Development of Bioluminescence Immunoassay Using Photoprotein, Aequorin and Site-directed Immobilization

  • The heterogeneous bioluminescence immunoassay for digoxin was developed using photoprotein, native aequorin as a label and the site-directed immobilization technique based on avidin/biotin interaction. Aequorin is a bioluminescence protein, originally isolated from the jellyfish Aequoria Victoria and an attractive label in analytical applications because of sensitive detection due to virtually no background bioluminescent signal. Digoxin is a cardioactive drug, and its therapeutic level in serum is at low concentration with very narrow therapeutic index. The aequorin-digoxigenin conjugates were synthesized by the N-hydroxysuccinimide ester method and characterized in terms of bioluminescent residual activity. The resulting dose-response curve shows that the detection limit is $1.0\;{\times}\;10^{-10}\;M$ and a dynamic range is three orders of magnitude, which was obtained by $1.0\;{times}\;10^{-10}\;M$ conjugate and 0.9 μg/mL anti-digoxin antibody. Three structurally similar molecules to digoxin were examined for their cross-reactivity. None of these three compounds showed any crossreactivity with digoxin antibody employed in this study. Standard amounts of digoxin corresponding to the therapeutic range were spiked into the each serum solution. Study of the serum matrix effect indicated that correlation coefficient shows good agreement between luminescence light intensity between in buffer and in serum.

Manganese(II) Ion-Selective Membrane Electrode Based on N-(2-picolinamido ethyl)-Picolinamide as Neutral Carrier

  • A new poly (vinyl chloride) (PVC) membrane electrode that is highly selective to $Mn^{+2}$ ions was prepared using N,N'-bis(2'-pyridinecarboxamide)-1,2-ethane ($bpenH_2$) as a suitable neutral carrier. This concentration range ($1.0{\times}10^{-5}$ to $1.0{\times}10^{-1}\;M$) with Nernstian slope of $29.3{\pm}0.5\;mV$ per decade. The detection limit and the response time of electrode were $8.0{\times}10^{-6}\;M$ and (${\leq}15\;s$) respectively. The membrane can be used for more than two months without observing any divergence. The electrodes exhibited excellent selectivity for $Mn^{+2}$ ion over other mono-, di- and trivalent cations. Selectivity coefficients were determined by the matched potential method (MPM). The electrode can be used in the pH range from 4.0 - 9.0. The isothermal coefficient of this electrode amounted to 0.00023 V/$^{\circ}C$. The stability constant (log $K_s$) of the $Mn^{+2}$ - $bpenH_2$ complex was determined at $25^{\circ}C$ by potentiometric titration in mixed aqueous solution. The proposed electrode was applied to the determination of $Mn^{+2}$ ions in real samples.

Determination on the Optimal Sample Size in the Aquatic Insect Community Analysis - Pangtae Creek Model (수서곤충 군집분석에 있어서 최적표본크기의 결정 - 방태천 모형)

  • A molel study was conducted to determine the optimal sample size for the analysis of the aquatic insect community in a stream reach of the Pangtae Creek, Kangwon-do in October 1995 and may 1996. The results showed that the required minimum sample size varied and depended on the purposes of the community analysis. Acoording to the Species: Area Curve method, at least 16 Surber samplings ($30{\times}30cm$) were required in a stream reach in each spring and fall survey. The species diversity index did not vary significantly as the sample size increased. Based on the coefficient of variation analysis, the minimum sample sizes of 10 were required in order to compare seasonal differences of the community in the study area. Considering the static community structure of aquatic insects, including both species numbers and individual numbers of aquatic insects, 11 and 7 samplings were optimal sizes for the fall and spring survey, respectively. We concluded that 12 Surber samplings from 3 riffle-pool sequences (4 samplings at each riffle-pool sequence) would be required in a stream reach (length 1 km) to obtain reliable as well as cost efficient data. Our model showed that the optimal sample size should be determined by interactions between minimum sample size, the degree of data reliability, and cost efficiency.

Effect of Swirl Flow Disturbance on Uncertainty of Flow Rate Measurement by Venturi (선회유동 교란에 따른 벤투리 유량측정의 불확실성 해석)

  • Venturi has long been an attractive method of measuring flow rate in a variety of engineering applications since pressure loss is relatively small compared with other measuring methods. The current study focuses on making detailed uncertainty estimations as the upstream flow disturbance affects uncertainty levels of the flow rate measurement. Upstream flow disturbance can be determined by 9 different swirl generators. Measurement uncertainty of flow rate has been estimated by a quantitative uncertainty analysis which is based on the ANSI/ASME PTC 19.1-2005 standard. The results of flow rate uncertainty analysis show that the case with systematic error has higher than that without systematic error. Especially the result with systematic error exhibits that the uncertainty of flow rate was gradually increased by swirl flow disturbance. The uncertainty of flow rate measurement can be mainly affected by differential pressure and discharge coefficient. Flow disturbance can be also reduced by increasing of the upstream straight length of Venturi.

Development of Korean Infant Developmental Assessment Scale (한국형 영아발달사정도구의 개발)

  • Purpose: To develop an Infant Developmental Assesment Scale appropriate for Korean infants that includes cultural context. Method: The total sample included 990 infants aged birth to 24months. The developmental tests were conducted at hospital well baby clinic, daycare centers, public health centers and homes located in city, urban and rural areas. Item analysis was done to examine content validity of the scale. Test responses were analyzed by SPSS to examine the inter-rater reliability, and construct validity. Result: Based on the naturalistic observation and analysis of other developmenta l instruments, 260 Korean infant developmental items were created. The mean score of the developmental quotient of 966 infants was 101.63±10.89. Correlation coefficient of inter-rater reliability was .99. In addition, 84.6% of total items showed Kappa .70 and above. The development quotient was significantly different depending upon the residential area and father's occupation, therefore, construct validity using known group technique was supported. Conclusion: The developed instrument can be used to assess the development of handicapped infants, as well as normal infant's general abilities and to study individual differences within the normal range for Korean.

  • This paper deals with a new mathematical formulation of nonlinear wave profile based on Banach fixed point theorem. As application of the formulation and its solution procedure, some numerical solutions was presented in this paper and nonlinear equation was derived. Also we introduce a new operator for iteration and getting solution. A numerical study was accomplished with Stokes' first-order solution and iteration scheme, and then we can know the nonlinear characteristic of Stokes' high-order solution. That is, using only Stokes' first-oder(linear) velocity potential and an initial guess of wave profile, it is possible to realize the corresponding high-oder Stokian wave profile with tile new numerical scheme which is the method of iteration. We proved the mathematical convergence of tile proposed scheme. The nonlinear strategy of iterations has very fast convergence rate, that is, only about 6-10 iterations arc required to obtain a numerically converged solution.

Development of a Wave Absorbing System Using a Liquefied Sandbed

  • A new wave-absorbing system, called the liquefied sandbed wave barrier (LSWB) system, is currently under development at the Port and Airport Research Institute (PARI) of Japan. The wave damping effect by the LSWB system is substantial, as confirmed by small-scale experiments and FEM numerical calculations, i.e., the wave transmission coefficient of the system is less than 0.2. Here, the results of large-scale experiments arediscussed in view of practical application. Although the LSWB system provides high wave damping, nearly equal to theoretical values, difficulty exists in obtaining a homogeneously liquefied sandbed, due to the occurrence of liquefied sandbed compaction by cyclic wave loading, which in turn, reduces excess pare pressure and the wave damping effect. These two phenomena primarily occur when the sandbed is composed of fine sand with small permeability. Based on experimental results, we propose a design method that includes countermeasures against such problems, and a prototype LSWB system is constructed in a very large wave flume at PARI. Wave damping by the prototype LSWB system is confirmed to be quite stable and high, as predicted by theoretical calculations.

A Study on Flow and Creation and Dissipation of Vorticity around Rectangular Floating Breakwater (부방파제의 유동과 와의 생성 및 소멸에 관한 연구)

  • In this study, flow and creation and dissipation of vorticity around rectangular floating breakwater is investigated both experimentally and numerically. The PIV system(Particle image velocimetry) is employed to obtain the velocity field in the vorticity of rectangular structure. The numerical model, combined with ${\kappa}-{\varepsilon}$ turbulence model and the VOF method based on RANS equation, is used to analyze the turbulence structure. In the results of this study, the vorticity is found around conner of rectangular structure at all time domain, and creation and dissipation of vorticity are closely related to wave period. Separation points of phase of vortex due to flow separation for longer period waves are faster then for shorter period waves.

New Interference Alignment Technique using Least Square Method in Multi-User MIMO Systems (다중 사용자 MIMO 시스템에서 최소 제곱 기법을 이용한 새로운 간섭 정렬 기법)

  • In this paper, the scheme for designing optimal beamforming matrix for interference control is proposed. The optimal beamforming matrix is found though linear combination of interference alignment conditions and renewal of linear combination coefficient. The proposed scheme has advantages that the complexity is reduced and there is no multiplying operation in matrix calculations even if proposed scheme has the form similar to that of existing least square based scheme. The simulation results show that proposed scheme has about 4bps/Hz higher gain than existing least square scheme. Also there is no additional multiplying calculation and increase of matrix size when the number of transmit and receive antennas is increased.

An Experimental Study on Hydration Heat Characteristics for Thermal Crack Analysis Based on FEM of Urea Mixed Mass Concrete (Urea 혼입 매스콘크리트의 FEM 온도균열 해석을 위한 수화발열특성에 관한 실험적 연구)

  • In domestic construction industry progress, construction and quality control of large structures are considered to be important as the superstructure and mass scale of structures. In the case of mass concrete, high hydration heat caused by cement hydration generates temperature stress by generating internal temperature difference with the concrete surface. These temperature stresses cause cracks to penetrate the concrete structure. A method of lowering the heat generation by incorporating Urea in order to reduce the concrete temperature crack has been proposed. In this study, the heat function coefficient for the FEM temperature crack analysis of the mass concrete containing the element was derived and the adiabatic temperature rise test was carried out according to the incorporation of the element. As a result of this experiment, the maximum temperature of 41 ± 1℃ was obtained irrespective of the amount of urea, and the maximum temperature decreased by 16.9℃ in concrete containing 40kg/㎥ of urea.
