식이섬유가 고콜레스테롤혈증 흰쥐의 혈청 지질대사에 미치는 영향 (Effect of Fiber on Serum Lipid Metabolism in Rats with Diet-Induced Cholesterolemia)

  • The study was conducted to investigate the effect of cholesterol and fiber on serum lipid metabolism in rats. Male Sprague Dawley rats were administrated 1% cholesterol and 0.25% sodium cholate to induce hypercholesterolemia and were fed on diet containing three concentrations(0%, 5%, 10%) of cellulose and pectin, respectively. The rats were sacrified after 5 weeks of feeding periods. Serum triglyceride concentration was significantly decreased in proportion to level of fiber. Phospholipid concentration was more decreased in 10% fiber groups than 5% fiber groups but did not show any difference by fiber types. Free cholesterol and cholesteryl ester concentrations in serum were significantly decreased by fiber supplement and pectin was more effective than cellulose in cholesterol lowering effect. Whereas HDL cholesterol concentration and HDL C/TC were increased in proportion to level of fiber and the effect of pectin was not more significant than that of cellulose. Serum chylomicron cholesterol concentration also showed significant decrease by pectin supplement and LDL , VLDL cholesterol concentrations were more significantly decreased in 10% fiber groups than those of 5% fiber groups. The results indicate that 10% pectin may have beneficial roles in hypercholesterolemia.

지방의 섭취량과 첨가된 섬유소의 종류가 흰쥐의 체내 지질 수준에 미치는 영향 (The Effect of Dietary Fat Levels and Sources of Dietary Fiber on Serum and Liver Lipids of Rats)

  • 식이중 지방 수준과 fiber 종류가 흰 쥐의 혈청과 간 조직내의 지질 함량에 미치는 영향을 알아보기 위해 지방 수준을 0% , 5% , 20% 수준으로 달리 하였으며 fiber 종류는 고지방 식이군( 20% 지방 )에 pectin과 cellulose를 각각 10% 수준으로 첨가하였다. 또 식이내 cholesterol첨가의 효과를 관찰하기 위해 고지방 식이군에 0.5% cholesterol을 첨가한 후 pectin과 cellulose를 10%씩 넣어 전체적으로 7군으로 나누어 각 각 다른 식이로 4주간 사양한 후 혈청과 간에서 지질 함량을 조사하였으며 간 조직의 형태학적 조사를 실시하여 다음과 같은 결과를 얻었다. 1) 식이 섭취량은 지방 수준이 낮을수록 높았으나 뚜렷한 차이는 보이지 않았으며 pectin과 cellulose 첨가로 인해서 섭취량이 줄었다. 증체량은 지방 수준이나 fiber종류에 따라 큰 차이를 보이지 않았다. 변의 무게는 지방 수준에 따른 차이는 없었으나 fiber 첨가로 인해서는 pectin은 약간의 감소를 cellulose 는 뚜렷한 증가를 보였다. 또 cholesterol을 첨가한 군 간의 비교에서도 pectin첨가군은 감소를 cellulose첨가군은 증가를 나타냈다. 간과 epididymal fat pad의 무게는 지방 수준이 높아짐에 따라 증가하였으며 fiber 종류에 따라서는 pectin첨가군이 cellulose 첨가 군 보다 낮았다. 2) 혈청 분석 결과는 식이내 지방 수준이 높을수록 total lipids수준이 높아지는 경향을 보였으며 pectin이 cellulose보다 혈청내 지질 수준을 뚜렷하게 낮추었다. Total cholesterol 및 phospholipids 량에 있어서도 지방 수준이 증가함에 따라 높아졌으며 cellulose보다 pectin이 첨가 되었을때 현저한 감소를 나타냈다. HDL- cholesterol수준은 무지방군에서는 매우 높게 나타났으나 표준군과 고지방군간에 는 명확한 결과를 얻을 수 없었다. 3) 간 분석 결과는 지방수준이 높을수록 total lipids량이 높아지는 경향을 보였다. 또 pectin 첨가가 cellulose첨가군보다 현저히 낮은 lipids수준을 나타냈다. 간의 total cholesterol함량은 지방 수준이 높아짐에 따라 증가하였으나 뚜렷한 차이는 아니었으며 pectin첨가가 cellulose첨가보다 cholesterol 수준이 상당히 낮았다. 4) 간 조직의 현미경 관찰 결과는 지방 수준이 높아짐에 따라 조직내 지방 침착이 심해졌으며 fiber 의 첨가로 인해서는 pectin은 뚜렷한 지방 침착의 감소를 보였으나 cellulose는 거의 감소 효과를 보이지 않았다.

  • This study was conducted to investigate the effects of dietary fibers (cellulose ; polydextrose ; pectin ; ricebran) supplementation on the kidney fucntion of hyperlipidemic-induced rats on a high cholesterol diet (5% cholesterol). Serum lipid concentrations were not influenced by dietary fiber sources, but the pectin group was effective in reducing serum lipid levels. Total serum protein and albumin concentrations tended to higher in the polydextrose group, and serum creatinine and urea-N concentrations were higher in the pectin group than those in the other groups. Even though total urinary protein was high in the pectin group, GFR was reduced (18~30% decrease) as compared to other groups. The urea-N level was elevated in the polydextrose group as compared to that of cellulose group(25% increase). Total lipid, triglyceride and cholesterol concentrations in the liver of pectin group were lower than those of other groups. Total lipid and cholesterol concentrations in the kidneys were reduced in pectin and ricebran groups, respectively. The total cholesterol concentration infecal was significantly high in the polydextrose and pectin groups(p<0.05), and the triglyceride was highest in the pectin group. These results indicate no significant effects of dietary fiber supplementation on the kidney function of hyperlipidemic-induced rats on a high cholesterol diet, but pectin was very effective to improve lipid metabolism and to reduce GFR.

두류의 첨가가 당뇨병 쥐의 내당능과 지질대사 및 지질과산화에 미치는 영향 (Effects of Legume Supplementation on the Glucose and Lipid Metabolism and Lipid Peroxidation in Streptozotocin-Induced Diabetic Rats)

  • The present study was conducted to evaluate the usefulness of common Korean legumes as a high-fiber supplement in therapeutic diets for diabetic patients. Streptozotocin-induced diabetic rats were used as animal models and four kinds of legumes, black soybean (BS), yellow soybean (YS), green pea (GP) and soybean curd residue (SCR) were tested as high-fiber supplements. Seven groups of normal and streptozotocin-induced diabetic rats were fed isocaloric experimental diets containing 8% dietary fiber from one of four legumes or purified cellulose and pectin for 6 weeks. The effects of the legumes on the glucose and lipid metabolism of diabetic rats was examined and compared with the effects of cellulose and pectin. The legume supplementation did not show any beneficial effect on glucose tolerance, however, it exhibited a plasma cholesterol-lowering effect in diabetic rats. The cholesterol-lowering action was especially strong in BS and the degree of the effect was comparable to that of pectin. The levels of total lipids, cholesterol, and triglyceride in the hepatic tissues of rats fed legume diets were similar to those of the pectin group. All legume supplements induced an increase in fecal steroid excretion. The fecal cholesterol contents were significantly high following the supplementations of YS and SCR (p < 0.05). The excretion of fecal bile acids in the BS and YS groups was significantly higher than it was in the pectin group (p < 0.05). Concentration of lipid peroxidation products in the blood and urine of diabetic animals was lower in the legume groups than in the cellulose group. The levels of hepatic lipid peroxidation products were significantly lower in the BS and YS groups than in the pectin group (p < 0.05). From the results of this study, the plasma cholesterol-lowering effect of BS is possibly due to the significant (p < 0.05) in-crease in fecal steroid excretion, which suggests that BS could be beneficial in improving abnormal lipid metabolism in diabetic rats.   • The study was conducted to investigate the effect of cholesterol and fiber on liver lipid metabolism in rats. Male Sprague Dawley rats were administrated 1% cholesterol and 0.25% sodium cholate to induce hypercholesterolemia and were fed diet containing three levels(0%, 5% and 10%) of cellulose and pec-tin respectively. The rats were sacrified after 5 weeks of feeding period. Net weight gain and feed ef-ficiency ratio were increased in 10% fiber groups as compared to those of 5% fiber groups. Feed intake did not show significancy by fiber kinds and levels. Liver and kidney weights tended to be decreased in proportion to level of fiber. Heart weight was lowered in 10% fiber groups as compared to that of 5% fi-ber groups. Liver triglyceride concentration was significantly increased in pectin groups. Total-free-cholesterol and cholesteryl ester concentrations in liver were significantly decreased by fiber and were decreased in proportion to level of fiver. Phospholipid concentration was significantly decreased by fiber and were decreased in proportion to level of fiber. Phospholipid concentration was significantly decreased in 5% cellulose group. the results indicate that 10% pectin may have beneficial roles in hypercholester-olemia.

식이중에 첨가된 섬유소의 종류와 수준이 흰쥐의 체내 지질함량에 미치는 영향 (Influence of Sources and Levels of Dietary Fiber on Lipid Composition in Rats)

  • 식이 중의 섬유소 종류와 급여수준이 흰쥐의 혈청과 간 조직내의 지질함량에 미치는 영향을 검토하기 위하여 섬유소중 pectin과 cellulose를 선택하여 급여수준을 5%, 10%, 20%로 달리하여 식이중에 참가하였다. 각 군의 콜레스테롤 함량은 0,5%로 고정시켰고 이와는 별도로 콜레스테롤 무첨가군을 설정하였다. 각 실험식이로 4주간 사양한 후 혈청과 간에서의 지질함량을 분석하여 다음과 같은 결과를 얻었다. 1) 체중 증가량은 식이섬유소의 종류에 따라 현저한 차이를 나타내어 pectin 첨가군은 cellulose 첨가군에 비하여 유의적으로 감소하는 경향이었고 cellulose첨가군에서는 대조군과 별다른 차이를 나타내지 않았다. 사료효율에 있어서도 pectin 10%와 20% 첨가수준에 있어서 대조군에 비하여 점차로 감소되었다. 간장의 무게는 체중 증가량의 변화와 같은 경향을 나타내었다. 2) 간장내의 총 지질수준에 있어서는 pectin군은 첨가수준이 증가함에 따라 점차로 감소되었고 cellulose 첨가군에서는 첨가수준이 증가함에 따라서 오히려 증가되는 경향을 나타내었다. 간의 콜레스테롤 수준도 총 지질과 마찬가지로 pectin 첨가군의 경우에 첨가수준의 증가에 따라 유의적으로 감소되었고 20% 첨가수준에서는 cholesterol 무첨가군과 차이를 나타내지 않았다. Cellulose첨가군에서는 첨가수준이 감소됨에 따라 유의적으로 감소하였다. 또한 인지질은 pectin 5%, 10%군에서는 대조군에 비해 점차로 증가하였으나 20%군에서는 감소된 경향을 보였고 cellulose 첨가군에서는 따라서 일치된 경향을 보이지는 않았다. 3) 혈청내 콜레스테롤 수준은 pectin첨가군에서는 첨가수준의 증가에 따라 대조군과 별다은 차이를 보이지 않았고 cellulose 처가군에서는 10%, 20% 수준에서 대조군에 비하여 유의적으로 증가되었다. 형청내 인지질 함량은 pectin 첨가의 경우에는 첨가수준에 관계없이 대조군과 유의적인 차이를 나타내지 않았고 cellulose 첨가의 경우에는 모든 첨가수준에 있어서 대조군보다 증가하였다. 중성지바의 함량은 pectin첨가군에서는 모두 대조군에 비하여 유의적으로 감소하였으나 cellulose첨가군에서는 5% 준에서는 약간의 증가를 보였으나 10%, 20% 수준에서는 유의적인 차이를 나타내지 않았고 콜레스테롤 무첨가군에서는 대조군에 비해 유의적으로 감소되었다.

식이중 지방 수준과 Fiber 종류가 흰쥐의 지방대사에 미치는 영향 (Influence of fat levels and types of dietary fiber on lipid metabolism of rats)

  • The influence of fat level and dietary fiber on serum and liver lipid concentration of the rat was investigated. Groups of rats were fed respectively 3%, 5%, 20% fat containing and free fiber diet until 5th week. From 5th week to 9th week high fat diet groups of rates were devided high fat diet group, high fat pectin supplemented diet group and high fat celulose supplemented diet group. Pectin and cellulose at 10% was supplemented to high fat diets. After rats were fed for 9 weeks, all rats were sacrificed to collect the liver and blood samples by heart puncture. Serum and liver lipids were determined on all rats and compared the histochemical lipid staining method with the biochemical analysis of serum and liver lipids. A high fat diet caused increased level of the total lipids, total cholesterol and free cholesterol contents in the liver and the serum. Rats fed pectin reduced body weight, werum lipid and liver lipid but cellulose had no effect to reduce serum and liver lipid concentration. These results indicate that dietary fat level led to changes in the lipid metabolism of rats and that pectin was more effective in lowering serum and liver lipid than cellulose.

고지방식이와 식이섬유가 흰쥐의 체내 지질대사에 미치는 영향 (The effect of High Fat Diet and Dietary Fiber on Lipid Metabolism in Rats)

  • 이유기를 지난 웅성흰쥐를 대조식이 (C), 고지방식이 (HF) 및 고지방 고에너지식이 (HFHE) 로 4 주간 사양하고 일부를 희생시킨 후 , HF 군과 HFHE 군 일부에식이섬유 pectin 과 cellulose를 각각 첨가한 HF-P , HF-C, HFHE-P , HFHE-C 식이를 C, HF ,HFHE 식이와 함께 4 주간 더 사양한 후 희생시켜 체중증가량, 총 에너지섭취량, 총식이섭 취량, 식이효율, 간장과 비장의 무게, 혈중 지질의 성상 및 간세포에 미치는 영향을 비교 관 찰한 결과 체중증가는 C 군, HF 군, HFHE 군간의 유의적 차이는 없었으며, 식이섬유 첨가 시 모두 체중이 감소되었다. 총에너지 섭취량은 C 군이 가장 많았고, 식이효율은 HF-HE군 이 가장 높았으며, 식이섬유 첨가에 의해서는 영향을 받지 않았다. 간장의 무게는 C군 , HF 군, HFHE 군 모두 식이의 영향이 없었고, pectin 과 celulose 첨가에 의한 변화도 나타내지 않았다. 비장의 무게는 C군 보다 HF 군과 HFHE 군이 감소하였으나, pectin 과 cellulose 첨가에 의해서는 변화가 없었다. 혈중 총 지질, 총콜레스테롤, 중성지방 및 인지질의 함량은 각 실험군모두 실험기간이 경과함에 따라 증가되었으며, 특히 HFHE 군의 유의성있게 증가 하였고 pectin 과 cellulose를 첨가하였을때는 감소하였으며 pectin이 cellulose 보다 감소효 과가있다.

식이내 섬유질의 종류가 성장기 흰쥐의 납 흡수 및 체내대사에 미치는 영향 (Effect of dietary fiber on lead absorption and metabolic changes in growing rats)

  • This study was performed to investigate nutritional effect of various dietary fibers on lead absorption and metabolism of protein and lipid in growing rats. Forty eight male rats of Sprague-Dawley strain weighing 75.7$\pm$0.7g were blocked into six groups according to body weight and fed six kinds of diet different with fiber source(non-fiber, cellulose, pectin) and lead level(0%, 1% ) for 4 weeks. Results are summerized as follows: 1) Food intake, weight gain, FER and PER were remarkably decreased in lead(Pb) added groups, and FER and PER in Pb-added pectin group were significantly lower than those in Pb-added non-fiber group. 2) Weight of liver, kidney and epididymal fat pad, bone weight and length, hematocrit, and hemoglobin content were decreased in Pb-added groups. 3) Total protein content in serum was tended to be decreased in Pb-added groups, but total lipid and cholesterol contents in serum were not different with dietary Pb level and fiber source. 4) Nitrogen, lipid and cholesterol content in liver were tended to be deceased in Pb-added groups, and especially those of the Pb-added pectin group were the lowest among groups. 5) Daily urinary and fecal excretions of nitrogen, lipid and cholesterol were decreased in Pb-added groups. Especially fecal excretions of nitrogen, lipid and cholesterol in Pb-free groups were significantly increased by dietary cellulose and pectin. 6) Pb content in blood was significantly increased in Pb-added pectin group. There was no significant decrease in Pb contents of liver, kidney and tibia, and increase in excretion of Pb by feeding dietary fibers. In conculsion, dietary fibers had no effect on the absorption of Pb, and dietary pectin seemed to increase Pb poisoning by decreasing bioavailibility of protein, lipid and other nutrients in the diet.

식이섬유 첨가식이 Streptozotocin-유도 당뇨쥐의 장기능과 지질 및 당질대사에 미치는 영향 (The Effects of Dietary Fiber Feeding on Gastrointestinal Functions and Lipid and Glucose Metabolism in Streptozotocin-induced Diabetic Rats)

  • This study was conducted to compare the effects of four types of dietary fiber supplementations (cellulose, pectin, guar gum, and polydextrose) on gastrointestinal function, diabetic symptom amelioration and lipid & glucose metabolism in streptozotocin-induced diabetic rats. Six groups of male rats were fed ad libitum dietary fiber-free control diet or one of experimental diets containing 5% dietary fiber for four weeks. All types of dietary fiber supplementation seemed to protect the diabetic animals from the loss of body weight. The primary diabetic symptoms such as polydipsia, polyphasia, polyuria and urinary glucose excretion were ameliorated by cellulose, pectin, and guar gum, but not by polydextrose. Gastrointestinal transit time was significantly shortened and fecal dry weight was significantly increased in all the dietary fiber-supplemented groups except the polydextrose group. Large intestine was significantly lengthened by dietary fiber feeding. The serum triglyceride and total cholesterol levels were effectively lowered by pectin, guar gum and polydextrose. Regardless of their types, the fiber supplementation had no effect on serum HDL-cholesterol. Whereas fasting blood glucose level was significantly lowered by all types of fiber supplementations, glucose tolerance was more effectively improved by pectin and guar gum.
