식용 및 외용색소의 유전독성에 관한 연구 (1) (Mutagenicity studies of food and cosmetic dyes (1))

  • 국내에서 실제사용되고 있는 22가지의 식용 및 외용색소에 대하여 유전독성실험을실시하였다. salmonella typhimurium을 이용한 유전자 복귀돌연변이 시험(Ames test)과 Chiness hamster lung cell을 이용한 염색체이상 시험을 시행한 결과 Ames test에서는 등색 203호 (D&C Orange No.17)가 대사활성계의 존재 유무와 관계없이 돌연변이 유발성을 보였고, 적색 204호 (D&C Red No.9)는 대사활성계의 존재하에서 돌연변이 유발성을 나타내었다. 염색체이상 시험에서는 적색104-1호 (Phroxin B)의 시약급과 적색 215호 (D&C Red No. 37)가 의양성을 나타내었다.

  • Pendimethalin [N-(1-ethyl-propyl)-2, 6-dinitro-3, 4-xylidine, $C_{13}$H$_{19}$N$_3$O$_4$, M.W. = 281.3, CAS No. 40481-42-1]는 제초제의 일종으로, 본 연구에서는 박테리아 복귀 돌연변이 시험과 포유동물 세포를 이용한 염색체 이상 시험 및 마우스를 이용한 in vivo 소핵 시험을 수행하여 pendimethalin의 유전독성을 평가하였다. 박테리아 복귀 돌연변이 시험에서 pendimethalin은 Salmonella thphimurium TA98, TA1537 균주의 경우, 대사 활성계 존재와 부재시,TA100의 경우는 대사 활성계 부재시에만 313∼5,000 $\mu\textrm{g}$/p1a1e의 범위에서 농도의존적인 돌연변이율의 증가를 보여주었고, TA1535의 경우에는 대사 활성계 존재시 약간의 돌연변이가 증가되는 것을 관찰할 수 있었다. 그러나 대사 활성계 부재시 TA1535와 대사 활성계 존재시 TA100균주의 경우에는 돌연변이 유발능을 관찰할 수 없었다. 한편 포유동물 세포인 Chinese hamster lung(CHL) fibroblast를 이용한 염색체 이상 시험에서 pendimethalin은 대사 활성계 존재 및 부재시 2.32∼9.28 $\mu\textrm{g}$/ml 농도에서 clastogenicity를 보이지 않았다. 또한 203∼810 mg/kg의 pendimethalin을 구강 투여한 마우스의 골수세포를 이용한 in vivo소핵 시험의 결과에서도 통계적으로 유의한 소핵 유발능을 관찰할 수 없었다.다.

DK1002에 대한 급성독성시험 및 유전독성에 관한 연구 (Acute and Genetic Toxicity Study of DK1002, a Drug Candidate for Analgesics)

  • The acute and genetic toxicity of DK1002 was subjected in this study. DK1002 which is a morphine-like new drug candidate synthesized by Dong-Kook Pharmaceutical Co. Ltd. is now under developing as a analgesics that have better drug efficacy and least addictive property. In acute toxicity study, the 50% lethal doses ($LD_{50}$) of DK1002 were determined as>2000mg/kg (p.o.), 237.0mg/kg(i.p.), 57.5mg/kg(i.v.), and 1266.9mg/kg (s.c.). And also, to study the genotoxicity of DK1002, we performed bacterial reversion assay with Salmonella typhimurium TA98, TA100, TA1535, and TA1537, and in vitro chromosomal aberration assay with Chinese hamster lung cells in the presence and absence of S-9 metabolic activation system. In vivo micronucleus assay using mouse bone marrow cells was also performed. From these results, DK1002 was revealed nonmutagenic potential in S. typhimurium TA98, TA100, TA1535, and TA537 both in the absence and presecne of metablic activation system. No clastogenicity of DK1002 was observed in chromosomal aberration assay in vitro as well as in micronucleus assay in vivo.

Lactobacillus plantarum AF1와 Lactobacillus plantarum HD1이 생성한 조항균 물질의 유전학적 독성평가 (A Genotoxicological Safety Evaluation of Crude Antifungal Compounds Produced by Lactobacillus Plantarum AF1 and Lactobacillus Plantarum HD1)

  • This study investigates the genotoxicity of crude antifungal compounds produced by Lactobacillus plantarum AF1 (L.plantarum AF1) and Lactobacillus plantarum HD1 (L. plantarum HD1) isolated from kimchi. The genetic toxicity of crude antifungal compounds was evaluated in bacterial reverse mutation in Salmonella and Escherichia spp., chromosome aberrations in Chinese hamster lung cells, and micronucleous formations in mice. In bacterial reversion assays with Salmonella Typhimurium TA98, TA100, TA1535, TA1537, and WP2uvrA, crude antifungal compounds did not increase the number of revertant colonies in both the absence and presence of the 59 metabolic activation system. In the chromosome aberration test with Chinese hamster lung cells, crude antifungal compounds showed no increase in the frequency of chromosome aberrations in the short-period test with/without the S9 mix or in the continuos test. In the in vivo mouse micronucleus assay, crude antifungal compounds showed no increase in the frequency of polychromatic erythrocytes with micronuclei. The results show that crude antifungal compounds produced by L. plantarum AF1 and L. plantarum HD1 did not induce any genotoxicity.

  • The detection of many synthetic chemicals used in industry that may pose a genetic hazard in our environment is of great concern at present. Since these substances are not limited to the original products, and enter the environment, they have become widespread environmental pollutants, thus leading to a variety of chemicals that possibly threaten the public health. In this respect, to regulate and to evaluate the chemical hazard will be important to environment and human health. The clastogenicity of 18 synthetic chemicals was evaluated in Chinese hamster lung fibroblast cells in vitro. 4-Chloro-3,5-dimethyl phenol (CAS No. 88-04-0) induced chromosomal aberrations with significance at the concentration of 15.7 $\mu\textrm{g}$/$m\ell$ both in the presence and absence of metabolic activation system. Phenoxybenzene (CAS No. 101-84-8) which is one of the most cytotoxic chemical among 18 chemicals tested revealed no clastogenicity in the range of 0.11-0.43 $\mu\textrm{g}$/$m\ell$ both in the presence and absence of metabolic activation system. From the results of chromosomal aberration assay with 18 synthetic chemicals in Chinese hamster lung cells in vitro, 4-chloro-3,5-dimethyl phenol (CAS No. 88-04-0) revealed weak positive clastogenic results in this study.

Since these substances are not limited to the original products, and enter the environment, they have become widespread environmental pollutants, thus leading to a variety of chemicals that possibly threaten the public health. In this respect, to regulate and to evaluate the chemical hazard will be important to environment and human health. The clastogenicity of 11 synthetic chemicals was evaluated in Chinese hamster lung fibroblast cells in vitro. 1-Chloro-3-bromopropane CAS No. 109-70-6) induced chromosomal aberrations with significance at the concentration of $185.0\;{\mu}g/mL\;and\;1,600\;{\mu}g/mL$ both in the presence and absence of metabolic activation system, respectively. Triphenyl phosphite (CAS No. 101-02-0), which is one of the most cytotoxic chemical among 11 chemicals tested revealed no clastogenicity in the range of $95.0-4.9\;{\mu}g/mL$ both in the presence and absence of metabolic activation system. From the results of chromosomal aberration assay with 11 synthetic chemicals in Chinese hamster lung cells in vitro, 1-chloro-3-bromopropane revealed a positive clastogenic result in this study.

  • To identify nongenotoxic carcinogen determined as negative by ICH guideline-recommended standard genotoxicity test battery; Ames test, chromosome aberration assay, mouse lymphoma $tk^{+/-}$ assay, in vivo micronucleus assay, we picked bisphenol A as a model compound. In this study, we applied in vitro BalB/c 3T3 cell transformation assay and Syrian hamster embryo (SHE) cell transfarmation assay. Bisphenol A was treated upto $769.2 ug/m{\ell}$ in BalB/c 3T3 cells and upto $125 ug/m{\ell}$ in SHE cells. bisphenol A didn't induced morphological transformation both with one stage treatment protocol and with two stage treatment protocol. But, treated far 48 hr, Bisphenol A induced morphological transformation significantly in SHE cells.

The clastogenicity of 17 synthetic chemicals was evaluated in Chinese hamster lung fibroblast cells in vitro. Two most cytotoxic chemicals, dodecyl methacrylate (CAS No. 142-90-5) and 2-ethylhexyl methacrylate (CAS No. 688-84-6), among 17 chemicals tested revealed no clastogenicity in the range of 0.0165-0.066 $\mu\textrm{g}$/$m\ell$ and 0.006-0.024 $\mu\textrm{g}$/$m\ell$ both in the presence and absence of metabolic activation system, respectively. All 17 chemicals revealed no significant induction of chromosomal aberration both in the presence and absence of metabolic activation system in this assay. From the results of chromosomal aberration assay with 17 synthetic chemicals in Chinese hamster lung cells in vitro, we did not observed positive clastogenic results in this study.

In this respect, administrative authorities has great concern to regulate and to evaluate the chemical hazard to environment and human health. The clastogenicity of 28 synthetic chemicals was evaluated in Chinese hamster lung fibroblast cells in vitro. Glycidylacrylate which is one of the most cytotoxic chemical among 28 chemicals tested revealed clastogenicity in the range of 0.31-1.25 $\mu\textrm{g}$/$m\ell$ both in the presence and absence of metabolic activation system. Neopentyl glycol (340-1360 $\mu\textrm{g}$/$m\ell$) also revealed weak positive result both in the presence and absence of metabolic activation system. Cyanoguanidine (/$420.5-841 $\mu\textrm{g}$m\ell$) and N-butylchloride ($231.5-926 $\mu\textrm{g}$/m\ell$) revealed weak positive result only in the absence of S-9 metabolic activation system. Nevertheless total aberration percentages of N-butylchloride in the presence of metabolic activation system, and 3,4'-dichlorobenztrifluoride in the absence of S-9 metabolic activation revealed above 5% aberration, there is no statistical significance. From the results of chromosomal aberration assay with 28 synthetic chemicals in Chinese hamster lung cells, glycidylacrylate (CAS No. 106-90-0), neopentyl glycol (CAS No. 126-30-7), N-butyl chloride (CAS No. 109-69-3) and cyanoguanidine (CAS No. 461-58-5) revealed positive clastogenic results in this study.

  • This study was carried out to perform genotoxicological safety evaluation of crude antifungal compounds produced by Bacillus subtilis SN7 (B. subtilis SN7) isolated from meju. Bacterial reverse mutation assay with Salmonella typhimurium TA98, TA100, TA1535, and TA1537 or Escherichia coli WP2uvrA in the presence and absence of the S9 metabolic activation system was carried out, and the crude antifungal compounds produced by B. subtilis SN7 showed no significant increase in the number of revertant colonies. In the chromosomal aberration tests using Chinese hamster lung (CHL) cells, sample treatment groups showed no increase in the frequency of chromosome aberrations compared to the negative control group. Furthermore, in the micronucleus formation test, the crude antifungal compounds showed no significance increase in the frequency of polychromatic erythrocytes with micronuclei. These results suggest that the crude antifungal compounds produced by B. subtilis SN7 isolated from meju showed no harmful genotoxic effects.
