  • It order to extract the soluble organic substances of Korean ginseng effectively, the ginseng extract have been made by using amylase. The investigation on the optimum condition of enzyme reaction was carried out, and the amounts of gained extract and its saponin pattern were compared among the ethanol extract, water extract and enzyme extract. The results obtained are summerized as follows. 1. The gaining ratio or ginseng extract was the highest value when the raw ginseng and dried ginseng were extracted in the concentration of 7.5% and 5% with 0.3%∼0.6% enzyme for 25 hour. 2. The amounts of ethanol extract, water extract and enzyme extract were 9.14%, 17.23% and 23.73% in case of raw ginseng and 64.09%, 72.52% and 74.36% in case of dried ginseng, respectively. The amount of enzyme extract was increased as much as 6∼14% in case of raw ginseng, and 2∼10% in case of dried ginseng compared with that of ethanol and water extract. 3. The absolute content of saponin was nearly constant in spite of the different extraction method and all of the ginseng saponin pattern of thin-layer chromatograms were almost same.

  • Optimal operation in chromatography is needed to save operation time and the solvent used in multiple chromatographic runs. To this end, many simulation studies of chromatography process have been performed. The relationship between the distribution coefficient and the ionic strength is important in gradient elution ion chromatography. Experimental runs and computer simulations were carried out under linear gradient elution condition in order to compare the experiments and the simulation. Experiments were performed with formic acid under isocratic conditions to determine the simulation equation parameters. Computer simulation was based on three equations which related distribution with ionic strength as follows; K=${\alpha}$I(sup)-${\beta}$, K=A+BI+Cl$^2$and K=y(sub)0+A$_1$$.$e(sup)(-I/m$_1$). The effects of gradient slope on the chromatograms are discussed, and good agreement between the experimental and the simulated results is shown.

  • The aroma components of Korean green tea in the south western part of Mt. Chiri prepared by the traditional method from native variety were analyzed using GC and GC-MS. The patterns on GC chromatograms of the three samples from the flushes plucked in early spring were similar, though they are prepared by different producers in the area of Hadong-kun, Kyung sang nam-do. A total of 51 aroma compounds were identified in all samples. Main component in the aroma compounds of these teas were geraniol, benzyl alcohol, 2-phenylethanol, $\beta$-ionone, benzyl cyanide and linalool oxides. The aroma components of green teas manufactured by the different plucking periods were also compared. The amounts of geraniol, typical rose floral aroma were particularly decreased in the final plucking period. The amounts of pyrazines and furfuryl alcohol, typical roasted aroma and nutty aroma were slightly increased in later plucking period.

  • In Cnidium officinale $M_{AKINO}$ and Platycodon grandiflorum A. $D_E\;C_{ANDOLE}$, chlorogenic acid was identified by Rf values, color reactions on paper chromatograms and UV-absorption spectra of the eluate of phenolic spots. And isochlorogeni acid-like substance was also found in the former. $1-Phenylalanine-U-C^{14}$ and sodium $acetate-2-C^{14}$ werse fed to both plants and their incorporation ratio to chlorogenic acid and isochlorogenic acid-like substance was compared. Phenylalanine was better precursor for chlorogenic acid in both plants than acetate. But acetate showed higher incorporation ratio to isochlorogenic acid-like substance in Cnidium officinale than that of phenylalanine.

  • Evaluation method of crude drug preparations was studied in Saengmaek-san. Zig-zag TLC scanning profiles and high performance liquid chromatograms were obtained from Saengmaek-san and its each crude drug. A method using TLC densitometry and high performance liquid chromatography was established for the precise determination of ginsenoside $Rb_1$ in Saengmaek-san containing Ginseng Radix. Consequently, ginsenosicle $Rb_1$ content was 0.45-0.48 mg per g of Saengmaek-san. This method was found to be useful for the quality evaluation of oriental medicinal preparations.

  • At the different growing stages of Fagopyrum esculentum Moench planted at the green house and out doors, the contents of free amino acids in the whole plants, were determined quantitatively by spot-extract colorimetric method (J. Awapara method) using the chromatograms obtained by the ion-exchange resin and paper chromatographic method. And the contents of carbohydrate in the whole plant were determined by Bertrand method. The results were summarized as follows. 1. Almost same kinds of free amino acids were detected in the both plants. 2. Concentrations of most amino acids was increased progressively during growth; further growth was accompanied by decreased concentrations. 3. Rate of growth and increasing of total amino acid content of green house plant is greater than that of out dorrs plant. 4. At the same stages of gwoth, total free amino acid content of green house plant is higher than that of out doors plant but sugar content is lower. 5. There was rapidly increasing of sugar content and decreasing of total free amino acid content except asparagine during mature of seeds.

  • Pantothenic acid and biotin contents in Panax Ginseng roots were determined microbiologically with L. arabinosus 17-5. Detection of the vitamins was achieved by the thin-layer chromatography and PPC. Pantothenic acid and biotin were found at the Rf values of 0.42 and 0.55 respectively on the thin-layer chromatograms. In order to find out whether or not the L. arabinosus growth promoting factors contained in the respective ginseng extracts, as shown by the microbiological assay, were really the vitamins respectively, PPC was carried out. Microbiological assay of the vitamins met with results that the average pantothenic acid content in the roots was 6.6r/g, calculated as Ca-pantothenate, and the average biotin content 9.24 mr/g.

  • Protein ion exchange chromatography was studied experimentally in order to prove the theoretical prediction from the linear model of Yamamoto, S. et al (1). Adsorption isotherms were measured as a function of ionic strength in a batch experiment. The relationship between the characteristics of chromatogram and the operating conditions of ionic strength, flow rate, length of column, concentration and amount of protein sample were studied. At the higher ionic strength, the lower flow rate and the longer column conditions, the higher number of plate was obtained. Satisfactory agreement was observed between the experimental and the calculated chromatograms except for the case of high protein concentration.

  • Strong anti-inflammatory saponins Phytolacca americana (Phytolaccaceae) wereobtained from callus mass derived from the stems and also from that derived from the roots of cultivated Phytolacca americana (which were designated as PAS and PAR, respectively). The callus were grown on Linsmair and Skoog's agar medium supplemented with 1ppm OF 2, 4-dichlorophenoxyacetic acid. Phytol accoside B and D were obtained from PAS and phytolaccoside A and B from PAR. The thin layer chromatograms of the crude saponins from PAS and PAR were similar to those of original plants. PAS contained phytolaccoside B as a major component while phytolaccoside E was a major saponin in original plants.
