  • 식육의 소비형태가 변화함에 따라 생체 기능성물질의 탐색과 이용에 관한 연구가 활성화 되어지고 있다. 식품에 소량 존재하며 지방세포분화를 촉진하는 물질로 밝혀진 conjugated linoleic acid (CLA)중 9c-11c(9c)가 3원 교잡종 암퇘지(Yorkshire ${\times}$ Landrace ${\times}$ Duroc)의 어떤 유전자에 영향을 줌으로써 지방세포의 분화를 촉진하는지 DNA chip을 이용하여 분석하였다. 분석도구로는 Genepix 6.0과 Vector Xpression 3.0을 사용하였다. 분석결과 $Na^+,\;K^+$-ATPase, ciliary neurotrophic factor, complement cytolysis inhibitor, prolactin receptor, type II collagen alpha1 등 과 같은 분화 관련 유전자가 나타났다. 이 연구는 나아가 DNA chip을 이용해서 지방세포 분화억제에 관여하는 유전자를 찾아내고, 이 유전자들은 지방세포 분화 조절과 비만관련 연구에 활용될 것이다.

  • The concrete mixer truck which is in charge of raw materials in civil engineering construction of the concrete loading, transport, placement, is used $6m^3$, $7m^3$ class in domestic(Korea). But in the case of the international construction fields are utilized $9m^3$ or above class because of the large-scale engineering and construction circumstances. In this paper, to develop a large $9m^3$ class mixer drum and the mixer drum in order to complement the technical and discharge that is responsible for stirring the blades by applying optimal design through implementation of the optimal shape of the concrete in the drum maintenance and placement of high-quality effects on increasing discharge such as advanced conventional drum mixer is to secure and differentiated technology. Large, heavy weight in development and uphold the drum mixer vehicle sub-frame is required to settle the design of the existing class mixer drum frames per $6m^3$ changed to account for changes in slope and length using CATIA V5 3D modeling work was performed.

Cloning of the Genomic DNA Which Complements the Drug-Hypersensitivity of Saccharomyces cerevlsiae

  • The yeast Saccharomyces cerevisiae, mutant CH117, shows a drug-hypersensitivity (dhs) to cycloheximide, bleomycin, actinomycin D, 5-fluorouracil. nystatin, nigericin and several other antibiotics. CH 117 was also temperature-sensitive (ts). being unable to grow at $37^{\circ}C$ and secreted more invertase and acid phosphatase into the medium than the parent yeast. CH117 grows very slowly and the cell shape is somewhat larger and more sensitive to zymolyase than the wild type cells. Light microscopic and electron microscopic observation also revealed abnormality of the mutant cell wall. These characteristics indicate that CH117 has a defect in an essential component of the cell surface and that the cell wall which performs barrier functions has become leaky in the mutant. We screened a genomic library of wild type yeast for clones that can complement the mutation of CH117. A plasmid, pCHX1, with an insert of 3.6 kilobases (kbs) could complement the dhs and ts of CH117. Deletion and subcloning of the 3.6 kb insert showed that a gene for the complementation of mutant phenotypes was located in 1.9 kbs Puvll-Hindlll fragment.

Studies on the Control of Sex Ratio by Treatment of Protein Column and H-Y Antibody in Ewes (양에 있어서 Protein Column 및 H-Y항체의 처리에 의한 성비조절에 관한 연구)

  • This experiment was carried out to investigate the sex ratio of produced lamb after artificial insemination into the cervix with spermatozoa from the top and bottom portion of ram semen separated by diluting semen a column of protein, and after implantation into the uterus with normal morulae and blastocyst cultured in BMOG-3 and Ham F-10 medium containing H-Y anbibody and complement treated with spleen and testis, respectively. All embryos developed to morulae and blastocyst were cultrued in medium under gas phase of 5% CO2 in air at 37$^{\circ}C$, 24 hrs. Estrus of ewes induced by a MAP vaginal sponge and 750 IU PMSG during the non-breeding season. The result obtained in these experiments were summarized as follows: 1. The ratio of heating, lambing, and prolificacy after artificial inseminatin into the cervix of ewes induced by a MAP vaginal sponge and 750 IU PMSG during the non-breeding season were 100%, 40% and 2.74 heads, respectively. 2. Involving 44 ewes, spermatozoa from the top of the protein column produced 23.1% male and 76.9% female lambs, while spermatozoa from the bottom of the column produced 81.3% male and 18.8% female offspring, respectively. 3. The sex ratio (male-to-female) of progeny produced after implantation with normal morulae and blastocyst cultured in medium containing H-Y antibody and complement treated with spleen and testis were 21.0%, 79.0% and 17.4%, 82.6%, respectively.

Recent progress in using Drosophila as a platform for human genetic disease research

  • As advanced sequencing technologies continue to uncover an increasing number of variants in genes associated with human genetic diseases, there is a growing demand for systematic approaches to assess the impact of these variants on human development, health, and disease. While in silico analyses have provided valuable insights, it is essential to complement these findings with model organism studies to determine the functional consequences of genetic variants in vivo. Drosophila melanogaster is an excellent genetic model for such functional studies due to its efficient genetic technologies, high gene conservation with humans, accessibility to mutant fly resources, short life cycles, and cost-effectiveness. The traditional GAL4-UAS system, allowing precise control of gene expression through binary regulation, is frequently employed to assess the effects of monoallelic variants. Recombinase medicated cassette exchange or CRISPR-Cas9-mediated GAL4 insertion within coding introns or substitution of gene body with Kozak-Gal4 result in the loss-of-function of the target gene. This GAL4 insertion strategy also enables the expression of reference complementary DNA (cDNA) or cDNA carrying genetic variants under the control of endogenous regulatory cis elements. Furthermore, the CRISPR-Cas9-directed tissue-specific knockout and cDNA rescue system provides the flexibility to investigate candidate variants in a tissue-specific and/or developmental-timing dependent manner. In this review, we will delve into the diverse genetic techniques available in Drosophila and their applications in diagnosing and studying numerous undiagnosed diseases over the past decade.

Screening and characteristics of anti-complementary polysaccharides from Chinese medicinal herbs (한약재로부터 항보체 활성 다당의 검색 및 특성)

  • We conducted screening on Chinese medicinal herbs to examine their anti-complementary activity by hemolytic complementary assay $(TCH_{50})$. Among 55 kinds of herbs, several herbs showed relatively potent anti-complementary activity which decreased $TCH_{50}$, more than 70% in comparison with control. Then, hot water extracts of the following herbs, Curcuma aromatica, Areca catechu, Gleditsiae spina, Euonymus alata, Acanthopanax senticous. Lonicera japonica, Aconitum carmichaeli, Curcuma zedoaria and Cinnamoum cassia, which were shown relatively potent anti-complementary activity were partially purified and analyzed their chemical properties. These activities were resistant to digestion with pronase but decreased by treatment with $NaIO_4$. These results may indicate that the complement activating ability in their herbs is due to polysaccharide. Furthermore, the anti-complementary activity of Areca catechu which was showed the most potent activity, was reduced partially in the absence of the $Ca^{++}\;ion$. After incubation of the normal human serum with partially purified polysaccharide of A. catechu in the absence of $Ca^{++}\;ion$, a cleavage of C3 in the serum was found to have occurred through immunoelectrophoresis using rabbit anti-human C3 serum. These results indicate that the mode of complement activation by polysaccharide of A. catechu is via both the alternative and classical pathway.

Nutritional Status and Indicators of Immune Function of Elderly Residing in Low Income Urban Area (도시생활보호노인들의 영양상태와 면역지표에 관한 연구)

  • This study was conducted to assess the nutritional status of the elderly from low income areas and its relationship with the indices of immunity. Ninety nine(male : 28, female : 71) healthy, subjects aged over 65 years staying in homes in Seoul were assessed with regard to their dietary intake and biochemical measurements. Their mean daily energy intakes were 1256kcal and 109kcal for males and females respectively. Mean serum IgG were 2356mg/dl in men and 2161mg/dl for women, higher than the normal range. Mean serum $C_3$ were 120.9mg/dl and 130.6mg/dl for males and females respectively and mean $C_4$ were 30.6mg/dl and 31.9mg/dl. The mean $C_3$ and $C_4$ were within the normal range and the proportion of subjects whose $C_3$ and $C_4$ were at a higher level than the upper limit were 15.4$\%$ and 16.3$\%$ respectively. Complement $C_3$ showed significant correlation with serum albumin(p〈0.01), serum cholesterol(p〈0.00l, serum TG(p〈0.05), LDL-C(p$\ll$$\ll$$\ll$$\ll$$\ll$$\ll$$\ll$$\ll$$\ll$$\ll$$\ll$$\ll$$\ll$q0.001), VLDL-C(p$\ll$0.05), serum copper(p$\ll$0.05) and serum Ca(p$\ll$0.01). Purification of Anti-HLA Antibodies in Human Placenta Sera (사람 태반혈청내의 항HLA항체 정제)

  • To determine the existence of anti-HLA antibodies finally in 220 human placental extracts to be proved negative antiserum by previous anti-HLA A,B,C antibody screening procedure, the present study was performed by fractionation of immunoglobulins using saturated ammonium sulfate and by simple batch method on DEAE cellulose. Thereafter using known 150 T-lymphocyte panels, complement-dependent microlymphocytotoxicity test was performed to observe the existence of anti-HLA antibodies and the degree of the antibody response of the concentrates. The following results were obtained: 1. Of total 141 placental sera concentrated 45 cases(31.9%) were showed significant anti-HLA A,B,C antibody response after concentration(Excellent, 19(13.5%), Good, 3(2.1%), Weak, 23(16.3%)). 2. Anti-HLA specificities of placental sera obtained after concentration were A2, A24, B13, B27, B44, B51, CN1, C7. 3. A new type C new-1 anti-HLA antibody that is only expressed in Korean people, was obtained. 4. 79 placental sera purrified by simple batch method using DEAE cellulose were showed negative anti-HLA antibody responses.

Pure Isolation and Identification of Babesia ovata by Morphological Characteristics and Complement Fixation Test in Korea (Babesia ovata의 순수분리와 형태학적 및 혈청학적 동정)

  • This study was conducted to identify Babesia sp. strain isolated from the imported beef cattle, Aberdeen-angus heifers, in J farm of Jangsu Prefecture, Cheon-buk Province of Korea, morphologically, etiologically and serologically. Babesia sp. strain was purely isolated through the serial blood passages of three generations against splenectomized calves and one generation of blood passage against non-splenectomized calf(intact calf) by inhibiting the appearance of Theileria sergenti in the blood stream by means of injection of 20% oil pamaquine intramuscularly. In the splenectomized calves, the parasite multiplied markedly in blood stream soon after inoculation and parasitaemia was much severe. An elevated body temperature, anorexia, severe anaemia and icterus were observed clinically. Of three splenectomized calves, two were affected with haemoglobinuria and died. But in the non-splenectomized calf the clinical signs and parasitaemia were very mild. The means of the incubation period, the highest parasitaemia, the highest body temperature and the lowest PCV were 3.5 days, 41.1%, ${42^{\circ}C}$ and 9%, respectively, in the splenectomized calves. In calf erythrocytes Babesia sp. protozoa were polymorphic showing the round, oval, ameboid, piriform and paired piriform etc. The sizes of protozoa were $2.51{\sim}3.91{\times}1.32{\sim}2.19{{\mu}m}$ ($3.20{\times}1.60{\mu}m$). Serologically the isolated Babesia sp. were compared with other parasites, B. ovata, B. bigemina, B. bovis, T. sergenti, A. marginale and A. centrale by using the complement fixation(CF) test. As a result, the antibody titer between the homologous species were high. It was two tubes or more in the CF test. From the results obtained above the pure isolated Babesia species was identical with Babesia ovata Minami and Ishihara, 1980.

STRAINS OF FOOT-AND-MOUTH DISEASE VIRUS IN DIFFERENT DISTRICTS OF BANGLADESH

  • An investigation was carried out to find out the strains of foot-and-mouth disease (FMD) virus in 24 districts of Bangladesh. A total of 505 FMD virus samples were collected from June, 1989 to June, 1991 and tested by complement fixation test (CFT). Of these, 276 (54.7%) were found positive for different strains of FMD virus and the rest 45.3% were either negative or anticomplementary. Strains identified were O, C, Asia-1 and sub-strains $A_5$ and $A_{22}$. Strain O was found to be most prevalent(39.8%) followed by Asia-1 (5.7%), C (5.3%), $A_5$ (3.4%) and $A_{22}$ (0.4%). Prevalence of sub-strain $A_5$ was reported for the first time in Bangladesh. District-wise typing of FMD virus has been done which would be helpful for appropriate vaccination programme in different districts of Bangladesh for control of the malady.
