  • The demands of complicated data communications have issued a new challenge to port logistics systems. Customers expect ports to handle their generated administrative data while a vessel is docked in a port. One port logistics system, known as the Vessel Clearance Process (VCP), manages large numbers of documents related to port of entry. In the VCP, information flows through many organizations such as the port authority, shipping agents, marine offices, immigration offices, and others. Therefore, for effective management of the Business Process (BP) of the VCP, a standardized method of BP modeling is essential, especially in heterogeneous system environments. In a port, according to port policy, terms and data are sued that are similar to but different from those of other logistics partners, which hinders standardized modeling of the BP. In order to avoid tedious and time-consuming document customization work, more convenient modeling of BP for VCP is essential. This paper proposes an ontology-based process similarity measurement to assist designer for process modeling in port domain, especially VCP. We expect that this methodology will use convenient and quick modeling of port business processes.

Effect of Analytical Method on the Pharmacokinetic Evaluation of Bromosulfophthalein: Comparison of HPLC and UV Spectroscopy Method (Bromosulfophthalein의 체내동태 평가에 미치는 분석법의 영향: HPLC 법과 UV 흡광광도법의 비교)

  • The aim of this study was to evaluate the difference of analytical methods for the pharmacokinetic study of bromosulfophthalein (BSP), an indicator of hepatobiliary function. The plasma and bile concentrations of BSP after intravenous administration were measured according to custom UV spectroscopy and HPLC, respectively. Plasma concentration of BSP measured by UV spectroscopy was similar to that measured by HPLC. There was no significant difference in the distribution volume, total body clearance, area under the curve and mean residence time of BSP between different analytical method groups. However, bile concentration of BSP measured by UV spectroscopy was overestimated compared with concentration measured by HPLC method. Biliary clearance of BSP obtained from UV spectroscopy method was almost 3 times higher than that obtained from HPLC method. Thus, a feasibility of UV spectroscopy method for high throughput pharmacokinetic evaluation of BSP was limited to the study based on the plasma concentration of BSP, not bile concentration.

The Influence of Cimetidine on the Pharmacokinetics of Diltiazem and its Main Metabolite in Rabbits

  • The purpose of this study was to investigate the pharmacokinetic alteration of diltiazem and its main metabolite, deacetyldiltiazem, after oral administration of diltiazem in rabbits with or with-out cimetidine co-administration. The area under the plasma concentration-time curve (AUC) of diltiazem was significantly elevated in rabbits pretreated with cimetidine, suggesting that the oral clearance, an index of intrinsic clearance, may be decreased by the cimetidine treatment. Consistent with the increased AUC by the treatment, peak plasma concentration ($C_{max}$) for diltiazem was also elevated. Apparent volume of distribution normalized by the bioavailability (($V_{d}$/F) of diltiazem increased sigrificantly in rabbits pretreated with cimetidine increased. Taken together with the fact that the first pass metabolism for diltiazem is the primary determinant for the oral bioavailability, these observations indicate that increases in the oral clearance and (($V_{d}$/F may be a manifestation of the decreased first pass metabolism. Consistent with the hypothesis, the AUC of deacetyldiltiazem was significantly decreased in rabbits with cimetidine treatment. Ratio of deacetyldiltiazem to total diltiazem in the plasma was significantly decreased in rabbits with cimetidine treatment. These observations suggested that the metabolism of diltiazem to deacetyldiltiazem was reduced by cimetidine treatment and that the dosage of diltiazem should be adjusted when the drug is co-administered chronically with cimetidine in a clinical setting.

  • Bromphenol blue (BPB) was studied with rabbits in normal and disease states to understand the basic principles of hepato-biliary transport process, and the effect of disease states on the drug disposition. The time course of plasma concentration and of biliary excretion was studied in normal and $CCl_4$ intoxicated rabbits. A conspicuous retention of BPB clearance from the plasma was observed, and the slope of the first-phase of plasma curve was decreased in the intoxicated rabbits. The shape of biliary excretion was same in normal and intoxicated states, but the amount of BPB excreted into bile in the intoxicated states was much smaller than in normal states. A relationship was found which enables one to predict the pattern of uptake of BPB by the liver, and the pattern of excretion into the bile in normal states, but was not in $CCl_4$ intoxicated states. It may be that the application of this experiments would extend the effect of disease states on the drug disposition.

Tip Clearance Effects on Inlet Hot Streaks Migration Characteristics in Low Pressure Stage of a Vaneless Counter-Rotating Turbine

  • In this paper, three-dimensional multiblade row unsteady Navier-Stokes simulations at a hot streak temperature ratio of 2.0 have been performed to reveal the effects of rotor tip clearance on the inlet hot streak migration characteristics in low pressure stage of a Vaneless Counter-Rotating Turbine. The hot streak is circular in shape with a diameter equal to 25% of the high pressure turbine stator span. The hot streak center is located at 50% of the span and the leading edge of the high pressure turbine stator. The tip clearance size studied in this paper is 2.0mm(2.59% high pressure turbine rotor height, and 2.09% low pressure turbine rotor height). The numerical results show that the hot streak is not mixed out by the time it reaches the exit of high pressure turbine rotor. The separation of colder and hotter fluid is observed at the inlet of low pressure turbine rotor. Most of hotter fluid migrates towards the rotor pressure surface, and only little hotter fluid migrates to the rotor suction surface when it convects into the low pressure turbine rotor. And the hotter fluid migrated to the tip region of the high pressure turbine rotor impinges on the leading edge of the low pressure turbine rotor after it goes through the high pressure turbine rotor. The migration of the hotter fluid directly results in very high heat load at the leading edge of the low pressure turbine rotor. The migration characteristics of the hot streak in the low pressure turbine rotor are dominated by the combined effects of secondary flow and leakage flow at the tip clearance. The leakage flow trends to drive the hotter fluid towards the blade tip on the pressure surface and to the hub on the suction surface, even partial hotter fluid near the pressure surface is also driven to the rotor suction surface through the tip clearance. Compared with the case without rotor tip clearance, the heat load of the low pressure turbine rotor is intensified due to the effects of the leakage flow. And the numerical results also indicate that the leakage flow effect trends to increase the low pressure turbine rotor outlet temperature at the tip region.

Calculation of a First-In-Man Dose of 7-O-Succinyl Macrolactin A Based on Allometric Scaling of Data from Mice, Rats, and Dogs

  • 7-O-Succinyl macrolactin A (SMA) exerts several pharmacological effects including anti-bacterial, anti-inflammation, and anti-cancer activities. Recently, SMA has been extensively evaluated as an anti-cancer drug. Thus, the objectives of the present study were to characterise the pharmacokinetics of SMA via both non-compartmental and compartmental analysis in mice, rats, and dogs, and to derive an appropriate first-in-man dose based on allometric scaling of the animal data. The time courses of plasma SMA concentrations after intravenous administration to rats and dogs were analysed retrospectively, as were data collected after intraperitoneal SMA injection in mice. Pharmacokinetic parameters were estimated via both noncompartmental and compartmental analysis, and were correlated with body weight and/or the potential maximum life-span. The clearance and distribution volume of SMA in humans were predicted, and a first-in-man dose proposed. A two-compartment model best described the time courses of SMA plasma concentrations after a saturation elimination process was applied to fit the dataset obtained from rats. Incorporation of the maximum potential life-span during allometric scaling was required to improve the estimation of human clearance. The SMA clearance and the distribution volume in the steady state, in a 70-kg adult male, were estimated to be 30.6 L/h and 19.5 L, respectively. To meet the area under the curve (AUC) required for anti-tumour activity, a dose of 100 mg (~1.5 mg/kg) was finally proposed as the first dose for a 70-kg human. Although toxicological profiles derived from non-clinical studies must be considered before any final decision is made, our work will facilitate clinical studies on SMA.

Loganin Ameliorates Acute Kidney Injury and Restores Tofacitinib Metabolism in Rats: Implications for Renal Protection and Drug Interaction

  • Tofacitinib, a Janus kinase (JAK) inhibitor used to treat rheumatoid arthritis, is metabolized through hepatic cytochrome P450 (CYP), specifically CYP3A1/2 and CYP2C11. Prolonged administration of rheumatoid arthritis medications is generally associated with an increased risk of renal toxicity. Loganin (LGN), an iridoid glycoside, has hepatorenal regenerative properties. This study investigates the potential of LGN to mitigate acute kidney injury (AKI) and its effects on the pharmacokinetics of tofacitinib in rats with cisplatin-induced AKI. Both intravenous and oral administration of tofacitinib to AKI rats significantly increased the area under the plasma concentration-time curve from time 0 to infinity (AUC) compared with control (CON) rats, an increase attributed to the decelerated non-renal clearance (CLNR) and renal clearance (CLR) of tofacitinib. Administration of LGN to AKI rats, however, protected kidneys from severe impairment, restoring the pharmacokinetic parameters (AUC, CLNR, and CLR) of tofacitinib to those observed in untreated CON rats, with partial recovery of kidney function, as evidenced by an increase in creatinine clearance. Possible interactions between drugs and natural components should be considered, especially when co-administering both a drug and a natural extract containing LGN or iridoid glycosides to patients with kidney injury.

An Empirical Study on Trade Facilitation by the Korean Government's Single Window System

  • Purpose - Korea became a trillion-dollar trading country in 2011. With the exponential increase in Korea's trade volume over the past decades, trade-related administrative burdens per capita for Korea Customs became enormous, for which the government established the Single Window, a trade-facilitating system, in 2004 to enhance the efficiency of customs-clearing procedures for traders. This paper focuses on finding whether the Korean Single Window system affects the country's trade facilitation positively through an empirical methodology. Design/methodology - To find empirical evidence that Single Window affects trade facilitation for the customs-clearing procedure, this study assumes that a time-efficient environment enables the handling of the increase in trade volume, under which four independent variables related to import customs-clearing procedures and two dependent variables to import were adopted for empirical analysis. The import customs procedures are classified into four steps from port entry to declaration acceptance. To understand the relationship between variables, scattered plots and correlation coefficients were calculated. Eight hypotheses were set and underwent simple linear regression. The data for analysis were collected by Korea Customs, and were about the lead time of import, the volume of imports in million USD, and the number of import declarations reported to customs offices on a monthly basis from 2005 to 2013. Findings - Six of the eight hypotheses showed the statistically significant result that lead time in the import customs-clearing procedure positively affects the number of import declaration reports and import volume. Specifically, Hypothesis 1, Hypothesis 2, and Hypothesis 3 strongly support the assumption lead time in import customs declaration has an inverse relationship with the number of import declarations, which means that the shorter the import lead time, the more import declaration increases. Research Limitations/Implications - With limited data accessibility to the government's custom-sclearing procedures, only the import lead time for customs clearance were adopted as independent variables. This paper, however, successfully found that the Single Window system contributed to trade facilitation. Originality/value - This study found that the time-saving Single Window system of Korea Customs enables itself to manage an exponentially-increasing trade volume by creating a trade-facilitating environment for customs personnel and traders, which may be a unique implication found through quantitative methodology.

  • Application of air foil bearing (AFB) is widely using to cope with requirements of high speed and high efficiency system at turbo machinery. At same time, a wide variety of AFB design methods are required. In this paper, the AFB design factor influencing the most greatly in bearing performance was selected through theoretical analysis and vibration test was conducted using selected factor. Through this process, AFB system applicable for vehicle turbo machinery was developed.
