The Effects of Nonylphenol on Freshwater Phytoplankton and Zooplankton Communities

  • Recent studies reveal that the endocrine disrupter nonylphenol can also influence the growth of planktonic organisms. To clarify the effect of nonylphenol on the whole planktonic community, we monitored planktonic abundances after addition of nonylphenol using small-scale microcosms in a laboratory. Nonylphenol was added at final concentrations of 1.25 and $2.5{\mu}g\;L^{-1}$, close to the EC50 for the growth of the rotifer, Brachionus calyciflorus. Chlorophyll $\alpha$ concentration increased significantly between 2 to 5 days after nonylphenol treatment compared to the control. The abundance of the predominant phytoplankton, Stephanodiscus hantzschii, followed the same pattern as chlorophyll a concentration. While there was no negative effect on the abundance of ciliates and rotifers, crustacean zooplankton abundance was higher in nonylphenol treatments. Although the relationship did not reach significance, the growth rate of rotifers tended to decline with increasing nonylphenol dosing. It is likely that the decreased rotifer grazing on S. hantzschii caused significant increase in their abundance. This study emphasizes the importance of considering indirect effects of environmental pollutants when predicting the response of biological community to toxicant exposure.

On the utrophication in the Lake Changja (장자못의 부영양화에 관하여)

  • The process of eutrophication was studied in the lake Changja from November in 1974 to May in 1975. The stratification of water temperature was going to be formed in April and the content of dissolved oxygen varied a little but it was almost zero from January. Total phosphorus was increased very rapidly in February but total nitrogen and carbohydrate-like substances were increased a little during the experimental period. The amount of chlorophyll-$\alpha$ varied from 127mg/$m^2$ to 717 mg/$m^2$ with maximum value occuring in January. This phenomenon was an evidence that chlorophyll-$m^2$ and total phosphorus were correlated each other. The numbers of colony of the coliform bacteria in point A was 10 times more than in point B. In conclcesions, the lake Changja is a very eutrophic and the ecological equilibrium of the lake can not be recovered as far as origin of pollution is erased out.

Preliminary Study on the Toxicity and Transfer of Heavy Metals and Tributyltin to Seagrass Zostera marina (잘피의 광합성에 대한 중금속 및 TBT의 독성 영향과 중금속 흡수에 대한 연구)

  • Uptake kinetics of Cd and Zn by leaves and rhizome of the seagrass Zostera marina were examined in controlled laboratory radiotracer experiments. Subsequently, acute toxicity of Cd, Cu and TBT on photosynthetic quantum yield (ΔF/Fm’ of Z. marina were determined, and the differential sensitivities of rapid light curve (RLC) to those harmful substances were also compared. All measurements on photosynthetic activity were determined by chlorophyll a fluorescence method using pulse amplitude modulation (PAM). Metal uptake by Z. marina was saturated with increasing exposure time in leaves and rhizomes. Uptake of Zn by Z. marina was faster than that of Cd. Metal uptake rates in Z. marina decreased with the increase of dissolved metal concentrations and also with the increase of biomass. The adverse effect of TBT on effective quantum yield was stronger than other pollutants. Average acute toxicity on the RLC of the seagrass exposed to TBT and two heavy metals (Cd and Cu) was going to decrease as follows: TBT > Cd > Cu. Our preliminary results in this study suggested that Z. marina potentially can be used as a biomonitor of harmful substances contamination in coastal waters.

Dynamics of the Phytoplankton Community in the Coastal Waters of Chuksan Harbor, East Sea (동해 축산항 연안의 식물플랑크톤 군집 동태)

  • In order to investigate the distribution of phytoplankton community in the coastal waters of the Chuksan Harbor, East Sea, the abundance and biomass of phytoplankton have been evaluated through seasonal interval sampling from April 2000 to October 2002. A total of 363 different phytoplankton species was observed and most of them were composed of diatoms. The mean abundance and chlorophyll-a concentration of phytoplankton during the study period ranged from 56 ${\times}$ $10^3$ to 720 ${\times}$ $10^3$ cells $L^{-1}$ and from 0.78 to 3.29 μg chl-a $L^{-1}$, respectively. The relative contribution of the size-fractionated phytoplankton to phytoplankton community showed difference according to seasons. The average contribution of nano-phytoplankton(<20 $\mu$m) was over 50% in the total abundance and biomass of the phytoplankton. Our results show that nano-phytoplankton play an important role in the southern coastal waters of the East Sea. And the environmental factors such as suspended substances, phosphates and silicates were positively correlated with the abundances and biomass of phytoplankton.

  • Seashore joining with land and sea, which is typical habitat for marine macroalgae, is classified two types of shore as soft- and hard-bottom shore according to topographical (geological) and ecological features. We compared two of Ulva pertusa Kjellman from two contrasting habitats, sandy (soft-bottom, Haenam) and rocky shore (hard-bottom, Hadong) in terms of chlorophyll-a fluorescence and its parameters, and various photosynthetic pigment and nutrient content in the tissue of those. Both of habitats were different in the light environment such as light attenuation coefficient and even in nutrient concentration of ambient seawater. Electron transport rate (ETR) of Ulva from sandy shore was higher than from rocky shore. The range of photosynthetic pigment content in the tissue of U. pertusa was significantly much more in from sandy shore, and also nitrogen and phosphorus content were significantly higher except for carbon content. However, there were no significant differences in the ratio of among photosynthetic pigments, and N:P ratio was similar between each other, even though significantly different. Our result implied on the reason of why most of green tides in the worldwide concentrated and frequently occurred at sites with sandy, muddy and silty bottoms, being classified as soft-bottom shore.

Effects of CellCaSi and Bioflocculant on the Control of Algal Bloom (규산질다공체와 미생물응집제의 녹조제어 효과)

  • The effects of CellCaSi and bioflocculant on the control of algal bloom were investigated in enclosures in a small eutrophic pond. The bioflocculant produced by a bacterial strain S-2 was finally selected to remove Microcystis aeruginosa which was a dominant species of algal bloom in the pond. Enclosure experiment showed that phosphorus concentration decreased dramatically from $131\mu{g}\ell^{-1}$ (Control) to $1-14\mu{g}\ell^{-1}$ in three CellCaSi-enriched enclosures. Chlorophyll $-\alpha$ concentration also decreased from $215\mu{g}\ell^{-1}$ (Control) to $59\mu{g}\ell^{-1}$ by the addition of CellCaSi $(1g\ell^{-1}$, bioflocculant $(2ml\ell^{-1}$, calcium chloride $(1g\ell^{-1}$ and ferric chloride $(2mg\;Fe\ell^{-1})$ in Enclosure 4. From the results of the mouse acute toxicity test of the S-2 bioflocculant and the goldfish survival test in enclosures, it seems that both the S-2 bioflocculant and the CellCaSi do not show any severe toxicity in water system. Consequently, it was concluded that the bioflocculant and the CellCaSi could be used to control algal bloom in eutrophic waters by removing phosphorus and chlorophyll$-\alpha$.

Egg Production of the Copepod Acartia steueri in Ilkwang Bay, Southeastern Coast of Korea (한국 동해 남부 연안 일광만의 요각류 Acartia steueri의 알 생산력)

  • Egg production of Acartia steueri was estimated in llkwang Bay, located in the southeastern coast of Korea. The equation, combining fecundity, temperature and chlorophyll a, obtained under the laboratory experiments, was applied to the in situ temperature and chlorophyll a for the estimation of field egg production. Mean egg size was $80.52{\mu}m$. Prosome length of adult females was not correlated with egg size. Egg production of field population ranged from 0.32 to $63.32{\mu}gC\;m^{-3}d^{-1}$ with a mean of $13.33{\mu}gC\;m^{-3}d^{-1}$, which were equivalent to $7.1\~1407.1\;eggs\;m^{-3}d^{-1}$ and $296.1\;eggs\;m^{-3}d^{-1}$, respectively. Fecundity of adult females ranged from 5.4 to $12.5\;eggs\;female^{-1}d{-1}$ with a mean of $8\;eggs\;female^{-1}d^{-1}$. Specific egg production rates ranged from 0.028 to 0.117 $d^{-1}$ with a mean of 0.064 $d^{-1}$. Considering the egg Production ($\%$ female body carbon) as a function of temperature and chlorophlrll a concentration, our results showed rather low fecundity, which might be underestimated. Probable cannibalism of egg by the adults were thought to be pan of the reason for this low estimated fecundity.

Seasonal Variation of Microalgae in the Surface Water of Marian Cove, King George Island, the Antarctic 1998/1999 (1998/1999 남극 킹조지섬 마리안소만 표층수에 서식하는 미세조류의 계절적 변동)

  • We investigated seasonal variation of microalgal assemblages, sea water temperature, salinity and suspended solid and the parameters measured daily from January 1998 to October 1999 at a nearshore shallow-water in Marian Cove, Maxwell Bay, King George Island, the Antarctic. Annual mean surface water temperature was -0.3$0^{\circ}C$ and the highest water temperature was 4.53$^{\circ}C$ (22 January 1999) and the lowest water temperature was -2.07$^{\circ}C$ (23 August 1998). Annual mean salinity was 33.38 psu, ranging from 42.80 psu (6 January 1999) to 19.50 psu (6 June 1999). Annual mean suspended solid (SS) during two years was 34.14 mgㆍ1$^{-1}$, ranging from 60.62 mgㆍ1$^{-1}$(7 March 1998) to 12.90 mgㆍ1$^{-1}$ (26 December 1998). Chlorophyll $\alpha$ (Chl $\alpha$) concentrations were measured in order to know seasonal variations of microalgae in the surface seawater. Annual mean of total Chl a concentration was 0.55$\mu\textrm{g}$ㆍ1$^{-1}$, the highest Chl $\alpha$ concentration (12.16$\mu\textrm{g}$ㆍ1$^{-1}$) appeared in 4 October 1998, the lowest Chl $\alpha$ concentration appeared 0.19$\mu\textrm{g}$ㆍ1$^{-1}$, Monthly mean total Chl $\alpha$ concentration was high in October 1998 (1.32$\mu\textrm{g}$ㆍ1$^{-1}$) and low in July on 1998 (0.28$\mu\textrm{g}$ㆍ1$^{-1}$). Annual mean nano-sized Chl $\alpha$ concentration was 0.40$\mu\textrm{g}$ㆍ1$^{-1}$, monthly mean nano -sized Chl $\alpha$ concentration was high in November 1998 (0.90$\mu\textrm{g}$ㆍ1$^{-1}$), and low in July 1999 (0.22$\mu\textrm{g}$ㆍ1$^{-1}$). Annual mean micro-sized Chl $\alpha$ concentration was 0.15$\mu\textrm{g}$ㆍ1$^{-1}$ monthly mean micro-sized Chl $\alpha$ concentration was high in October 1998 (0.81$\mu\textrm{g}$ㆍ1$^{-1}$), and low July 1998, January, February and September 1999 (0.05$\mu\textrm{g}$ㆍ1$^{-1}$). More than 65% of total Chl $\alpha$ was concentrated during spring and summer time between October and March. Microalgal variation appeared to be due to physical factors of seawater in the Antarctic nearshore from 1998 to 1999. The reason why micro-sized Chl $\alpha$ did not increase during austral summer was the bay had been frozen by decrease of water temperature. We think that total microalgal abundance was decreased because the summer microalgal abundance was determined by variation of water temperature during winter season. Anti-inflammatory effects of fruit and leaf extracts of Lycium barbarum in lipopolysaccharide-stimulated RAW264.7 cells and animal model (염증유도 RAW264.7 세포와 동물모델에서 구기자와 구기엽의 항염 효능)

  • Purpose: Medicinal herbs have recently attracted attention as health beneficial foods and source materials for drug development. Recent studies have demonstrated that extracts of Lycium's fruits and roots have a range of physiologically active substances. The extract of Lycium's leaves has been reported to have excellent anti-oxidant and anti-microbial activity, but its anti-inflammatory efficacy is not known. The chlorophyll present in the leaves can act as an anti-oxidant or pro-oxidant depending on the presence of light. Therefore, this study analyzed the anti-inflammatory effects of Lycium's fruit extract (LFE), leaf extract (LLE), and leaf extract with chlorophyll removal (LLE with CR). Methods: This study examined the inhibitory effects of LFE, LLE, and LLE with CR on pro-inflammatory mediator production as well as on the expression of iNOS and COX-2 in lipopolysaccharide (LPS)-stimulated RAW264.7 cells and BALB/c mice. Results: LFE, LLE, and LLE with CR inhibited the production of pro-inflammatory mediators (NO, $TNF-{\alpha}$, IL-6, and $IL-1{\beta}$) and the expression of iNOS and COX-2 in LPS-stimulated RAW 264.7 cells in a dose-dependent manner. Furthermore, the administration of LLE and LLE with CR inhibited the serum pro-inflammatory cytokine levels and suppressed DNA damage in BALB/c mice. In particular, LLE with CR exhibited the highest anti-inflammatory activity. Conclusion: These results suggest that the fruit and leaves of Lycium are potential therapeutic agents against inflammation.

Changes in Radical Scavenging Activity and α-Glucosidase Inhibitory Activity of Dried Daraesoon (Shoot of Hardy Kiwi, Actinidia arguta) during Cooking (건조 다래순의 조리 중 라디칼 소거 활성과 알파글루코시데이스 억제 활성의 변화)

  • This study evaluated the in vitro radical scavenging and ${\alpha}$-glucosidase inhibitory activities of dried daraesoon (shoot of hardy kiwi) during cooking involving rehydration and subsequent heating at $180^{\circ}C$ with or without perilla oil. Pigments and antioxidants were quantified by HPLC and spectrophotometry. Unlike the tocopherol content, the polyphenol, flavonoid, chlorophyll, and carotenoid contents as well as the DPPH radical scavenging and ${\alpha}$-glucosidase inhibitory activities of daraesoon extract were significantly decreased by rehydration (p<0.05). Heating the rehydrated daraesoon for 10 or 20 min increased its radical scavenging activity irrespective of perilla oil addition, whereas the ${\alpha}$-glucosidase inhibitory activity increased significantly only after heating with perilla oil (p<0.05). During cooking, changes in both activities showed a similar pattern to that showed by polyphenol content changes. These results suggest that the health functionality of daraesoon can be enhanced by an appropriate cooking process that retains polyphenols.
