Development of Colorimetric Paper Sensor for Pesticide Detection Using Competitive-inhibiting Reaction

  • Contamination by pesticides is an everincreasing problem associated with fields of environmental management and healthcare. Accordingly, appropriate treatments are in demand. Pesticide detection methods have been researched extensively, aimed at making the detection convenient, fast, cost-effective, and easy to use. Among the various detecting strategies, paper-based assay is potent for real-time pesticide sensing due to its unique advantages including disposability, light weight, and low cost. In this study, a paper-based sensor for chlorpyrifos, an organophosphate pesticide, has been developed by layering three sheets of patterned plates. In colorimetric quantification of pesticides, the blue color produced by the interaction between acetylcholinesterase and indoxyl acetate is inhibited by the pesticide molecules present in the sample solutions. With the optimized paper-based sensor, the pesticide is sensitively detected (limit of detection =8.60 ppm) within 5min. Furthermore, the shelf life of the device is enhanced to 14 days after from the fabrication, by treating trehalose solution onto the deposited reagents. We expect the paper-based device to be utilized as a first-screening analytic device for water quality monitoring and food analysis.

Investigation of thermal annealing effects on the optical transparency and luminescent characteristics of Eu-doped Y2O3 thin films

  • The thermal annealing effects on the optical transparency and luminescent characteristics of the Eu-doped Y2O3 thin films have been investigated. The as-deposited Y2O3:Eu films exhibited an optical band gap of 5.78 eV with a transparency of 89 % at a wavelength of 550 nm. As the annealing temperature increased from 1000 to 1300 ℃, the optical band gap and transparency of the films decreased from 5.77 to 4.91 eV and from 86.8 to 64.5 % at 550 nm, respectively. The crystalline quality of the films was improved with increasing annealing temperature. The annealed Y2O3:Eu films emitted a red-color photoluminescence (PL) with the highest emission peak near 612 nm. The PL intensity was increased with increasing annealing temperature to 1200 ℃, resulting from the improvement in the crystalline quality of the films. The PL intensity was decreased with further increasing temperature above 1200 ℃ due to the formation of Y2SiO5 phase by the reaction of the film with the quartz substrate.

Anatomical, Chemical, and Topochemical Characteristics of Transgemic Poplar Down-regulated with O-methyltransferase

  • The present work was undertaken to investigate the anatomical and chemical characteristics of transgenic poplar down-regulated with antisense OMT gene. Also the distribution of lignin in transgenic poplar trees was investigated at cellular level. No visible abnormal phenotype was observed in the fibers and vessel elements of transgenic poplar. Any marked differences in the staining intensities of Wiesner and Mäule color reaction were not identified in the transgenic poplar. TEM micrographs did not show any staining intensities in the cell walls stained with KMnO4. Interestingly, the UV spectroscopy of semi-thin sections exhibited a distinct decrease of lignin absorption at 280 nm in the vessel walls, indicating transgenic poplar wood with lower amount of guaiacyl lignin in vessel elements. Chemical composition of antisense OMT poplar was almost identical to that of wild-type poplar. Klason lignin content of transgenic poplar did not show any significant difference from that of the controls. The solid state NMR spectra revealed the transgenic poplar with only slightly more syringyl lignin than the control. The present work showed that antisense OMT gene constructed in the poplar was not enough to reduce the overall content of Klason lignin, and suggested that the expression of transformation was confined to vessel walls.

Improved Resolution of Paper-based Sensor for Proline Detection by Low-temperature Drying of Ninhydrin Solution (닌히드린 용액의 저온 건조에 의한 프롤린 검출을 위한 종이기반 센서의 분해능 개선)

  • In this study, we describe the improvement of the resolution of a paper-based sensor by fabricating a high-concentration ninhydrin part using a low-temperature drying method to detect proline with high resolution. In the conventional paper-based sensor for detecting proline, the ninhydrin part is fabricated at room temperature, and in this process, the ninhydrin solution spreads around the ninhydrin part. Therefore, the concentration of the ninhydrin part becomes lower than that of the applied solution, lowering the resolution of the sensor. The proposed paper-based sensor better improved the sensitivity of the sensor compared to the existing sensor by fabricating a high-concentration ninhydrin part through drying the ninhydrin solution using a low-temperature drying method. Owing to the experiment, the intensity of the green color of the paper-based sensor with the integrated ninhydrin part fabricated at 10 ℃ is approximately 20% lower than the paper-based sensor with an integrated ninhydrin part fabricated at room temperature, indicating better sensor resolution. Therefore, the paper-based sensor with an integrated ninhydrin part fabricated at a high concentration could be useful for diagnosing drought.

Improved Flowability and Wettability of Whey Protein-Fortified Skim Milk Powder via Fluidized Bed Agglomeration

  • Recently, protein-fortified milk powders are being widely consumed in Korea to prevent sarcopenia, and the demand for high-protein food powders is continuously increasing in the Korean market. However, spray-dried milk proteins have poor flowability and wettability owing to their fine particle sizes and high inter-particle cohesive forces. Fluidized bed agglomeration is widely used to improve the instant properties of food powders. This study investigated the effect of fluidized bed agglomeration on whey protein isolate (WPI)-fortified skim milk powder (SMP) at different SMP/WPI ratios. The fluidized bed process increased the particle size distribution, and agglomerated particles with grape-like structures were observed in the SEM images. As the size increased, the Carr index (CI) and Hausner ratio (HR) values of the agglomerated WPI-fortified SMP particles exhibited excellent flowability (CI: <15) and low cohesiveness (HR: <1.2). In addition, agglomerated WPI-fortified SMP particles exhibited the faster wetting time than the instant criterion (<20 s). As a result, the rheological and physical properties of the WPI-fortified SMP particles were effectively improved by fluidized bed agglomeration. However, the fluidized bed agglomeration process led to a slight change in the color properties. The CIE L* decreased, and the CIE b* increased because of the Maillard reaction. The apparent viscosity (ηa,10) and consistency index (K) values of the rehydrated solutions (60 g/180 mL water) increased with the increasing WPI ratio. These results may be useful for formulating protein-fortified milk powder with better instant properties.

Enhancing the Oxygen Removal Rate for Its Application in Food Packaging Through the Impregnation of Porous Materials with the Non-metallic Oxygen Scavenger Sodium Metabisulfite (메타중아황산나트륨을 다공성물질에 함침하여 제조한 비금속류 산소제거제의 산소제거속도 향상 및 식품 포장 적용 연구)

  • The addition of oxygen scavengers to food products helps to reduce oxygen exposure, thereby mitigating deterioration, including changes in taste, odor, and color, as well as inhibiting microbial growth. Despite the advantages of the existing non-metallic oxygen removal materials in terms of safety for the human body and suitability for use in microwave ovens, their utilization has been limited due to their slow reaction initiation speed. Therefore, in the current study, sodium metabisulfite was impregnated into various porous media, including halloysite nanoclay, activated carbon, montmorillonite, and silica gel. The oxygen scavenger, produced by impregnating silica gel with sodium metabisulfite, demonstrated a 425% improvement in the initial oxygen removal rate compared to pure sodium metabisulfite. Additionally, sachets containing an oxygen-removing composition with an enhanced oxygen removal rate effectively decreased the oxygen concentration to less than 0.5% on the third day of storage in apple packaging, without elevating carbon dioxide levels. Moreover, it proved effective in preventing the browning of the apple surface. Therefore, the SM/SG oxygen-removal composition can be effectively applied to active food packaging by controlling the oxygen concentration within the packaging.

Non-Enzymatic Browning Reaction of the Dried File-Fish (건조(乾燥) 말쥐치의 비효소적(非酵素的) 갈변(褐變))

  • A kinetical approach for the non-enzymatic browning reaction of the dried file-fish was studied. The reaction rates revealed a tend to increase with increasing water activity and showed the maximum at $0.75\;a_w$ The activation energies obtained from the Arrhenius plot ranged 12.5 to 16.5 Kcal/mole. From these energies of activation, the $Q_{10}$ values at $45^{\circ}C$ showed 1.9 to 2.3 and both activation energy and $Q_{10}$ values were reduced with increase in $a_w$ Shelf-lives, the time to reach an 0.15 O.D./g solid at which severe brown color change could be de ectable, decreased rapidly as the temperature and water activity increase. A storage study under a square-wave fluctuating temperature condition (at 35 and $55^{\circ}C$ periodically with 7 days interval), the rate constants at all water activities used in the experiments were higher than those at $45^{\circ}C$, the mean temperature of the cycle which affects other kinetic parameters including activation energies, $Q_{10}$ values and finally the shelf-lives. The data obtained from the fluctuating temperaure storage study will be used in prediction of shelf-life. The shelf lives assessed at $25^{\circ}C$ from the accelerated shelf-life tests ranged from 179 daysat $0.75\;a_w$ to 302 days at $0.44\;a_w$.

Behavioral reaction of common squid Todarodes pacificus to different colors of LED Light (LED 색광에 대한 살오징어의 행동반응)

  • To study the behavioral reaction of common squid Todarodes pacificus, to different colors of LED light, individual distribution in the dark condition without LED light stimuli, individual distribution when four colors of light stimuli were at the sections of end and middle of water tank and the changes of gathering rates as the elapse of illuminating time were examined. When it was dark, the distribution of the fish in each section of the tank was U-shape regardless of the investigating time intervals, which means that Todarodes pacificus gathered at the both ends of the tank. The individual distribution when light stimuli were at the one end of the tank showed the tendency of gathering at the illuminated section and decreasing at the opposite section. There were 448 of them at the end of the tank when the light was blue, 352 when white, 302 when green and 132 when red. Thus the fish liked blue light the most, followed by white light. The variations of gathering rate at the both ends of tank as illuminating time elapsed showed the tendency of increasing at the light section (A section), however, it showed the tendency of decreasing at the darkest section (F section). The individual distribution showed the tendency of letter $\wedge$ of gathering at the center and decreasing at the both ends mostly when the light stimuli were at the middle section of tank. The gathering rates at the lighting section were 80.4% when red, 76.4% when white, 69.6% when green and 56.7% when blue. The fishes showed the affinity for the red light mostly, followed by the white light source. The red light and blue light showed the opposite when the light stimuli were at the one end of water tank. The variations of gathering rates as the elapse of time at the lighted section showed the tendency of gradual increasing in the four light sources. The color of light source showing the highest gathering rate within 25-30 minutes of light stimuli was the white, followed by the red.

Assessment of Availability of Korean Computerized Neurobehavioral Test in Patients with Brain Injury (뇌손상 환자에서 컴퓨터 신경행동검사의 유용성 평가)

  • This study was conducted to evaluate the correlation between the Korean version of Mini Mental State Examination (MMSE-K) and the Korean Computerized Neurobehavioral Test (KCNT) and the usefulness of KCNT for discriminating the patients with suspected dementia from normal persons in patients with a brain injury. The results of each items of KCNT had passed the test for normality. T test for group comparison and bivariate correlation analysis for correlation between two tests were used. There were no significant differences in characteristics of two groups. The differences in results of eight tests of KCNT between two groups were statistically significant (p<.05) and the Pearson correlation coefficients (r) between two tests were statistically significant (p<.05) : simple reaction time (-.580), choice reaction time (-.341), color word vigilance (-.661), digit classification (-.703), 2-digit addition (-.582), 3-digit addition (-.610), symbol digit substitution (-.642), and digit span (.807). These results suggested that simple reaction time and 2-digit addition were useful in discriminating the patients with suspected dementia from normal persons because the Pearson correlation coefficients of the two items of KCNT showed anticlastic association between two groups. And each items of KCNT may use in follow up cognitive function for patients in two groups.

Study of Stability and Shelf-life of Red Ginseng Beverage Emulsified by Homogenizer High Pressure (고압균질기로 유화된 홍삼음료의 안정성과 유통기한 변화에 대한 연구)

  • This study was carried out to predict the shelf-life of emulsified red ginseng beverage. To investigate the quality changes during the storage, physicochemical properties such as acidity, pH, droplet size, Hunter's color value and sensory evaluation test were measured periodically. It was found that acidity, pH, and droplet sizes were little changed in initial stage. After about 70 days of storage, however, they tended to be changed possibly due to Maillard reaction. From the results of correlation analysis between sensory evaluation results and physicochemical characteristics, a-value was chosen as a quality index of red ginseng emulsions. Using reaction constants at various temperature, the activation energy and the $Q_{10}$ value for the a-value from Arrhenius equation found to be 13.37 kcal/mol, 1.56-2.14, respectively. It could be concluded that the shelf-life of red ginseng emulsions estimated to be 730 days (approximately 2 years) when stored at $20^{\circ}C$.
