  • The interaction of the carcinogenic mycotoxin, Aflatoxin $B_1$, with the electron-donating molecule, benzene, was studied spectrophotometrically. The formation of charge-transfer complex between Aflatoxin $B_1$ and benzene in the presence of zinc chloride was observed and the apparent equilibrium constant of this charge-transfer complex was found to be 0.198 (liter $mole^{-1}$). It is assumed that, as the result of this study, some charge-transfer complexes could be formed between the weak electron-accepting Aflatoxin $B_1$ and strong electron-donating molecules, and the spectral changes occurred in the binding of Aflatoxin $B_1$ with proteins or DNA is attributed to the existence of charge-transfer type interaction.

Protective Effect of Vascular Endothelial Growth Factor on Focal Cerebral Ischemia in Rats

  • This study aimed to investigate the cerebroprotective effect of vascular endothelial growth factor (VEGF) on permanent focal cerebral ischemia in Sprague-Dawley rats. Right middle cerebral artery (MCA) was occluded for 6 and 24 hours by an intraluminal monofilament technique. An open cranial window was made on the right parietal bone for determination of continuous changes in regional cerebral blood flow (rCBF) by laser-Doppler flowmetry. The infarct size was morphometrically determined using the 2,3,5-triphenyltetrazolium chloride technique. Brain edema was determined by measuring brain water content. In normal rats, rCBF was significantly increased by intravenous infusion of VEGF for 10 minutes. The VEGF-induced increase in rCBF was significantly inhibited by pretreatment with suramin, a heparin-binding growth factor inhibitor as well as $N^{\omega}-nitro-L-arginine$, a nitric oxide synthase inhibitor. In focal cerebral ischemic rats, the amplitude of decrease in rCBF during ischemic period was significantly less in VEGF-treated group, compared with that in vehicle-treated group. The cerebral infarct size was reduced by VEGF in a dose-dependent manner. The brain edema formation was dose-dependently reduced by VEGF in 24-hour MCA occlusion group but not in 6-hour MCA occlusion group. It is suggested that VEGF not only improves the rCBF during cerebral ischemic period but also reduces the brain edema formation, and thereby exert a protective effect on focal cerebral ischemia in rats.

  • Pregnancy is a unique event in which a fetus develops in the uterus despite being genetically and immunologically different from the mother, and the underlying mechanisms remain poorly understood. To analyze the differential gene expression profiles in nonpregnant and 7 days post coitus (dpc) pregnant uterus of mice, we performed a global proteomic study by 2-D gel electrophoresis (2-DE) and MALDI-TOF-MS. The uterine proteins were separated using 2-DE, Approximately 1,000 spots were detected on staining with Coomassie brilliant blue. An image analysis using Melanie III (Swiss Institute for Bioinformatics) was performed to detect variations in protein spots between pregnant and nonpregnant uterus. Twenty-one spots were identified as differentially expressed proteins, of which 10 were up-regulated proteins such as alpha-fetoprotein, chloride intracellular channel 1, transgelin, heat-shock protein beta-1, and carbonic anhydrase II, while 11 were down-regulated proteins such as X-box binding protein, glutathione S-transferase omega 1, olfactory receptor Olfr204, and metalloproteinase-disintegrin domain containing protein TECADAM. Most of the identified proteins appeared to be related with catabolism, cell growth, metabolism, regulation, cell protection, protein repair, or protection. Our results uncovered key proteins of mouse uterus involved in pregnancy.

Flavonoidal constituent in Korean Lactuca dentata Makino (한국산 씀바귀의 Flavonoid 성분에 관한 연구)

  • The ethylacetate extract of Lactuca dentata Makino showed 6 flavonoidal components as detected by ferric chloride solution. The flavonoidal constituent of Lactuca dentata Makino was isolated and purified by the series of column chromatography. The chemical structure of one of the flavonoidal component named as compound E was identified by UV, IR and NMR spectrometry. The melting point range of compound E was $249.5^{\circ}C-251^{\circ}C$. The UV and IR spectra of purified compound E, and its genin were measured with the various shifting agents. The results of UV analysis showed the free state of hydroxy group at 3rd and 4th carbon and binding of sugar at the 7th carbon of compound. The sugar bound to the compound E was identified as glucose by TLC. The IR spectrum showed the presense of hydroxy group, conjugated carbonyl group and aromatic group. The analysis of NMR spectrum was done to the purified compound and its derivatives. The chemical shifts against hydrogen atom, hydroxy group, and the moiety of luteolin were observed in the NMR spectrum along with their position and number as well as type of sugar bound. The isolated and purified compound was identified as $luteolin-7-0-{\beta}-D-glucoside$.

Lithium ameliorates rat spinal cord injury by suppressing glycogen synthase kinase-3β and activating heme oxygenase-1

  • Glycogen synthase kinase $(GSK)-3{\beta}$ and related enzymes are associated with various forms of neuroinflammation, including spinal cord injury (SCI). Our aim was to evaluate whether lithium, a non-selective inhibitor of $GSK-3{\beta}$, ameliorated SCI progression, and also to analyze whether lithium affected the expression levels of two representative $GSK-3{\beta}$-associated molecules, nuclear factor erythroid 2-related factor-2 (Nrf-2) and heme oxygenase-1 (HO-1) (a target gene of Nrf-2). Intraperitoneal lithium chloride (80 mg/kg/day for 3 days) significantly improved locomotor function at 8 days post-injury (DPI); this was maintained until 14 DPI (P<0.05). Western blotting showed significantly increased phosphorylation of $GSK-3{\beta}$ (Ser9), Nrf-2, and the Nrf-2 target HO-1 in the spinal cords of lithium-treated animals. Fewer neuropathological changes (e.g., hemorrhage, inflammatory cell infiltration, and tissue loss) were observed in the spinal cords of the lithium-treated group compared with the vehicle-treated group. Microglial activation (evaluated by measuring the immunoreactivity of ionized calcium-binding protein-1) was also significantly reduced in the lithium-treated group. These findings suggest that $GSK-3{\beta}$ becomes activated after SCI, and that a non-specific enzyme inhibitor, lithium, ameliorates rat SCI by increasing phosphorylation of $GSK-3{\beta}$ and the associated molecules Nrf-2 and HO-1.

High Temperature Properties of Cement Mortar Using EVA, EVCL Redispersible Polymer Powder and Fly Ash (EVA, EVCL 분말수지와 플라이애시를 혼입한 시멘트 모르타르의 고온특성)

  • 3D printing technology of construction field can be divided into structural materials, interior and exterior finishing materials, and is mainly done by extruding and adapting. Particularly when it is applied as an exterior materials, it is mainly applied to an unstructured exterior materials and high accuracy is required. The exterior materials can be used as a cement composite materials, it is suitable also for a additive type manufacturing, and the role of a redispersible polymer powder is important. But, high temperatures, redispersible polymer cement base material beget dehydration and micro crack of cement matrix. In this research, we developed a EVA, EVCL redispersible polymer cement base material applicable as a 3D printing exterior materials, confirmed density and strength characteristics for application as an exterior materials, a flame retardancy test for improving the fire resistance of buildings and confirmed its possibility. From the test result, developed EVCL redispersible polymer cement mortar showed good stability in high temperatures. These high temperature stability is caused by the ethylene-vinyl chloride binding. Thus, this result indicates that it is possible to fire resistant 3D printing interior and exterior finishing materials.

Supplementation with psyllium seed husk reduces myocardial damage in a rat model of ischemia/reperfusion

  • BACKGROUND/OBJECTIVES: Myocardial infarction (MI) is caused by extensive myocardial damage attributed to the occlusion of coronary arteries. Our previous study in a rat model of ischemia/reperfusion (I/R) demonstrated that administration of arabinoxylan (AX), comprising arabinose and xylose, protects against myocardial injury. In this study, we undertook to investigate whether psyllium seed husk (PSH), a safe dietary fiber containing a high level of AX (> 50%), also imparts protection against myocardial injury in the same rat model. MATERIALS/METHODS: Rats were fed diets supplemented with PSH (1, 10, or 100 mg/kg/d) for 3 d. The rats were then subjected to 30 min ischemia through ligation of the left anterior descending coronary artery, followed by 3 h reperfusion through release of the ligation. The hearts were harvested and cut into four slices. To assess infarct size (IS), an index representing heart damage, the slices were stained with 2,3,5-triphenyltetrazolium chloride (TTC). To elucidate underlying mechanisms, Western blotting was performed for the slices. RESULTS: Supplementation with 10 or 100 mg/kg/d of PSH significantly reduces the IS. PSH supplementation (100 mg/kg/d) tends to reduce caspase-3 generation and increase BCL-2/BAX ratio. PSH supplementation also upregulates the expression of nuclear factor erythroid 2-related factor 2 (NRF2), and its target genes including antioxidant enzymes such as glutathione S-transferase mu 2 (GSTM2) and superoxide dismutase 2 (SOD2). PSH supplementation upregulates some sirtuins ($NAD^+$-dependent deacetylases) including SIRT5 (a mitochondrial sirtuin) and SIRT6 and SIRT7 (nuclear sirtuins). Finally, PSH supplementation upregulates the expression of protein kinase A (PKA), and increases phosphorylated cAMP response element-binding protein (CREB) (pCREB), a target protein of PKA. CONCLUSIONS: The results from this study indicate that PSH consumption reduces myocardial I/R injury in rats by inhibiting the apoptotic cascades through modulation of gene expression of several genes located upstream of apoptosis. Therefore, we believe that PSH can be developed as a functional food that would be beneficial in the prevention of MI.

Transcriptome Profiling of Differentially Expressed Genes in Cowpea (Vigna unguiculata L.) Under Salt Stress

  • Cowpea [Vigna unguiculata (L.) Walp] is one of the most important grain legumes that enhance soil fertility and is well-adapted to various abiotic stress. Also, it is cultivated worldwide as a tropical annual crop, and the semi-arid regions are known as the main cowpea-produced regions. However, accumulation of soil salinity induced by low rainfall in these regions is reducing crop yields and quality. In general, plants exposed to soil salinity cause an accumulation of high ion chloride, which leads to the degradation of root and leaf proteins. In this study, we identified candidate genes associated with salinity tolerance through an analysis of differentially expressed genes (DEGs) in four cowpea germplasms with contrasting salinity tolerance. A total of 553,776,035 short reads were obtained using the Illumina Novaseq 6000 platform for RNA-Seq, which were subsequently aligned to the reference genome of cowpea Vunguiculata v1.2. A total of9,806 DEGs were identified between NaCl treatment and control of four cowpea germplasms. Among these DEGs, functions related to salt stress such as calcium transporter and cytochrome-450 family were associated with salt stress. In GO analysis and KEGG analysis, these DEGs were enriched in terms such as the "phosphorylation", ''extracellular region", and "ion binding". These RNA-seq results will improve the understanding of the salt tolerance of cowpea and can be used as useful basic data for molecular breeding technology in the future.

Effects of 2-Ethylhexyl Diphenyl Phosphate on the Development and Growth Hormone Endocrine System in Zebrafish Larvae (2-Ethylhexyl Diphenyl Phosphate가 제브라피쉬 치어의 발달과 성장호르몬 내분비계에 미치는 영향)

  • Background: 2-Ethylhexyl diphenyl phosphate (EHDPP) is widely used as a flame-retardant plasticizer in the production of polyvinyl chloride, adhesives, and food packaging. This chemical has been frequently detected in water, sediment, and indoor environments, and its lipophilicity raises concerns about bioaccumulation. Objectives: In this study, the effects of EHDPP on the development, behavioral changes, and growth hormone (GH) endocrine system of zebrafish larvae were investigated. Methods: Fertilized embryos were exposed to various concentrations (control, solvent control, 0.07, 0.7, 7, 70, and 700 ㎍/L) of EHDPP for 96 h. Developmental toxicity endpoints were observed daily. Behavioral changes under light-dark-light conditions and changes in hormones and genes related to GH/insulin-like growth factors (IGFs) axis were determined. Results: Significant decreases in survival, body length and moving distance were observed in zebrafish larvae exposed to 70 and 700 ㎍/L EHDPP. The concentrations of GH and IGF-1 were significantly decreased in zebrafish larvae exposed to 70 and 700 ㎍/L EHDPP. This change was well supported by changes in the transcription of genes involved in GH, IGF, IGF receptors, and IGF binding proteins. Conclusions: Our observations showed that exposure to 70 and 700 ㎍/L EHDPP could disrupt the feedback circuits of the GH/IGFs axis, ultimately leading to developmental toxicity, hypoactivity, and mortality.

Capsicum annuum NAC4 (CaNAC4) Is a Transcription Factor with Roles in Biotic and Abiotic Stresses

  • Transcription factors (TFs) regulate gene expression by binding to DNA. The NAC gene family in plants consists of crucial TFs that influence plant development and stress responses. The whole genome of Capsicum annuum shows over 100 NAC genes (CaNAC). Functional characteristics of the most CaNAC TFs are unknown. In this study, we identified CaNAC4, a novel NAC TF in C. annuum. CaNAC4 expression increased after inoculation with the pathogens, Xanthomonas axonopodis pv. vesicatoria race 3 and X. axonopodis pv. glycines 8ra, and following treatment with the plant hormones, salicylic acid and abscisic acid. We investigated the functional characteristics of the CaNAC4 gene and its roles in salt tolerance and anti-pathogen defense in transgenic Nicotiana benthamiana. For salt stress analysis, the leaf discs of wild-type and CaNAC4-transgenic N. benthamiana plants were exposed to different concentrations of sodium chloride. Chlorophyll loss was more severe in salt stress-treated wild-type plants than in CaNAC4-transgenic plants. To analyze the role of CaNAC4 in anti-pathogen defense, a spore suspension of Botrytis cinerea was used to infect the leaves. The disease caused by B. cinerea gradually increased in severity, and the symptoms were clearer in the CaNAC4-transgenic lines. We also investigated hypersensitive response (HR) in CaNAC4-transgenic plants. The results showed a stronger HR in wild-type plants after infiltration with the apoptosis regulator, BAX. In conclusion, our results suggest that CaNAC4 may enhance salt tolerance and act as a negative regulator of biotic stress in plants.
