This study was performed to investigate nutritional effect of various dietary fibers on lead absorption and metabolism of protein and lipid in growing rats. Forty eight male rats of Sprague-Dawley strain weighing 75.7$\pm$0.7g were blocked into six groups according to body weight and fed six kinds of diet different with fiber source(non-fiber, cellulose, pectin) and lead level(0%, 1% ) for 4 weeks. Results are summerized as follows: 1) Food intake, weight gain, FER and PER were remarkably decreased in lead(Pb) added groups, and FER and PER in Pb-added pectin group were significantly lower than those in Pb-added non-fiber group. 2) Weight of liver, kidney and epididymal fat pad, bone weight and length, hematocrit, and hemoglobin content were decreased in Pb-added groups. 3) Total protein content in serum was tended to be decreased in Pb-added groups, but total lipid and cholesterol contents in serum were not different with dietary Pb level and fiber source. 4) Nitrogen, lipid and cholesterol content in liver were tended to be deceased in Pb-added groups, and especially those of the Pb-added pectin group were the lowest among groups. 5) Daily urinary and fecal excretions of nitrogen, lipid and cholesterol were decreased in Pb-added groups. Especially fecal excretions of nitrogen, lipid and cholesterol in Pb-free groups were significantly increased by dietary cellulose and pectin. 6) Pb content in blood was significantly increased in Pb-added pectin group. There was no significant decrease in Pb contents of liver, kidney and tibia, and increase in excretion of Pb by feeding dietary fibers. In conculsion, dietary fibers had no effect on the absorption of Pb, and dietary pectin seemed to increase Pb poisoning by decreasing bioavailibility of protein, lipid and other nutrients in the diet.
The research was designed to study the effect of different fat sources and levels on Body lipid metabolizm and immune responses in Sprague-Dawely strain male rats. These effect of different fat sources compared with sesame Oil, Perilla oil and Beet tallow. Fat sources were divided into 3 groups respectively 7%, 15%, 30% fat level on diet weight basis. The experimental period was 54days. 1) The body weight gain was significantly low in NF group. In Sesame oil group and perilla oil group, low fat level groups were higher than medium, high fat level groups. But in Beef tallow group, high fat level groups were higher than low and Medium groups. 2) The weight of liver, kidney and epididymal fat ped tend to increse with increasing body weight. 3) The contents of triglyceride and total lipid in serum were significantly different with dietary fat sources and Perilla oil group was the lowest. 4) The contents of triglyceride and total lipid in liver were significantly different with dietary fat levels and high fat level group was higher than low fat level group. 5) Perilla oil group, compared with Beef tallow group, showed the higher excretion of cholesterol through feces and the higher deposit of cholesterol in liver. Therefore serum cholesterol level of Perilla oil group was lower than that of Beef tallow group. But eventhough Sesame oil is vegetable oil, Sesame oil did not showed an effect like Perllia oil on serum cholesterol level. 6) Weight of thymus decreased with fat levels particularly in vegetale oil. And it had on effect on mitogen response, mitogen response decreased with fat level in vegetable oil. But in Beef tallow, there was no difference in fat level.
As diverse physiological functions of taurine have been reported, taurine-containing health drinks and products are marketed worldwide for the treatment of various conditions such as improvements of liver, heart and circulatory functions or as an aid to athletic performance. Although animal studies have shown that taurine is fairly safe when supplemented in the diet for an extended period, the effective dose range of taurine for dietary supplements is in controversy. Reports on dietary taurine intakes have been sparse, and would serve as a guideline for determining an appropriate taurine dosage. The present study was aimed to estimate dietary intake level of taurine using the taurine content database of commonly used food stuffs established recently in our laboratory, and also to evaluate plasma concentration and urinary excretion of taurine in adolescents and adults residing in Seoul area. Dietary taurine intakes of the subjects were 219$\pm$16.9mg/day for 16-19 years old(n=123), 177$\pm$18.1mg/day for adults older than 20 years old(n=123). Male subjects(n=115) consumed 216$\pm$21.1mg of taurine/day, while female subjects(n=131) consumed 181$\pm$14.3mg of taurine/day(p<0.05). The level of dietary taurine intake was positively correlated with the levels of dietary intakes of energy, carbohydrate, total lipids, cholesterol, vitamin A, vitamin B$_1$, niacin, vitamin C, calcium, phosphorous, sodium and potassium at p<0.01, and with dietary intakes of iron and animal lipids at p<0.05, respectively. Plasma taurine concentration of subjects were 135$\pm$5.9$\mu$mol/L, which is considered to be within a normal range for healthy subjects. The subjects excreted 1158$\pm$72.7nmol/ of tarine mg creatinine in their urine, which is approximated as 150-170mg of taurine/24hr urine based on the assumption that 18mg creatinine/kg/day is excreted in the urine of healthy adults, and this would be about 80% of the daily taurine intake observed in the same subjects. Dietary taurine intake level was positively correlated with plasma taurine concentration, as well as with urinary taurine excretion corrected by creatinine excretion at p<0.05. The present study was the first report of taurine intake, and plasma concentration and urinary excretion of taurine in a Korean population so far, and these results would serve as an index for the future study evaluating taurine status in a diverse population within and outside Korea. The cholesterol-lowering effects of soymilk fermented with Bacillus subtilis KCCM42923 were studied in apolipoprotein E knockout (ApoE KO) mice and compared to the cholesterol-lowering effects of soymilk to which cheonggukjang powder had been added. ApoE KO mice were divided into 3 groups (n=7/group). Animals were fed either an atherogenic diet only (AD, control group), an AD supplemented with fermented soymilk containing Corni fructus (FSM group), or an AD supplemented with soymik to which cheonggukjang (CGJ) powder had been added at 5% (w/v) (CPS group) for 8 weeks. The amount of FSM or CPS supplementing the AD was 20.8 mL/kg BW. There were no differences in either body weight gain or organ weights among three groups. In the FSM group, the concentration of plasma total cholesterol (TC) and LDL cholesterol (LDLC) were significantly decreased by 26.2% and 30.3% compared with the values of the control group (p<0.05). However, the triglyceride (TG) and HDL-cholesterol (HDLC) levels were not affected. These beneficial effects of FSM on suppressing the increase in plasma cholesterol level by AD were greater than those of CPS, which revealed 15.4% and 16.4% inhibition for TC and LDLC, respectively. However, these differences between FSM and CPS groups were not significantly different. A preventative effect of FSM or CPS on the accumulation of hepatic TC, but not on TG, was observed. FSM and CPS did not demonstrate any effects on fecal lipid excretion. In conclusion, the cholesterol-lowering effects of the soymilk fermented with Bacillus subtilis KCCM42923 were comparable to CGJ powder-added soymilk. These results suggest that drinking FSM might provide beneficial effects on controlling plasma cholesterol levels.
We assessed the effect of surimi gel, which is prepared from the king oyster mushroom (pleurotus eryngii) and cuttlefish meat paste (KCP) on lipid metabolism and antioxidant activity in high-cholesterol-fed rats. Three groups of 3-week-old male Sprague-Dawley rats were fed on a diet containing 1 g cholesterol/kg for 6 weeks. We administered only a high-cholesterol diet to the control group, one group was fed on surimi gel containing cuttlefish paste and king oyster mushrooms, and another group was fed with general boiled fish meat paste (GFP), which is commonly sold in marketplaces. Plasma and hepatic lipid profiles were measured, and the antioxidant status of the liver was assessed. The plasma triglyceride concentration did not differ significantly among the groups. Supplementation with KCP resulted in lower plasma and hepatic cholesterol concentrations and atherogenic index as compared to the control group and GFP, whereas the plasma high-density lipoprotein-cholesterol concentration was elevated. Moreover, the KCP-supplemented animals evidenced greater bile acid excretion. The KCP groups evidenced significantly lower plasma and hepatic levels of thiobarbituric acidreactive substances as compared to the control group. Besides, hepatic antioxidant enzyme activities, including catalase and superoxide dismutase, were significantly higher in the KCP group. In conclusion, KCP was quite effective in improving the lipid metabolism and reducing oxidative stress by upregulating the hepatic antioxidant enzymes in high-cholesterol-fed rats.

This study demonstrates that pork cholesterol levels are reduced in fattening stage swine fed $\beta-cyclodextrin({\beta}-cyclodextrin)$. The study subjects were 120 swine fed their respective chow diets containing 0, 5, 7, or 10% $\beta$-cyclodextrin for 35 consecutive days. Plasma total lipids, triglyceride and total cholesterol of the $\beta$-cyclodextrin treated group were significantly lower than those of the control group (p<0.05). The levels of plasma lipid were significantly decreased by 63.22 mg, 73.98 mg, and 82.12 mg in the fattening swine group fed $\beta$-cyclodextrin at 5%, 7%, and 10%, respectively, compared to those in the control group (p<0.05). When 5, 7, and 10% $\beta$-cyclodextrin was administered to fattening swine, the triacylglyceride levels were decreased by 56.24 mg, 55.48 mg, and 60.02 mg, and total cholesterol concentration was reduced by 25.05 mg, 27.17 mg, and 30.19 mg, respectively, compared to those in the control group (p<0.05). Excretion of total steroid significantly (p<0.05) increased with the increasing amount of $\beta$-cyclodextrin supplementation. The cholesterol levels of swine back fat, belly, loin, and ham were significantly decreased with increasing $\beta$-cyclodextrin supplementation (p<0.05). The pork cholesterol was significantly (p<0.05) reduced by 15.31% in the $\beta$-cyclodextrin treated group, compared to that of the control group. These results suggest that feeding $\beta$-cyclodextrin to fattening swine may produce novel functional pork with low cholesterol levels.
Ursodeoxycholic acid (UDCA) has long been used as an adjuvant or first choice of therapy for liver disease. Commonly, UDCA has been reported to play a role in improving hyperbilirubinemia and disorder of bromsulphalein. More commonly, UDCA has been used in reducing the rate of cholesterol level in bile juice that can cause cholesterol stone. The effects on the promotion of bile acid release that leads an excretion of toxic materials and wastes produced in liver cells as well as various arrays of liver disease such as hepatitis. Other than already reported in clinical use, immunosuppressive effect has been studied, especially in transplantation. In the study, we hypothesized that UDCA might have a certain role in anti-inflammation through a preventive effect of pro-inflammatory potentials in the brain macrophages, microglia. We found that the treatment of $200\;{\mu}g/ml$ UDCA effectively suppressed the pro-inflammatory mediators (i.e. nitric oxide and interleukin-$1{\beta}$) in rat microglia compared to comparators. Interestingly, RT-PCR analysis suggested that UDCA strongly attenuated the expression of $IL-1{\beta}$ that was comparable with cyclosporine A at 48 h incubation. Conclusively, we found that UDCA may playa cytoprotective role in microglial cells through direct or indirect pathways by scavenging a toxic compound or an anti-inflammatory effect, which are known as major causes of neurodegenerative diseases.
This work was conducted to investigate dietary supplementation of oyster mushroom fruiting bodies on biochemical and histological changes in hyper and normocholesterolemic rats. Six-week old female Sprague-Dawley albino rats were divided into three groups of 10 rats each. Feeding a diet containing a 5% powder of Pleurotus ostreatus fruiting bodies to hypercholesterolemic rats reduced plasma total cholesterol, triglyceride, low-density lipoprotein (LDL), total lipid, phospholipids, and LDL/high-density lipoprotein ratio by 30.18, 52.75, 59.62, 34.15, 23.89, and 50%, respectively. Feeding oyster mushrooms also significantly reduced body weight in hypercholesterolemic rats. However, it had no adverse effects on plasma albumin, total bilirubin, direct bilirubin, creatinin, blood urea nitrogen, uric acid, glucose, total protein, calcium, sodium, potassium, chloride, inorganic phosphate, magnesium, or enzyme profiles. Feeding mushroom increased total lipid and cholesterol excretion in feces. The plasma lipoprotein fraction, separated by agarose gel electrophoresis, indicated that P. ostreatus significantly reduced plasma ${\beta}$ and pre-${\beta}$-lipoprotein but increased ${\alpha}$-lipoprotein. A histological study of hepatic cells by conventional hematoxylin-eosin and oil red O staining revealed normal findings for mushroom-fed hypercholesterolemic rats. These results suggest that a 5% P. ostreatus diet supplement provided health benefits by acting on the atherogenic lipid profile in hypercholesterolemic rats.
This study was conducted to compare the effects of four types of dietary fiber supplementations (cellulose, pectin, guar gum, and polydextrose) on gastrointestinal function, diabetic symptom amelioration and lipid & glucose metabolism in streptozotocin-induced diabetic rats. Six groups of male rats were fed ad libitum dietary fiber-free control diet or one of experimental diets containing 5% dietary fiber for four weeks. All types of dietary fiber supplementation seemed to protect the diabetic animals from the loss of body weight. The primary diabetic symptoms such as polydipsia, polyphasia, polyuria and urinary glucose excretion were ameliorated by cellulose, pectin, and guar gum, but not by polydextrose. Gastrointestinal transit time was significantly shortened and fecal dry weight was significantly increased in all the dietary fiber-supplemented groups except the polydextrose group. Large intestine was significantly lengthened by dietary fiber feeding. The serum triglyceride and total cholesterol levels were effectively lowered by pectin, guar gum and polydextrose. Regardless of their types, the fiber supplementation had no effect on serum HDL-cholesterol. Whereas fasting blood glucose level was significantly lowered by all types of fiber supplementations, glucose tolerance was more effectively improved by pectin and guar gum.
This study was performed to elucidate the mechanism of dietary protein level on renal function through lipid and eicosanoids metabolism. Male rats of 337.8$\pm$5.7g body weight were undergone unilateral nephrectomy or sham-operation. The rats were divided into high protein(40% casein), normal protein(15% casein) and low protein(8% casein) diet groups and fed experimental diets ad libitum for 24 weeks. The results are summarized as follows. Serum total lipid, cholesterol and HDL-cholesterol of rats in 15% and 40% casein groups were higher than those of 8% casein group. But serum triglyceride was affected neither by uninephrectomy nor by dietary protein level. Serum thromboxane(TX) B2 and 6-keto prostaglandin F1$\alpha$ increased with increasing dietary protein level. Serum prostaglandin(PG) E2 was not affected by uninephrectomy nor by dietary protein level. Urinary PGE2 and TXB2 excretion tended to be lower in uninephrectomized groups. Renal tissue concentration of TXB2 was lower in uninephrectomized groups and in high protein group. These results suggest the possibility that the effects of dietary protein level on renal function could be due to changes in lipid and eicosanoids metabolism.
