Suppressive Effect of Chlorella Methanol Extract on Oxidative Stress and NFkB Activation in RAW 264.7 Macrophages

  • This study was designed to investigate whether a methanol extract of chlorella can suppress oxidative stress and nuclear factor kB (NFkB) activation in lipopolysaccharide (LPS)-stimulated RAW 264.7 murine macrophage cells. Treatment of RAW 264.7 cells with chlorella methanol extract (25, 50, and 100 $\mu$g/mL) significantly reduced LPS-stimulated nitric oxide production in a dose-dependent manner. Treatments with chlorella methanol extract at all concentrations also reduced thiobarbituric acid-reactive substances accumulation and enhanced glutathione level at 50 and 100 $\mu$g/mL levels. The specific DNA binding activities of NFkB on nuclear extracts in cells treated with 50 $\mu$g/mL and 100 $\mu$g/mL chlorella methanol extracts were significantly suppressed. These results suggest that chlorella methanol extract has mild antioxidative activity and the ability to suppress intracellular oxidative stress and NFkB activation.

해수클로렐라 [Chlorella elliposidea C020] 에탄올 추출물에 대한 생리 활성 (Biological activities of ethanol extract from the seawater algae, Chlorella elliposidea C020)

  • 해수클로렐라 (Chlorella elliposidea C020) 에탄올 추출물로부터 항균 및 항곰팡이 활성, 항산화활성, tyrosinase inhibitory 활성 (미백효과)과 용혈활성을 실험하였다. 효율적인 추출을 위해서 동결건조 후 95% 에탄올로 2시간 추출하는 것이 가장 좋다는 것을 알 수 있었다. 해수클로렐라 에탄올 추출물의 항균 및 항곰팡이에 대한 활성을 측정한 결과, B. subtilis PM125와 B. licheniformis, V. parahemolyticus와 E. tarda NUF251에서 활성을 나타내었다. 그러나, 곰팡이인 C. albicanse에 대하여서는 활성을 나타내지 않았으며, 인간의 적혈구로 용혈활성 실험한 결과, 아주 낮은 활성을 나타내었다. DPPH방법을 이용하여 항산화 능력을 측정한 결과, 2 mg/mL에서 75% 라디칼 소거능력을 나타내었다. 해수클로렐라 에탄올 추출물의 tyrosinase 저해활성 $IC_{50}$는 10.87 mg/mL로 나타났다. 이러한 결과를 바탕으로 해수클로렐라 에탄올 추출물로부터 다양한 생리활성물질을 개발하여 고부가가치 제품을 창출할 수 있을 것으로 판단된다.

Exploring the nutritional biochemical profiles and biological functions in the green microalga Chlorella fusca

  • Chlorella species of microalgae are utilized in the crop and food industries. The aim of this research was to investigate the metabolite profiles of Chlorella fusca for the first time and evaluate its biological properties. The two ion modes of UPLC-Q-TOF-MS/MS were used to identify a total of 62 components in the methanol extract of C. fusca, with 26 in the negative and 36 in the positive ion mode, including 10 identical ingredients. Fatty acids (negative mode) and combinations of chlorophyll and fatty acids (positive mode) were the most prevalent chemical structures, constituting over 80 and 70% of the total metabolites, respectively, followed by chlorophyll, polar lipids, carotenoids, and fatty alcohols. Moreover, this extract exhibited potent antioxidant and anti-aging benefits in decreasing order of potency at a concentration of 200 ㎍ mL-1: tyrosinase inhibition (100%), ABTS radical scavenging (90%), elastase inhibition (88%), and DPPH radical scavenging (34%). Notably, this extract protected the mobility of DNA fragments up to 5 ㎍ mL-1 (26%), with potential effects (> 60% at 200 ㎍ mL-1). These findings suggest that C. fusca may be a promising candidate for applications related to its biological functions, owing to the high accumulation of fatty acids and chlorophyll derivatives.

해수산 클로렐라(Chlorella ellipsoidea) 유기용매 추출물의 항염증 효과 (Anti-Inflammatory Effect of Chlorella ellipsoidea Extracted from Seawater by Organic Solvents)

  • Chlorella has been reported to have certain beneficial physiological effects, including hypocholesterolemic, antihypertensive, antioxidative, and anti-tumor activities in animal and human studies. The aim of this study was to determine the anti-inflammatory activities of an 80% methanol extract(CE-Met), hexane fraction (CE-Hex), and ethyl acetate fraction (CE-EA) of Chlorella ellipsoidea in lipopolysaccharide (LPS)-stimulated RAW 264.7 murine macrophage. Treatment with various concentrations of the C. ellipsoidea extract resulted m a significant, dose-dependent reduction in nitric oxide (NO) production by LPS-induced macrophages. The C. ellipsoidea extract significantly inhibited LPS-induced NO production accompanied by an attenuation of IL-6 and TNF-$\alpha$ formation in macrophages. These results suggest potent inhibit이y effects on the production of inflammatory mediators by a C. ellipsoidea extract. Thus, C. ellipsoidea extract may be a potent anti-inflammatory agent for troubled skin.
