Toxicity Assessment of PFOA and PFOS Using Freshwater Flea Hyalella azteca (담수산 물벼룩인 Moina macrocopa를 이용한 과불화화합물 PFOA와 PFOS의 독성평가)

  • Perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) are fully flurorinated organic compounds which are highly persistent in environment and accumulated in organism. These chemicals are released to the environment at their manufacture, during their use in industrial and consumer applications and from disposal after their use. The purpose of this study was to determine the effects of PFOA and PFOS on the freshwater flea (Moina macrocopa). Acute toxicity test and chronic toxicity test were performed for 2 days and 10 days, respectively. Acute toxicity was assessed on the basis of mortality, while chronic toxicity was assessed by fecundity as well as mortality. The acute toxicity studies on PFOA and PFOS showed that the values of $LC_{50}$ were $73.9\;mg/L\;and\;27.7\;mg/L$ respectively. In the chronic toxicity test, fecundity was reduced significantly at 24.1 mg/L of PFOA and 9.3 mg/L of PFOS, respectively. Conclusively, the results of this work suggest that Moina macrocopa could be a suitable model organism for screening and assessing of environmental pollutants in water.

Effects of Endocrine Disruptors, Nonylphenol in Daphnia magna (물벼룩 (Daphnia magna)에서 내분비계장애물질인 노닐페놀의 영향)

  • Nonylphenol is one of endocrine disruptors, as structurally stable, hydrophobic compounds exhibit high condensability and long-lasting in the natural environment. The purpose of this study was to determine the toxic effects of nonylphenol on Daphnia magna. In acute toxicity test, D. magna was exposed for 48 h at concentrations of 0, 10, 18, 32, 56 and $100{\mu}g\;L^{-1}$ nonylphenol. In chronic toxicity test, D. magna were exposed through water for 21 days at concentrations of 0, 1.0, 1.8, 3.2, 5.6 and $10{\mu}g\;L^{-1}$ nonylphenol. Acute toxicity was assessed on the basis of immobility, while chronic toxicity was assessed on the basis of fecundity. The acute toxicity test on nonylphenol was showed that the values of 24 h and 48 h $EC_{50}$ were $25.0{\mu}g\;L^{-1}$ and $13.7{\mu}g\;L^{-1}$, respectively. In chronic test, fecundity was reduced significantly at $5.6{\mu}g\;L^{-1}$ of nonylphenol. These results indicated that nonylphenol have some hazard for acute or chronic toxicity to freshwater invertebrate organism.

Prioritization of Chemicals for Chronic/Carcinogenic Inhalation Testing (만성발암성 흡입독성시험 우선순위 물질 선정 연구)

  • Objectives: In this study, we seek to perform a priority selection for test substances for chronic inhalation toxicity studies, including acute and subchronic inhalation toxicity studies, which are to be performed after the construction of a chronic/carcinogenicity inhalation toxicity study facility and enactment of pertinent legislation. Methods: Through this study, qualitative and quantitative priority evaluation of test substances according to acute, subchronic and chronic categories were respectively performed and priorities were suggested by expert group review, redundancy and other methods. Meanwhile, a draft on test substance selection criteria, procedures and methods referring to the National Toxicology Program (NTP) system was proposed. Results: This study selected priorities for candidate substances for chronic inhalation toxicity studies to be conducted from 2016. Conclusions: In the future, by assessing in advance the toxicological effects of chemicals to which workers can be potentially exposed in the workplace via long-term inhalation, expected health disturbances among workers will be reduced and it is anticipated that occupational disease induced by chemicals will be effectively prevented.

Toxicity Evaluation of Perfluorinated Compounds Using Daphnia magna (Daphnia magna를 이용한 과불화화합물의 독성평가)

  • Perfluorooctane sulfonic acid (PFOS) and Perfluorooctanoic acid (PFOA) are the principal chemicals known as perfluoroalkyl acids (PFAs). Despite the widespread use of these compounds, relatively little is known about their fate and effects. The purpose of this study was to determine the toxic effects of PFOS and PFOA on Daphnia magna. In the acute toxicity test, D. magna were exposed for 48 hours at concentrations of 0, 30, 45, 67.5, 101.25 and 151.88 mg/L PFOS, and 0, 100, 160, 225, 337.5 and 506.25 mg/L PFOA, respectively. In the case of chronic toxicity test, D. magna were exposed through water for 21 days at concentrations of 0, 0.375, 0.75, 1.5, 3 and 6 mg/L PFOS, and 0, 1.25, 2.5, 5, 10 and 20 mg/L PFOA, respectively. Acute toxicity was assessed on the basis of immobility, while chronic toxicity was assessed on the basis of fecundity. The acute toxicity test on PFOS and PFOA showed that the values of $EC_{50}$ were 50.90 mg/L and 253.47 mg/L, respectively. In the chronic test, fecundity was reduced significantly at 1.5 mg/L of PFOS and 10 mg/L of PFOA, respectively. These results indicated that PFOS is more toxic to zooplankton than PFOA, and both chemicals have some hazard demonstrates risk for acute or chronic toxicity to freshwater organism.

  • The toxicity test of ammonium persulfate was conducted to ensure of its potential toxic effects according to the single-dose acute oral toxicity study (OECD Guideline 423) and 90-day repeated dose sub-chronic oral toxicity study guideline (OECD Guideline 408) for establishing national chemical management system, and matching in the Globally Harmonized Classification System (GHS) category. In acute oral toxicity study, pasty stool, perineal contamination and temporary body weight decrease were observed after dosing 1st and 2nd challenge (300 mg/kg body weight). All test animals were dead within 6 hours after dosing at 3rd challenge (2000 mg/kg body weight). Therefore, the GHS class of test substance is considered class 4. In sub-chronic toxicity study, body weight changes, food consumptions, hematological, biochemical and pathological examination did not show any noticeable and significant differences between the administered (5, 20, 80 mg/kg body weight) and control (vehicle only) group animals. Based on these results, the no observed adverse effect level (NOAEL) is considered above 80 mg/kg body weight.

The Acute Toxicity Effect of Triotganotin on the Growth of Microalgae and Shellfish and A Design of A Chemostat System for the Chronic Toxicity Experiment (미세조류와 패류의 성장에 미치는 Triorganotin의 급성 독성영향 및 만성독성 실험을 위한 Chemostat System의 설계)

  • The acute toxicity effect of triorganotin of trioganotin on the growth of microalgae and shellfish was investigated through flask culture. The value of 120 hr-LC$_{50}$ that is the median lethal concentration of TBTO on the shellfish (R. philipinarum) was found to be 6 $\mu$g/L. The acute toxicity effect of TBTO on T. suecica was obviously shown even at the concentration of 0.5 $\mu$g/L, and the effect diminished as the initial cell density increased. The effect also diminished less in the experiment done under aeration than in that done under non-aeration. To design a chemostat system for the test of chronic toxicity, the culture of T. suecica was executed in photobioreactor. In batch culture, the profiles of chlorophyII a and D.C.W. showed the growth of T. suecica very well, and the maximum specific growth rate was estimated to be 0.54 d$^{-1}$. with this value, as a dilution rate in contimuous culture, pH was nicely maintained between 7 and 9 when air was supplied with 3% CO$_{2}$. From all results and the natural environment of clam, a novel chemostat system was invented. Through this system, we can observe each independent toxicity effect of TBTO and plankton and combined toxicity effect as well.

The Toxicity Test of Water Flea by Heartbeat Measurement (물벼룩의 심장박동을 이용한 독성실험)

  • The water flea has been used as a test organism of toxicity test for surface water. Toxicity test with water flea is categorized into two parts. One is acute toxicity test with observing immobility and mortality and the other is chronic toxicity test determined by survival and reproduction of water flea. Heartbeat measurement of water flea was designed as a short-term toxicity test in this study. Direct measurement of heartbeat under microscope by aid of video camera gives and early diagnosis of mortality in short time. Therefore, the effects of measuring illumination, measuring time, and non-feeding during the test on heartbeat of water flea was evaluated to establish a new test approach. Test organisms used in this study are Daphnia magna, a well standardizes toxicity test organism, and Simocephalus mixtus, a newly refined organism $IC_{50}$ values of these test organism by heartbeat measurement were compared and discussed. It was found that toxicity test by heartbeat measurement was a reproducible, easy and simple method accomplished in a few hours.

Subacute Inhalation Toxicity of 3-Methylpentane

  • 3-Methylpentane ($C_6H_{14}$, CAS No. 96-14-0), isomer of hexane, is a colorless liquid originating naturally from petroleum or natural gas liquids. 3-Methylpentane has been used as a solvent in organic synthesis, as a lubricant, and as a raw material for producing carbon black. There is limited information available on the inhalation toxicity of 3-methylpentane, and the aim of this study was to determine its subacute inhalation toxicity. According to OECD Test Guideline 412 (subacute inhalation toxicity: 28-day study), Sprague Dawley rats were exposed to 0, 284, 1,135, and 4,540 ppm of 3-methylpentane for 6 hr/day, 5 days/week for 4 weeks via whole-body inhalation. Mortality, clinical signs, body weights, food consumption, hematology, serum chemistry, organ weights, and gross and histopathological findings were compared between control and all exposure groups. No mortality or remarkable clinical signs were observed during the study. No gross or histopathological lesions, or adverse effects on body weight, food consumption, hematology, serum chemistry, and organ weights were observed in any male or female rats in all exposure groups, although some statistically significant changes were observed in food consumption, serum chemistry, and organ weights. In conclusion, the results of this study indicate that no observable adverse effect level (NOAEL) for 3-methylpentane above 4,540 ppm/6 hr/day, 5 days/week for rats.

A Study on Subchronic Toxicity Test and Method of Increasing Output of Scolopendrid Pharmacopuncture (오공약침의 아만성독성 시험 및 생산량 증가방안에 관한 연구)

  • Purpose The purpose of this study was to investigate sub-chronic toxicity of scolopendrid pharmacopuncture in mouse and method of increasing output of scolopendrid pharmacopuncture. Methods In order to prove the clinical safety of scolopendrid pharmacopuncture during 90 days, We have observed the physical reaction(side effect) and clinical pathology test after scolopendrid pharmacopuncture treatment and investigated method of increasing Output of scolopendrid pharmacopuncture for 90%, 80%, 70% ethanol. Results In subchronic toxicity test, there was no significant sign in clinical sign, opthalmological values, body weights, hematological values and urinalysis values. And we could see that food consumptions and water consumptions increased significantly, albumin, triglycerides, GPT in blood chemical values and Liver, Testis(right) in organ weights changed significantly in some groups, compared with those in the S1 group. But these changes were observed within the scope of physiology. So there was no sign of toxication in subchronic toxicity test, and we can tell that NOAEL(No Observed Adverse Effect Level) is above 0.286mg/kg/day. And 70% ethanol solution of scolopendrid was yielded the most amount of substance. Conclusions This study demonstrates that scolopendrid pharmacopuncture is to treatment of safety for a long time and we can obtain much amount from 70% ethanol solution of scolopendrid.

Toxicity Assessment of Titanium (IV) Oxide Nanoparticles Using Daphnia magna (Water Flea)

  • Objectives: Titanium dioxide ($TiO_2$), a common nanoparticle widely used in industrial production, is one of nano-sized materials. The purpose of this study was to determine the acute and chronic toxicity of $TiO_2$ using different size and various concentrations on Daphnia magna. Methods: In the acute toxicity test, four concentrations (0, 0.5, 4, and 8 mM) for $TiO_2$ with 250 or 500 nm and five concentrations (0, 0.25, 0.5, 0.75, and 1 mM) for $TiO_2$ with 21 nm were selected to analyze the toxic effect to three groups of ten daphnia neonates over 96 hours. In addition, to better understand their toxicity, chronic toxicity was examined over 21 days using 0, 1, and 10 mM for each type of $TiO_2$. Results: Our results showed that all organisms died before the reproduction time at a concentration of 10 mM of $TiO_2$. In addition, the exposure of anatase (21 nm) particles were more toxic to D. magna, comparing with that of anatase (250 nm) and rutile (500 nm) particles. Conclusions: This study indicated that $TiO_2$ had adverse impacts on the survival, growth and reproduction of D. magna after the 21days exposure. In addition, the number of test organisms that were able to reproduce neonates gradually were reduced as the size of $TiO_2$ tested was decreased.
