Antimicrobial Effect of Extract of Glycyrrhiza uralensis on Methicillin-Resistant Staphylococcus aureus (감초 추출물이 항생제 내성균주의 항균활성에 미치는 영향)

  • Antimicrobial drug-resistance is natural response to antimicrobial stress based on selection, which weakens chemotherapy effect. Introduction of large numbers of chemotherapeutic agents to clinical practice has generated strains of microorganisms that survive and multiply in vivo with high-drug concentrations. Methicillin-resistant Staphylococcus aureus (MRSA), bacteria found in normal daily life, can be easily ingested through milk vegetables, and meats, etc. MRSA emerged in many port of the world, increasing complex clinical problems. Therefore, new agents are needed to treat MRSA. Glycyrrhiza uralensis was extracted using 80% MeOH to investigate its antimicrobial activity against MRSA stains KCCM 11812, 40510, and 40512 through bacterial measurement, disc diffusion, and O.D. methods, MIC values, MRSA gene expression investigation, and scanning electron microscope observation. Results revealed MecA, Mecl, MecRI, and FemA were the most highly manifested MRSA genes. Methanolic extract of G. uralensis significantly inhibited MRSA and thus could be used in development of antibacteria.

Fretting Wear Test of Inconel 690 Tubes Employing Piezoelectric Actuator (압전 구동기를 이용한 인코넬 690 튜브의 프레팅 마멸시험)

  • A fretting wear test rig for dry ambient condition, which employs a piezoelectric actuator, has been developed. It is driven and loaded in a very simple manner with acceptable experimental accuracy. By using the rig, Inconel 690 tube has been tested under the normal load of 10 and 15N with sliding amplitude of less than $100{\mu}m$ during $10^6$cycles. The wear resistance of the material has been characterized in terms of the wear coefficient based on the work rate model. SEM micrographs show the complex structures of the scars, which consist of risen peaks, plate-type thin layers and locally exposed bare surfaces. The cracks spread over the layers give clue to the fretting wear mechanism of the material.

  • In this paper, we perform a numerical analysis to evaluate the potential of fault reactivation during gas production from hydrate bearing sediments and the moment magnitude of induced seismicity. For the numerical analysis, sequential coupling of TOUGH+Hydrate and FLAC3D was used and the change in effective stress and consequent geomechanical deformation including fault reactivation was simulated by assuming that Mohr-Coulomb shear resistance criterion is valid. From the test production simulation of 30 days, we showed that pore pressure reduction as well as effective stress change hardly induces the fault reactivation in the vicinity of a production well. We also investigated the influence of stress state conditions to a fault reactivation, and showed that normal fault stress regime, where vertical stress is relatively greater than horizontal, may have the largest potential for the reactivation. We tested one simulation that earthquake can be induced during gas production and calculated the moment magnitude of the seismicity. Our calculation presented that all the magnitudes from the calculation were negative values, which indicates that induced earthquakes can be grouped into micro-seismic and as small as hardly perceived by human beings. However, it should be noted that the current simulation was carried out using the highly simplified geometric model and assumptions such that the further simulations for a scheduled test production and commercial scale production considering complex geometric conditions may produce different results.

  • Globalization has been the buzzword during the past couple of decades, embroiling humankind into the inescapable maelstrom of homogenization. Some critics identify the globalization process undergoing in the realms of politics, economy, culture and ecology with Americanization which entails Neoconservatives' scheme to stretch out the hegemony of the United States. The top-down transcontinental movement, however, deems to confront localization or a reasoned resistance from the local driven by the spiritual attachment to places and, in that sense, regional identity. What is needed to cope with this complex circumstance of glocalization is to be acquainted with beth the global sense of the local and the localized sense of the global at once. For this, it seems indispensible to do justice to area studies. As far as American studies are concerned, this research field has so far been tainted by an overdose of politics, economics, business administration, law and sociology. Regional analysis which constitutes the reason d'etre of the discipline of geography has remained marginal to the political economic mainstream. It is the mounting concerns about America and enhanced research caliber that raises the regional geographical studies of America on the right track. Particularly, a number of articles have been published since the mid-1990s owing to the combined efforts of practitioners, institutions and periodicals. It is expected that systematic training of new generation of American specialists, advance in research infrastructure, and generous funding will stimulate geographical investigation of America in the coming future.

Characterization of alkaline cellulase from Bacillus subtilis 4-1 isolated from Korean traditional soybean paste (전통 장류에서 분리된 알칼리성 Cellulase 생성 Bacillus subtilis 4-1 균주의 효소학적 특성)

  • In this study, we isolated a cellulase-producing bacterium isolated from traditional Korean fermented soybean paste and investigated the effect of culture conditions on the production of cellulase. This bacterium, which was identified as Bacillus subtilis 4-1 through 16S rRNA gene sequence analysis, showed the highest cellulase activity when the cells were grown at $45^{\circ}C$ for 24 hours in the CMC medium supplemented with 1.0% of soluble starch and 0.1% yeast extract. The initial optimum pH of the medium was observed in the range of 5.0~9.0. The optimal pH and temperature for the production of cellulase from B. subtilis 4-1 were pH 9.0 and $60^{\circ}C$ respectively. In addition, the enzyme showed significant activity in the temperature range of $20{\sim}90^{\circ}C$, which indicates that B. subtilis 4-1 cellulase is an alkaline-resistance and thermo-stable enzyme. This enzyme showed higher activity with CMC as the substrate for endo-type cellulase than avicel or pNPG as the exo-type substrates for exo-type cellulase and ${\beta}$-glucosidase. These results suggest that the cellulase produced from B. subtilis 4-1 is a complex enzyme rather than a mono-enzyme.

Antibiotics Resistance and Molecular Analysis of Enterococcus Isolated from the Han-river in Korea (한강에서 분리한 장구균의 항생제 내성과 분자생물학적 분석)

  • Identification was performed in March 2008 for the 76 Enterococcus strains isolated from the Han-river, which is used as water supply for Seoul citizens. The antibiotic susceptibility, antibiotic resistant structural analysis, trans-conjugation, pulsed-field gel electrophoresis (PFGE) and multi-locus sequence typing (MLST) were also carried out for the isolated strains. Among the isolated strains, 25 strains were E. casseliflavus, 4 strains were E. faecalis and 1 strain was E. hirae. Investigation of antibiotic susceptibility indicated that 15 strains demonstrated tolerance against vancomycin, and that 11 strains of E. faecium and 4 strains of E. casseliflavus were VRE. The vanA gene detection of the VRE strains revealed that 6 E. faecium strains were vancomycin-resistant Enterococcus faecium (VREF) possessing vanA. Analyses of transposon Tn1546 structure containing vanA demonstrated that Km36 and Km37 belonged to Tn1 type, Km20 and Km38 was Tn2 type, and Km39 and Km40 was Tn3 type. PFGE disclosed that among the 6 VREF strains, Km36 and Km37 exhibited equivalent subtype, while the rest 4 strains showed subtypes different to each other. MLST for the 6 VREF strains disclosed that 3 strains were ST78, while the rest 3 strains were ST18, ST192 and ST230, respectively. All these clonal complexes were derived from CC17 which has been isolated from clinical sources. 4 strains belonged to CC78, while the rest 2 strains were CC18 and CC192, respectively.

Impedance Parameters of Electrical Double Layer I. A Determination Method of Electrolytic Cell Impedance Parameter on the Platinum Electrode (전기이중층의 임피던스 파라미터 I. 백금전극을 사용한 전해쎌 임피던스 파라미터의 결정방법)

  • This study is focused on the correct measurement of the equations for the determination of the impedance parameters-the differential capacity of the double layer $C_d$, solution resistance $R_Q$, transfer resitance $R_i$, and adsorption pseudcapacity $C_{\phi}$/ The application of only an imaginary part of complex function of ${\omega}$ at the sinusoidal steady state indicates the following equations of total impedance: at low frequency $|Z_{LF}|=1/{\omega}_1\;C_{\phi}\;{\sqrt{1+{{\omega}_1}^2/{\omega}^2}$, at high frequency $|Z_{HF}|={\omega}_2/({\omega}_1{\omega}_3C{\phi})({\omega}^2+{{\omega}_2}^2)\;{\sqrt{{({\omega}^2+{\omega}_2{\omega}_3)}^2+{({\omega}_2{\omega}-{\omega_3{\omega})^2}}$. The values of the total impedance of cell, phase angle, and cell current that are necessary for the calculations of impedance parameters were experimentally measured from 200 to 6000Hz for the following supporting electrolytes, 0.5M $Na_2SO_4$, 1M NaCl, 19.373% sea water, 1M HCl, 1M $KNO_3$ and for $10^{-2}M$ KI and 60mM DBNA (Di-iso-Butylnitrosoamine) in these supporting electrolytes. The derived equations in this study shows that the values of impedance parameters of $C_d,\;C_{\phi},\;R_i\;and\;R_Q\;are\;15{\sim}40\;{\mu}F/cm^2,\;162{\sim}758\;{\mu}F/cm^2\;11.5{\sim}57.6\;ohm{\cdot}cm^2\;and\;0.5{\times}10^{-2}{\sim}4.1{\times}10^{-2}\;ohm{\cdot}cm^2$ respectively.

The Association of HLA-DRB1 and DQB1 Alleles and a Study of Anticardiolipin Antibody and Anti-β2 Glycoprotein I Antibody in Korean SLE Patients (한국인 전신성홍반성루푸스 환자에서 HLA-DRB1, DQB1 대립유전자의 연관성 및 항인지질 항체와 항β2 Glycoprotein I 항체에 관한 연구)

  • Background: Systemic lupus erythematosus (SLE) is a complex autoimmune disease characterized by diverse clinical manifestations and autoantibody production, which is known to be strongly influenced by genetic factors. Previous studies have revealed the associations of SLE with HLA class II alleles and antiphospholipid antibody system (anticardiolipin antibody (aCL) and anti-${\beta}_2$ glycoprotein I antibody (anti-${\beta}_2$ GPI)). Therefore, we studied the associations of HLA class II alleles with SLE and antiphospholipid antibody system. Methods: The genotyping for HLA-DRB1 and DQB1 alleles were performed in 61 SLE patients and 100 controls by the polymerase chain reaction (PCR)-sequence specific oligonucleotide probe method. ELISA tests for aCL and anti-${\beta}_2$ GPI were performed in 39 of the 61 SLE patients. The results were evaluated statistically by Chi-square test. Results: The frequencies of the HLA-$DRB1^*15$ and $DQB1^*06$ in SLE patients were significantly higher than those in controls. HLA-$DRB1^*12$ was significantly lower in SLE patients than controls. Nine of 39 patients were positive for aCL (IgG) and three were positive for aCL (IgM). One of 39 patients were positive for anti-${\beta}_2$ GPI (IgG) and none of them positive for anti-${\beta}_2$ GPI (IgM). Association of aCL with HLA class II alleles was not observed in our study. Conclusion: According to our results, it was found that HLA-$DRB1^*15$ and $DQB1^*06$ were associated with genetic susceptiblility and $DRB1^*12$ was associated with resistance to SLE in Korean population. No Association of aCL with HLA class II alleles was observed and the positive rate for anti-${\beta}_2$ GPI was very low.

Development of Nonlinear Spring Modeling Technique of Group Suction Piles in Clay (점성토 지반에 근입된 그룹 석션파일에 대한 비선형 스프링 모델링 기법 개발)

  • Recently, several researches on the development of new economical anchor systems have been performed to support floating structures. This study focused on the group suction piles, which connect mid-sized suction piles instead of a single suction pile with large-diameter. The group suction pile shows the complex bearing behavior with translation and rotation, so it is difficult to apply conventional design methods. Therefore, the numerical modeling technique was developed to evaluate the horizontal bearing capacity of the group suction piles in clay. The technique models suction piles as beam elements and soil reaction as non-linear springs. To analyze the applicability of the modeling, the horizontal load-movement curves of the proposed modeling were compared with those of three-dimensional finite element analyses. The comparison showed that the modeling underestimates the capacity and overestimate the displacement corresponding to the maximum capacity. Therefore, the correction factors for the horizontal soil resistance was proposed to match the bearing capacity from the three-dimensional finite element analyses.

  • Simulated Moving Bed(SMB) process consists of multiple chromatographic columns, which are usually partitioned into four zones. Such a process characteristic allows a continuous binary separations those are impracticable in conventional batch chromatographic processes. Compared with batch chromatography, SMB has advantages of continuity, high purity and productivity. Various researches have been reported for the integration of reaction and recovery during process operation on the purpose of economics and effectiveness. Simulated Moving Bed Reactor(SMBR) is introduced to combine SMB as a continuous separation process and reactor. Several cases of SMBR have been reported for diverse reactions with catalytic, enzymatic and chemical reaction on ion exchange resin as main streams. With an early type of fixed bed using catalyst, SMBR has been developed as SMB using fluidized enzyme, SMB with immobilized enzyme and SMB with discrete reaction region. For simple modeling and optimization of SMBR, a method considering convection only is possible. A complex method considering axial dispersion and mass transfer resistance is needed to explain the real behavior of solutes in SMBR. By combining reaction and separation, SMBR has benefits of lower installation cost by minimizing equipment use, higher purity and yield by avoiding the equilibrium restriction in case of reversible reaction.
