Comparison of TLC and GC-MS Method in the Analysis of Dye Extracted from Madder Plant (꼭두서니 추출염료에 대한 TLC와 GC-MS 분석법의 비교 연구)

  • This research was aimed to investigate and compare the effectiveness of TLC and GC-MS methods in the analysis of chromophoric substances extracted from madder plant. Alizarin and purpurin 0.3% solution were used as comparative standards; madder extraction was prepared by heating the solution of powdered madder at 80℃, pH 1.5, for 90 min. Best elution solvent for TLC in silica gel plate was toluene:ethyl acetate=9:1, which resulted in red and yellow spots from madder extraction each of which showed R/sub f/ values 0.32-0.43 and 0.07-0.11. Although the red spot in particular exhibited similar characteristics as standard purpurin in color, shape, and R/sub f/ values, the result was inconsistent throughout different TLC trials. GC-MS analysis showed only small amount of alizarin and no purpurin in the madder extraction. Other chromophoric substance such as 2-furancarboxaldehyde, 5-(hydroxymethyl)-, anthralin, and danthron were also detected in small amounts. The result indicated that TLC was less sensitive to detecting and identifying the natural dyestuff which is generally constituted with a number of similar but chemically different chromophoric substances.

Separation of Chromophoric Substance from Madder Plant under Different Extraction and Analytical Conditions (염료추출 및 분석 조건에 따른 꼭두서니의 색소성분 분리 거동)

  • This research was aimed to establish the standard extraction and analytical procedures for examining the chromophoric substance in madder root with the ultimate goal of identifying the dyes in badly faded textiles of archaeological origin. The separation temperature of gas chromatography, pH and other extraction conditions were tested. The results were as follows: The suitable separation temperature for the GC cappillary column was 50∼305$^{\circ}C$, and methanol was a good GC solvent for both standard alizarin and madder extraction. The best extraction of madder was achieved by 90 min soaking in room temperature followed by filtration and the actual heat extraction procedure. The best pH for extracting alizarin was pH 3 and above pH 5 alizarin was not detectible. Only alizarin and no purpurin was found in the extraction of the currently used madder plant.

  • A cationic demand(CD) is a very useful indication for determining the optimum dosage rate of retention aids to the papermaking stock at the wet end. Polyelectrolyte titration has been most often used to determine a CD. Highly accurate results can be obtained by this method when the ionic strength of sample is low. But this is accompanied by the serious errors when the ionic strength is higher than that corresponding to 20 milli molar(mM) monovalent, 2 mM divalent or 0.2 mM trivalent ions because of no occurrence of the end point of titration. Therefore, it is very difficult or almost impossible for the conventional method to be applied to the industrial suspensions such as papermaking stock and industrial waste water. Then a novel method using chromophoric labeled cationic polymer which can be applicable to the sample with high ionic strength has been developed.

GC-MS Analysis of Dyes Extracted from Turmeric

  • Standard extraction procedure for examining chromophoric substances of turmeric was investigated. Acetone and methanol were used as extracting solvents with different extraction procedures and pH levels. GC-MS analysis identified curcumene 2 (6.7 min), feruloylmethane 3 (8.3 min), coumaran 4 (6.09 min), vanillin 5 (6.2 min), and zingiberene 6 (10.5 min) as the major products. Curcumin 1 which has been known as the major chromophoric substance of turmeric was not detected in any samples. The maximum amount of curcumene 2, which was used as the fingerprint product for turmeric dye, was obtained by utilizing presoaking and decanting step with methanol prior to actual extraction step using a waterbath shaker (WMM). The highest relative abundance of curcumene 2 was detected in pH 6 sample followed by pH 5 indicating that the most appropriate pH level was in the range of pH 6-5.

  • This study was performed to investigate the effect of Artemisiae argyi Herba extract on the treatment of chromaticity and colorfastness. Artemisiae argyi Herba has been used as a Korean medicine. It is effective in removing skin disorders and suppressing pain. It is also good as a fungicide and in treating pruritis. In the long history of Korea, dyeing has been applied for a means representing the grace of natural and inner esthetic consciousness of man. Vegetable dyes give us such great benefits, diversified color; but no pollution. And ramie fabric has distinctive features such as beautiful brilliance, elegance, and strong durability. So, it is regarded as a special product of Korea traditionally. These studies were carried out to treat with acetate iron, dichloride copper and alum with a mordant to ramie fabric. The ramie fabric was died with Artemisiae argyi Herba extract. The results of experiment showed as follows: First, the chromophoric degree was the highest in acetate iron but not distinction in another mordants. Seocond, the light colorfastness was the highest in non treated and dichloride copper, but alum was the lowest. Third, the discoloration was alum and dichloride copper showed first grade in washing colorfastness. Abrasion colorfastness was not significant in this test. According the previous results, Artemisiae argyi Herba has an efficiency in removing skin disorders and suppressing pain. So it is considered that Artemisiae argyi Herba can be applied effectively to theraphy of fungicide and in treating pruritis.

Chromophoric Structures of Alkali Lignin (알카리리그닌의 착색구조(着色構造)에 관(關)한 연구(硏究))

  • To investigate the formation of the chromophoric structures taking place during the alkaline pulping vanillyl alcohol [${\alpha}-^{13}C$] guaiacylglycerol-${\beta}$-aryl ether [${\alpha}-^{13}C$ or ${\gamma}-^{13}C$] and phenylcoumarn [${\alpha}-^{13}C$] units as model lignins were treated with 1N sodium hydroxide at 165$^{\circ}C$ for 1.5-3 hours. From the chemical structures of the isolated products and $^{13}C$-NMR Spectra of the reaction mixtures, the main conclusion is as follows; 1) Condensation products of II-1-5 were identified from the reaction mixture of vanillyl alcohol treated with alkali and theses compounds afforded the quinonmethide structure(Fig. 3-7) by air oxidation. 2) Treatment of guaiacylglycerol-${\beta}$-aryl ether unit gave ${\varphi}$-aryl-${\beta}$-aroxy quinone structures (IV-15, IV-16), diguaiacyl-1, 4-penta-diene ${\beta}$, ${\beta}$'-diaroxyl distyrene methane unit, ${\beta}$-aroxy distyrene methane. These distyrene methanes of the compounds are transformed by air oxidation into the corresponding o-quinonemethide units (V-8, V-9). 3) On the treatment of phenylcoumaran, the stilbene derivative was formed in quantitative yield and dimerized(VI-11) in preference to oxidation to the corresponding extended quinone structures. The chromophoric structures taken place during the alkaline treatment of the model lignins are thought to be some important types in alkaline pulping on the basis of the reaction mechanism in this experiment.
