Selecting Appropriate Seedling Age for Restoration Using Comparative Analysis of Physiological Characteristics by Age in Abies koreana Wilson

  • The aim of this study was to investigate the sensitivity to environmental stress, and changes in the photosynthesis capacity in Abies koreana seedlings by age and to suggest the most effective age for restoration. To identify these physiological characteristics of A. koreana, the chlorophyll fluorescence and photosynthetic capacity of 1-, 2-, 3-, 5- and 6-year-old A. koreana seedlings were observed from June 2020 to June 2021. The maximum quantum efficiency of Photosystem II (Fv/Fm), a chlorophyll fluorescence measurement parameter, was strongly positively correlated with the monthly average temperature (1-year-old seedling: r=0.8779, 2-year-old seedling: r=0.8605, 3-year-old seedling: r=0.8697, 5-year-old seedlings: r=0.8085, and 6-year-old seedlings: r=0.8316). The Fv/Fm values were the lowest in winter (November 2020-March 2021). In addition, the Fv/Fm values of 1-, 2-, and 3-year-old seedlings in winter were lower than that of 5- and 6-year-old seedlings, while the Fv/Fm values in summer were relatively higher than those in winter. Further, the Fv/Fm values of seedlings of all ages decreased in August 2020, when the monthly average temperature was the highest. In particular, 1-year-old to 3-year-old seedlings showed Fv/Fm values less than 0.8. Further, the photosynthetic capacity measured in August 2020 increased with increasing seedling age. The analysis of variance results for summer Fv/Fm values showed significant differences in age-specific averages (p<0.05), and Duncan's multiple range test showed significant differences between 5- and 6-year-old seedlings and 1-, 2-, and 3-year-old seedlings (p<0.05). These results suggested that the 5- and 6-year-old seedlings were less sensitive to environmental stress and showed better photosynthetic capacity than the 1-, 2-, and 3-year-old seedlings. Therefore, 5-year-old or older A. koreana seedlings can be used as restoration materials because they can show increased adaptability and stable growth during transplantation due to their relatively high environmental resistance and photosynthetic capacity.

Growth and Physiological Characteristics of Containerized Seedlings of Sageretia thea at Different Fertilization Treatments (시비처리에 따른 상동나무 용기묘의 생장 및 생리특성)

  • This study aims to optimize the appropriate concentration of fertilizers for Sageretia thea by analyzing growth performances (height and root collar diameter) and physiological characteristics (photosynthesis, chlorophyll contents, and chlorophyll fluorescence reaction). As fertilizer concentration was increased to 1.5 g·L-1, growth increased, but it decreased at 2.0 g·L-1 treatment. Root collar diameter growth was reduced because of higher fertilizer concentrations. Photosynthesis reactions showed the highest CO2 reaction curves, maximum photosynthesis rate, and maximum carboxylation rate in the 1.5 g·L-1 fertilizer treatment. The chlorophyll fluorescence reaction and SPAD values revealed that fertilizer treatment improves photosynthesis efficiency and robustness compared with untreated control. Therefore, the appropriate fertilizer concentration for producing good seedling quality of Sageretia thea is 1.0~1.5 g·L-1.

The Effects of Fertilization on Growth Performances and Physiological Characteristics of Liriodendron tulipifera in a Container Nursery System (시비 처리가 백합나무 용기묘의 생장 및 생리적 특성에 미치는 영향)

  • Fertilization is essential to seedling production in nursery culture, but excessive fertilization can contaminate surface and ground water around the nursery. The objective of this study was to find optimal fertilization practice of container seedling production for reducing soil and water contamination around the nursery without compromising seedling quality. This study was conducted to investigate growth performance, photosynthesis, chlorophyll fluorescence, and chlorophyll contents of Liriodendron tulipifera growing under three different fertilization treatments (Constant rate, Three-stage rate, and Exponential rate fertilization). Root collar diameter, height, and biomass of L. tulipifera were the highest at Constant treatment. Like growth performance, seedling quality index (SQI) were higher at Constant than at other treatments, but not significantly different among treatments. L. tulipifera showed good photosynthetic capacity at all treatments. Photochemical efficiency and chlorophyll contents were significantly lower at Exponential than at other treatments. Therefore, Exponential fertilization which is 50% fertilizer of other treatments would maximize seedling growth and minimize nutrient loss.

Effect of Calcium Chloride($CaCl_2$) on Chlorophyll Fluorescence Image and Photosynthetic Apparatus in the Leaves of Prunus sargentii (염화칼슘 처리가 산벚나무 엽의 엽록소형광반응과 광합성기구에 미치는 영향)

  • There is a little information on the effect of calcium cloride ($CaCl_2$) which is used as deicing salt in Korea on the physiological responses of the street trees. Prunus sargentii is one of the most widespread tree species of street vegetation in Korea. In this study, the effect of $CaCl_2$ on photosynthetic apparatus such as chlorophyll fluorescence image and light response curve of P. sargentii in relation to their leaf and root collar growth responses were investigated. To study the effect of $CaCl_2$ treatment in the early spring, we irrigated twice in rhizosphere of P. sargentii (3-year-old) planted plastic pots with solution of 0.5%, 1.0%, 3.0% $CaCl_2$ concentration before leaf expansion. Results after treatments, total chlorophyll contents and the chlorophyll a/b, photosynthetic rate, quantum yield, dark respiration decreased with increasing $CaCl_2$ concentration. On the contrary, light compensation point increased with increasing $CaCl_2$ concentration. Through the linear regressions of correlation of photosynthetic rate with photosynthetic parameters (quantum yield, dark respiration and light compensation point), we found a significant relationship (p<0.05) between photosynthetic rate and quantum yield and light compensation point except dark respiration. Calcium cloride ($CaCl_2$) induced inhibition of photochemical efficiency ($F_v/F_M$) and non-photochemical quenching (NPQ) were found in treatments of $CaCl_2$, and these reduction rates between control and CaCl2 treatments were drastically showed at 80 days. We suggest that physiological activities are limited from treatment of $CaCl_2$. These reductions of photosynthetic apparatus ability caused eventually the reduction of leaf and diameter at root collar growth.

Introduction of Stay Green Mutant for the Development of Black Seed Coat and Green Cotyledon Soybean Variety (녹색자엽 검정콩 품종 육성을 위한 Stay green 변이체 활용)

  • The soybean stay green mutant genotype (SSG) derived from the nuclear gene, d1d2, and cytoplasmic gene, cytG, inhibit the breakdown of chloroplast in the leaves, pod walls, seed coats, and embryos during maturity. Soybean seed with black seed coat and green cotyledon (SBG) are preferred than black seed coat with yellow cotyledon (SBY) especially for cooking with rice and as source of traditional food in Korea. The researchers evaluated the seed's chlorophyll content of SSG and introduced SSG to the SBG variety breeding program. The seed chlorophyll content of SSG with d1d2 was $39.93{\sim}60.80\;{\mu}g/g$ and SSG with cytG $38.08{\sim}39.89\;{\mu}g/g$. The Korean SBG variety which was derived from SSG with cytG, contains $16.35{\sim}37.73\;{\mu}g/g$. The composition of seed chlorophyll differs according to the genetic background of SSG genotype. Inheritance study showed that cotyledon color was segregated 15:1 (yellow:green) at $F_2$ seed indicating two recessive genes control green cotyledon as revealed by previous study. Only less than 3% soybean lines showed black seed coat with green cotyledon among crosses SBY and SSG (d1d2). Results showed that SSG with d1d2 can be used as a good source for SBG with high chlorophyll content in the seed cotyledon, but due to the complex genetic behavior, breeding resource of SBG with d1d2 should be prepared to improve the breeding efficiency for development SBG variety.

Shading Effects on the Growth and Physiological Characteristics of Osmanthus insularis Seedlings, a Rare Species (희귀 식물 박달목서 유묘의 생장 및 생리적 특성에 대한 차광 효과)

  • This study was conducted to determine the optimal light conditions for the in situ and ex situ conservation and restoration of Osmanthus insularis, a rare plant species in South Korea. Evaluations included the growth performance, leaf morphological features, photosynthetic characteristics, and photosynthetic pigment contents of seedlings grown from April to November under different light conditions (100%, 55%, 20%, and 10% relative light intensity). The shoot lengths and root collar diameters did not differ significantly with relative light intensity. The dry weights of leaves, stems, and roots and the leaf number were highest at 55% relative light intensity. The leaf shape showed morphological acclimation to light intensity, with leaf area decreasing and thickness increasing as the relative light intensity increased. Several leaf parameters, including photosynthetic rate and stomatal conductance at light saturation point, net apparent quantum yield, and dark respiration, as well as chlorophyll a, chlorophyll b, and carotenoid contents, were all highest at 55% relative light intensity. Under full light conditions, the leaves were the smallest and thickest, but the chlorophyll content was lower than at 55% relative light intensity, resulting in lower photosynthetic ability. Plants grown at 10% and 20% relative light intensity showed lower chlorophyll a, chlorophyll b, and carotenoid contents, as well as decreased photosynthetic and dark respiration rates. In conclusion, O. insularis seedlings exhibited morphological adaptations in response to light intensity; however, no physiological responses indicating enhanced photosynthetic efficiency in shade were evident. The most favorable light condition for vigorous photosynthesis and maximum biomass production in O. insularis seedlings appeared to be 55% relative light intensity. Therefore, shading to approximately 55% of full light is suggested for the growth of O. insularis seedlings.

Effect of Controlled Light Environment on the Growth and Ginsenoside Content of Panax ginseng C. A. Meyer (광환경 조절이 인삼의 생육과 진세노사이드 함량에 미치는 영향)

  • Background: The photosynthetic efficiency cool-season, semi-shade ginseng is normal at low morning temperatures, but drops at high afternoon temperatures. Therefore, optimal plant performance would be ensured if it were possible to control daily light transmission rates (LTR). Methods and Results: Plants were grown in a controlled light environment that replicated 11 AM conditions and comparatively analyzed against plant grown under normal conditions. Growth in the controlled light environment resulted in a 2.81 fold increase in photosynthetic efficiency with no change in chlorophyll content, although LTR were high due to low morning temperatures. Increased aerial plant growth was observed in the ginseng plants adapted to the controlled light environment, which in turn influenced root weight. An 81% increase in fresh root weight (33.3 g per plant on average) was observed in 4-year-old ginseng plants grown in controlled light environment compared to the plants grown following conventional practices (18.4 g per plant on average). With regard to the inorganic composition of leaves of 4-year-old ginseng plants grown in controlled light environment, an increased in Fe content was observed, while Mn and Zn content decreased, and total ginsenoside content of roots increased 2.37 fold. Conclusions: Growth of ginseng under a favorable light environment, such as the condition which exist naturally at 11 AM and are suitable for the plant's photosynthetic activity creates the possibility of large scale production, excellent-quality ginseng.

Regeneration of Plants from EMS-treated Immature Embryo Cultures in Soybean [Glycine max(L.) Merr.]

  • Since somatic embryogenesis combined with ethylmethane sulfonate(EMS) treatments is the most efficient technique for mutagenesis, the embryogenic capacity of four soybean cultivars was evaluated at different EMS concentrations, treatment times, and preculture durations. Two to 4 mm long immature cotyledons were placed in induction medium after EMS treatment, and the numbers of somatic embryos formed per explant were counted four weeks after culture initiation. We observed genotypic differences in the efficiency of somatic embryogenesis from immature embryos among four cultivars treated with different concentrations of EMS for six hours. Cultivars, Sinpaldalkong 2 and Jack, displayed highly efficient somatic embryogenesis regardless of EMS concentration, whereas very low efficiency or no survival was observed in Jinju 1 and Iksannamulkong cultivars. Preculture duration did not influence the efficiency of somatic embryogenesis. Because Sinpaldalkong 2 exhibited the best somatic embryogenesis, much higher concentrations of EMS were used to test somatic embryo formation under different periods of time in this cultivar. Three and six hour treatments with both 1 and 2 mM EMS yielded higher embryo formation than longer periods of time. Increasing the time with embryos in 2 mM EMS caused a reduction in somatic embryogenesis in Sinpaldalkong 2, but many chlorophyll-deficient soybean variants were identified in the $M_1R_0$ and $M_2R_1$ generations. In addition to Jack, Sinpaldalkong 2 is a good genotype for plant regeneration from EMS-treated immature embryo cultures.

Increased Microalgae Growth and Nutrient Removal Using Balanced N:P Ratio in Wastewater

  • Microalgal cultivation using wastewater is now regarded as essential for biodiesel production, as two goals can be achieved simultaneously; that is, nutrient removal efficiency and biomass production. Therefore, this study examined the effects of carbon sources, the N:P ratio, and the hydraulic retention time (HRT) to identify the optimal conditions for nutrient removal efficiency and biomass production. The effluent from a 2nd lagoon was used to cultivate microalgae. Whereas the algal species diversity and lipid content increased with a longer HRT, the algal biomass productivity decreased. Different carbon sources also affected the algal species composition. Diatoms were dominant with an increased pH when bicarbonate was supplied. However, 2% $CO_2$ gas led to a lower pH and the dominance of filamentous green algae with a much lower biomass productivity. Among the experiments, the highest chlorophyll-a concentration and lipid productivity were obtained with the addition of phosphate up to 0.5 mg/l P, since phosphorus was in short supply compared with nitrogen. The N and P removal efficiencies were also higher with a balanced N:P ratio, based on the addition of phosphate. Thus, optimizing the N:P ratio for the dominant algae could be critical in attaining higher algal growth, lipid productivity, and nutrient removal efficiency.
