Effects of Dietary Xylooligosaccharide on Lipid Levels of Serum in Rats Fed High Cholesterol Diet (고콜레스테롤 식이를 급여한 흰쥐에서 Xylooligo당의 혈중 지질개선 효과)

  • The purpose of this study was to investigate the effects of dietary xylooligosaccharide on lipid levels of serum in rats fed high cholesterol diet. Male Sprage-Dawley rats weiging 100$\pm$10g were randomly assigned to groups of two normal(N, N+10X) and four high cholesterol diets which contained 1%(w/w) cholesterol. High cholesterol diet groups were classified to xylooligosaccharide free diet (C group), 5% xylooligosaccharide diet(C+5X group), 10% xylooligosaccharide diet(C+10X group) and 15% xylooligosaccharide diet(C+15X group) according to the level of dietary xylooligosaccharide supplementation. These experimental diets were fed ad libidum for 4 weeks. The weight gain of high cholesterol diet group were significantly increased more than that of normal group, but those of 10% and 15% dietary xylooligosaccharide groups were significantly decreased more than that of high cholesterol diet(C)group. The higher content of xylooligosaccharide, the more food intake was increased. The food efficiencies of 10%, 15% cholesterol diet groups were lower than that of high cholesterol diet(C)group. The levels of serum triglyceride(TG) and total cholestoral were significantly high in cholesterol diet groups compared with normal diet group but were decreased in groups fed 5% and 10% dietary xylooligosaccharide. Especially, the lowest level showed in group fed high cholesterol diet containing 10% xylooligosacchride. High cholesterol diet group containing 10% xylooligosaccharide increased HDL-cholesterol level and then decreased LDL-cholesterol level and atherogenic index compared with other groups. The level of TBARS(thiobarbituric acid reactive substances) in serum was decreased in rat group fed xylooligosaccharide in high cholesterol diet. The higher content of xylooligosacchride, the more gastrointestinal transit time was decreased. The results indicate that dietary xylooligosaccharide can improve status of TG and total cholesterol and repress lipid peroxidation in serum lipid at hypercholesterolemia induced by high cholesterol diet.

  • Casein or soybean protein was subjected to there action with caffeic acidtyrosinase system at 30-35$\circ$C, pH 6.8 with aeration for 5hr. The resulting brown proteins were washed with acetone until the washings were on longer colored. However, modified protein still retained a light brown. The effects of the modified proteins and brown compounds on male Wistar strain rats were studied by pair-feeding of a cholesterol-free diet for 14days. Significant decrease in protein digestibility for the rats fed with the modified proteins were observed. Weight gain and protein digestibility were not influenced by feeding brown compounds, but the feeding of brown compound from casein caused an enlargement of caecum. The concentrations of serum cholesterol and triglyceride in the rats fed with modified proteins and brown compounds were mostly unchanged against the rats fed with untreated proteins. These results suggest that the decrease in protein digestibility induced by enzymic browning-reaction did not cause the decrease in concentration of serum cholesterol.

Hyperlipidemic Inhibitory Effects of Phellinus pini in Rats Fed with a High Fat and Cholesterol Diet

  • This study evaluated the in vitro and in vivo hypolipidemic effects of the medicinal mushroom Phellinus pini. The methanol extract (ME) of the fruiting body of Ph. pini was active against pancreatic lipase and cholesterol esterase with 99.14% and 67.23% inhibited activity at 1.0 mg/mL, respectively. It also inhibited 81.81% and 55.33% of ${\alpha}$-glucosidase and ${\alpha}$-amylase activities, respectively, at 2.0 mg/mL. Hyperlipidemia as induced by feeding rats with a high fat and cholesterol diet (HFC). HFC supplemented with a 5% fruiting body powder of Ph. pini (HFC + PhP) significantly reduced plasma total cholesterol, low-density lipoprotein cholesterol, and triglycerides in rats compared with HFC. The reduced levels were comparable to rats fed the normal control diet (NC). The atherogenic index of HFC + PhP rats was significantly lower than that of the HFC rats. The excretion of fecal total lipid and cholesterol in the HFC + PhP rats was significantly higher than those in the NC and HFC rats. Histopathological examinations demonstrated scant deposition of lipids in the liver of rats fed HFC + PhP. The dietary supplementation with the fruiting body powder provided natural plasma lipid and glucose lowering effects in experimental rats without adverse effects on the plasma biochemical parameters and liver function related enzyme activities. Therefore, the hypolipidemic effects of Ph. pini may be due to the inhibitory effects on pancreatic lipase, cholesterol esterase, ${\alpha}$-glucosidase, and ${\alpha}$-amylase, and excretion of excess lipids and cholesterol in the feces.

Effects of Yellow and Black Soybeans on Plasma and Hepatic Lipid Composition and Fecal Lipid Excretion in Rats (노란콩과 검정콩 식이가 흰쥐의 체내 지질 함량과 분변으로의 지질 배설에 미치는 영향)

  • This study was undertaken to determine the effect of yellow and black soybeans on plasma and hepatic lipid composition and fecal lipid excretion in Sprague Dawley rats. Animals were fed with diets containing 52% yellow soybean, 50% black soybean, or 20% casein for 7 weeks. Feeding efficiency was significantly increased in the animals fed soybeans(p<0.05). Plasma cholesterol concentration was significantly lowered in the group fed yellow soybean compared with black soybean and casein-fed control (p<0.05). Hepatic triglyceride concentration was significantly lowered in soybean groups compared with casein-fed control(p<0.05), whereas hepatic cholesterol concentration was not affected by diet treatments. Soybean feeding significantly increased fecal weight, triglyceride and bile acid contents compared with casein feeding(p<0.05). It is concluded that soybean feeding in rats affects plasma and hepatic lipid levels by increasing the excretion of triglyceride and bile acids.

Effects of Yam on Lowering Cholesterol Level and Its Mechanism (마(Dioscorea)의 콜레스테롤 저하작용 및 그 작용기전)

  • The effects of yam(Dioscorea) on lowering cholesterol level and its mechanism were investigated. The concentrations of plasma and liver lipids, and the excretions of fecal neutral sterol and bile acid were measured in male Sprague-Dawley rats. Five Groups of 8 rats were fed hypercholesterolemic diet(1% cholesterol, 10% lard ; control), hypercholesterolemic diet plus 15% or 30% dried yam powder prepared by either hot-air(15HY, 30HY) or freeze dry(15FY, 30FY) for 4wk. Plasma total lipid, total cholesterol and cholesterol level was also significantly lower(28%, p<0.05), buy HMG-CoA reductase activity was higher in 30FY(230%, p<0.05) than in control. Although no significant differences in fecal neutral sterols were observed among groups, the yam-fed rats apparently had less bacterial degradation of cholesterol as indicated by a significantly greater of fecal cholesterol to coprostanol than in controls. Total fecal bile acids were significantly greater in rats fed yam(15HY : 5 folds, 15FY ; 12,30HY ; 12, 20FY ; 22) than in controls. The ratio of secondary to primary bile acids was almost 8 times lower in 30FY than in control. These data indicate that yam lowers cholesterol both in plasma and in liver through increasing fecal bile acid excretion as well as HMG-CoA reductase activity. Freeze-dried yam, which possesses viscosity, was more effective in cholesterol-lowering action than hot-air dried one.

  • The effectsof garlic vinegar supplementation on body weight, blood blucose, and plasma triglyceride and cholesterol were investigated in streptozotocin -induced diabetic rats fed high-cholesterol(1%) diet for 4 wk. The garlic vinegar was made by fermenting 20% frsh garlic juice. There was no effect of garlic vinegar on body weight, plasma glucose or triglyceride concentrations in the diabetic rats. Plasma total -cholesterol concentrations were unaffected by garlic vinegarr supplementation. However, plasma LDL-cholesterol concentrations and atherogenic index were significantly lower in the diabetic rats supplemented with garlic vinegar diet than tin the cotnrol rats. The diabetic-rats supplemented with garlic vinegar not only had increased HDL-cholesterol levels but had decreased LDL-cholesterol. This alteration in the HDL/LDL-cholesterol ratio in the diabetic rats may decrease the risk of atheroscelrosis. Therefore, the over-all effect of garlic vinegar supplement may contribute to the antiatherogenic role in streptozotocin-induced diabetic rats.

  • The present study was designed to investigate the serum cholesterol lowering effect oj the phytosterol derivative (LPSS) on high cholesterol (HC) diet-induced hypercholesterolemia in male weaning Sprague-Dawley (SD) rats. Rats were fed with HC diet containing 1 % cholesterol and 0.5% cholic acid for 1 week. After 1 week, the LPSS oil suspension (0.32 g/kg B. W.) was orally administered to the rats fed with either basal diet or HC diet groups for 7 days. In addition, the LPSS powder (0.14%) mixed with basal diet or HC diet was Jed to the rats for 7 days. Serum total cholesterol and LDL-cholesterol contents were not altered by administration of the LPSS oil suspension with basal diet. However, they were significantly decreased by administration of the LPSS oil suspension with HC diet at day 14. Also, they were significantly decreased by the LPSS powder mixed with basal diet or HC diet at day 9, 11, 14. HDL-cholesterol contents were not altered by the LPSS oil suspension or LPSS powder. These results indicated that the phytosterol derivative(LPSS) might decrease serum total cholesterol and LDL-cholesterol contents in rats.

Effects of Buckwheat on Super Oxide Dismutase Activity and Histopathological Change of Rats Fed with Hypercholesterolemic Diet (메밀이 흰주 SOD 활성도와 장기에 미치는 영향)

  • This study was carried out to investigate the effects of rutin from buckwheat vegetable on rats fed with hypercholesterolemic diets. Sprague-Dawley male rats were fed the diets providing various level of buckwheat vegetable for five weeks. Body weight increased during the experimental period. Body weight gain decreased by addition of cholesterol group (C-C). Rutin feeding group (T-l) and buckwheat vegetable feeding group (T-2, T-3, and T-4) showed the reduction of weight gain. Total cholesterol level decreased whereas HDL-cholesterol level increased with the increasing amount of buckwheat vegetable added. The value of thiobarbituric acid in serum decreased with increasing buckwheat vegetable added. The histopathological changes of the kidney were different among the experimental groups. Kidney of the rats fed with cholesterol added diet showed severe calcification.

Effects of Indigestible Dextrin on Lipid Metabolism in Rats Fed Normal or High Fat Diet (식이지방 수준이 다른 식이를 섭취한 흰쥐의 체내 지질대사에 대한 난소화성 덱스트린의 섭취 효과)

  • The effects of indigestible dextrin on lipids in serum, tissue, and feces were investigated in two experiments carried out with 64 male Sprague-Dawley rats for eight weeks. Experiment I diet(normal fat diet) containing 5% corn oil and experiment II diet(high fat diet) containing 18% fat(13% beef tallow and 5% corn oil) and 1% cholesterol were divided into four groups with 0.5% cellulose(0.5CL : control), 10% cellulose(10CL), 10% Indigestible dextrin I(ID-I) and 10% indigestible dextrin II(ID-II), respectively. ID-I and ID-II diets produced a decrease in serum total lipid and cholesterol concentrations in both normal and high fat diets but more significantly in the groups fed high fat diet. ID-I, ID-II, and 10CL of high fat diet decreased serum glucose concentration. ID-I and ID-II diets also decreased liver cholesterol and triglyceride concentrations in rats fed normal and high fit diets. The fecal excretion of cholesterol in ID-II/high fat diet was significantly higher than that of 0.5CL diet. Bile acid excretion was increased in the rats fed 10CL and ID-II diets of normal and high fat diets. These observations indicat the hypolipidemic and hypocholesterolemic effects of indigestible dextrin on the rats. These effects are mediated by the increase of facal cholesterol and bile acid excretion. (Korean J Nutrition31(6) : 981-990, 1998)

Effects of the Dietary Boiled Eggs on the Antithrombotic Activity and Cholesterol Metabolism in Rats (항혈전작용 및 콜레스테롤 대사에 관한 흰쥐 식이내 삶은 계란 급여 효과)

  • The influence of the ditary containing boiled eggs on the plasma cholesterol level and antithrombotic activity in rats was studied. Rats were fed basal diet(0% boiled eggs) as a control group or diets containing 25% and 50% boiled eggs or a mixed diet with 95% boiled eggs plus 5% $\alpha$-cellulose powder as a experimental groups for 30 days. The bleeding time and whole blood clotting time were significantly(P<0.05) increased by feeding diet containing 25% boiled eggs compared to groups of basal diet, 50% or 95% boiled eggs diets. The plasma clotting time was high in group of 25% boiled eggs diet. However, there were no difference in plasma clotting time among rats fed the dietary boiled eggs. The levels of plasma total cholesterol(TC) and low density plus very low density lipoprotein cholesterol(LDL, VLDL-C) were significantly(P<0.05) highest in group 95% boiled eggs diet compared to others. There were no differences in high density lipoprotein cholesterol(HDL-C) among rats fed the dietary boiled eggs. The levels of plasma TC, HDL-C, LDL$.$VLDL-C and the ratios of HDL-C/TC were not significant among the basal diet, 25% and 50% boiled eggs diets. These results suggest that the intakes of the dietary boiled eggs have the antithrombotic activity and plasma cholesterol lowering effect.
