight. The specimen obtained from the lower lobe of left lung were sliced into $1mm^3$ and prefixed with 2% glutaraldehyde -2.5% paraformaldehyde solution prepared with Millonig's phosphatae buffer solution (pH 7.4) at $4^{\circ}C$ for 3-4 hours. After postfixation with 1% osmium tetroxide solution all specimens were embedded in Epon 812. Ultrathin sections about $600-800{\AA}$ in thickness were stained with uranyl acetate and lead citrate and observed with Hitachi-600 electron microscope. The results obtained were as follows: 1. Local swellings with increase of electron density and number of pinocytic vesicles in the cytoplasms of the type I pneumocyte and endothelial cell of the blood air barrier in interalveolar septum of cis-platin treated mice were observed. 2. Cisternae of rough endoplasmic reticulum were dilated and sacculated in association with detachment of membrane bound ribosomes of the type II pneumocyte in interalveolar septum of cis-Platin treated mice. 3. Swollon mitochondria with uneven electron density of their matrix were observed in the type II pneumocyte of interalveolar septum in the cis-Platin treated mice. 4. The lamellae of lammelar bodies in type II pneumocyte of interalveolar septum in cis-Platin treated mice were devoided or transformed into homogeneous electron dense material. It is consequently suggested that cis-Platin would induce the cellular edema of type I pneumocyte and endothelial cell, and degenerative changes of cytoplasmic organelles of the type II pneumocyte in the interalveolar septum of the mouse lung.
Prostaglandin $E_2$ ($PGE_2$), a major product of cyclooxygenase-2 (COX-2), plays an important role in the carcinogenesis of many solid tumors, including colorectal cancer. Because $PGE_2$ functions by signaling through $PGE_2$ receptors (EPs), which regulate tumor cell growth, invasion, and migration, there has been a growing amount of interest in the therapeutic potential of targeting EPs. In the present study, we investigated the role of EP4 on the effectiveness of cordycepin in inhibiting the migration and invasion of HCT116 human colorectal carcinoma cells. Our data indicate that cordycepin suppressed lipopolysaccharide (LPS)-enhanced cell migration and invasion through the inactivation of matrix metalloproteinase (MMP)-9 as well as the down-regulation of COX-2 expression and $PGE_2$ production. These events were shown to be associated with the inactivation of EP4 and activation of AMP-activated protein kinase (AMPK). Moreover, the EP4 antagonist AH23848 prevented LPS-induced MMP-9 expression and cell invasion in HCT116 cells. However, the AMPK inhibitor, compound C, as well as AMPK knockdown via siRNA, attenuated the cordycepin-induced inhibition of EP4 expression. Cordycepin treatment also reduced the activation of CREB. These findings indicate that cordycepin suppresses the migration and invasion of HCT116 cells through modulating EP4 expression and the AMPK-CREB signaling pathway. Therefore, cordycepin has the potential to serve as a potent anti-cancer agent in therapeutic strategies against colorectal cancer metastasis.
Urine is a widely used matrix in biomonitoring studies on the assessment of human exposure to environmental chemicals such as phthalate esters and bisphenol A (BPA). In addition to the need to apply valid analytical techniques, assurance of specimen integrity during collection and storage is an important prerequisite for the presentation of accurate and precise analytical data. One of the common issues encountered in the analysis of non-persistent contaminants is whether shipping and storage temperature and time since collection have an effect on sample integrity. In this study, we investigated the stability of phthalate metabolites and BPA in spiked and unspiked urine samples stored at room temperature ($20^{\circ}C$) or at $-80^{\circ}C$ for up to 8 weeks. Concentrations of phthalate metabolites declined, on average, by 3% to 15%, depending on the compounds, and BPA declined by ~30% after 4 weeks of storage of spiked urine samples at $20^{\circ}C$. In a test of 30 unspiked urine samples stored at $20^{\circ}C$ and at $-80^{\circ}C$ for 8 weeks, the concentrations of phthalate metabolites and BPA decreased by up to 15% to 44%, depending on the compound and on the samples. It was found that the small reduction in phthalate concentrations observed in urine, varied depending on the samples. In a few urine samples, concentrations of phthalate metabolites and BPA did not decline even after storage at $20^{\circ}C$ for 8 weeks. We found a significant relationship between concentrations of target analytes in urine stored at $20^{\circ}C$ and at $-80^{\circ}C$ for 8 weeks. We estimated the half-lives of phthalate metabolites and BPA in urine stored at $20^{\circ}C$. The estimated half-life of monoethyl phthalate (mEP) and mono (2-ethyl-5-carboxyphentyl) phthalate (mECPP) in urine stored at $20^{\circ}C$ was over two years, of mono (2-ethyl-5-oxohexyl) phthalate (mEOHP) and monobenzyl phthalate (mBzP) was approximately one year, and of other phthalate metabolites was approximately 6 months. The estimated half-life of BPA in urine stored at $20^{\circ}C$ was approximately 3 months, which is much longer than that reported for aquatic ecosystems.

This paper is on the modeling of two-dimensional groundwater flow, which is the first step of the development of Dynamic System Model for groundwater flow and pollutant transport in subsurface porous media. The particular features of the model are its versatility and flexibility to deal with as many real-world problems as possible. Points as well as distributed sources/sinks are included to represent recharges/pumping and rainfall infiltrations. All sources/sinks can be transient or steady state. Prescribed hydraulic head on the Dirichlet boundaries and fluxes on Neumann or Cauchy boundaries can be time-dependent or constant. Sources/sinks strength over each element and node, hydraulic head at each Dirichlet boundary node and flux at each boundary segment can vary independently of each other. Either completely confined or completely unconfined aquifers, or partially confined and partially unconfined aquifers can be dealt with effectively. Discretization of a compound region with very irregular curved boundaries is made easy by including both quadrilateral and triangular elements in the formulation. Large-field problems can be solved efficiently by including a pointwise iterative solution strategy as an optional alternative to the direct elimination solution methed for the matrix equation approximating the partial differential equation of groundwater flow. The model also includes transient flow through confining leaky aquifers lying above and/or below the aquifer of interest. The model is verified against three simple cases to which analytical solutions are available. The groundwater flow model shall be combined with the model of pollutant transport in subsurface porous media. Then the combined model, with the applications of the Eigenvalue technique and the Dynamic system theory, shall be improved to the Dynamic System Model which can simulate the real groundwater flow and the pollutant transport accurately and effectively for the analyses and predictions.
The studies for the derivatization of 3-monochloropropane-1,2-diol (3-MCPD) were performed mainly as acylation with HFBI (heptafluorobutyrylimidazole), alkylation with PBA (phenylboric acid) and silylation with BSTFA (N,O-bis[trimethylsilyl]trifluoroacetamide). Also silylation with MTBSTFA(N-methyl-N-[tert.-butyldimethylsilyl] trifluoroacetamide) and acylation with MBTFA (N-Methyl-bis[trifluoro-acetamide]) were also considered. Except the TBDMS derivative of 3-MCPD, all the derivatives were detected well. The derivatives of 3-MCPD with HFBI, PBA and BSTFA showed below 10 ${\mu}g/kg$ which was sensitive enough to satisfy Korea maximum residue limit 0.3 mg/kg. Among the tested adsorbents, Extrelut20 and Florisil were evaluated as the proper adsorbents to eliminate the soy sauce matrix for 3-MCPD. Ethyl acetate was the most efficient eluent with good recovery rate. The desired surrogate compound and internal standard were 1,2-butanediol and 1,2-dibromo-3-chloropropane, respectively. The limit of detection for PB-MCPD and TMS-MCPD were 10.16 and 7.06 ${\mu}g/kg$ on GC/MSD, respectively. HFB-MCPD derivative showed the lowest detection limits 2.98 and 5.32 ${\mu}g/kg$ by GC/ECD and GC/MSD, respectively.

Niemann Pick type C disease (NPC) is an inherited progressive neurodegenerative disorder, due to defects of intracellular lipid trafficking and storage. Hepatosplenomegaly may prevail, while progressive neurodegenerative symptoms such as cerebellar involvement, dystonia, vertical supranuclear ophthalmoplegia, cataplexy, and eventually seizures starting at juvenile or late infantile period may accompany after normal early development. Here we describe a 3-year-old Korean boy with NPC who presented with splenomegaly at age 3. Liver biopsy showed characteristic foamy cell stained by periodic acid-schiff, and molecular analysis for NPC1 identified the compound heterozygous mutations, novel mutation of c.1631G>A (p.Trp544Ter) and c.2662C>T (p. Pro888Ser) as a known mutation. Filipin was strongly stained with unesterified cellular cholesterol in the patient's skin fibroblasts. The patient has received migulstat since age 3 years and his long-term outcome is needed to be observed.

Twelve kinds (1set) standard materials of chemical ingredients of lead zirconate titanate[$Pb(ZrTi)O_3$] have been developed in order to determine fast and accurate measurement of X-ray fluorescence spectrometry. Especially, we used diluted(ahout sixteen times) filling compound($Li_2B_4O_7/LiBO_2=4/1$) to consider removal effect of matrix, storage convenience, and homogenous characteristics. As a result from the four different laboratories, we obtained extremly good agreement about the standard curve on twelve standard materials which containing eleven elements, PbO, $ZrO_2$, $TiO_2$, SrO, $WO_3$, $La_2O_3$, $Cr_2O_3$, MgO, $Nb_2O_5$, and $MnO_2$. The correlation factor of standard curve was over 0.998. However, ZnO has relatively low correlation factor, 0.977, because the concentration was 10ppm lower than other original materials. This analysis reveals that ZnO has shown the poor linearity as well as low fluorescence intensity. In present work, XRF standard materials are useful for determining a rapid and accurate results for major and minor elements concentration among PZT.

Recently, the applications of carbon fiber reinforced plastics (CFRPs) have become broader than ever when it comes to such industries as automotive, ships, aerospace and military because of their lightweight-ness and high mechanical properties. Thermosetting plastics like epoxy are frequently used as the binding matrix in CFRPs due to their high hardness, wetting characteristics and low viscosity. However, they cannot melted and remolded. For this reason, thermosetting plastic wastes have caused serious environmental problems with the production of fiber reinforced plastics. Thus, many studies have focused on the carbon fiber reinforced thermoplastics (CFRTPs) and recycling carbon fiber. In this study, recycled carbon fiber (RCF) was prepared from CFRPs using a pyrolysis method, which was employed to separate resin and carbon fiber. The degree of decomposition for epoxy resin was confirmed from thermal gravimetric analysis (TGA) and scanning electron microscope (SEM). The RCF was cut and ground to prepare a carbon fiber composite sheet (CFCS). CFCS was manufactured by applying recycled carbon fibers and various thermoplastic fibers. Various characterizations were performed, including morphological analyses of surface and cross-section, mechanical properties, and crystallization enthalpy of CFCS at different cooling conditions.

Yuza (Citrus junos Sieb ex TANAKA) is a citrus fruit that is cultivated in northeast Asia. Citron is known for containing abundant antioxidants such as vitamin C, flavonoids, for example hesperidin and hesperetin, and terpenoids such as limononin. When mature citron is processed for tea or other beverage food products in Korea, massive amounts of seeds and pericarp are remained as waste. This study aimed to exploit the processed remnant of Citron for developing functional cosmetic applications. Ethanol extracts of Yuza seed and pericarp did not show significant radical scavenging activities measured by DPPH (2,2-diphenyl-1-picrylhydrazyl) method. But they contained significantly high phenolic compounds. Cultured human dermal fibroblasts and HaCaT keratinocytes were irradiated with 25 mJ UVB and the citron extracts were added to the medium of each culture. Cellular damages caused by UVB irradiation were prevented by the addition of the Yuza extract. In addition, the reduction of the enhanced MMP-1 expression after irradiation of UVB in human dermal fibroblasts was observed. Also the increased level of pro-inflammtory TNF-$\alpha$ in the UVB irradiated HaCaT cells was decreased. The collagen expression was enhanced by the extract. Yuza extract markedly inhibited melanin production from $\alpha$-MSH treated B16F1 melanoma cells. Melanin assay, tyrosinase zymography results indicated that Yuza extract had strong depigmenting activity. In conclusion, Yuza ethanol extracts have good anti-photoaging and strong anti-melanogenic efficacies.
The protective effects of extract powder of Angelica gigas on the degeneration of the articular cartilage in rats was investigated with monosodium iodoacetate (MIA)-induced osteoarthritis, The treatment of high concentration (50 μg/mL) of Angelica gigas effectively inhibited nitric oxide (NO) production induced by interleukin-1α (IL-1α) without any cytotoxicity. Specifically, mRNA and protein expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) were dose dependently reduced by extract powder of Angelica gigas. Importantly, mRNA expression in articular cartilage of inflammatory cytokines, tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β) and interleukin-6 (IL-6) were clearly reduced. The inflammatory cytokines in blood were also reduced as well. These results suggested that the protective effects on the degeneration of the articular cartilage was derived from the inhibitory effects of mRNA and protein expression of tested inflammatory cytokines which is linked to prevent the degradation of proteoglycan (PG), the main matrix content in articular cartilage. Meanwhile, the 2 hrs incubation of decursin, a major compound of extract powder in rat whole blood rapidely converted decursin into decursinol which shows string anti-inflammatory activity. The coverted decursinol was detected after 8 hrs in whole blood by LC-MS/MS. Conclusively, the inhibitory effects of inflammatory cytokines production in osteoarthritis may be derived from the production of decursinol, which performs against inflammatroy cytokines like TNF-α, IL-1β, and IL-6.
