Toxicoproteomic identification of $TiO_2$ nanoparticle-induced protein expression changes in mouse brain

  • A proteomic analysis of the proteins in mouse brain that were differentially expressed in response to $TiO_2$ nanoparticles was conducted to better understand the molecular mechanism of $TiO_2$ nanoparticle-induced brain toxicity at the protein level. A total of 990 proteins from mouse brain were resolved by two-dimensional gel electrophoresis. A comparative proteomic analysis revealed that the expression levels of 11 proteins were changed by more than 2-fold in response to $TiO_2$ nanoparticles: eight proteins were upregulated and three were downregulated by $TiO_2$ nanoparticles. In addition, the activities of several antioxidative enzymes and acetylcholine esterase were reduced in $TiO_2$ nanoparticle-exposed mouse brain. The protein profile alterations seem to be due to an indirect effect of $TiO_2$ nanoparticles, because $TiO_2$ nanoparticles were not detected in the brain in this investigation.

Comparative Analysis between a Real-Life Explosion Case and A Damage Prediction Program (J도 LPG충전소 가스 누출로 인한 폭발사례와 피해예측 프로그램의 비교 분석)

  • This study aims to estimate the scope of damage impact with a real-life explosion case and a damage prediction program (ALOHA) and suggest measures to reduce risk by comparing and analyzing the results using a Probit model. After applying it to the ALOHA program, the toxicity, overpressure, and radiant heat damage of 5 tons of storage scopes between 66 to 413 meters, and the real-life case also demonstrated that most of the damage took place within 300 meters of the LPG gas station. In the Probit analysis, the damages due to radiant heat were estimated as first-degree burns (13-50%), while structural damage (0-75%) and glass window breakage (94-100%) were expected from overpressure, depending on the storage volume. After comparing the real-life case and the damage prediction program, this study concluded that the ALOHA program could be used as the scope of damage impacts is nearly the same as the actual case; it also concluded that the analysis using the Probit model could reduce risks by applying calculated results and predicting the probability of human casualties and structural damages.

  • The reproduction toxicity of several pesticides on two Korean water flea was investigated to develop a new standard species used for ecological risk assessment of pesticide. Moina macrocopa and Daphnia sp. were exposed to 4 different types of pesticides over 10 and 21 days, respectively. No-ohserved effect concentration (NOEC) for synthetic pyrethroid, fenpropathrin on Moina macrocopa and Daphnia sp. were 0.17 and $0.06\;{\mu}g\;L^{-1}$, respectively. Diazinon, carbofuran and myclobutanil were in the order of their reproduction toxicity to cladocerans tested. There were large differences between Moina macrocopa and Daphnia sp. in their susceptibility to fenpropathrin, diazinon and carbofuran except myclobutanil. Daphnia sp. is more sensitive than M. macrocopa to pesticides tested. Therefore Daphnia sp. may be a good surrogate species to assess the reproduction effect of pesticides on aquatic invertebrates. M. macrocopa also be a good surrogate species because it is one of the most abundant cladocera in agricultural environment, especially rice paddy in Korea. In addition to it's ecological importance of wide spread distribution, it has also economical importance to make possible to shorten period for reproduction test using M. macrocopa due to it's short life cycle.

A Comparative Toxicological Study of Dried Mylabris phalerata Extract and it's Modifier : Single Dose Toxicity on Male Mice (반묘(斑猫)와 가공반묘(加工斑猫)의 단회투여(單回投與) 독성에 대한 비교연구)

  • Objectives : The objective of this study was to compare, the potency of toxicity of Cantharidin containing dried Mylabis phalerata (MP) extract and it's modifier. Methods : They were monitored at dosage level 2,000, 1,000, 500, 250 and 125 mg/kg, respectively. Changes of body weight, clinical signs, mortality, LD50, macroscopic changes of gastrointestinal tract and liver were observed after single oral dose of test articles with changes of serum Gastrin and Somatostatin levels. Results : Dosage-dependent decrease of body weight and/or gains were demonstrated in dried MP extract-dosing groups, were also detected in modified and dried MP extract-dosing groups at 2,000 and 1,000 mg/kg-dosing group. However, below 500 mg/kg-dosing group, the body weights were significantly increased compared to that of equal dosage group of dried MP extract-dosing group. Dosage-dependently detected clinical signs in dried MP extract-dosing groups, were also detected in modified and dried MP extract-dosing groups at 2,000 and 1,000 mg/kg-dosing group. However, below 500 mg/kg-dosing group, these clinical signs dramatically were decreased compared to that of equal dosage group of dried MP extract-dosing group. Dosage-dependent increase of mortality rates were observed in dried MP extract-dosing groups, were also detected in modified and dried MP extract-dosing groups at 2,000 and 1,000 mg/kg-dosing group. However, below 500 mg/kg-dosing group, the mortalities were significantly decreased compared to that of equal dosage group of dried MP extract-dosing group. The LD50 of dried MP extract in male mice was dramaticlly increased in their modify, 265.86 vs 426.99 mg/kg. Dosage-dependently increase of number of hemorrhagic and/or erythematous spots detected in the gastrointestinal tracts of dried MP extract-dosing groups, were also detected in modified and dried MP extract-dosing groups at 2,000 and 1,000 mg/kg-dosing group. However, below 500 mg/kg-dosing group, these abnormal spots were dramatically decreased compared to that of equal dosage group of dried MP extract-dosing group. Dosage-dependently increase of degrees of enlargement and congestion detected in the liver of dried MP extract-dosing groups, were also detected in modified and dried MP extract-dosing groups at 2,000 and 1,000 mg/kg-dosing group. However, below 500 mg/kg-dosing group, these abnormal signs were dramatically decreased compared to that of equal dosage group of dried MP extract-dosing group. Dosage-dependently increase of serum gastrin levels of dried MP extract-dosing groups, were also detected in modified and dried MP extract-dosing groups at 2,000 and 1,000 mg/kg-dosing group. However, below 500 mg/kg-dosing group, these abnormal increase were dramatically decreased compared to that of equal dosage group of dried MP extract-dosing group. Dosage-dependently increase of serum somatostatin levels of dried MP extract-dosing groups, were also detected in modified and dried MP extract-dosing groups at 2,000 and 1,000 mg/kg-dosing group. However, below 500 mg/kg-dosing group, these abnormal increase were dramatically decreased compared to that of equal dosage group of dried MP extract-dosing group. Conclusions : The toxicity of dried MP extract was reduced by their modify.

A Comparative Study of Standard Methods for Assessing Ecotoxicity of Endocrine Disrupting Chemicals (내분비계장애물질의 생태독성평가를 위한 표준시험법 비교연구)

  • Endocrine disrupting chemicals (EDCs) interfere with endocrine system in organisms, and have different mode of actions compared to conventional chemicals. Therefore, EDCs specific ecotoxicity tests and assessments have been globally developed by some organizations such as OECD, ASTM, and USEPA. In Korea, researches on EDCs and monitoring of EDCs in domestic environments were also continued. However, Korean ecotoxicity test guidelines for EDCs are not suggested till date. The purpose of this study is to review and analyze international ecotoxicity test guidelines for EDCs and the compare ecotoxicity methods and toxicity endpoints among standard test guidelines. We found that there are very limited EDCs specific soil ecotoxicity test guidelines (only in ASTM) compared to aquatic ecotoxicity test guidelines. Currently, fish, amphibian, waterflea, copepoda, earthworm, white worm, springtail, nematode, mite, and midge are suggested as standard ecotoxicity test species for EDCs. Reproduction, hormones, growth, vitellogenin, sex retio and development were proposed as endpoints for EDCs ecotoxicity. This study provided the comparison of EDCs specific ecotoxicity methods and endpoints between standard test guidelines, and suggested the further research to develop the method for assesseing ecotoxicity of EDCs.

Comparative physiological and proteomic analysis of leaf in response to cadmium stress in sorghum

  • Cadmium (Cd) is of particular concern because of its widespread occurrence and high toxicity and may cause serious morpho-physiological and molecular abnormalities in in plants. The present study was performed to explore Cd-induced morpho-physiological alterations and their potentiality associated mechanisms in Sorghum bicolor leaves at the protein level. Ten-day-old sorghum seedlings were exposed to different concentrations (0, 100, and $150{\mu}M$) of $CdCl_2$, and different morpho-physiological responses were recorded. The effects of Cd exposure on protein expression patterns in S. bicolor were investigated using two-dimensional gel electrophoresis (2-DE) in samples derived from the leaves of both control and Cd-treated seedlings. The observed morphological changes revealed that the plants treated with Cd displayed dramatically altered shoot lengths, fresh weights, and relative water content. In addition, the concentration of Cd was markedly increased by treatment with Cd, and the amount of Cd taken up by the shoots was significantly and directly correlated with the applied level of Cd. Using the 2-DE method, a total of 33 differentially expressed protein spots were analyzed using MALDI-TOF/TOF MS. Of these, treatment with Cd resulted in significant increases in 15 proteins and decreases in 18 proteins. Significant changes were absorbed in the levels of proteins known to be involved in carbohydrate metabolism, transcriptional regulation, translation and stress responses. Proteomic results revealed that Cd stress had an inhibitory effect on carbon fixation, ATP production and the regulation of protein synthesis. In addition, the up-regulation of glutathione S-transferase and cytochrome P450 may play a significant role in Cd-related toxicity and stress responses. Our study provides insights into the integrated molecular mechanisms involved in response to Cd and the effects of Cd on the growth and physiological characteristics of sorghum seedlings. The upregulation of these stress-related genes may be candidates for further research and use in genetic manipulation of sorghum tolerance to Cd stress.

A Study on the Applicability of Acrylic Water Leak Repair Materials used to Repair Cracks in Conduits and Underground Structures (관거 및 지하구조물 균열 보수에 사용되는 아크릴 누수 보수재의 적용성에 대한 연구)

  • Various injection materials, such as asphalt-based injection materials, urethane-based injection materials, cement- based injection materials, and acrylic-based injection materials, are used for the repair of aged conduits and underground structures with cracks. In this study, research was conducted on an environmentally friendly acrylic- based leak repair material that exhibits good curing properties even in humid conditions and stability in temperature fluctuations. To compare the performance of the improved acrylic leak repair material with the existing acrylate injection material, experiments were conducted using KS standard methods, including underwater length change rate tests, underwater leakage resistance tests, and chemical performance tests. The comparative experiments revealed that the improved acrylic leak repair material showed no changes in shrinkage due to humidity, temperature variations, or chemical reactions compared to the existing acrylate injection material. In the underwater resistance test, the improved acrylic leak repair material did not show any leakage. Additionally, to assess the environmental impact of the improved acrylic leak repair material, acute fish toxicity tests and acute oral toxicity tests were conducted, and the results showed no mortality and no specific concerns with the test specimens. The experimental results led to the conclusion that the improved acrylic leak repair material is considered to be superior in performance, environmentally safe, and harmless to the human body. Based on various experimental results, it is inferred that the improved acrylic leak repair material is suitable for use as a repair material for cracks in manholes and underground structures compared to the existing acrylate repair material. This study aims to propose valuable data for future technological development by evaluating the applicability of acrylic leak repair materials.

The Comparative Study of Oriental Medicine in Korea, Japan and China (한국(韓國)과 일본(日本) 및 중국(中國)의 동양의학(東洋醫學)에 대한 비교연구(比較硏究))

  • During these days of new understanding, western medicine has developed remarkably and a revaluation of traditional medicine has been achieved. This appears to have resulted from the sound criticisms of what western medicine has achieved up to now; excessive subdivisions of clinical medicine, severe toxicity of chemical drugs, lack of understanding about patients complaints which cannot be understood objectively, and etc. It is thought that the role of traditional medicine will be more important in the future than it is now. Someone said that the research methods of traditional medicine depends on the way of experimental science too much. That there was no consideration of a system for traditional medicine and the critic also went so far as to assert that in some cases the characteristics of eastern ideas is to permit irrationalism itself. In view of this thinking, the term traditional medicine seems to have been used somewhat too vaguely. However, traditional medicine is a medical treatment which has existed since before the appearance of modern medicine and it was formed from a traditional culture with a long history. One form of traditional medicine, oriental medicine based upon ancient Chinese medicine, was received in such countries as Korea, Japan, Thailand, Vietnam, Tibet, and Mongolia. Oriental medicine then developed in accordance with its own environment, race, national characteristics, and history. Although there are some simultaneous differences between them, three nations in Eastern Asia; Korea, Japan, and China, have especially similar features in their clinical prescriptions and medical literature. These three nations are trying to understand each others unique traditional medicines through numerous exchanges. Even though many differences in their ways of studying have developed over history exist, recent academic discussions have been made to explore new ways into oriental medicine. Therefore a comparative study of oriental medicine has gradually been thought to be more important. In Korea the formation of a new future-oriented paradigm for oriental medicine is being demanded. The purpose of the new paradigm is to create a new recognition of traditional culture which creates an understanding of oriental medicine to replace the diminished understanding of oriental medicine that was brought about by the self-denial of traditional culture in modem history and cultural collisions between oriental and occidental points of view. Therefore, to make a new paradigm for oriental medicine which is suitable for these days, and fortifies the merit of oriental medicine while compensating its defects, the author has compared the characteristics of oriental medicines in Korea, Japan, and China. The conclusions of this research are as follows: 1. The fundamental differences of the traditional medicines of these three nations are caused by the differences in the systems of Naekyung and Sanghannon. 2. The pattern-identification of illnesses is generally divided into two categories; the pattern identification of Zang-Fu and the pattern identification of prescription. 3. There are many differences in the definition of terms, such as Yin and Yang, Deficiency and Excess, and etc. 4. Chinese traditional medicine has some new concepts about pattern identification and epidemic febrile disease. 5. Japanese traditional medicine has some characteristics about pattern identification of the whole bodys condition and signs of abdominal palpation. 6. In terms of the effects of herbal drugs, Chinese traditional medicine attaches great importance to the experiential efficacy of the herb, and Japanese traditional medicine is taking a serious view of the effects of experimental medical actions.

  • The technical requirements to the propellant of rocket engine using chemical energy are developed from the point of its performance, design and the exploration. The propellant, i.e. to get higher energy, to increase the cooling effect, and to be used in safety and comfort should be made. So from these aspect the technical requirements were written by the factors of physical chemistry. And the propellant combination used recently in Launch vehicles is liquid Oxygen and Kerosene, the main characteristics of this Kerosene as a fuel were reviewed. The toxicity and the safety of Kerosene, in specially, require the attention of the users. Also, it would be used in the development of Korean Kerosene by the comparative review of the several Kerosenes in Abroad.

The Latest Research Trends on Woohwangchungsim-won : A Review in Korean Articles (우황청심원 관련 최근 연구 동향 : 국내 논문에 대한 고찰)

  • Objectives : Woohwangchungsim-won has been used for acute diseases such as cerebrovascular disease (stroke), heart disease. The aim of this study was to determine the effects, significance & necessity of Woohwangchungsim-won in acute stages of these disease by reviewing Korean articles about Woohwangchungsim-won. Methods : Article searches were performed on 8 major Korean web article search engine from January 1980 to August 2010. There were no restrictions on the types of publication, including grey articles. Results : Forty-three articles were included. Twenty one were efficacy tests, thirteen were comparative efficacy tests, eight were toxicity tests, and one was new method development. Eighteen articles were multicenter studies and twenty five were single center studies. Thirty two articles were animal testing, only ten articles were clinical articles. Thirty seven articles presented the composition and quantity of Woohwangchungsim-won, but six did not. Conclusions : The results of Korean articles about Woohwangchungsim-won can not strongly support the effects, significance & necessity of Woohwangchungsim-won. More rigorous studies are required for determination of the effects of Woohwangchungsim-won.
