Optical-effect Analysis of Nanoscale Collagen Fibers

  • To understand the cause of the high light transmittance of the human eye, the optical effects of the collagen fibers of the stroma layer, which constitute the majority of the cornea, were analyzed. These collagen fibers, approximately 20 nm in diameter, have a regular arrangement. Accordingly, the optical properties of the collagen fibers and the fiber layer were analyzed by simulation. A standing wave was formed in the incident space by the overlapping incident light and the light reflected by the plate. In addition, it was confirmed that when the collagen fibers are arranged in a layer, the light transmittance periodically changes, depending on the number of fiber layers. The standing wave was formed in the incident space, and the light's intensity distribution was changed by the nanoscale collagen fibers in the section with the collagen layer, which affected the transmittance. To explain this phenomenon, the collagen fiber was defined as a second light source, and an attempt was made to describe the simulation results in terms of overlap of the incident light with the light emitted from the collagen fiber.

Reconstruction of Collagen Using Tensor-Voting & Graph-Cuts

  • Collagen can be used in building artificial skin replacements for treatment of burns and towards the reconstruction of bone as well as researching cell behavior and cellular interaction. The strength of collagen in connective tissue rests on the characteristics of collagen fibers. 3D confocal imaging of collagen fibers enables the characterization of their spatial distribution as related to their function. However, the image stacks acquired with confocal laser-scanning microscope does not clearly show the collagen architecture in 3D. Therefore, we developed a new method to reconstruct, visualize and characterize collagen fibers from fluorescence confocal images. First, we exploit the tensor voting framework to extract sparse reliable information about collagen structure in a 3D image and therefore denoise and filter the acquired image stack. We then propose to segment the collagen fibers by defining an energy term based on the Hessian matrix. This energy term is minimized by a min cut-max flow algorithm that allows adaptive regularization. We demonstrate the efficacy of our methods by visualizing reconstructed collagen from specific 3D image stack.

  • Collagen and chitosan are used in medical and cosmetic materials as natural polymers. In order to utilize the advantages of the materials, collagen/chitosan conjugated wet-spun fibers were prepared. The analysis of surface, optical, thermal and mechanical properties was carried out on the various composition of collagen and chitosan. As a result of images analysis, it was verified that the collagen/chitosan conjugated fibers were stably spun. In addition, the optical and thermal properties of fibers were observed to be changed by hydrogen bond. As a result, an optimized composition could be found at an appropriate content. Moreover, the optimized fibers have mechanical properties similar to chitosan fibers, while improving the structural and thermal stability by its hydrogen bond. In addition, the wet-spun collagen/chitosan conjugated fibers can be applied to medical and various fields through mechanical properties according to content control.

Positive Effects of Diphlorethohydroxycarmalol (DPHC) on the Stability of the Integument Structure in Diet-Induced Obese Female Mice

  • Diphlorethohydroxycarmalol (DPHC) is a known to modulate the expression of extracellular matrix (ECM) components in 3T3-L1. However, the possible role of DPHC in integument stability during obesity induction is not clear yet. We evaluated the effects of DPHC on collagen or elastic fiber quantity in integument during obesity induction with high-fat diet. The dorsal back integument sections were stained with hematoxylin-eosin, Masson trichrome, and Verhoff-Van Gieson. The intensities of collagen fibers and elastin fibers were analyzed with ImageJ. The number of fibroblasts was counted at ${\times}1,000$ fields. The number of fibroblast was increased by obesity induction, but DPHC suppressed it in a concentration-dependent manner both in lean and obese mice. On the other hand, the intensities of collagen fibers were increased by DPHC treatment in obese mice groups but not in lean mice groups. The intensities of collagen fibers of obese mice were lower than that of the lean mice in 0% group. However, the number became similar between lean and obese mice by the treatment of DPHC. The intensity of elastic fibers was increased in the lean mice with the concentration of DPHC. In the obese mice group, there were increasing patterns but only significant at 10% DPHC group. The intensity of elastic fibers of obese mice was higher than lean mice in 0%, 1%, and 10% groups. Histologically epithelial cells and follicle cells which were diffused nuclear staining forms were increased by DPHC treatment. The results suggest that the activity of integument cells during obesity induction can be modulated by DPHC.

Interplay of collagen and mast cells in periapical granulomas and periapical cysts: a comparative polarizing microscopic and immunohistochemical study

  • Objectives: This pilot study aimed to establish the interrelationship between collagen and mast cells in periapical granulomas and periapical cysts. Materials and Methods: An observational cross-sectional study was conducted on the paraffin-embedded tissue sections of 68 specimens (34 periapical granulomas and 34 periapical cysts). The specimens were stained with picrosirius to observe collagen fiber birefringence and anti-tryptase antibody to evaluate the mast cell count immunohistochemically. The mean number and birefringence of collagen fibers, as well as the mean number of mast cells (total, granulated, and degranulated), and the mean inflammatory cell density were calculated. The data obtained were analyzed using the Kruskal Wallis test, Mann Whitney U test, and Spearman correlation test (p < 0.05). Results: The mean number of thick collagen fibers was higher in periapical cysts, while that of thin fibers was higher in granulomas (p = 0.00). Cysts emitted orange-yellow to red birefringence, whereas periapical granulomas had predominantly green fibers (p = 0.00). The mean inflammatory cell density was comparable in all groups (p = 0.129). The number of total, degranulated, and granulated mast cells exhibited significant results (p = 0.00) in both groups. Thick cyst fibers showed significant inverse correlations with inflammation and degranulated mast cells (p = 0.041, 0.04 respectively). Conclusions: Mast cells and inflammatory cells influenced the nature of collagen fiber formation and its birefringence. This finding may assist in the prediction of the nature, pathogenesis, and biological behavior of periapical lesions.

양식 및 자연산 도미와 넙치 어육 중의 콜라겐 DSC 측정 및 근육 조직 관찰 (Observation of Muscle Structure and DSC Measurement of Collagen of the Cultured and Wild Red Sea Bream and Flounder.)

  • Thermal measurements were made for connective tissues of 5 different fish muscles by using a differential scanning calorimeter(DSC), and connective tissues between muscle fibers and the cross sections of muscle fibers were observed by a light microscope. Red sea bream(cultured and wild) and flounder(cultured, cultured with obosan and wild) were used in this study. It was found that the connective tissues of cultured and frozen fish muscle required less endothermic enthalpy and the endothermic peak temperature was lower than those of wild and fresh ones when they were shrunken and denatured. Therefore, it is likely that the former are more unstable to heat than the latter. The cultured flounder fed with obosan and wild flounder which contained more collagen than cultured flounder and the wild red sea bream showed clear connective tissues between fibers. The cross-section of cultured fish muscle fiber was larger than that of wild one. From these results, collagen content and thermal properties of collagen, cross section of muscle fibers seemed to contribute to the textural difference between wild and cultured fish.

각막실질 콜라겐섬유의 배열구조에 따른 광투과율 분석 (Analysis of Light Transmittance according to the Array Structure of Collagen Fibers Constituting the Corneal Stroma)

  • 목적 : 각막실질 내 다발 구조로 이루어진 콜라겐섬유의 크기와 규칙적인 배열은 투명성과 매우 밀접한 상관성을 가지고 있다. 시뮬레이션을 이용하여 배열구조 및 콜라겐섬유층 두께에 따른 광투과율의 변화를 확인하고자 하였다. 방법 : 시뮬레이션 소프트웨어인 OptiFDTD로 각막실질 내 콜라겐섬유를 정육각형, 육각형, 사각형 및 자유형으로 각각 배열하였고 이에 따른 광투과율을 분석하였다. 사각형 배열에 대하여 시뮬레이션 공간상에 있는 콜라겐섬유의 개수가 동일할 때 밀도변화에 따른 광투과율을 확인하고 콜라겐섬유의 개수와 밀도가 변화할 때 광투과율을 조사하였다. 결과 : 콜라겐섬유의 개수가 동일할 때 사각형, 정육각형, 자유형 및 육각형의 배열구조 순서로 밀도가 작아지고, 섬유층의 두께가 두꺼워진다. 배열구조를 변화시켜 광투과율을 측정한 결과 동일한 위치의 검출기에서 측정된 광투과율은 배열구조에 관계없이 거의 유사하였다. 검출기 D0, D1, D2 및 D3에서 각각 사각형, 육각형과 사각형, 정육각형 및 정육각형 배열구조에서 최대투과율로 나타났으며, 육각형, 자유형, 육각형과 사각형 및 사각형 배열구조에서 최소 투과율로 나타났다. 하지만, 최대 투과율과 최소 투과율의 차이는 1% 이내로 거의 유사하였다. 콜라겐섬유의 개수가 동일할 때 사각형 배열구조에서 밀도변화에 따른 광투과율은 섬유층 두께가 증가할수록 광투과율은 감소하였다. 또한, 두께가 증가하면서 콜라겐섬유의 개수가 감소하였을 때 광투과율이 더 많이 감소하였다. 결론 : 콜라겐 배열구조가 변화하여도 광투과율은 배열구조와 관계없이 거의 유사하게 나타났다. 하지만, 배열구조의 변화에 따라 콜라겐섬유층의 두께가 변화하였고, 두께가 증가할수록 광투과율이 감소하였다. 즉, 광투과율은 배열구조보다는 콜라겐섬유층의 두께와 더 밀접한 관계를 가지고 있음을 확인하였다.

Cyclosporin A에 의한 치은 과증식의 형태학적 특성 (Morphological features of Cyclosprin A-induced Gingial Hyperplasia)

  • Cyclosporin A(CsA) is now widely used to treat organ transplant recipients. But CsA has various short-and long-term side effects. Especially, gingival hyperplasia is not easy to resolve since its nature is still unknown. This study discusses the pathogenesis of CsA-induced gingival hyperplasia on the basis of data obtained from light and electron microscopic studies of biopsis from patients on CsA treatment after kidney transplantation. Light microscopically, the multilayered squamous epithelium showed an irregular surface of parakeratosis and deep invaginations in the subepithelial tissue. At lamina propria, we observed bundles of irregularly arranged collagen fiber, some fibroblasts, numerous capillary vessels and a large diffuse infiltration of plasma cells. Ultrastructurally, many fibroblasts, collagen fibers, collagen fibrils were present in lamina propria. On the basis of the data collected, we propose that the morphological features of the dimensional increase in gingival tissue associated with CsA treatment in kidney transplant patients may be considered proliferative fibroblasts, collagen fibers, collagen fibrils in lamina propria.

Optical Characteristics of Corneal Nanostructure According to the Angle of Collagen-fiber-layer Arrangement

  • Collagen fibers tens of nanometers in size, which constitute most of the corneal volume of the human eye, are layered in a uniform direction, and adjacent fiber layers are arranged at an angle of 90° to each other. According to the results of this study, the transmittance at 45° of interlayer rotation angle is highest, and higher than that of the 90° body structure. The transmittance is examined, concerning the polarization state of the incident light; circularly polarized light case shows higher transmittance than linearly polarized. Through this, a simulation to confirm the deformed structure of collagen fibers, which show higher transmittance than the anatomical structure of the cornea, is attempted.

  • GMI (Fibrel${(R)}$) is one of the dermal filling substances which have been successfully used for the treatment of depressed cutaneous scar and wrinkles. It's major components are; Gelatin powder, which provides a framework for the clot to form and remains stable under the scar, and ${\varepsilon}$-aminocaproic acid, which inhibits the production of fibrinolysin, and Plasma, which provides the necessary ingredients for collagen synthesis. GMI has advantages of low immunogenicity and increased longevity. It has been known to induce fibroblast activity and promote new collagen synthesis. We used 34 Sprague-Dawley rats which were bred under the same condition and duration. 18 of experimental animals were undergone cardiac puncture, and their blood were collected, centrifugated, and stored in freezer. Out of 16 animals, control group were injected with 2ml plasma into the subcutaneous tissue of Lt. scapular, while experimental group were implanted of 2 ml GMI into the Rt. same area. Experimental animals were sacrificed at the 3rd day, 5th day, 1st week and 2nd week respectively after implantation of GMI. To observe the histopathologic change of GMI and surrounding tissue reaction of GMI, we had examined with H&E staining, immunohistochemical staining with vimentin, ${\alpha}$-SMA, S-100 under LM and SEM. The obtained results were as follows ; 1. In LM study, the inflammatory cell infiltrations and granulation tissue formation were observed, and muscle tissues were well attached with adipose tissues in the control group. In the experimental group, inflammatory cell infiltrations had been observed by the 2nd week and irregular adipiose tissues and well differentiated mesenchymal tissues were examined. 2. In immunohistochemical study, the experimental group of ${\alpha}$-SMA study, there were a prominent positive response on endothelial development of granulation tissues and mesenchymal tissues compare with the control group. In vimentin study, positive response on mescenchymal fibroblast continued to 2nd week, but negative in the control group. In S-100 study, both groups were positively responded on irregular adipose tissues. 3. In SEM study, collagen fibers were embedded by the plasma by the 5th day in the control group, and in the 3rd day experiment GMI were resorved but communited with collagen fiber till the 1st week. Collagen fibers were infilt-rated into GMI at the 2nd week and the infilltrated GMI were conglomerated with the mature adipose cells and the collagen fibers. From the above results, GMI implantation in the subcutaneous tissue of Sprague-Dawley rat, the mild infiltration of inflammatory cells were showed till 2nd week and the granulation tissues were observed. GMI were nearly resorbed till 2nd week, but well attached with adipose tissue and collagen fibers. The endothelium and fibroblasts were actively proliferated. Adipose tissues and mesenchymal tissue cells were observed. As already expressed, GMI showed resorptive change in course of time without any early immune reaction, and seemed to induce fibroblast activity and promote new collagen synthesis.
