Enhancement of Seed Germination by Aging, Cold-stratification, and Light Quality during Desiccation in Burcucumber (Sicyos Angulatus L.)

  • Seeds of burcucumber were treated with accelerated aging, cold-stratification, and light quality illuminated during desiccation to enhance their germination and seedling emergence. The germination was increased by aging and cold-stratification although the latter treatment showed greater effectiveness than the former one. In the combined treatment of aging 6 days at $45^{\circ}C$ and cold-stratification, the germination was promoted under longer period of cold-stratification to reach nearly 100% in 3 week cold-stratification on the ninth day from sowing. In the sequentially combined treatment of aging, cold-stratification, and light quality during 24 hour desiccation at $35^{\circ}C$, no-stratified seeds showed the highest rate in red light treatment but the lowest in far-red light. This implies that the phytochrome action run during the desiccation of imbibed seeds. The red light exposure during drying for the cold-stratified seeds after aging accelerated the germination even more than the dark treatment and germinated 100% on the next day of sowing. It is concluded that the sequential treatment of aging, cold-stratification, and red light illumination during desiccation can highly promote percentage and speed of burcucumber seed germination.

Seed Germination Response to Temperature, Cold Stratification Period, and Gibberellin Treatment in Spiraea fritschiana

  • To improve the germination of Spiraea fritschiana seeds for mass propagation, we evaluated the effect of a range of temperatures, cold stratification periods, and gibberellic acid ($GA_3$) treatments on three germination characteristics. Final germination percentage (FGP) increased as the temperature for seed germination increased, up to $30^{\circ}C$, while the mean germination time (MGT) and the mean number of days to 30% germination ($T_{30}$) decreased when seeds were germinated at $25-30^{\circ}C$. The optimum germination temperature of S. fritschiana seeds is approximately $30^{\circ}C$ considering FGP, MGT, and $T_{30}$ together. FGP increased with the duration of cold stratification up to a period of 6 weeks, but declined after 8 weeks of cold stratification, as prolonged cold stratification can induce dormancy with a resultant decline in germination. Pretreatment with 6-8 weeks of cold stratification or soaking seeds in distilled water or $500mg{\cdot}L^{-1}$ $GA_3$ for 24 h accelerated and increased the germination of S. fritschiana seeds, regardless of temperature. However, further study might be required to evaluate the effect of $GA_3$ concentrations lower than $500mg{\cdot}L^{-1}$ on the promotion of germination in S. fritschiana seeds.

  • When a cold HPSI (High Pressure Safety Injection) fluid associated with an design basis accident, such as LOCA (Loss of Coolant Accident), enters the cold legs of a stagnated primary coolant loop, thermal stratification phenomena will arise due to incomplete mixing. If the stratified flow enters a reactor pressure vessel downcomer, severe thermal stresses are created in a radiation embrittled vessel wall by local overcooling. Previous thermal-mixing analyses have assumed that the thermal stratification phenomena generated in stagnated loop of a partially stagnated coolant loop are neutralized in the vessel downcomer by strong flow from unstagnated loop. On the basis of these reasons, this paper presents the thermal-mixing analysis results in order to identify the fact that the cold plume generated in the vessel downcomer due to the thermal stratification phenomena of the stagnated loop is affected by the strong flow of the unstagnated loop.

Thermal-Mixing Analyses for Safety Injection at Partial Loop Stagnation of a Nuclear Power Plant

  • When a cold HPSI (High pressure Safety Injection) fluid associated with an overcooling transient, such as SGTR (Steam Generator Tube Rupture), MSLB (Main Steam Line Break) etc., enters the cold legs of a stagnated primary coolant loop, thermal stratification phenomena will arise due to incomplete mixing. If the stratified flow enters the downcomer of the reactor pressure vessel, severe thermal stresses are created in a radiation embrittled vessel wall by local overcooling. As general thermal-hydraulic system analysis codes cannot properly predict the thermal stratification phenomena, RG 1.154 requires that a detailed thermal-mixing analysis of PTS (pressurized Thermal Shock) evaluation be performed. Also. previous PTS studies have assumed that the thermal stratification phenomena generated in the stagnated loop side of a partially stagnated primary coolant loop are neutralized in the vessel downcomer by the strong flow from the unstagnated loop. On the basis of these reasons, this paper focuses on the development of a 3-dimensional thermal-mixing analysis model using PHOENICS code which can be applied to both partial and total loop stagnated cases. In addition, this paper verifies the fact that, for partial loop stagnated cases, the cold plume generated in the vessel downcomer due to the thermal stratification phenomena of the stagnated loop is almost neutralized by the strong flow of the unstagnated loop but is not fully eliminated.

Seed Dormancy and Germination Characteristics of Endemic Elder Species (Sambucus racemosa subsp. pendula) and Common Elder Species (S. williamsii) in Korea

  • This study was conducted to determine the seed dormancy types of Sambucus racemosa subsp. pendula Nakai and S. williamsii Hance. Low temperature stratification (1, 2, 3, 4 months) and GA3 treatment (1,000 mg/L) were performed on seeds to determine the type of seed dormancy. After the treatment, seeds were placed on a petri dish at 25℃ under light conditions. The germination rate and mean germination time were investigated. Results showed that cold stratification was effective in breaking the dormant state of the seed in both species. In the low temperature stratification treatment, the seed germination performance was improved as the treatment period was prolonged. Gibberellin treatment was effective in breaking the dormant state of S. racemosa subsp. pendula without low temperature stratification. However, S. williamsii did not break the dormant state of the seed by gibberellin treatment without low temperature stratification treatment. In the gibberellin treatment, germination performance was improved according to the low temperature stratification treatment period. As a result of this study, the seeds of S. williamsii have both an intermediate complex and a deep complex morphophysiological dormancy (MPD). In comparison, it was found that the S. racemosa subsp. pendula had intermediate composite MPD.

GA3처리와 저온습윤처리에 의한 흑노호(Kadsura coccinea)의 종자발아 및 유묘생장 촉진 (Gibberellic Acid and Cold Stratification Promotes Seed Germination and Seedling Growth in Kadsura coccinea)

  • Kadsura coccinea (Lem.) A.C. Smith is used as a medicinal plant and cosmetic material in China and Southeast Asia. To mass-produce Kadsura coccinea seedlings, the effects of gibberellic acid (GA3) and cold stratification treatments on seed germination were investigated. Seed germination rate with GA3 treatment was most effective at concentrations of 250 or 500 mg/L. With respect to mean germination time (MGT), mean daily germination, and T50 (days to reach 50% seed germination), the germination-promoting effect was improved as the concentration of GA3 increased. Stem growth of seedlings was the highest following GA3 treatments of 250 and 500 mg/L, and the growth promoting effect gradually decreased as the concentration of GA3 decreased. Root growth was stimulated at GA3 concentrations of 250-1,000 mg/L. Examination of the effect of stratification treatment for 15, 30 and 60 days at temperatures of 0, 5 and 10℃ on the germination rate revealed that the most stratification treatment temperature was 10℃, and the results improved with longer treatment periods. Altogether, GA3 and stratification treatments improved the seed germination rate, shortened the MGT, improved germination uniformity, and produced healthy seedlings.

저온습윤 및 지베렐린 처리가 눈잣나무의 종자발아에 미치는 영향 (Effect of Wet Cold and Gibberellin Treatments on Germination of Dwarf Stone Pine Seeds)

  • 눈잣나무는 우리나라에서 산림청 지정 멸종위기종으로 구분되어 있다. 눈잣나무는 설악산 대청봉 지역(해발고 1550–1700 m)에만 제한적으로 분포하고, 대부분의 구과가 조류와 설치류의 피해를 받기 때문에 종자를 확보하기 어렵다. 본 연구는 눈잣나무의 현지외보존림 조성을 위하여 습사, 예냉 및 GA3 처리가 눈잣나무 종자발아에 미치는 영향을 알아보기 위해 수행되었다. 눈잣나무 종자에 대해 습사 및 예냉처리를 각각 1, 2, 3, 4, 5개월 처리하고 GA3는 0, 100, 500, 1,000, 2,000, 3,000 ㎎/L 농도로 처리한 뒤 페트리디쉬에 치상하였으며 25℃의 광조건에서 실험하였다. 종자 발아율, 평균발아일수, 발아속도를 조사한 결과, 습사와 예냉처리는 모두 종자 발아를 향상시키는데 효과적으로 나타났다. 그러나 두 저온처리의 방법에 따른 차이는 나타나지 않았다. 각각의 처리에서 처리기간이 길어짐에 따라 종자 발아가 향상되었으나, 3개월 이상의 처리에서는 종자 발아의 향상이 통계적으로 유의하게 나타나지 않았다. GA3 처리에서도 눈잣나무의 종자 발아가 효과적으로 향상되었는데, 100 ㎎/L 처리 조건에서 발아율이 79.0%로 나타났다. 그러나 GA3 고농도 처리조건(2000 ㎎/L 이상)에서는 오히려 종자 발아가 감소하였다. 결론적으로 3개월 이상의 습사처리와 GA3 100 ㎎/L 처리가 눈잣나무의 종자의 발아향상에 적절한 방법으로 나타났다.

비상노심냉각계통 주입에 따른 저온관 및 강수관에서 단상 열성층 수치해석 : 부력항 고려 필요성에 관한 연구 (Numerical Analysis of Single Phase Thermal Stratification in both Cold Legs and Downcomer by Emergency Core Cooling System Injection : A Study on the Necessity to Consider Buoyancy Force Term)

  • When emergency core cooling system (ECCS) is operated during loss of coolant accident (LOCA) in a pressurized water reactor (PWR), pressurized thermal shock (PTS) phenomenon can occur as cooling water is injected into a cold leg, mixed with hot primary coolant, and then entrained into a reactor vessel. Insufficient flow mixing may cause temperature stratification and steam condensation. In addition, flow vibration may cause thermal stresses in surrounding structures. This will reduce the life of the reactor vessel. Due to the importance of PTS phenomenon, in this study, calculation was performed for Test 1 among six types of OECD/NEA ROSA tests with ANSYS CFX R.17. Predicted results were then compared to measured data. Additionally, because temperature difference between the hot coolant at the inlet of the cold leg and the cold cooling water at the inlet of the ECCS injection line is 200 K or more, buoyancy force due to density difference might have significant effect on thermal-hydraulic characteristics of flow. Therefore, in this study, the necessity to include buoyancy force term in governing equations for accurate prediction of single phase thermal stratification in both cold legs and downcomer by ECCS injection was numerically studied.

Germination of Hemerocallis Seeds as Influenced by Seed Development and Temperature Treatments

  • Germination of Hemerocallis seeds as influenced by cold stratification at $5^{\circ}C$ (CS) and maturity of seeds evaluated using X-ray imaging has not been well investigated. Seeds of H. lilioasphodelus, H. citrina, H. citrina 'April Flower', and H. minor collected from China and H. thunbergii collected from Korea were germinated at $20^{\circ}C$ without pre-temperature treatment, while H. hongdoensis, H. dumortieri, H. minor, and H. vespertina seeds were treated with CS. Harvesting 'Stella de Oro' capsules at 35-40 days after anthesis yielded mature seeds with well-developed embryo and cotyledons analyzed by X-ray images with a 92% germination in 17 days after sowing. Seeds of H. thunbergii and H. citrina germinated in less than 13 days without CS; two weeks of CS did not accelerate seed germination. Seeds of H. hongdoensis germinated in 24 days when seeds were stored at $25^{\circ}C$ without CS and in less than 27 days when cold stratified. Therefore, 'Stella de Oro' capsules should be harvested at 35-40 days after anthesis to harvest mature seeds. Cold stratification is not required to accelerate seed germination in the Hemerocallis taxa evaluated in this study.
