Effects of Onion Kimchi Extract Supplementation on Blood Glucose and Serum Lipid Contents in Streptozotocin-induced Diabetic Rats (양파김치 추출물 투여가 Streptozotocin 유발 당뇨병 흰쥐의 혈당강하 및 혈중지질 함량에 미치는 영향)

  • The purpose of this study was carried out to examine the effects of onion kimchi extract supplementation on blood glucose level and serum lipid components in streptozotocin (STZ)-induced diabetic rats for 4 weeks. STZ was administered as a single dose (50 mg/kg BW) to induce diabetes, and the diabetic rats were divided into eight groups (normal, diabetic control, and six treatment groups). The dose of onion kimchi extract 100 (OK-100), 200 (OK-200), and 400 (OK-400) mg/kg/day or quercetin as a main compound of onion 5 (Q-5), 10 (Q-10), and 20 (Q-20) mg/kg/day were orally administered daily to STZ-induced diabetic rats for 4 weeks after STZ injection. The diabetic control rats (465.6 mg/dL) showed significantly higher blood glucose level than the normal rats (76.3 mg/dL) after 4 weeks, but was significantly reduced with onion kimchi extract and quercetin supplementation (p<0.001). Changes in body weight, kidney weight and urine volume were not significantly different in diabetic control rats, and in onion kimchi extract and quercetin treated rats. The serum total cholesterol levels of control were significantly decreased in onion kimchi extract and quercetin supplementation groups, respectively (p<0.001). The blood urea nitrogen level and urinary protein excretion in diabetic rats were not significant different among the groups. These results suggest that onion kimchi extract supplementation in STZ-induced diabetic rats may be a very important factor for the reduction of blood glucose and serum cholesterol profiles.

  • This study were investigated the effects of physiologically active compounds isolated from Platycodon grandiflorum on streptozotocin-induced diabetic rats. The experimental groups were divided into six groups, normal, diabetic control (CM), and four experimental groups (inulin (IN), inulin + saponin (IS), oligosaccharide (OS), and total extract group (WE)). The rats have free access to water and diet. Experimental groups were administered orally with inulin (630 mg/mL), inulin (630 mg/mL) + saponin (25.2 mg/mL), oligosaccharide (367.5 mg/mL), and total extract (225 mg/mL) into the diabetic rats for 5 weeks after STZ injection; The levels of fasting blood glucose in diabetic control increased by 10.0% for 5 weeks, but the level of blood glucose in inulin and total extract groups decreased by 21.3% and 21.2%, respectively. The concentrations of total cholesterol and triglyceride in serum of diabetic rats fed physiologically activity compounds were lower than those in diabetic control rats. HDL-cholesterol was similar among all the groups. Weights of testicle and heart were lighter, while weight of kidney was significantly increased in diabetic groups than normal group. The concentrations of serum protein insulin and albumin in diabetic groups were significantly decreased compared with those of the normal group. Urinary glucose excretion was decreased in inulin group than other diabetic groups.

The Effects of Levan on Blood Lipids and the Absorption of Calcium in Rats Fed a Low Calcium Diet (레반의 혈장지질 감소와 칼슘흡수율 증가 효과)

  • Levan, the fructose polymer is an indigestible carbohydrate regularly consumed by humans. Its physiological functions, in terms of hypocholesterolemic effect and calcium metabolism, have not been well documented. The objective of this study was to investigate the effects of levan on blood lipids and the calcium absorption ratio in rats fed a 0.1% low calcium diet. Thirty rats were divided into three groups and fed a 0.1% low calcium diet (control) or low calcium diets containing either 2.5% levan or 5% levan for eight weeks. The blood lipid and biomarkers relevant to Ca metabolism (urinary Ca and hydroxyproline), the femoral weight and the Ca contents were determined. The body weight gains were lower in the 5% levan group than the control group. Plasma concentrations of triacylglycerol and LDL-cholesterol decreased in the 5% levan group, compared to the control group, but the atherogenic indice were not affected. Blood alkaline phosphatase activity, Ca and urinary hydroxyproline excretion levels were not different in the three groups. The net calcium absorption in rats fed a 5% levan diet was greater than rats fed the control diet, while the femoral weights and Ca contents were not significantly different in the three groups. We concluded that a 5% levan diet could both enhance the calcium absorption and improve the lipid profiles in rats fed a low calcium diet.

Effects of Using Monensin and Different Levels of Crude Protein on Milk Production, Blood Metabolites and Digestion of Dairy Cows

  • Twenty-four Holstein dairy cows were used to evaluate the single and combined effects of different levels of crude protein (CP) and monensin treatment during early lactation on blood metabolites, milk yield and digestion of dairy cows. The experiment was designed as a completely randomized block with a $3{\times}2$ factorial arrangement of treatments. The factors were three concentrations of CP supplement (19.5, 21.4, and 23.4% of dry matter) and two levels of monensin (0 and 350 mg per cow per day). The experiment consisted of three phases and each phase was 3 wk in length. Monensin did not affect milk yield, lactose, solids-non-fat (SNF), blood glucose, triglyceride and DMI, but increased blood cholesterol, blood urea nitrogen (BUN), insulin and reduced blood ${\beta}$-hydroxybutyrate (BHBA), milk fat and protein percentage. Monensin premix significantly decreased rumen ammonia, but rumen pH and microbial protein synthesis were not affected by monensin treatment. Increasing dietary CP improved milk and protein production, but did not alter the other components of milk. Digestibility of NDF, ADF, CP were improved by increasing dietary CP. Increasing dietary CP from 19.5 to 21.4% had no significant effect on ruminal ammonia, but increasing CP to 23.4% significantly increased ruminal ammonia. There was a linear relationship between level of crude protein in the diet and volume of urine excretion. Microbial protein synthesis was affected by increasing CP level; in this way maximum protein synthesis was achieved at 23.4% CP.

A Study on Nutrition Education for Rural elderly of Kyungki province in Korea (경기일부 농촌노인의 식생활 향상을 위한 영양교육 효과)

  • The purpose of this study was to assess the nutrition education program for older adults at rural area. Eighty three adults (mean age ;55.6$\pm$11.9) participated in nutrition education program. Data collection includes serum and urine collections for health state. The impact of nutrition KAP (knowledge attitude and practice) and retention was examined among participants who were assessed at program entry 3 times of education and 1 month of follow-up. The results were as follows : The participants were composed 52 persons of under 60-year-old and 31 persons over 60 years. Of the 83 participants, 60 were females and 23 were males. Serum albumin and calcium content of females (3.5$\pm$1.2g/dl, 7.5$\pm$3.9mg/dl) were significantly lower than males(4.7$\pm$0.9g/dl, 11.2$\pm$4.6mg/dl). Serum TG and cholesterol contents of females (143$\pm$117mg/d1, 205$\pm$31mg/d1) were higher than males (101$\pm$86mg/dl, 192$\pm$19mg/dl) but not significantly different. The calcium (13.4$\pm$9.0mg/dl) and urea (743$\pm$440mg/dl) contents of urine excretion were also significantly lower in female than in males (19.7$\pm$11mg/dl, 1041 $\pm$535mg/dl). Via the question of nutrition attitude, the participants improved significantly between pre and post education (the score of pre, post and after 1 month, 6.7$\pm$1.8, 7.2$\pm$1.5, 7.2$\pm$1.8), and were also attributed to better personal feeling health by modified CMI test but food habit was not significantly improved. As the differences of age and gender groups were compared, under 60-year-old group had better nutrition practice score than over 60 years group.

The Effects of Korean Wild Vegetables on Blood Glucose Levels and Liver-muscle Metabolism of Streptozotocin-induced Diabetic Rate (한국산 야생식용식물이 당뇨유발 흰쥐의 혈당 및 간과 근육내 에너지원 조성에 미치는 영향)

  • Attempts were made to determine the effects of 5-Korean wild vegetabel consumptions on blood glucose leveles and orgen-energy metabolisms in streptozotocin-induced diabetic rats. The 5-Korean wild vegetables were : Cassia tora L.(C.t), Lycium chinese Mill(L.c), Trichosanthes kirilowii Max(T.k), Polygonatum odoratum var. Pluriflorum Ohwi(P.o) and Arctium lappa L(A.l). Sixty male Spargue-Dawley rats(160-220g) were induced diabetes mellitus by streptozotocin injection into the tail vein and were devided into 6 groups : a diabetic control and 5-experimental groups. All groups of the rats were fed on a AIN-76 diet, and the 5-experimental groups were fed with each wild vegetable (10%) for four weeks. An increased tendency in body weight of all the groups was observed and the tendency was more significant in L.c, T.k. and P.o. groups. The organ weight of liver and kidney were higher in L.c. and A.l. groups and lower in T.k. group which has shown the improvement from diabetes. Plasma glucose levels were markedly decreased from the 1st week in C.t, T.k. and P.o. groups and the tendency has lasted throughout the four weeks experimental period. The consumption of P.o. has decreased plasma cholesterol level while any significant difference was not seen in plasma protein levels from all the experimental groups. The level of plasma triglyceride was decreased in P.o. group and the levels of plasma free fatty acids were also significantly lower in P.o. and T.k. groups. The liver protein levels were significantly higher in P.o. and T.k. groups and these two groups also showed the negative or relatively small amount of urinary glucose excretion. The experimental group of T.k. has revealed the decreased level of muscles protein and the increased level of muscle glycogen. The 5-Korean wild vegetables contained dietary fiber and 9-analyzed minerals comparable to the ordinary use vegetables.

Lactic Acid Fermentation of Chinese Yam (Dioscorea batatas Decne) Flour and Its Pharmacological Effect on Gastrointestinal Function in Rat Model

  • To develop a health-aid preparation of Chinese yam (Dioscorea batatas Decne), lactic acid fermentation was attempted using a mixed starter comprising of Lactobacillus acidophilus, Streptococcus thermophilus, and Bifidobacterium bifidus. The anaerobic fermentation of a 5% Chinese yam flour suspension gave a uniform suspension of pH 4.35, containing $7.76{\times}10^6 CFU/mL$ lactic acid bacteria (LAB), and which was found to be acceptable to the panel from a sensory assessment. During the administration of the lactic acid fermented (LAF) Chinese yam to Sprague Dawley rats for 6 weeks, a smaller body weight gain, but greater excretion of feces were observed, implying the creation of a healthy gastrointestine on the administration of LAF Chinese yam, which was also confirmed by the gastrointestinal motility of the feed in rats fed on LAF Chinese yam. The constipation induced by loperamide was further suppressed in a rat group fed on a LAF Chinese yam diet, which was qualified from healthy gastrointestinal flora established by LAB. A serochemical analysis revealed a slight improvement in the blood glucose, neutral lipid and total cholesterol concentrations on administration of LAF Chinese yam, suggesting LAF Chinese yam could be served as a healthy-aid preparation, even for hyperglycemia or hyperlipidemia patients.

A Study of the Effect on Obesity and dyslipidemia in Kidney-hypofunction Animal Model Induced by Unilateral Ureteral Obstruction (신장절제로 유발한 신약(腎弱) 동물 모델에서의 비만 및 지질대사에 대한 영향 평가)

  • Objectives: The objective of this study is to develop a new animal model with Kidney-hypofunction for Sasang Constitutional Medicine, especially for partial Soyangin(one of four constitution which has good digestive function and poor renal function) by Unilateral Ureteral Obstruction, and to estimate the factor related to obesity, dyslipidemia, and metabolic syndrome. Methods: The C57BL/6J mice were divided into 3 groups : normal group, high fat diet(HFD) control group, and HFD group with Unilateral Ureteral Obstruction(UUO). Then, the HFD control group and the experimental group were fed with high fat diet for 6 weeks. Food intake and body weight were measured at regular time by week. After the final experiment, blood was gathered for bloodchemical examination and organs(liver, fatty tissue) were remoed, weighted, and mRNA was analyzed with real-time PCR. Results: The weight growth rate with High fat diet went down by 8.35% in experimental group and had similar FER with the normal group, while HFD control group had higher weight growth rate and FER than any other groups. Also The experimental group had lower triglyceride and LDL cholesterol rate and higher glucose rate in serum. and in mRNA expression, GLUT-9, the protein related to excretion of uric acid and metabolic syndrome, expressed lower rate than that of HFD control group. and IL-6, a kind of cytokine related to obesity and metabolic syndrome, expressed more than HFD control group. Conclusions: It was found that Kidney-hypofunction animal-experimental model is susceptible to metabolic syndrome.

Effect of Feeding Chitosan, Hibiscus Extract and L-Carnitine Mixture on Body Weight and Lipid Metabolism in Rats (키토산, 히비스커스 추출물 및 L-카르니틴 함유 식이가 흰쥐의 체중과 지질대사에 미치는 영향)

  • This study was peformed to investigate effect of feeding experimental mixture containing chitosan, hibiscus extract and L-carnitine on body weight and lipid metabolism in rats. Forty-eight male rats(Charles River CD) of eight weeks old and weighing $336.5{\pm}2.3g$ were raised for five weeks with high fat diet(40% fat as calorie) to induce obesity. After induction of obesity, rats weighing $560.4{\pm}5.6g$ were blocked into four groups according to body weight and raised for eight weeks with diet containing either 0.09%(+1D group), 0.9%(+10D group) or 4.5%(+50D group) of experimental mixture. Aspartate aminotransferase(AST) and alanine aminotransferase(ALT), total protein and albumin were normal levels in plasma. Body weight gain and epididymal fat pad weight were lower in experimental mixture groups than control group However, weights of perirenal fat pad and brown adipose tissue were not significantly different among all groups. There was no significant difference in plasma and hepatic lipid levels among all groups. Liver citrate lyase and camitine acyltransferase activities were not significantly different among all groups, however, citrate lyase activity was tended to be decreased with increasing experimental mixture level in diet. Fecal total lipid and total cholesterol excretions were highest in +50D group, and triglyceride excretion was highest in +1D group. in conclusion, intake of experimental mixture containing chitosan, hibiscus extract and L-camitine was effective in reducing body weight and body fat, and its inhibitory effects might lead to obesity improvement.

The Effects of Kimchi Intake on Lipid Contents of Body and Mitogen Response of Spleen Lymphocytes in Rats (김치의 급여가 흰쥐의 체내 지질함량과 비장 면역세포 증식능력에 미치는 영향)

  • Effects of kimchi on lipid metabolism and immune function were studied in experiments using 63mals SD rats fed 6 inds of Baechu-kimchi containing diet during 4 weeks. Three kinds of freeze dried kimchi differ in fermentation period (not fermented, 3-, 6-week-fermented at 4$^{\circ}C$) were added at 5%, 10% of the diet containing 15% lard. The levels of serum total lipid and triglyceride and the content of liver total lipid and triglyceride of all kimchi groups were lower than those of a control group. But the levels of serum and liver cholesterol is not affected by kimchi intake. The triglyceride concentration of epididymal fat pad and feces of kimchi groups were higher than those of a control group. The food efficiency ratio, epididymal fat pad weight of 3-, 6-week-fermented kimchi 10% groups were significantly lower than control and not-fermented kimchi groups. Especially 6-week-fermented kimchi groups showed adipocytes, less in number and larger in size than those of other groups. The blastogenesis of spleen lymphocytes to LPS was higher in rats fed fermented kimchi diets than rats fed control and not-fermented kimchi diet. These results suggest that kimchi stimulates lipid mobilization to epididymal fat pad and lipid excretion via feces, so lower serum and liver triglyceride concentration. The fermented kimchi stimulate the proliferation of B cell and lower the lipid accumulation in epididymal fat pad, especially kimchi fermented for 6 weeks at 4$^{\circ}C$ lower the adipose cell number.
