Outbreak of Fire Blight of Apple and Pear and Its Characteristics in Korea in 2019 (2019년 국내 사과와 배 화상병 대발생과 그 특징)

  • To find out the cause of the fire blight outbreak in apples and pears of Korea in 2019, we investigated disease appearing situation of thirty fire blight infected orchards, and interviewed farmers to determine the cultivation characteristics. Fire blight occurred mostly in orchards that had infected more than 2 years before. The cause of this were as follows: farmers did not know the symptoms of the disease properly. It is presumed that it has spread from the first occurrence to the surrounding orchards by flower-visiting insects or farmers and to a short distance or a long distance by the same cultivator or co-farmer. These series of processes repeated in the newly spreading area, and then disease reports increased as farmers became aware of fire blight. To minimize the spread of fire blight in Korea, it suggested that thorough education of farmers for early diagnosis and quantitative detection technology that can diagnose even in no symptom showing plants. And chemical or biological spraying systems suitable for domestic cultivation methods, which are producing large fruits, and molecular epidemiological studies of pathogens.

Evaluation of Antithrombosis and Antioxidant Activities of the Ethanol Extract of Different Parts of Hibiscus cannabinus L. cv. 'Jangdae' (케나프 장대 품종의 부위별 에탄올 추출물의 항혈전 및 항산화 활성)

  • Kenaf (Hibiscus cannabinus L.), one of the four major fiber crops, is attracting attention for its efficient CO2-absorbing ability and versatility for producing daily supplies, including textiles. In Korea, a new cultivar 'Jangdae' was established in 2013. The ease of cultivation and seed gathering of 'Jangdae' has led to its nationwide cultivation. However, evaluation of the bioactivities of the different parts of kenaf, and especially the 'Jangdae' cultivar, remains rudimentary. In this study, the antithrombosis and antioxidant activities of extracts prepared from different parts of the 'Jangdae' cultivar were evaluated by determining their effects on blood clot formation. Extracts prepared from seeds (HC-SD), seedpods (HC-SP), leaves (HC-L), stems (HC-S), and roots (HC-R) of the 'Jangdae' cultivar strongly inhibited blood clot formation. In particular, the HC-SD, HC-SP, and HC-S extracts showed strong inhibition against the coagulation factors prothrombin, and thrombin. The HC-SP extract showed strong antioxidant activities, such as scavenging ability against DPPH anion, ABTS cation, nitrite, and reducing power. Since blood clot formation is closely related to oxidative stress, the HC-SP extract could be developed as a novel anticoagulation and antioxidant resource. This is the first report of the antithrombosis activities of different parts of H. cannabinus L. cv. 'Jangdae'.

Growth-promoting effect on Tricholoma matsutake mycelium by Terrabacteria isolated from pine mushroom habitats in Korea (국내 송이 자생지에서 분리된 Terrabacteria에 의한 송이균사체 생장촉진 효과)

  • To cultivate pine mushroom (Tricholoma matsutake) artificially, co-cultivation with microorganisms has been introduced. Here, experiments were performed to assess the growth-promoting effect of bacteria on T. matsutake mycelia. Bacteria were isolated from soil samples collected in Yangyang County, Korea. Four of the bacterial isolates (Y22_B06, Y22_B11, Y22_B18, and Y22_B22) exhibited a growth-promoting effect on T. matsutake mycelia (154.67%, 125.91%, 134.06%, and 158.28%, respectively). To analyze the characteristics of the bacteria, especially the antifungal activity, 𝛼-amylase and cellulase activity assays were performed. In comparison with the controls, the isolated bacteria exhibited low 𝛼-amylase and cellulase activity. 16S rRNA gene sequencing was performed to identify the four bacterial isolates. The isolates belonged to the Terrabacteria group and were identified as Microbacterium paraoxydans, Paenibacillus castaneae, Peribacillus frigoritolerans, and P. butanolivorans. These bacterial isolates are expected to have contributed to the growth promotion of T. matsutake mycelia and the artificial cultivation of T. matsutake.

Growth promoting effect on Tricholoma matsutake mycelium by bacteria from fairy Ring in Bonghwa-gun, Korea (국내 봉화 송이 자생지 내 균환 유래 토양세균의 송이균사체 생장촉진 효과)

  • As a member of ectomycorrhizal fungi, Tricholoma matsutake has a symbiotic relationship with its host, Pinus densiflora. To cultivate T. matsutake artificially, the co-cultivation of T. matsutake mycelia and bacteria from shiro was introduced. In this study, bacteria were isolated from soil samples in Bonghwa-gun, and seven bacterial isolates (B22_7_B05, B22_7_B06, B22_7_B07, B22_7_B08, B22_7_B10, B22_7_B13, and B22_7_B14) promoted the growth of T. matsutake mycelia (147.48, 232.11, 266.72, 211.43, 175.17, 154.62, and 177.92%, respectively). Sequencing of the 16S rRNA region of the isolated bacteria was performed. B22_7_B05 and B22_7_B10 were identified as Bacillus toyonensis, B22_7_B06 and B22_7_B08 as Paenibacillus taichungensis, B22_7_B07 and B22_7_B14 as P. gorilla, and B22_7_B13 as P. odorifer. These bacterial isolates were associated with the shiro community and are expected to contribute to the cultivation of T. matsutake.

Growth-promoting effect of microorganisms from a fairy ring in Yangyang, Korea on Tricholoma matsutake mycelium (국내 양양 송이 자생지 내 균환 유래 토양미생물과 송이균사체 생장촉진 효과)

  • Tricholoma matsutake is a traditional favorite food in East Asia, cultivated in fairy rings called "shiro," which are found near Pinus densiflora. For effective artificial cultivation of Tri. matsutake, microorganisms from symbiotic fairy rings are co-cultivated. In this study, one bacterial isolate (Y22_B35) and two fungal isolates (Y22_F64 and Y22_F68) displayed growth-promoting effects on Tri. matsutake mycelium (158.47, 125.00, and 122.26% enhanced growth, respectively). For identification, 16S rRNA or ITS regions from the microorganisms¡¯ genomes were sequenced. Other sequences, including BenA, CaM, and RPB2 were sequenced in the fungal isolates. The bacterial isolate Y22_B35 was identified as Bacillus cereus. Y22_F64 and Y22_F68 were identified as Umbelopsis nana and Aspergillus parvulus, respectively. To identify the effects of the dominant microorganisms on Tri. Matsutake cultivation, metagenomic analyses were performed. Discovery of these Tri. matsutake mycelium growth-promoting microorganisms and metagenomics analyses are expected to contribute to our understanding of Tri. matsutake fruiting body growth and construction of biomimicry.

Analysis of Microbial Communities of Salted Cabbage and Kimchi according to Cultivation Areas (재배지역에 따른 절임배추 및 김치의 미생물 군집 분석)

  • In this study, the microbial distribution and diversity of kimchi manufactured in the same method as salted cabbage manufactured from Pyeongchang, Andong, and Haenam cabbage according to the storage period were compared. Among Pyeongchang, Andong, and Haenam salted cabbages, the Haenam salted cabbage microbial community showed the highest diversity on the 1st day of storage. As the storage period of salted cabbage increased, the alpha diversity value increased, the proportion of cyanobacteria decreased, and bacteria derived from sea salt and water increased. Principal coordinates analysis(PCoA) and unweighted pair group method with arithmetic mean(UPGMA) trees showed that Andong salted cabbage on the 5th day of storage had a microbial community close to salted cabbage on the 10th day of storage. At the species level, Sinocapsa zengkensis was 78.65%, 90.64%, and 63.44%, respectively, in Pyeongchang, Andong, and Haenam salted cabbages on the 1st day of storage. Marinomonas primoryensis was showed in Pyeongchang, Andong, and Haenam salted cabbage on the 5th day of storage at 24.39%, 26.60%, and 21.75%, respectively, and at 42.17%, 31.43%, and 45.21%, respectively, on the 10th day of storage. Kimchi made from Pyeongchang, Andong, and Haenam salted cabbages showed Janthinobacterium lividum at 30.47%, 29.60%, and 25.97%, respectively. In addition, Leuconostoc spp. involved in fermentation were showed from the 5th day of storage, but Andong salted cabbage on the 10th day of storage was not showed. These results show to be due to differences in soil, climate, and cultivation methods of cabbage.

Swinging Effect of Salicylic Acid on the Accumulation of Polyhydroxyalkanoic Acid (PHA) in Pseudomonas aeruginosa BM114 Synthesizing Both MCL- and SCL-PHA

  • A bacterium, Pseudomonas aeruginosa BM114, capable of accumulating a blend of medium-chain-length (MCL)- and short-chain-length (SCL)-polyhydroxyalkanoic acid (PHA), was isolated. Salicylic acid (SA), without being metabolized, was found to specifically inhibit only the accumulation of MCL-PHA without affecting cell growth. An addition of 20 mM SA selectively inhibited the accumulation of MCL-PHA in decanoate-grown cells by 83% of the control content in one-step cultivation, where overall PHA accumulation was inhibited by only ${\sim}11%$. Typically, the molar monomer-unit ratio of the PHA for 25 mM decanoate-grown cells changed from 46:4:25:25 (=[3-hydroxybutyrate]:[3-hydroxycaproate]: [3-hydroxyoctanoate]:[3-hydroxydecanoate]) at 0 mM SA (dry cell wt, 1.97 g/l; PHA content, 48.6 wt%) to 91:1:4:4 at 20 mM SA (dry cell wt, 1.85 g/l; PHA content, 43.2 wt%). Thus, the stimulation of SCL-PHA accumulation was observed. Growth of P. aeruginosa BM114 on undecanoic acid also produced a PHA blend composed of 47.4% P(3HB-co-3-hydroxyvalerate) and 52.6% P(3-hydroxyheptanoate-co-3-hydroxynonanoate-co-3-hydroxyundecanoate). Similar to the case of even-carboxylic acids, SA inhibited the accumulation of only MCL-PHA, but stimulated the accumulation of SCL-PHA. For all medium-chain fatty acids tested, SA induced a stimulation of SCL-PHA accumulation in the BM114 strain. SA could thus be used to suppress only the formation of MCL-PHA in Pseudomonas spp. accumulating a blend of SCL-PHA and MCL-PHA.

Development of Land Fill Gas(LFG)-MGT Power Generation and Green House Design Technology (쓰레기 매립지 MGT 발전 및 유리온실 설계기술개발)

  • The high fuel flexibility of Micro Gas Turbine(MGT) has boosted their use in a wide variety of applications. Recently, the demand for biogas generated from the digestion of organic wastes and landfill as a fuel for gas turbines has increased. We researched the influence of firing landfill gas(LFG) on the performance and operating characteristics of a micro gas turbine combined heat and power system. $CH_4$ and $CO_2$ simultaneous recovery process has been developed for field plant scale to provide an isothermal, low operating cost method for carrying out the contaminants removal in Land Fill Gas(LFG) by liquid phase catalyst for introduce into the green house for the purpose of $CO_2$ rich cultivation of the plants. Methane purification and carbon dioxide stripping by muti panel autocirculation bubble lift column reactor utilizing Fe-EDTA was conducted for evaluate optimum conditions for land fill gas. Based on inflow rate of LFG as 0.207 $m^3$/min, 5.5 kg/$cm^2$, we designed reactor system for 70% $CH_4$ and 27% $CO_2$ gas introduce into MGT system with $H_2S$ 99% removal efficiency. A green house designed for four different carbon dioxide concentration from ambient air to 1500 ppm by utilizing the exhaust gas and hot water from MGT system.

A Study on Effects of Oil Contaminated Soil on the Growth of Plant (유류오염토양이 식물식생에 미치는 영향에 관한 연구)

  • Oil contamination soil has been one of the most environmental social issues for decades in the inside and outside of country. The law of soil environmental preservation was carried out in the 1990s and the government controlled not only soil environment management and the remediation of contaminated soil but also promoted the development of remedial technology and cleanup business of contaminated soil by national policy. In addition to agriculture areas, the main oil contaminated sites are a gas station, oil reservoir, petro-chemical complex, site of railway carriage base and military camp. The contamination-frequency of agriculture area and effect sites are low but it has significantly important area on account of producing food for human beings. Therefore, we should be concerned about oil contamination damage of agriculture area. The oil contamination damage of agriculture area influenced drop of birth and breeding since the oil directly adheres to seeds and farm products even diffusion of contaminated soil to cultivation area. The studies of the crops and the food vegetation has not enough detailed data caused by the incident of oil contamination. This study investigated the effect of oil in germination and growth of selected plant seeds. In this study, we try to verify whether the oil contamination by accidents on farmland influenced the damage of farm produce and the mutual relation both oil contaminated soil or the vegetation of crops. The impact of oil on plant development was followed by phytotoxicity assessments. The plants exhibited visual symptoms of stress, growth reduction and perturbations in developmental parameters. The increase of the degree of pollution induced more marked effects in plants, likely because of the physical effects of oil. The relationships between the phytotoxicity contents of plants and growth reduction suggest a chemical toxicity of fuel oil. In addition, while cleaned up the contaminated soil under the standard of contaminated soil we examined it was suitable for region standard and it may have practical possibility for fill material of construction of afforestation and molding soil of landfill.

Dual Coating Improves the Survival of Probiotic Bifidobacterium Strains during Exposure to Simulated Gastro-Intestinal Conditions (위장관내 조건에서 이중코팅 처리 된 프로바이오틱 비피도박테리움의 생존력 향상)

  • Probiotics have been reported to benefit human health by modulating immunity, lowering cholesterol, improving lactose tolerance, and preventing some cancer. Once ingested, probiotic microorganisms have to survive harsh conditions such as low pH, protease-rich condition, and bile salts during their passage through the gastro-intestinal (GI) tract colonize and proliferate to exert their probiotic effects. The dual coating technology, by which the bacteria are doubly coated with peptides and polysaccharides in consecutive order, was developed to protect the ingested bacteria from the harsh conditions. The aim of the study was to evaluate the viable stability of a doubly coated blend of four species of Bifidobacterium by comparing its bile/acid resistance and heat viability in vitro with that of the non-coated blend. After challenges with acid, bile salts, heat, and viable cell counts (VVCs) of the dual coated and non-coated blend were determined by cultivation on agar plates or flow cytometric measurement after being stain with the BacLigtht kit$^{TM}$. The results showed that the dual coated blend was much higher resistant to the acidic or bile salt condition than the non-coated blend and heat viability was also higher, indicating that the dual coating can improve the survival of probiotic bacteria during their transit through the GI tract after consumption.
